Neurotropic alphavirus infection induces PARP-1 hyperactivation-mediated energy collapse in motor neurons.

UNLABELLED: Motor neurons are highly vulnerable to metabolic stress, yet the mechanisms driving their degeneration during neurotropic alphavirus infections remain unclear. Venezuelan equine encephalitis virus (VEEV) causes motor neuron injury, but the intrinsic pathways underlying this susceptibility are not fully defined. Previous work suggests alphavirus-infected motor neurons may die through caspase-independent mechanisms. Here, we show that VEEV infection induces sustained activation of the DNA repair enzyme poly(ADP-ribose) polymerase-1 (PARP-1), leading to depletion of NAD+ and ATP in mu

A Phase 1, Open Label, Safety Study of HBV-002 (West Nile Recombinant Subunit Vaccine) in Healthy Adults.

Since its introduction in 1999, West Nile virus (WNV) has become endemic to the United States and many other countries around the world and is a leading cause of viral encephalitis each year. Despite the need, there are no licensed vaccines or therapies to address West Nile virus disease. The HBV-002 vaccine, which consists of the recombinant WNV envelope subunit protein (WN-80E) and Alhydrogel '85' (Croda Pharma, Princeton, NJ), was tested in an open-label, dose-escalation Phase 1 clinical trial (NCT00707642) alongside a WN-80E cohort without adjuvant. HBV-002 was found to be generally safe a

The Human Threat From West Nile Virus and Usutu Virus in a Changing Climate.

In recent years, the closely related arboviruses West Nile virus (WNV) and Usutu virus (USUV) have spread farther north in Europe. The viruses often co-circulate and are maintained in a natural cycle between ornithophilic mosquitoes and birds. Humans and other mammals can become infected with these viruses, but are considered dead-end hosts. Both viruses can cause fever and encephalitis and may potentially be fatal to humans, birds, and horses. Usutu virus was discovered in blackbirds in Denmark for the first time in the fall of 2024 and again in 2025. Denmark is currently a European border co

Self-limiting population suppression gene drive design in the West Nile vector mosquito, Culex quinquefasciatus.

Culex quinquefasciatus is a major vector of West Nile virus and other pathogens, yet genetic population suppression tools for this species remain limited. Here, we develop a self-limiting, CRISPR-based suppression gene drive system targeting doublesex, close to the male-determining locus, promoting male transmission. A recoded dsxM sequence converts females into sterile intersexes, preventing population-level spread. The drive achieves super-Mendelian inheritance ( ~ 71%) and generates resistance alleles that are fully or partially dominant female sterile. Single-release cage trials show exten

Widespread circulation of Alongshan virus in Austria and serological evidence for infection in humans: a nationwide molecular and serological observational study.

BACKGROUND: Alongshan virus, a segmented RNA virus in the Jingmenvirus group of flaviviruses, was first identified in 2017 in China in patients with tick-borne encephalitis-like illness. Alongshan virus has since been detected in ticks and mammals in several European countries. In this study, we aimed to characterise the temporal and geographical distribution of Alongshan virus in Austria through nationwide tick surveillance combined with serological and molecular screening in individuals with suspected tick-borne encephalitis or tick exposure. METHODS: In this nationwide molecular and serolo

Immunological Interactions Between Gammadelta T Cells and West Nile Virus in the Infected Host.

Between mosquitoes and birds, West Nile virus (WNV) is a neurotropic flavivirus, an arthropod-borne pathogen involved in an enzootic cycle. Additionally, it can infect both people and horses, leading to severe illness. Since 1999, WNV has spread across North and South America, including Mexico and the Caribbean. It is endemic in several regions of Europe, Africa, the Middle East, and Asia. WNV affects the central nervous system (CNS), causing severe disease in a small percentage of infected individuals, especially in immunocompromised or elderly hosts. This re-emerging pathogen was identified

Long-term sequelae after tick-borne encephalitis in Germany: a nationwide claims-based matched cohort study.

BACKGROUND: Tick-borne encephalitis (TBE) remains a public health concern in many parts of Europe despite the availability of effective vaccines. Acute disease is well described, but real-world data on the frequency, timing, and economic impact of long-term sequelae are limited. METHODS: A retrospective cohort study was performed using nationwide statutory health insurance claims covering up to 6.8 million insured individuals in Germany (2013-2023). Incident TBE cases were matched 1:5 to TBE-free controls by age, sex, region, comorbidities and healthcare utilization prior to index event. Neur

Cognitive outcome in relation to cerebrospinal fluid markers of neuronal and glial cell damage: A prospective study of tick-borne encephalitis.

OBJECTIVES: Tick-borne encephalitis (TBE) often results in long-term cognitive sequelae. While biomarkers of neuronal and glial injury are established in other neurological conditions, their role in predicting cognitive outcomes in TBE remains unclear. This study investigated whether acute-phase levels of neurofilament light (NfL), glial fibrillary acidic protein (GFAP), and S100B (calcium-binding protein B) in both cerebrospinal fluid (CSF) and serum, and tau and chitinase-3-like protein 1 (YKL-40) in CSF only, were associated with cognitive functioning at follow-up. METHODS: Ninety-two adul

Climate-based forecasting from national Culex mosquito surveillance to support West Nile and Usutu virus preparedness in England and Wales.

