The Clinical Significance of Streptococcus mitis Bacteremia.

BACKGROUND: Except for their role in bacterial endocarditis, nonpneumococcal Streptococcus mitis group (SMG) bacteria have traditionally been regarded as commensals of the oropharynx. Widespread adoption of matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry has increased identification of these organisms and has led to increased concern about their clinical significance. We evaluated SMG bacteremia in a large multicenter cohort to characterize associated clinical syndromes and better define their clinical relevance. METHODS: We performed a retrospective r

A Two-Year Retrospective Study of Blood Cultures in a Secondary Western Greece Healthcare Setting.

Blood culture remains the gold standard for identifying bloodstream infections caused by bacteria and fungi. Isolation of the culprit microorganism onto agar plates also facilitates antimicrobial susceptibility testing. The purpose of this study was to determine the contamination rates, pathogen profile, and antimicrobial resistance in a secondary healthcare setting in a two-year timeframe. In this study, data regarding blood cultures of the years 2023 and 2024 were retrospectively analyzed to address the above questions. Blood cultures were incubated for seven days before being discarded as n

Inference of ceftobiprole susceptibility through surrogate testing of ceftaroline.

UNLABELLED: Ceftobiprole is an advanced-generation cephalosporin active against methicillin-resistant Staphylococcus aureus and other clinically relevant bacteria. Ceftobiprole was recently (April 2024) approved for clinical use in the United States, but susceptibility testing is still not available on commercial automated susceptibility testing systems. We evaluated the accuracy and error rates when using susceptibility to ceftaroline to predict susceptibility to ceftobiprole. A total of 42,363 clinical isolates were collected through the SENTRY Antimicrobial Surveillance Program from 34 US m

A modular genetic toolbox for precise gene regulation and multi-color imaging in streptococci.

Fluorescent labeling is a powerful tool in microbiology allowing live cell imaging and providing insights into dynamic cellular processes, quantification of gene expression, and protein subcellular localization. Although multicolor imaging is widely used in Streptococcus pneumoniae and S. mutans, this is less common in other streptococcal species. To address this gap in the streptococcal molecular toolbox, we benchmarked five different fluorescent proteins. They were fused to the C-terminus of S. pneumoniae HlpA, a small non-specific DNA binding histone-like protein. These reporters, combined

tRNA modification landscapes in streptococci: shared losses and clade-specific adaptations.

tRNA modifications are central to bacterial translational control. Here, we integrated genetics, mass spectrometry, epitranscriptomics and comparative genomics to map the tRNA modification genes of the Gram-positive pathogens Streptococcus mutans and Streptococcus pneumoniae. Both species show a marked loss of modifications dependent on Fe-S enzymes, consistent with a broader trend of Fe-S enzyme reduction in Streptococcus central metabolism. In addition, the D, m1A, m7G, t6A and i6A modifications were mapped in S. pneumoniae tRNAs, and we confirmed that a unique DusB1 enzyme is responsible fo

Evidence of transfer of antimicrobial resistance genes from the porcine pathogen Streptococcus suis to human clinical isolates of Streptococcus agalactiae in a major pig-producing region of Spain.

The emergence of antimicrobial resistance (AMR) is a global threat, with livestock antibiotic use proposed as a significant contributor. We investigated Streptococcus suis, a multidrug-resistant porcine pathogen, as a potential source of AMR gene dissemination to human-associated streptococci in Aragón, Spain. Among 91 porcine isolates recovered across Spain, erythromycin and tetracycline resistance was linked to erm(B) and tet(O) genes, frequently co-located on Tn5252-family integrative and conjugative elements (ICEs) and integrative and mobilizable elements (IMEs). Tn5252-family ICEs shared

Differential susceptibility of cariogenic and commensal oral Streptococcus species to chemically distinct disinfectants including ozone water in vitro.

BACKGROUND: Streptococcus mutans is the primary causative agent of dental caries. Many Streptococcus species present in the oral cavity are thought to increase the risk of infective endocarditis and pneumonia. Conversely, S. sanguinis, S. oralis, S. salivarius, S. gordonii, and S. mitis are streptococci frequently isolated from the oral cavity and are thought to suppress the colonisation of cariogenic bacteria. Recently, mouthwashes have gained attention as a preventive strategy against dental caries. Therefore, we conducted an experiment to verify the caries prevention effect of various disin

Two Rare Cases of Neonatal Meningitis Caused by Globicatella sanguinis at the Marrakech University Hospital: Diagnosis and Therapeutic Implications.

