Diagnosis of Pediatric Myositis Ossificans Based on Cytomorphology and Molecular Analysis From FNAB Sample: A Case Report.

Myositis ossificans (MO) is a benign soft tissue lesion, characterized by ectopic ossification due to inappropriate fibroblast differentiation, most commonly affecting skeletal muscles. It often occurs in young adults after muscle trauma, predominantly in male patients and very rarely in children. We describe the case of a previously healthy 3-year-old boy who developed a lesion in his deltoid muscle after vaccination against tick-borne encephalitis. During an MRI scan, performed under general anesthesia, fine needle aspiration biopsy (FNAB) and core needle biopsy (CNB) were performed. While t

A case report and mini-review of Crimean-Congo hemorrhagic fever with encephalitis: an unexpected complication.

Crimean-Congo hemorrhagic fever is a severe tick-borne viral infection with high mortality rates. While Crimean-Congo hemorrhagic fever primarily presents as a hemorrhagic fever, central nervous system involvement, including encephalitis, is rare. The virus, transmitted through tick bites or direct contact with infected animal blood or bodily fluids, can lead to multi-organ failure. Neurological manifestations of Crimean-Congo hemorrhagic fever remain poorly understood. We report a 40-year-old man from Hormozgan province, Iran, who presented with fever, hematemesis, abdominal pain, and neurolo

Seroprevalence of IgG Antibodies Against Borrelia burgdorferi Sensu Lato, Anaplasma phagocytophilum, and Tick-Borne Encephalitis (TBE) Virus in Horses in Southern Norway.

Ixodes ricinus ticks play a crucial role as carriers of diseases, transmitting pathogens to vertebrate hosts, including horses. This study aimed to investigate the seroprevalence of IgG antibodies against Borrelia burgdorferi sensu lato (s. l.), Anaplasma phagocytophilum, and tick-borne encephalitis virus (TBE-virus) in equine sera collected in southern Norway. In total, sera from 331 horses stabled in four counties (Agder, Vestfold and Telemark, Vestland, and Viken) were analyzed by immunoblot. In total, 66% of the horses were IgG-seropositive for antibodies against one or multiple tick-borne

Tick-borne flavivirus exoribonuclease-resistant RNAs contain a double loop structure.

Viruses from the Flaviviridae family contain human relevant pathogens that generate subgenomic noncoding RNAs during infection using structured exoribonuclease resistant RNAs (xrRNAs). These xrRNAs block progression of host cell's 5' to 3' exoribonucleases. The structures of several xrRNAs from mosquito-borne and insect-specific flaviviruses reveal a conserved fold in which a ring-like motif encircles the 5' end of the xrRNA. However, the xrRNAs found in tick-borne and no known vector flaviviruses have distinct characteristics, and their 3-D fold was unsolved. Here, we verify the presence of x

Tick hazard in a Central European country: Mapping Europe's principal tick-borne disease vector across Germany.

The most common European tick species, Ixodes ricinus, is the principal vector of Borrelia and tick-borne encephalitis virus and several other pathogens of public health relevance in Europe. Comprehensive data on tick abundance and the underlying ecological drivers are crucial for developing awareness and control strategies and to assess future changes in tick-borne disease risk. We aimed to provide a Germany-wide map of I. ricinus abundance to aid in disease transmission risk assessment. During 2018-2020, questing tick density was assessed at 83 sites across the whole country by drag flagging

Cecropin anisaxin-2S has in vitro immunomodulatory, but not antiproliferative and antiviral properties.

Helminthic host defense peptides (HDP) are pleiotropic, multifunctional effector molecules of helminth immunity, efficient against Gram-negative and Gram-positive bacteria. Among them, anisaxin-2S (A-2S), membranolytic cecropin-like HDPs produced by the zoonotic nematodes of the genus Anisakis, shows remarkable efficacy even against multidrug-resistant Gram-negative bacteria, yet its immunomodulatory, antiproliferative and antiviral properties have not been elucidated. Therefore, we tested A-2S immunomodulation in the common carp (Cyprinus carpio) blood cells exposed to two pathogens, the zoon

Lack of serologic evidence of orthoflavivirus infection in dogs with meningoencephalitis of unknown origin and steroid-responsive meningitis-arteritis in the Netherlands.

The pathogenesis of meningoencephalomyelitis of unknown origin (MUO) and steroid-responsive meningitis-arteritis (SRMA) in dogs remains enigmatic. Numerous studies have attempted and failed to identify (viral) pathogens in samples from MUO- or SRMA-diagnosed dogs. Orthoflavivirus-associated meningoencephalitis or meningoencephalomyelitis has been diagnosed in dogs in several European countries. We investigated serologic evidence for orthoflavivirus infection in dogs with clinical diagnoses of MUO or SRMA in the Netherlands. Twelve dogs with a clinical diagnosis of MUO based on signalment, neur

Development of Langat virus infectious clones as a platform for live-attenuated tick-borne encephalitis vaccine.

Tick-borne encephalitis (TBE) is one of the most important tick-transmitted diseases in Europe and Asia. With no specific antiviral treatment available, vaccination remains the most effective protective strategy for TBE. Unlike currently available inactivated TBE vaccines that require repeated boosters, live-attenuated vaccines could offer lifelong immunity with a single dose. Langat virus (LGTV) is a naturally attenuated strain of TBE virus (TBEV). In this study, we engineered and rescued four infectious clones (ICs) of LGTV using RNA- and DNA-based reverse genetics methods. The ICs rescued b

Molecular Typing of Tick-Borne Pathogens in Ixodids of Bosnia and Herzegovina.