Culex mosquitoes are widespread in temperate regions and play a key role in transmitting veterinary and human vector-borne diseases. In the United Kingdom, Culex pipiens s.l. is highly prevalent and a competent vector of West Nile and Usutu viruses. Coupled with the northward expansion of West Nile virus in Europe, this raises concerns about emergence in the UK. For public and animal health preparedness, and effective vector control planning, it is essential to better understand the distribution of Culex mosquitoes in this area. This study developed species distribution maps for Culex pipiens p

Evaluation of CXCL12 Diagnostic Performance in Tick-Borne Encephalitis and Other Central Nervous System Infections.

CXCL12 is a chemokine acting via CXCR4 that plays a key role in inflammatory processes by regulating leukocyte migration and immune responses, making it an important focus of research in neuroinfections. In this study, the CXCL12 chemokine was evaluated as a potential diagnostic marker of different neuroinfections with particular emphasis on tick-borne encephalitis (TBE). A group of 214 patients with confirmed meningitis and/or encephalitis was included and divided according to etiology into TBE, aseptic meningitis, neuroborreliosis, and purulent meningitis. CXCL12 concentration and other infl

Brain endothelial cells orchestrate a neuroprotective antiviral state in the CNS in response to peripheral viral pattern sensing.

West Nile virus (WNV) and other neurotrophic arboviruses are human pathogens that can cause life-threatening encephalitis. To uncover determinants of severe disease progression, we employed a murine infection model of WNV and experimentally uncoupled peripheral pathogen sensing from active viral replication. Peripheral sensing of a viral dsRNA mimic in the footpad led to robust induction of antiviral type I interferon (IFN-I)-stimulated genes and establishment of an antiviral state within the brain. Systematic approaches combining cytokine profiling, single-nuclei transcriptomics, immune modul

West Nile Virus and Other Nationally Notifiable Arboviral Diseases - United States, 2024.

In the United States, arthropodborne viruses (arboviruses) are primarily transmitted by infected mosquitoes or ticks; rarely, they can be transmitted through blood transfusion or organ transplantation. Whereas a majority of arboviral infections are asymptomatic, symptomatic infections range from mild febrile illness to severe neuroinvasive disease. This report summarizes data for nationally notifiable domestic arboviral diseases with illness onset in 2024. Forty-eight states and the District of Columbia reported 1,955 arboviral disease cases, including 189 (10%) deaths. Although the number of

Spatial dynamics of viral replication drive biphasic lymphoid activation and systemic alphavirus immune responses.

Systemic type I interferon (IFN-I) and pro-inflammatory cytokine responses are critical for limiting mosquito-borne RNA virus viral replication and disease, yet mechanisms initiating these responses and their linkage to viral tropism for lymphoid tissues are poorly understood. Here, we reveal that virus-cell interactions and specific host signaling pathways in lymphoid tissues are determinants of systemic cytokine induction. Using Venezuelan equine encephalitis virus (VEEV) mutants with defined lymph node (LN) tropism in mice, we show that VEEV LN infection positively associates with systemic

Tick-borne encephalitis virus in Dermacentor reticulatus ticks, Poland.

We report the detection of tick-borne encephalitis virus (TBEV) in Dermacentor reticulatus ticks collected in Poland during the 2022 season. Both Ixodes ricinus and D. reticulatus ticks were surveilled. There was limited TBEV detection found in one pool out of 106 pools (418 ticks) that were shotgun genome sequenced. The TBEV positive tick pool consisted of N = 3 adult D. reticulatus ticks. Additional hybrid-capture sequencing of this pool resulted in the first full-length genome sequence of TBEV from this tick species. The assembled genome was classified as the European subtype through phylog

Pathogenesis and neurotropism of a contemporary Powassan virus lineage II strain in mice.

Powassan virus (POWV) is an emerging tick-borne flavivirus that can cause neurologic disease in humans. POWV exhibits substantial genetic and phenotypic diversity, including marked variability in replication in vitro and pathogenesis in mice. However, most experimental studies have relied on historical lineage I and II strains to assess POWV pathogenesis. Here, we characterized a contemporary lineage II strain, NY.19.12, in mice and investigated the potential viral determinants of disease. NY.19.12 caused earlier onset of clinical signs and earlier detection of viral RNA (vRNA) in the spleen a

Encephalitic alphavirus infection induces long-term humoral and cell-mediated responses in seropositive participants from Panama.

Encephalitic alphaviruses are an emerging viral threat with a history of large-scale outbreaks. Evaluating the immune response to natural infection in survivors is critical to the rational development of medical countermeasures (MCM), and yet little is known thus far. Here, we describe a comprehensive analysis of adaptive immune responses from a cohort of 37 convalescent seropositive individuals of Venezuelan equine encephalitis and/or Madariaga virus from Panama. We demonstrate persistent and polyfunctional B cell responses, as well as memory T cell cytokine production in response to viral an

Field-Collected Belgian Culex pipiens and Culex torrentium Mosquitoes are Competent for West Nile Virus Under Different Temperature Conditions.

Belgium detected West Nile virus (WNV) for the first time on its territory in August 2025 during a monitoring of wild birds. To support risk assessments, we determined the vector competence of field-collected Belgian Culex pipiens and Culex torrentium mosquitoes, which are considered important WNV vectors. Females fed on a WNV-spiked (lineage 1 Israel 98 strain) blood meal were kept for 14 days at (i) a constant 25°C, (ii) a 25/15°C day/night gradient reflecting current Belgian summers, or (iii) a 25/20°C gradient simulating a climate change scenario.
