THE MOSQUITO FAUNA OF K'GARI (FRASER ISLAND), AUSTRALIA, THE WORLD'S LARGEST SAND ISLAND.

K'gari (Fraser Island) is the largest sand island in the world and a popular tourist destination. Despite its importance as a United Nations Educational, Scientific and Cultural Organization (UNESCO) World Heritage Site, there is a paucity of information regarding its mosquito fauna. From 1968 to 1976, several qualitative entomological surveys were carried out on K'gari, but no published work has been undertaken since. This study presents quantitative findings from three surveys undertaken using CO2-baited light traps in April and October 2024, and May 2025. In total, 3,356 mosquitoes, represe

The RNA promoter for pathogenic orthoflaviviruses replication is universal and serves as target for viral inhibition.

Orthoflaviviruses comprise a diverse genus of positive-strand RNA viruses that includes major human pathogens such as dengue, yellow fever, Zika, Japanese encephalitis, tick-borne encephalitis, and West Nile viruses. Despite their global impact, the molecular constraints that preserve viral replication across distinct vectors and host environments remain incompletely understood. Viral RNA replication depends on Stem-Loop A (SLA), a structured RNA element located at the 5' end of the genome that recruits the viral polymerase NS5. Here, we examine the structural diversity of this RNA promoter ac

[Flavivirus Infection (Japanese Encephalitis)].

Japanese encephalitis remains a clinically relevant neuroinfectious disease in Japan. Adults in middle- to older- age groups are at higher risk due to the low prevalence of neutralizing antibodies. Clinicians should consider this disease when encountering acute encephalitis during the summer-to-autumn season. High fever, impaired consciousness, and extrapyramidal signs are characteristic clinical features and correspond to frequent involvement of the thalamus and substantia nigra. A definitive diagnosis is achieved through appropriate sampling of acute- and convalescent-phase specimens togethe

Unravelling the distribution of vectors of major vector-borne diseases in Koshi Province of Nepal: A concern of expansion in diverse geo-ecological and climatic regions.

BACKGROUND: Vector-borne diseases (VBDs), including malaria, visceral leishmaniasis, lymphatic filariasis, Japanese encephalitis, and dengue, are major public health concerns and are either slated for elimination or projected for control in Nepal. One of the major challenges in controlling these VBDs is halting their emergence and expansion in diverse geo-ecological and climatic regions. In this study, we collected vectors of major VBDs to assess their distribution, diversity, and associations with ecological variations and to provide an updated understanding of the current situation. Japanese Encephalitis Vaccine Decision Aid for Travelers: A Randomized Clinical Trial.

IMPORTANCE: Japanese encephalitis (JE) is a mosquito-borne disease with low infection risk but high consequences. Low uptake of JE vaccines among travelers persists despite effective vaccines. Tools that improve decision quality may help address this gap. OBJECTIVE: To evaluate whether a web-based JE vaccine decision aid (JEVaDA) improves decision-making and vaccine uptake among Australian travelers compared with an online government JE resource. DESIGN, SETTING, AND PARTICIPANTS: This parallel-group, single-blind randomized clinical trial was conducted online across Australia from November

Rab4b is associated with inflammatory responses and TLR3-related signaling during Japanese encephalitis virus infection.

Japanese Encephalitis Virus (JEV) is a mosquito-borne zoonotic pathogen that triggers severe inflammatory responses upon infection. Previous research has demonstrated that the interaction between Ras-associated GTP-binding protein 4B (Rab4b) and the JEV E protein is essential for the virus's entry into early endosomes. However, the precise mechanism by which Rab4b contributes to JEV-induced inflammatory responses remains inadequately understood. In this study, we developed a Rab4b-knockout U251 cell model and found that Rab4b deficiency markedly inhibited viral replication and concurrently red

Detection and genetic characterization of Tembusu virus and other flaviviruses from mosquitoes in Lao PDR.

