Diffuse cerebral microhemorrhages in West Nile virus neuroinvasive disease: a case report.

West Nile virus neuroinvasive disease (WNND) most commonly presents as meningitis, encephalitis, or acute flaccid paralysis, with variable neuroimaging findings. While MRI abnormalities classically involve deep gray matter and brainstem structures, hemorrhagic manifestations are rarely reported. We report a 77-year-old woman who presented with rapidly progressive altered mental status following a brief febrile prodrome. Cerebrospinal fluid studies demonstrated a lymphocytic pleocytosis consistent with aseptic meningitis. Brain MRI revealed an acute lacunar infarct and diffuse cerebral microhem

[Test systems for the laboratory detection and differentiation of Powassan virus from tick-borne encephalitis virus (Flaviviridae: Orthoflavivirus) using RT-PCR and RT-qPCR].

INTRODUCTION: Powassan virus (POWV) is a tick-borne orthoflavivirus capable of causing neurological diseases. In the Russian Far East, the area affected by POWV overlaps with that of another neurotropic orthoflavivirus, tick-borne encephalitis virus (TBEV). Currently, there are no differentiated test systems for specific detection of POWV in the presence of TBEV. The aim of this work is to create differentiating test systems for the detection of POWV in a mixture with TBEV and other orthoflaviviruses. MATERIALS AND METHODS: We constructed a genome alignment of eight POWV and five TBEV strains

Omsk Hemorrhagic Fever Virus and Powassan Virus Infections at Different Times After Immunization with the TBE Vaccine in In Vivo Experiments.

Ranges of distribution of some orthoflaviviruses, including the Omsk hemorrhagic fever virus (OHFV) and the Powassan virus (POWV), have been identified in the range endemic for the tick-borne encephalitis virus (TBEV). In contrast to TBEV, no registered vaccine against OHFV and POWV is currently available. Nevertheless, recent studies have indicated that the anti-TBEV vaccine may offer partial protection against other orthoflaviviruses. The present study assesses OHFV and POWV infection in an experimental model on BALB/c mice one, three- and twelve-months post-immunization with inactivated TBE

Prevalence of West Nile Virus in Children Being Evaluated for Lyme Disease.

BACKGROUND: West Nile virus (WNV) is a flavivirus typically transmitted between mosquitoes and birds with occasional spill-over into other animals, including humans. Children evaluated for suspected Lyme disease may share environmental risk factors for WNV and present with overlapping symptoms. Our goal was to determine the prevalence of WNV infection in children with suspected Lyme disease. METHODS: We performed a nested case-control study within a prospective cohort study of children with suspected Lyme disease aged 1-21 years enrolled at 1 of 8 Pedi Lyme Net emergency departments. Lyme dis

A case study of Powassan virus encephalitis in an immunocompromised host.

BACKGROUND: Powassan virus (POWV) is a tick-borne, neurotropic arbovirus endemic to Canada and the United States and is transmitted by Ixodes ticks, which also carry Lyme disease. It is an increasingly recognized cause of neuro-invasive infection in humans, for which treatment is supportive. Prognosis in POWV encephalitis is variable, ranging from complete recovery to death. As with other arboviral encephalitides, the mainstay of diagnosis is serological testing. However, molecular diagnostics play an increasingly important role, particularly in the case of immunocompromised hosts. CASE SUMMA

Fulminant Powassan Virus Encephalitis Presenting as Acute Ischemic Stroke: A Case Report.

Powassan virus (POWV) is a rare tick-borne flavivirus that can cause severe neuroinvasive disease with high morbidity and mortality. Early clinical and radiographic features are often nonspecific and may mimic acute ischemic stroke, leading to diagnostic delay. We report the case of a 77-year-old woman with recent tick exposure who presented with acute-onset focal neurologic deficits and fever. Initial neuroimaging demonstrated focal diffusion restriction, raising concern for acute ischemic stroke, and dual antiplatelet therapy was initiated. Over subsequent days, the patient developed progres

Eastern equine encephalitis virus infection in an American pygmy goat.

Eastern equine encephalitis virus (EEEV; family Togaviridae, taxon species Alphavirus eastern) is a mosquito-borne alphavirus that is endemic to the Western Hemisphere and well established in freshwater hardwood swamps. EEEV is environmentally maintained through a mosquito-and-avian cycle, primarily by Culiseta melanura mosquitoes. Transmission to mammals, which are considered dead-end hosts, is well known and seasonal through C. melanura or other bridging mosquito vectors. EEE is well described in horses and humans, with relatively few reports in ruminants. Information regarding infection and

A Modular Design of a Cholera Toxin B Subunit-Scaffolded Sub-Virion Nanoparticle Vaccine Against West Nile Virus.