Ticks are key vectors of zoonotic pathogens, and their expanding distribution in Europe heightens public health concerns. In Bosnia and Herzegovina, while tick distribution is well documented, molecular data on tick-borne pathogens remain limited. This study aimed to illustrate the presence and diversity of these pathogens, focusing on areas with high human activity. Ticks (n = 556) were collected in April 2022 from eight diverse locations, including urban parks, private properties, and rural sites. PCR-based screening was employed to detect Anaplasmataceae, Borrelia, Francisella, Piroplasmida

A Case Series of Powassan Meningoencephalitis in a Single Institution in Connecticut With One Case Resembling Herpes Simplex Virus (HSV) Meningoencephalitis on MRI.

Powassan virus (POWV) is a single-stranded RNA virus that belongs to the genus Orthoflavivirus, which is transmitted to humans through the bite of infected deer tick (Ixodes scapularis), with peak transmission during summer and fall. Most patients are asymptomatic; those who develop symptoms usually present with flu-like symptoms or neuroinvasive disease, including encephalitis, meningoencephalitis, or aseptic meningitis. The gold standard test for diagnosis is serological testing. The treatment is usually supportive care. In this case series, we have reported two cases that presented with fev

Exploring approaches and advancements in the development and evaluation of multi-epitope subunit vaccines against tick-borne viruses.

Tick-borne viruses (TBVs) pose significant public health challenges worldwide, causing severe diseases such as hemorrhagic fevers and encephalitis. Given the limitations of existing therapeutic and preventive measures, the development of effective vaccines represents a critical strategy for controlling TBV-related infections. This review explores recent advancements in the design and evaluation of multi-epitope subunit vaccines targeting key TBVs, including hemorrhagic fever viruses (e.g., Crimean-Congo hemorrhagic fever virus, Severe fever with thrombocytopenia syndrome virus), encephalitis v

Ticks and Tick-Borne Pathogens Encountered by Dogs and Cats: A North European Perspective.

Climate change is increasing the worldwide burden of tick-borne diseases (TBDs). Dramatic increases in human cases of borreliosis have been reported during the past few decades, including from Finland, located in North Europe. As human exposure to ticks carrying pathogens is increasing, so likely is exposure of dogs and cats. However, feline or canine TBD cases are not notifiable. Likewise, no combined databases of cases exist in Finland, hindering assessment of related trends. Here, we utilize crowdsourced tick samples to reveal how commonly and to which species of TBPs dogs and cats are expo

Viral Isolation and Culture from Field-Collected Ticks: In Vitro.

Ticks may contain a diverse array of both pathogenic and invertebrate-specific viruses. Culture methods can be performed in vitro or in vivo to isolate and culture these viruses. Here, we demonstrate the culture of Deer tick virus (DTV) and Powassan virus (POWV) from ticks collected in Upstate New York. Research into medically significant tick-borne pathogens can be facilitated through in vitro models using cultured mammalian cells. This is especially true of viruses, which require amenable host cells to replicate. This chapter describes the isolation of viruses from field-collected ticks util

Virus Isolation from Field-Collected Ticks: In Vivo.

As tick-borne viruses such as Powassan virus (POWV) and Deer tick virus (DTV) increase in prevalence, having methodology to isolate circulating strains from field-collected ticks grants an opportunity to better understand viral evolution, to more accurately assess human health risks, and to make research into these viruses more translational. POWV and DTV are genetically similar but distinct emerging tick-borne viruses in North America that can cause severe encephalitic disease with high morbidity and mortality. Research into these viruses typically use prototypical lab strains, which may not

Human Skin Ex Vivo Culture for Tick-Borne Virus Infection Studies.

Tick-borne viral infections, like those described in this book, pose real threats to the health of humans across the world. Powassan virus (POWV) and Deer tick virus (DTV) are two examples of tick-borne flaviviruses that are endemic to North America and cause neuroinvasive disease. These two viruses are both genetically similar and are spread by similar vectors but have differing tropism and clinical outcomes. Studies involving these viruses have routinely consisted of mouse models and in vitro studies. These models, effective as they are, are limited in their use as they are either costly or

Immunohistochemical Assays to Visualize Tick-Borne Flaviviruses at the Tick Feeding Site.

Traditional immunohistochemistry (IHC) involves chromogenic detection of target antigens by antibodies that are conjugated to enzymes. Such immunostaining of thin tissue sections enables visualization of the distribution and localization of target antigens within their proper histological context. In this chapter, we describe how tick-borne flaviviruses can be visualized at the tick feeding site via IHC. We use antibodies specific to the North American tick-borne flavivirus, Powassan virus (POWV), to demonstrate how viral antigen can be visualized in the skin of a vertebrate host that is fed u

RNA In Situ Hybridization to Visualize Tick-Borne Viruses.

Visualization of tissue and cellular architecture is essential for understanding the pathobiology of tick-borne virus infection. RNA in situ hybridization (ISH) utilizes specific oligonucleotide probes to detect the presence and specific location of complementary RNA transcript targets within a tissue section. Chromogenic or fluorescent staining technologies allow researchers to visualize multiple RNA targets within a single tissue. Herein, we describe an RNA ISH methodology based on a commercially available kit to hybridize specific probes to POWV RNA within infected tissues.
