Modelling the risk of West Nile virus infection in seven European countries from published serological and case notification data, 2008 to 2022.

BACKGROUNDWest Nile virus (WNV) is a zoonotic mosquito-borne pathogen increasingly reported in Europe.AIMWe aimed to characterise heterogeneities in the average annual human risk of WNV infection (force of infection, FOI) and in WNV surveillance across Europe.METHODSWe conducted a systematic review following the PRISMA guidelines to identify serological studies on WNV in humans with IgG-based assays in Europe. We then used mathematical models fitted to both age-stratified serosurvey and case data to reconstruct spatially explicit FOI estimates, the sensitivity of syndromic surveillance and age

Modelling serological cross-reactivity to disentangle the dynamics of West Nile and Usutu viruses in an emerging area.

Zoos may serve as sentinel sites for zoonotic vector-borne diseases. West Nile virus (WNV) and Usutu virus (USUV) are closely related orthoflaviviruses transmitted between Culex mosquitoes and a bird reservoir. Both viruses can also infect mammals, including humans, where they may cause symptoms and, more rarely, hospitalization and death. However, serological cross-reactivity between WNV and USUV complicates their differential diagnosis. Here, we aimed to reconstruct the dynamics of emergence of WNV in a zoo located in a newly affected area in Europe, using ELISA and Virus Neutralization Test

RNF138-Mediated Ubiquitination and Degradation of NS5 Restricts Tick-Borne Encephalitis Virus Infection.

Tick-borne encephalitis virus (TBEV) causes severe neurological disease. However, whether host restricts its replication and the underlying mechanisms remain incompletely delineated. Here, we identify the E3 ubiquitin ligase RNF138 as an intrinsic restriction factor targeting the viral RNA-dependent RNA polymerase (RdRp) NS5. Mechanistically, RNF138 directly interacts with the RdRp domain of NS5 through its ubiquitin-interacting motif, catalyzes K48-linked polyubiquitination and induces degradation of TBEV NS5. Ubiquitin-remnant profiling and mutational analyses identify K372, K462, and K470 w

Molecular Mimicry by the Tick-Borne Encephalitis Virus E Protein: A Hidden Link to Autoimmunity.

In this study, we combined computational predictions with experimental validation as a hybrid strategy to explore whether the E protein of tick-borne encephalitis virus (TBEV) possesses autoimmune potential. Using in silico homology searches, we identified two viral epitopes (evglekl and vtgtqgt) within the TBEV E protein that share sequence identity with fragments of the human proteins DNAH7 and CSMD2. Antibodies against these epitopes were detected in the plasma of a subset of patients after natural TBEV infection. Notably, no such antibodies were found in recipients of the Tick-E-Vac vaccin

Spatio-temporal evaluation of EpiDCA: Case studies on avian influenza, African swine fever, and West Nile virus disease.

Event-Based Surveillance (EBS) systems play an important role in the early detection of disease outbreaks by monitoring unstructured data sources. However, they face key challenges, including handling the overwhelming volume of collected articles, detecting false positives, and the lack of explainability in detected events. To address these limitations, we previously developed EpiDCA, an unsupervised model that integrates epidemiological and environmental data, as well as expert-defined parameters, into the classification process. Its initial application to avian influenza (AI) in Asia demonst

Fine-scale heterogeneity and local amplification of West Nile virus in urban environments in Berlin.

Climate change can intensify mosquito-borne disease risks through rising temperatures and more frequent extreme weather events. To mitigate effects of climate change, cities are adopting nature-based solutions, such as urban greening and rainwater management, yet their implications for vector-borne diseases and host community composition remain poorly understood. West Nile virus (WNV), an emerging mosquito-borne human pathogen in Europe, is primarily transmitted between birds and mosquitoes. Using mosquito sampling at five sites within a one-square-kilometre area in Berlin, Germany, we examine

Economic burden estimation to inform prevention policies against emerging zoonotic diseases: The case of tick-borne encephalitis in France.

OBJECTIVES: Economic evidence on health losses can guide prioritisation of prevention policies for emerging zoonotic risks. Tick-borne encephalitis (TBE) is gaining attention in Western Europe, and notifications are rising in France. Our goal is to scope existing European burden estimates and to generate the first national figures for France using a standardized framework that can inform health policies. STUDY DESIGN: This study was designed as a nationwide model-based cost-of-illness analysis combining a systematic literature review with a decision-tree simulation to estimate the costs of TB

Tissue tropism and mRNA expression profiles of selected innate immunity-related genes following experimental tick-borne encephalitis virus and louping ill virus infection of sheep.

UNLABELLED: Tick-borne encephalitis virus (TBEV) and louping ill virus (LIV) are genetically and antigenically closely related tick-borne flaviviruses responsible for different disease outcomes in sheep. In order to determine whether key components of the innate immunity could contribute to the observed difference in virus dissemination and disease outcome, 8-month-old sheep were inoculated intradermally with TBEV Neudoerfl strain or LIV LI/31 strain. Only LIV-infected sheep developed fever and clinical signs. No TBEV RNA was detected in serum and examined tissues, except for low amounts in th

Río Negro virus (Venezuelan equine encephalitis virus complex) shows pathogenic potential in a permissive murine model.

Río Negro virus (RNV), an enzootic member of the Venezuelan equine encephalitis virus (VEEV) complex, has historically been regarded as avirulent based on studies in adult immunocompetent animals. However, its pathogenic potential under biologically relevant conditions has not been systematically examined. Here, we investigated RNV infection in infant C57BL/6 mice as a permissive developmental model. Infection resulted in a transient viremic phase followed by systemic dissemination, disruption of splenic immune architecture, and pronounced neuroinflammatory and neurodegenerative alterations in

The ER-Resident Protein PDZD8 Inhibits Virus Production of West Nile Virus.