Two infants, aged 3 and 5 months, were admitted to the neurosurgery ward with febrile meningeal signs and underwent lumbar punctures, which revealed turbid cerebrospinal fluid (CSF) with a predominantly neutrophilic pleocytosis, decreased CSF glucose levels, and elevated CSF protein levels. CSF cultures revealed alpha-hemolytic colonies. Identification by time-of-flight mass spectrometry with matrix-assisted laser desorption and ionization (MALDI-TOF) confirmed Globicatella sanguinis (G. sanguinis) in both cases. G. sanguinis is a pathogenic bacterium that is rarely isolated in clinical practi

Hemolytic Anemia From Native Aortic Valve Infective Endocarditis Due to Streptococcus gordonii in a Patient With End-Stage Renal Disease on Hemodialysis: A Case Report.

Infective endocarditis (IE) complicated by non-immune hemolytic anemia is a rare but clinically significant presentation, particularly in native valve disease, and often reflects severe valvular destruction leading to mechanical red blood cell fragmentation. Patients with end-stage renal disease (ESRD) on hemodialysis are at increased risk of bloodstream infections due to chronic vascular access, making early recognition essential. We report the case of a 36-year-old man with ESRD on hemodialysis via an arteriovenous (AV) graft who presented with acute dyspnea, hypoxia, and severe transfusion-

Elucidating the genetic architecture of yield components in pyramidale wheat (Triticum turgidum L.): a comparative approach using QTL analysis and genomic selection.

BACKGROUND: Diabetic retinopathy (DR) is a major microvascular complication of diabetes mellitus and a leading cause of visual impairment. While dyslipidemia has been implicated in DR pathogenesis, the relationship between high-density lipoprotein cholesterol (HDL-C) and DR remains controversial. This study aimed to investigate the association between HDL-C levels and DR prevalence in diabetic patients type 2 diabetes mellitus (T2DM). METHODS: This study is the second analysis based on a cross-sectional studv. A total of 2001 (858 men and 1143 women) diabetic patients who visited the diabetic

Alternative quadruplex real-time PCR reactions for detection and discrimination of Streptococcus pneumoniae serotypes within serogroup 6.

UNLABELLED: Streptococcus pneumoniae causes significant morbidity and mortality worldwide, and serotyping is important to assess the burden of disease that is vaccine preventable. For serotyping, the Centers for Disease Control and Prevention (CDC) use a series of 12 real-time multiplex PCRs (rmPCRs) performed in quadruplex reactions; however, rmPCR reaction 5 (rmPCR-5) for serotypes 6A, 6B, 6C, and 6D often failed at low DNA concentrations. This study investigated the cause of rmPCR-5 failure and provided alternative rmPCRs to resolve this issue. Quadruplex rmPCR target sequences were compare

Precise Manipulation of Bubble Cavitation and Blood-Brain Barrier Modulation using Temporal Interference Ultrasound.

Objective: To determine whether temporal interference (TI) ultrasound can enhance microbubble (MB) cavitation and provide transient modulation of blood-brain barrier (BBB) permeability. Impact Statement: By integrating a time-dependent multiphysics MB-vessel model with in vivo validation, this study links TI-enhanced cavitation dynamics to vessel-wall mechanical responses and a transient BBB permeability window, providing a mechanistic basis for pressure-efficient ultrasound-mediated brain delivery. Introduction: Focused ultrasound combined with MBs can transiently increase BBB permeability, b

Update on the evolving landscape of pneumococcal capsule types: new discoveries and way forward.

SUMMARYStreptococcus pneumoniae (the "pneumococcus") is a significant human pathogen. The key determinant of pneumococcal fitness and virulence is its ability to produce a protective polysaccharide (PS) capsule, and anti-capsule antibodies mediate serotype-specific opsonophagocytic killing of bacteria. Notably, immunization with pneumococcal conjugate vaccines (PCVs) has effectively reduced the burden of disease caused by serotypes included in vaccines but has also spurred a relative upsurge in the prevalence of non-vaccine serotypes. Recent advancements in serotyping and bioinformatics survei

Structural analysis of extracellular ATP-independent chaperones of streptococcal species and protein substrate interactions.

UNLABELLED: During infection, bacterial pathogens rely on secreted virulence factors to manipulate the host cell. However, in gram-positive bacteria, the molecular mechanisms underlying the folding and activity of these virulence factors after membrane translocation are not clear. Here, we solved the protein structures of two secreted parvulin and two secreted cyclophilin-like peptidyl-prolyl isomerase (PPIase) ATP-independent chaperones found in gram-positive streptococcal species. The extracellular parvulin-type PPIase, PrsA in Streptococcus pneumoniae and Streptococcus mutans maintain dimer