BACKGROUND: Lao People's Democratic Republic (Lao PDR), located in Southeast Asia and known for its rich biodiversity, is part of a region recognized as a hotspot for emerging and re-emerging infectious diseases. Among flaviviruses, dengue virus (DENV) and Japanese encephalitis virus (JEV) are recognized public health threats. However, other reemerging mosquito-borne flaviviruses may also infect humans and cause diseases. Despite that, their distribution and public health impact in Lao PDR are not well understood due to limited past surveillance. METHODOLOGY: Mosquitoes were collected using C

Epitope-level insights for immuno-recognition of JEV NS1 via gold nanoflower- MXene composite.

Early and accurate identification of Japanese encephalitis virus (JEV) is vital for effective outbreak control and surveillance. An electrochemical immunosensor was developed for the detection of JEV non-structural protein 1 (NS1). The JEV NS1 antigen was expressed, purified and subsequently refolded in vitro to recover soluble, functional protein. High-affinity monoclonal antibodies (mAbs) were generated through hybridoma technology. To gain molecular-level insights, antibody gene sequences were decoded using next-generation sequencing (NGS), while molecular docking analysis enabled predictiv

Comparative dynamics of Japanese encephalitis virus adaptation in porcine macrophages and insect cells.

BACKGROUND: Japanese encephalitis virus (JEV) is a zoonotic mosquito-borne Orthoflavivirus that circulates primarily in birds and pigs. Previous observations of vector-free transmission between pigs indicates the possibility of single-host cycling in swine. Therefore, the aim of this work was to investigate the evolutionary pressure of single host cycling using a relevant primary cell culture model. METHODS: To investigate whether such single-host cycles affect viral infectivity, fitness and genomic adaptations, two strains and a reverse genetic cDNA-derived clone of JEV were serially passage

Myeloid miR-155 deficiency exacerbates viral encephalitis by hindering M1 macrophage polarization due to impaired NLRP3 inflammasome activation in extraneural tissues.

INTRODUCTION: miR-155 regulates diverse inflammatory responses; however, its role in neurotropic virus-induced CNS inflammation, particularly in specific immune cell subsets, remains unclear. This study aimed to identify the immune cell population in which miR-155 is required for protection against Japanese encephalitis virus (JEV) and to elucidate the underlying mechanisms. METHODS: Conditional miR-155 knockout mice with cell type-specific deletion in myeloid cells, dendritic cells (DCs), T cells, or B cells were generated using Cre-LoxP technology. Following JEV infection, we assessed survi

Arbovirus Circulation among Patients with Acute Encephalitis: West Nile Virus Infection among Humans in Bangladesh, 2022.

West Nile virus (WNV) is an emerging infectious disease causing neuroinvasive diseases, including meningitis and encephalitis. Although infection has been documented in birds in Bangladesh, WNV has not been detected in humans. Since 2007, the Institute of Epidemiology, Disease Control and Research in Bangladesh has conducted hospital-based acute encephalitis syndrome (AES) surveillance to monitor Japanese encephalitis (JE), which is typically detected in 2% to 3%. Our study aimed to determine the presence of WNV, dengue virus (DENV), and chikungunya virus (CHIKV) among persons enrolled in AES

Japanese Encephalitis Virus and Host Innate Immunity: Insights Into TLR Signaling, PRR Crosstalk, and Viral Immune Evasion.

Japanese encephalitis virus (JEV), a neurotropic flavivirus and a major cause of viral encephalitis, poses a significant global health threat due to its neuroinvasive potential. Host innate immune responses, particularly those mediated by pattern-recognition receptors such as Toll-like receptors (TLRs) and RIG-I-like receptors, play critical roles in detecting JEV infection in neurons and glial cells, triggering antiviral defenses through induction of type I interferons (IFNs), inflammatory cytokines, and interferon-stimulated genes. However, dysregulated inflammatory responses may contribute

Japanese encephalitis virus hijacks the host purine biosynthetic network to promote viral replication in neurons.

Japanese encephalitis virus (JEV) is an important neurotropic orthoflavivirus that poses a threat to both human and animal health. However, the mechanism underlying its rapid replication in the central nervous system (CNS) remains poorly understood. In this study, we conducted metabolomic profiling of JEV-infected mouse brains and neurons, revealing a profound reprogramming of central carbon metabolism, particularly an enhancement in nucleotide synthesis. Integrated multi-omics analyses confirmed that JEV infection transcriptionally upregulates key enzymes involved in de novo purine biosynthes

Astrocyte-derived HMGB1 compromises the integrity of the blood-brain barrier through the CaM/CaMKII/AQP4 pathway and the protective function of trifluoperazine.

