Scenario-driven shifts in future Usutu and West Nile virus outbreak characteristics in the Netherlands.

Mosquito-borne virus transmission is shaped by its ecological context, including land use, climate, and population dynamics. Future changes in these factors may therefore affect the risk and intensity of Usutu virus (USUV) and West Nile virus (WNV) outbreaks in the Netherlands. We compared a reference scenario to four future Shared Socio-economic Pathway scenarios developed for the Netherlands, which differed in land use, host distribution, mosquito distribution, and temperature. Temperature (between + 1.0 °C and + 1.7 °C) and mosquito abundance (between + 5% and + 10%) were predicted to incre

Association of avian biodiversity and West Nile Virus circulation in Culex mosquitoes in Emilia-Romagna, Italy.

BACKGROUND: West Nile Virus (WNV) is a zoonotic arbovirus maintained in a transmission cycle between Culex mosquitoes and birds, occasionally spilling over into humans. The impact of avian biodiversity on WNV circulation remains debated, with studies reporting both negative and positive correlations (dilution and amplification effects respectively) across different settings. In Europe, this relationship remains largely unexplored, particularly in regions with high WNV transmission, such as Emilia-Romagna in Northern Italy. METHODS: We explored the association between avian biodiversity and WN

A human cerebral organoid model of West Nile virus encephalitis shows innate immunocompetency.

West Nile virus (WNV), an arbovirus of emerging global interest, can cause neuroinvasive disease in humans. Currently, no protective vaccine or specific treatment is available for human WNV encephalitis. The virus induces neuronal cell death, while astrocytes and microglia cells are suspected to contribute to WNV pathology. Hence, understanding their role is crucial for future treatment approaches. In this study, we establish a WNV encephalitis model using human cerebral organoids, generated with male iPSCs. Infection results in heterogeneous kinetics with an early strong replication potential

A novel approach to determine mosquito trap placement for West Nile virus surveillance.

The rapid spread of West Nile virus (WNV) is a growing concern. With no vaccines or specific medications available, prevention through mosquito control is the only solution to curb the spread. Mosquito traps, used to detect viral presence in mosquito populations, are essential tools for WNV surveillance. But how do we decide where to place a mosquito trap? And what makes a good trap location, anyway? We present a robust statistical approach to determine a mosquito trap's ability to predict human WNV cases in the Chicago metropolitan area and its suburbs. We then use this value to detect the la

Langat Virus Biology and Infection.

Flaviviruses pose a major threat to global health and can cause severe disease in animals and humans. Among them, tick-borne encephalitis virus (TBEV) and related members of the tick-borne encephalitis (TBE) serocomplex are transmitted primarily by ticks and can lead to neurological disease, including encephalitis and death. Despite many years of research on the TBE-serocomplex, no specific antiviral treatment has been developed. Langat virus (LGTV), another member of this serocomplex typically causes asymptomatic infections in humans, but may cause severe neurological disease in rare cases. M

Antiviral effect of chelerythrine chloride against tick-borne encephalitis virus.

Tick-borne encephalitis virus (TBEV) is a neurotropic Orthoflavivirus that causes severe encephalitis and long-term neurological sequelae. Due to the limited protection from existing vaccines and the absence of effective antiviral drugs, the prevention and management of tick-borne encephalitis (TBE) remain a serious challenge. Given the homology of conserved targets and shared pathogenic mechanisms among orthoflaviviruses, we screened anti-orthoflavivirus compounds from existing research for candidates against TBEV. In this study, we identified chelerythrine chloride (CHE), a quaternary ammoni

High burden of autochthonous arboviral infections during the summer season in Verona province, Italy, during 2025.

OBJECTIVES: We aimed at estimating the frequency of local transmission of arboviral infections in Verona province, Italy, between June and September 2025. METHODS: People presenting with acute fever and without recent travel history (and less than WBC 10,000 per µL, unless there was a history of tick bite) were screened for arboviruses following a multi-step protocol. Cases were confirmed by a molecular assay and/or virus-specific IgM antibodies, including rising antibody titre in a second serum sample collected 15-20 days later and a positive seroneutralization test. RESULTS: One hundred an

Multiscale modeling of vector-borne diseases: The role of dose-dependent transmission.

The transmission of infectious diseases involves complex interactions across multiple biological scales, from within-host immunological processes to between-host transmission dynamics. While multiscale models have the potential to capture these interactions more accurately, they are often hindered by increased complexity and limited data availability. In this study, we develop a multiscale epidemic model linking host-vector population-level transmission dynamics to within-host and within-vector pathogen dynamics. Our model captures key features of within-vector viral progression and allows bid

Projecting tick-borne encephalitis risk in Sweden under climate change scenarios: a high-resolution spatio-temporal modeling approach.