West Nile virus (WNV) remodels the endoplasmic reticulum (ER) membrane in cells to aid virus production. However, the molecular mechanisms underlying this process remain unclear. PDZD8 is an ER-resident protein that mediates ER-mitochondria contact and lipid transfer, both of which are associated with membrane remodeling. In this study, we found that WNV infection suppressed the expression of PDZD8, leading to a reduction in ER-mitochondria contact sites. By contrast, PDZD8 suppressed WNV production by inhibiting the processes of viral particle assembly and release. These findings suggest that

Acute confusional states and functional recovery after West Nile virus infection in hospitalized older adults.

The functional consequences of symptomatic West Nile virus (WNV) infection in hospitalized older adults remain poorly characterized. We aimed to describe the clinical presentation, acute confusional states, and six-month functional recovery trajectories of older adults hospitalized with symptomatic WNV infection. We conducted a retrospective case series of consecutive patients aged ≥65 years with laboratory-confirmed WNV infection admitted to a secondary-care hospital in Sardinia, Italy. Functional status was assessed using the Barthel Index at admission, discharge, and six-month follow-up. Se

A distinct E dimer epitope underlies selective recognition by a protective human West Nile virus antibody.

Human outbreaks of West Nile virus (WNV) are an imminent threat in North America, with many annual infections and numerous cases of severe neuroinvasive disease. There are no licensed treatments for WNV disease. Previous research identified WNV-86 as an ultrapotent neutralizing human antibody that binds domain II of the major envelope (E) glycoprotein in mature virions. Here, we report the structure of mature WNV in complex with the Fab of WNV-86, at a resolution of 3.8 Å, solved using cryogenic electron microscopy. Structure-based epitope mapping identifies a new class of E dimer epitope (EDE

In silico design and evaluation of a multi-epitope vaccine targeting eyach virus for the prevention of tick-borne encephalitis in humans.

Eyach virus is a tick-borne pathogen associated with neurological complications resembling encephalitis, and its increasing emergence highlights a growing public health concern. The absence of specific antiviral therapies and limited surveillance data emphasize the urgent need for effective preventive strategies such as vaccine development. This study presents an immunoinformatics-driven design and in silico evaluation of a multi-epitope vaccine candidate targeting Eyach virus. Structural proteins VP5 and VP7 were analyzed to identify highly antigenic, non-allergenic, and non-toxic B-cell, CTL

Seroprevalence of West Nile virus among blood donors in mainland France, 2021 to 2022.

BACKGROUNDWest Nile virus (WNV) is an orthoflavivirus maintained in an enzootic cycle between birds and ornithophilic Culex mosquitoes. Human infections are usually asymptomatic or paucisymptomatic, while severe neurological disease occurs mainly in children, elderly and immunocompromised individuals. Despite evidence of WNV circulation in France since the 1960s, nationwide seroprevalence data are unavailable.AIMWe wanted to determine the seroprevalence of WNV among blood donors in mainland France to characterise the epidemiology of WNV and assess risk factors and potential implications for tr

Patterns of emergence and circulation of West Nile Virus in Algeria.

In recent years, many countries have experienced increased outbreaks of vector-borne diseases, such as West Nile virus (WNV). WNV is primarily transmitted to humans by the bite of infected Culex mosquitoes and has an enzoonotic transmission cycle involving birds. In Algeria, serological evidence of WNV in humans dates back to the 1970s, but circulation patterns are poorly characterized. To address this, we conduct a cross-sectional seroprevalence survey of WNV in humans and fit serocatalytic models to the data. We also analyze data on severe WNV cases. In this study, we report a seroprevalence

CSF Testing for Neuroinvasive West Nile Virus and Measures to Improve Guideline Adherence.

BACKGROUND AND OBJECTIVES: Diagnosis of West Nile virus (WNV) neuroinvasive disease, an important cause of neurologic disability in endemic areas, requires appropriate testing given its often nonspecific presentation. Guidelines recommend CSF testing of WNV-specific immunoglobulin M (IgM) in all patients, with additional reverse transcription-polymerase chain reaction (RT-PCR) testing only in those who are unable to mount a humoral response due to significant immunosuppression. WNV testing is known to be underused, but real-world guideline adherence has not been described. METHODS: We analyze

Human West Nile virus lineage 2 neuroinvasive infection as a cause of fatal Guillain-Barré syndrome.

BACKGROUND: West Nile virus (WNV) is now endemic in Europe and continues to spread to several countries, with strains 1 and 2 primarily responsible for outbreaks. According to the European Centre for Disease Prevention and Control (ECDC), Italy accounts for the majority of locally acquired cases and West Nile neuroinvasive disease (WNND). CASE REPORT: We present the first case described in Italy and Europe of WNND-related Guillain-Barré syndrome (GBS). Phylogenetic analysis showed that the genome belonged to the WNV-2, sublineage 2a, clustering within sequences from genomes originating mainly

[From Bite to Loss of Vision-Ocular Complications of West Nile Virus].

Ocular involvement related to West Nile Virus infection is receiving increasing attention in Europe as the virus expands northward due to climate change. Next to the well-known neurological complications, the virus can also affect the eye, often in cases of severe neuroinvasive disease, and lead to retinal changes. The most common ocular manifestation is multifocal chorioretinitis with nummular lesions distributed in a distinctive linear pattern. Uveitis, optic neuritis, retinal vasculitis, and in some cases permanent vision loss are reported as well. Here, we present a case of West Nile Virus
