Introduction. Recently, flow cytometry has gained the attention of clinical microbiologists for its ability to characterize bacterial species. This article shows how acoustic-enhanced flow cytometry, combined with the fluorescent dye SYTO 9, can differentiate between Gram-positive and Gram-negative bacteria.Gap Statement. SYTO 9 is a cell membrane-permeable dye with a high affinity for nucleic acids in both living and non-living prokaryotic and eukaryotic cells and has been used as a counterstain to discriminate between live and dead cells in combination with other dyes. However, the consisten

Klebsiella pneumoniae is a gram-negative, facultative anaerobic bacillus, that represents a significant global health concern due to its role as a major pathogen in multidrug-resistant infections. The growing prevalence of carbapenem-resistant K. pneumoniae infections among human necessitates the development of alternative and complementary therapies. Phages can effectively lyse drug-resistant pathogenic bacteria. In this study, stability tests confirmed that phage P560 was stable over a wide range of pH (3 to 10) and temperatures (4 to 50 °C) conditions. The growth characteristics showed that

BACKGROUND: Klebsiella pneumoniae continues to be a problem worldwide. This study aimed to elucidate the evolution of a K. pneumoniae strain from harbouring blaKPC−2 to blaKPC−90. METHODS: Three K. pneumoniae strains (Kpn_XM11, Kpn_XM12, and Kpn_XM13) belonging to the same clone were isolated from a patient, followed by identification, antimicrobial susceptibility test, whole genome sequencing, and gene mutations. RESULTS: Kpn_XM11 and Kpn_XM12 strains harbouring blaKPC−2 were resistant to carbapenems but susceptible to ceftazidime-avibactam (CZA) while Kpn_XM13 possessing blaKPC−90 was resist

Many bacteria harbor an ATP-binding cassette (ABC) transporter named EgtU specific for the human dietary antioxidant and 2-thioimidazole-containing low-molecular weight thiol ergothioneine (ET). How the solute binding domain, EgtUC, discriminates among ET and other similar molecules is unknown. Here, we use a "chimeric" mutagenesis strategy and two distantly related EgtUCs from Streptococcus pneumoniae and Helicobacter pylori to show that a suite of EgtUC alkyl CH•••S hydrogen bonds to the ET thione S atom are central determinants of molecular recognition. Small perturbations in CH•••S distanc

BACKGROUND: Klebsiella pneumoniae (Kpn) is a hospital-acquired pathogen of critical importance due to extensive resistance to last-resort antibiotics like carbapenems and colistin. The emergence of hypervirulence and its convergence with antibiotic resistance in some Kpn lineages is a public health threat. Understanding the regional prevalence of different lineages of Kpn and their resistomes and virulomes, in the backdrop of global data, will be valuable in tackling Kpn infections. METHODS: A collection of 22,569 publicly available genomes of human Kpn isolates, from 83 countries, including t

Stenotrophomonas maltophilia is intrinsically resistant to carbapenems and most β-lactam antibiotics. Empiric therapy for bloodstream infections often includes either trimethoprim-sulfamethoxazole or levofloxacin. However, there are currently no European Committee on Antimicrobial Susceptibility Testing (EUCAST) breakpoints available for interpretation of susceptibility testing for levofloxacin. We examined the relationship between levofloxacin minimum inhibitory concentration (MIC) and clinical outcomes in S. maltophilia bacteraemia. We retrospectively examined 79 consecutive patients with S.

BACKGROUND: Klebsiella pneumoniae is a major cause of severe healthcare- and community-associated infections, and both hypervirulent and carbapenem-resistant forms are increasingly reported worldwide. However, the prognostic value of virulence factors remains uncertain. We conducted a systematic review and meta-analysis to evaluate their association with mortality.
METHODS: We searched PubMed, Scopus, and Cochrane Library for studies (2000-2024) reporting virulence factors in K. pneumoniae infections. Meta-analysis compared the prevalence of factors reported in ≥ 5 studies between survivors a

In recent years, the numbers of tularemia cases have risen. The rarest but most lethal form is pulmonary. Diagnosis is particularly difficult in immunosuppressed patients because clinical symptoms and radiological findings are nonspecific. Clinically and radiologically, the disease can mimic other causes of atypical pneumonia or pulmonary infiltrates, especially bacterial or viral pneumonia. Due to these nonspecific features, diagnosis is often delayed. More accurate diagnostics and properly administered therapy prevents complications and lowers lethal courses.