The integrity of the blood-brain barrier (BBB) is crucial for maintaining the function and homeostasis of the central nervous system (CNS), with astrocytes playing a key role in this process. Our study found that infection with the Japanese encephalitis virus (JEV) promoted the translocation of high-mobility group box 1 (HMGB1) from the nucleus to the extracellular space of astrocytes, a process directly associated with BBB disruption. Through bioinformatics analysis, we identified potential targets of encephalitis and constructed a protein-protein interaction (PPI) network. Subsequent functio

Alignment-free k-mer-guided design of a pan-Orthoflavivirus RT-qPCR assay.

The co-circulation and rapid expansion of the genus Orthoflavivirus, including dengue virus (DENV), Zika virus (ZIKV), West Nile virus (WNV), Yellow fever virus (YFV), and Japanese encephalitis virus (JEV), pose significant global health challenges. Developing inclusive pan-genus molecular diagnostics is hindered by high nucleotide divergence (>25-30%) and the computational limitations of traditional multiple sequence alignment in detecting conserved motifs across large data sets. To overcome these limitations, we developed a systematic alignment-free design pipeline that uses rigorous k-mer a

Oral lipid prodrug V2043 (1-O-octadecyl-2-O-benzyl-sn-glyceryl-P-remdesivir nucleoside) protects mice from lethal dengue infection.

Dengue virus and related flaviviruses are significant threats to global health, with no approved antiviral treatments available. Remdesivir triphosphate (RVn-TP) is a nucleotide analog that inhibits RNA-dependent RNA polymerases from many clinically important RNA viruses, including dengue virus. An orally bioavailable prodrug that efficiently delivers RVn-TP could facilitate early treatment for non-hospitalized dengue patients. To that end, we developed 1-O-octadecyl-2-O-benzyl-sn-glyceryl-P-remdesivir nucleoside (V2043, ODBG-P-RVn), a phospholipid prodrug that achieves RVn-TP delivery through

A target product profile and roadmap for Japanese encephalitis diagnostics in endemic areas in the context of other flaviviruses.

Japanese encephalitis (JE) virus (JEV) is a leading cause of neurological infection in the Asia-Pacific region with devastating socioeconomic consequences. We aimed to develop a target product profile (TPP) for JE diagnostics in endemic areas in the context of other flaviviruses, to better define improvements needed for patient management and public health. Thematic experts were identified from a scoping review, World Health Organization and Encephalitis International databases, as well as author networks. Initial interviews were performed with selected experts to inform a draft TPP document a

Analysis of West Nile disease convalescents identifies human monoclonal antibodies protective against West Nile and related orthoflaviviruses.

West Nile virus (WNV) is a mosquito-borne pathogen of global concern that can cause fatal neuroinvasive disease, and no specific prophylaxis or treatment exists for infections by WNV and most related orthoflaviviruses. Here, we isolated and characterized antibodies from WNV convalescent individuals and report that neutralizing autoantibodies against type I interferons did not impair antiviral antibody development. Among the monoclonal antibodies with potent neutralizing activity against WNV that were identified, W010 targeted a distinct epitope within the envelope protein domain III (EDIII) an

Underrecognized Tick-Borne Encephalitis in Serbia: Evidence from Patients with Suspected West Nile Virus Neuroinvasive Disease.

Tick-borne encephalitis (TBE) is an emerging vector-borne disease in Europe, but its epidemiology remains poorly defined in Serbia. In orthoflavivirus-endemic settings, diagnostic challenges may contribute to underrecognition of TBE, particularly among patients with suspected West Nile virus (WNV) infection. We conducted a multicenter retrospective study including patients hospitalized between 2018 and 2023 with suspected WNV neuroinvasive disease or viral encephalitis of unknown etiology. Serum samples were tested for TBEV-neutralizing antibodies using a microneutralization assay. Among 79 pa