BACKGROUND: Tick-borne encephalitis (TBE) is a serious vector-borne neurological disease in Europe, with a complex transmission cycle involving ticks of genus Ixodes, animal hosts, environmental and climatic determinants. METHODS: We modelled annual Geocoded Swedish TBE case data for the period 2005–2023 as a log-Gaussian Cox process in relation to population, environmental and climate data, and wildlife citizen science reports at high spatial resolution. We used the computationally efficient Integrated Nested Laplace Approximation (INLA) and projected the future TBE incidence using fifteen re

Confusion beyond the usual suspects.

Acute confusion in older adults is frequently attributed to systemic infections, often delaying the recognition of central nervous system involvement. West Nile virus (WNV) is an emerging cause of viral encephalitis in Europe, particularly during summer and autumn. We report the case of an 82-year-old man admitted in late summer in Northern Italy with fever, agitation, and fluctuating confusion. Initial investigations suggested a systemic infection, and empirical antibacterial therapy was started. Persistent fever, sterile cultures, and ongoing cognitive fluctuations prompted lumbar puncture,

Evaluation of the safety and insecticidal efficacy of ivermectin-treated bird feed formulations in different avian species.

BACKGROUND: West Nile virus (WNV) is maintained in an enzootic cycle between reservoir host birds and Culex (Cx.) spp. mosquitoes. This relationship presents a potential target for vector control strategies. Ivermectin (IVM), an endectocidal drug that selectively affects invertebrates while remaining safe at high concentrations in mammals and birds, can be delivered to Culex tarsalis via blood meals from birds fed IVM-treated bird feed. In this study, we evaluated the safety, efficacy, and utility of IVM-treated bird feed as a novel vector control strategy by assessing its impact on multiple b

West Nile Virus in Europe: Epidemiology, Vector Ecology, Environmental Drivers, and the Role of Equine Sentinel Surveillance in a One Health Framework.

West Nile virus (WNV) is a mosquito-borne flavivirus that remains an important public and veterinary health concern across Europe. Periodic outbreaks affecting humans, horses, and wildlife highlight the complex ecological interactions underlying viral circulation. This narrative review aims to synthesize current knowledge regarding WNV epidemiology, transmission dynamics, and surveillance strategies in Europe, with particular attention to the Romanian context. Available surveillance data indicate recurrent seasonal transmission in several European regions; however, reported case numbers may be

Reframing West Nile Virus in Latin America: From Enzootic Evidence to Human Risk-Surveillance Gaps and One Health Actions.

West Nile virus (WNV) is a mosquito-borne flavivirus with one of the widest global distributions. Since its discovery in Uganda in 1937, it has become a major zoonotic pathogen, and after its introduction into the United States in 1999, it spread rapidly across the Americas, becoming the leading cause of neuroinvasive arboviral disease. Its expansion illustrates a remarkable ecological adaptability, further intensified by climate change. In Latin America and the Caribbean, WNV circulation has been consistently documented in birds, horses, and mosquitoes; however, confirmed human cases remain d

Spatiotemporal and Demographic Patterns of West Nile Neuroinvasive Disease in Vojvodina, Serbia, 2012-2025.

West Nile neuroinvasive disease (WNND) causes substantial morbidity in endemic regions, yet data on its burden in Serbia remain limited. We conducted a retrospective, population-based study of WNND cases reported in Vojvodina Province, Serbia, from 2012 to 2025. Incidence and mortality trends were analysed by year, residence, age, sex, and week of symptom onset. Multivariable logistic regression was used to identify predictors of fatal outcome. Of 1337 suspected cases, 557 (41.66%) met the WNND case definition (530 confirmed, 27 probable cases) and 98.9% were autochthonous. The mean annual inc

Relative proportion and contrasting host preference of Culex pipiens biotypes across Europe.

BACKGROUND: The primary vector of West Nile virus (WNV) in Europe, Culex pipiens, has two morphologically identical but behaviorally and genetically distinct biotypes, here referred to as Pipiens and Molestus. Pipiens and Molestus, and their hybrids, are differentially distributed across Europe and display variable patterns of blood-feeding on birds and humans across the continent, but whether host choice correlates with host preference is unclear. METHODS: Samples of mosquitoes were collected and subsequently biotyped using real-time PCR, following which the relative proportions of each biot