Carbapenem-resistant hypervirulent Klebsiella pneumoniae (CR-hvKP) presents a major challenge for clinical treatment due to its characteristics of drug resistance and hypervirulence. This study aims to investigate the therapeutic effect of a novel tetracycline antibiotic, eravacycline, on CR-hvKP pneumonia. We isolated three CR-hvKP strains from clinical samples and examined their associated phenotypes. Subsequently, we infected mice with strains. The results indicated that the clinical isolates KP78, KP98, and KP100 carried multiple drug resistance genes and virulence genes, exhibiting high v

Klebsiella pneumoniae is an important nosocomial pathogen, considered a critical threat to public health, but its genomic data are scarce in Brazil. Therefore, the aim of this study was to identify the mechanisms of virulence and antimicrobial resistance and to evaluate the molecular epidemiology of K. pneumoniae isolates from an outbreak in a university hospital in Brazil. The DNA of the 25 isolates was sequenced, the functional annotation of the genome was performed in Prokka, for virulence and resistance analyses PanViTa was used, and for Multilocus Sequence Typing analyses pyMLST was used.

BACKGROUND/OBJECTIVES: The pancreatoduodenectomy is a high-risk surgery, and its high morbidity rate is mostly related to postoperative infectious complications. Positive bile cultures are associated with both increased frequency and severity of pancreatic fistulas; this is more common in previously drained or infected bile ducts. The currently used antibiotic regimens for surgical prophylaxis appear insufficient to cover the microorganisms identified in bile cultures of patients previously submitted to biliary drainage. This study aims primarily to compare the rates of positive bile cultures

Background: Cefiderocol, a siderophore cephalosporin, is approved for the treatment of infections caused by multi-drug-resistant Gram-negative bacteria (MRGN). At present, few data are available on the pharmacokinetics of this substance in critically ill patients, particularly for the treatment of central nervous system infections. Patients and Methods: Here, we reported on a 22-year-old male patient after severe open head trauma. Initial screening revealed colonization with 4MRGN A. baumannii (OXA-23) (perianal) and 4MRGN K. pneumoniae (KPC) (tracheal). Unfortunately, he developed ventriculit

BACKGROUND: In 2019, pneumonia caused 740,180 deaths in children under five years of age, representing 22% of global mortality in this age group. During the COVID-19 pandemic, public health interventions markedly reduced the circulation of most respiratory viruses other than SARS-CoV-2, leading to significant post-pandemic shifts in respiratory pathogen epidemiology. This study aimed to characterize the epidemiology, clinical features, and risk factors associated with respiratory viruses and bacteria causing pneumonia in Mexican children during the late pandemic and post pandemic periods.

UNLABELLED: Detection of viable bacteria in complex samples is a long-standing challenge in microbiology and public health. Traditional culture methods are slow and often fail to capture viable but non-culturable cells, while PCR-based methods can overestimate bacterial loads by amplifying DNA from non-viable (dead) cells. To address this challenge, we evaluated a novel, photoactivation-free viability PCR (vPCR) reagent from Promega Corporation that selectively crosslinks DNA from non-viable cells, rendering it unamplifiable by qPCR. This approach enables accurate, culture-independent quantifi

OBJECTIVES: Healthcare-associated infections affect nearly one-third of intensive care unit (ICU) patients and contribute substantially to mortality. Most surveillance systems overlook the diversity of organisms and site-specific resistance patterns that guide empirical therapy. We aimed to describe the distribution of pathogens, resistance trends, and related outcomes across ICUs in the largest single-institution analysis to date.
METHODS: We conducted a retrospective cohort study using the MIMIC-IV database covering 2008 to 2022. The study included adult ICU patients with at least one posit

BACKGROUND: Antimicrobial-resistant pathogenic microorganisms are disease causing microbes that cause several diseases in humans, animals, and plants. Currently, these microorganisms cause more than a quarter of the reported diseases worldwide. The other current global health risk is the imbalance generation of free radicals in human physiology, which exposes several millions to oxidative stress diseases like Alzheimer, Cancer, and Diabetes. OBJECTIVE: The objective of this study was to evaluate the antibacterial and antioxidant activities of Inula confertiflora roots, stems, and leaves extrac

BACKGROUND: The rise of antibiotic-resistant bacteria is a major public health concern. Physical plasma can generate reactive oxygen and nitrogen species (RONS) with antimicrobial activity. Plasma-activated liquid (PAL) can be used as a carrier for RONS. This study investigated the antibacterial effects of PAL against clinically relevant Gram-negative (Escherichia coli, multidrug-resistant (MDR) Escherichia coli, Pseudomonas aeruginosa, Acinetobacter baumannii, Enterobacter cloacae, Klebsiella pneumoniae, and Helicobacter pylori) and Gram-positive (Staphylococcus aureus and Enterococcus faeciu

BACKGROUND: Healthcare-associated infections (HAIs) in Europe affect millions of patients annually, driving antimicrobial resistance and imposing significant economic burdens. Carbapenem-resistant Enterobacterales (CRE) are particularly concerning due to their ability to spread, making genomic surveillance essential for tracking their transmission.
METHODS: From October 2022 to September 2023, we sequenced and analysed the genome of 340 CRE obtained from patients admitted to Fondazione IRCCS Policlinico San Matteo ( Italy). Sequencing was performed using a short-read approach and genomes were