Although veterinary vaccines against West Nile virus (WNV) have been developed, no approved human vaccine is currently available, highlighting the need for scalable and safer WNV vaccine candidates. In this study, a recombinant WNV subunit nanoparticle vaccine was developed by displaying the envelope protein domain III (ED3) on a cholera toxin B subunit (CTB) pentameric scaffold. The resulting recombinant protein comprising CTB-ED3 was expressed predominantly as soluble nanoparticles in Escherichia coli. Immunized mice produced strong humoral responses with balanced IgG1/IgG2a ratios, and some

Clearance mechanisms of the glymphatic/lymphatic system in the brain: new therapeutic perspectives for cognitive impairment.

The glymphatic/lymphatic system of the brain has been an important discovery in the field of neuroscience in recent years. As the "waste clearance network" of the central nervous system, it clears metabolic products and neurotoxic substances through the cerebrospinal fluid-interstitial fluid circulation, which is crucial for maintaining the homeostasis of the intracerebral environment and plays important roles in learning, memory and other advanced cognitive functions. The glymphatic/lymphatic system is crucial for the clearance of beta-amyloid and tau proteins, and thus the abnormal function

Distribution status and influencing factors of antibiotic resistance genes in the Chaohu Lake, China.

BACKGROUND: Chaohu Lake (CL) is one of the most polluted areas in China due to its high content of antibiotics. However, the distribution and influencing factors of antibiotic resistance genes (ARGs) in this lake are still controversial. METHODS: To solve this problem, we used metagenomic sequencing to investigate the distribution and in-fluencing factors of ARGs in CL. RESULTS: Our findings revealed the existence of nine kinds of ARGs, including 45 specific genes. The most abundant types were multidrug, bacitracin, polymyxin, macrolide lincosamide streptogramin, and aminoglycoside. Multiple

Muscle contraction type-specific association of acceleration and deceleration performance with rates of force development.

BACKGROUND: Many sports require rapid acceleration and deceleration, particularly when changing direction. These movements require a large impulse, highlighting the importance of rates of force development (RFDs). However, the relationships between acceleration and deceleration performance and concentric and eccentric RFDs have remained uncertain. This study evaluated the correlation between RFDs in different muscle contraction types and acceleration and deceleration performances. METHODS: This study included 28 healthy subjects (13 males and 15 females; age: 21 ± 2 years; height: 1.66 ± 0.09

Ecological risk factors for the establishment of West Nile virus in Britain.

West Nile virus (WNV) is a zoonotic mosquito-borne virus which is emerging across Europe, largely due to climate and other environmental changes. Detection of WNV at increasingly northern latitudes raises concern that WNV may be introduced to Britain, where ecological conditions could eventually support sustained transmission. Establishment of WNV depends on spatial and temporal overlap between infectious migratory birds and native vectors. However, understanding of the distributions and phenology of key vector species in Britain is incomplete and must be updated to prioritise activities for W

Emergence of West Nile virus infection in Kerala, South India: A case of concern.

West Nile virus (WNV) infection is emerging as a disease of public health concern in Kerala state, India with recurring outbreaks since 2011. With its tropical climate, biodiversity hot spots of Western ghats, forest cover, plenty of water bodies and bird sanctuaries, Kerala provides an ideal ecological niche for vector breeding and transmission of WNV. In this work, we reflect on the peculiar features of the outbreaks of WNV fever in Kerala and highlight the knowledge gaps, research priorities and the need for effective control measures. Information on transmission patterns of WNV, its circul

Usutu virus-induced meningoencephalitis in immunocompetent mice is characterized by the recruitment of mononuclear cells and a proinflammatory T helper 1 response.

Usutu virus (USUV) is an arbovirus and has emerged as a potential cause of encephalitis in humans and other vertebrates. The increasing detection of USUV in mosquitoes and birds across Africa and Central Europe, along with the lack of specific treatments or vaccines for many encephalitic orthoflaviviruses, underscores the need for focused research. In this study, we developed a USUV infection model in immunocompetent C57BL/6 mice (8-12 weeks old) to characterize disease development and associated inflammatory mechanisms. Mice were intracranially infected with 104 PFU of USUV, leading to neurol

West Nile virus non-structural protein 1 promotes amyloid Beta deposition and neurodegeneration.

Recent observations highlight a notable surge in West Nile Virus (WNV) infections in Europe that can lead to neuroinvasive consequences associated with neurodegeneration, mainly triggered by WNV Non-Structural protein 1 (NS1). During viral replication, various protein-protein interactions take place, allowing viral proteins to interact with host factors. NS1 is actively secreted in the bloodstream by infected cells and is known to affect endothelial permeability and host immune response. Focusing on the recently discovered antimicrobial roles of Amyloid-Beta (Aβ) in the context Central Nervous
