Seroprevalence study for selected zoonotic vector-borne pathogens in sheep from endemic areas of Croatia.

Surveillance is crucial in controlling and preventing vector-borne zoonotic diseases (VBDs). We analyzed the seroprevalence of selected vector-borne zoonotic pathogens in sheep from endemic areas and their role as possible sentinels for VBDs. A total of 300 sheep from seven farms at three micro-locations were tested for the presence of IgG antibodies against tick-borne encephalitis virus (TBEV), West Nile virus (WNV), Usutu virus (USUV), Borrelia burgdorferi s.l., and Crimean-Congo hemorrhagic fever virus (CCHFV) using ELISA with confirmation of borderline/positive results by VNT. Seropositivi

First report of the taiga tick Ixodes persulcatus in Norway.

The distribution range of the taiga tick, Ixodes persulcatus, has increased in the last decade in Northern Europe. We performed samplings at nine locations in Nordland County, east and northeast of Brønnøysund, south of the Arctic Circle in July 2023. Four of 62 ticks collected were identified as I. persulcatus (one female, two males and one nymph), the remaining were identified as Ixodes ricinus (9 females, 2 males and 47 nymphs). This is the first report of I. persulcatus in Norway. Given the importance of I. persulcatus as vector of Borrelia burgdorferi sensu lato and more pathogenic subtyp

Clinical Spectrum and Dynamics of Sequelae Following Tick-Borne Encephalitis Virus Infection: A Systematic Literature Review.

BACKGROUND: Infection with the tick-borne encephalitis virus (TBEV) can affect the nervous system and lead to significant morbidity. To summarize current knowledge of long-term outcomes following TBEV infection, we systematically reviewed the prevalence of TBEV infection sequelae after hospital discharge across different age groups and follow-up time points. METHODS: Studies of adults, children, and "all-age" populations with laboratory-confirmed TBEV infection were identified via electronic database searches. Study categorization was based on follow-up time after hospital discharge: ≤6, 7 to

Differential Impact of Simultaneous or Sequential Coinfections With Borrelia afzelii and Tick-Borne Encephalitis Virus on the Ixodes ricinus Microbiota.

Ticks, particularly Ixodes ricinus, are significant vectors of pathogens such as Borrelia spp. and tick-borne encephalitis virus (TBEV), which cause Lyme borreliosis (LB) and tick-borne encephalitis (TBE), respectively. Understanding how these pathogens interact within the tick microbiome is essential for developing vector control strategies. This study investigates the impact of Borrelia afzelii and TBEV, as well as their coinfection, on the microbiota composition and structure of I. ricinus nymphs. Using a network-based approach, we analyzed the microbial communities of ticks exposed to infe

An Emerging Cause of Arboviral Encephalitis in Pennsylvania.

Powassan virus (POWV) is a rare neuroinvasive flavivirus transmitted through the bite of an infected Ixodes scapularis tick. Although infections are infrequent, they have become increasingly recognized in recent years, particularly in the northeastern United States. With the expanding geographic range of Ixodes ticks due to climate change, the risk of POWV transmission is rising, making awareness of the virus crucial for timely diagnosis and effective management. Preventive measures, such as avoiding tick exposure, using repellents, and performing thorough tick checks, are essential in reducin

The Role of Lysine Tyrosylquinone Containing Oxidases in Progression of Solid Tumors.

Lysine tyrosylquinone (LTQ), the cofactor formed through copper-assisted tyrosine oxidation and subsequent intramolecular cross-linking, is inherent in all members of the lysyl oxidase family. Lysyl oxidases are unique among amine oxidases in that they maintain the LTQ coenzyme in a relatively surface-exposed position, making it accessible for the oxidative deamination of lysine side chains in various proteins, especially in the extracellular matrix. This process facilitates the formation of intramolecular cross-links, which are vital for the normal development of skin, bones, aorta, and other

Atomic-resolution structure of a chimeric Powassan tick-borne flavivirus.

Powassan virus (POWV) is an emerging tick-borne flavivirus for which no vaccine or antiviral treatment exists. The incidence of human infections of POWV in North America has been increasing because of the expanding distribution of the tick vector Ixodes scapularis that transmits POWV to humans. To mitigate the dangers of handling a risk group 3 human pathogen, a chimeric virus was constructed from the genetic backbone of a yellow fever virus vaccine strain 17D (YFV-17D) and the external structural proteins of POWV lineage II. The chimera had a comparable phenotype as POWV. The atomic resolutio

Exploration of effective pharmacological inhibitors for NS5 protein through computational approach: A strategy to combat the neglected Kyasanur forest disease virus.

Kyasanur Forest Disease Virus (KFDV) poses a significant public health threat due to the limited efficacy of existing vaccines, necessitating the development of effective antiviral therapeutics. The nonstructural protein 5 (NS5), essential for viral RNA synthesis and methylation, serves as a promising drug target. This study employs computational approaches to identify and evaluate potential NS5 inhibitors that may contribute to the development of antiviral compounds against KFDV. The 3D structure of NS5 was predicted using Robetta, SwissModel, and I-TASSER, with the Robetta model (ERRAT score

Viral Infection Induces Alzheimer's Disease-Related Pathways and Senescence in iPSC-Derived Neuronal Models.

INTRODUCTION: The Pathogen Infection Hypothesis proposes that β-Amyloid (Aβ) functions as an antimicrobial peptide, with pathogen-induced aggregation potentially contributing to Alzheimer's disease (AD) pathology. METHODS: We used human iPSC-derived 2D neurons and 3D cerebral organoids from wild-type and familial AD (PSEN1/2 mutant) lines to model acute infections with HSV-1 and TBEV and Aβ aggregation. Transcriptomic and proteomic analyses were conducted to assess molecular responses. RESULTS: HSV-1, but not TBEV, induced robust Aβ clustering, which was, however, dependent on extracellular

Mosquito- and tick-borne orthoflaviviruses cross an in vitro endothelial-astrocyte barrier.

INTRODUCTION: The genus Orthoflavivirus of the Flaviviridae family includes several notable pathogens such as mosquito-borne West-Nile virus (Orthoflavivirus nilense, WNV) and Tick-borne encephalitis virus (Orthoflavivirus encephalitidis, TBEV) that are highly neurotropic and may cause severe neurological disease leading to lifelong disabilities, coma and death. These viruses have developed mechanisms to breach the compact blood-brain barrier (BBB) and establish infection within the central nervous system (CNS). Nevertheless, neuroinvasive mechanisms of orthoflaviviruses remain poorly understo

New record of Ixodes pavlovskyi (Acari: Ixodidae) from the Osaka Prefecture, western Japan: An update on the southernmost distribution record.

Ixodes pavlovskyi inhabits cool temperate zones, especially in Russia, north-eastern China and northern Japan. This tick species is thought to transmit various pathogens such as Borrelia spp., Rickettsia spp. and tick-borne encephalitis virus. Hence, understanding the geographical distribution of I. pavlovskyi is important for estimating risk of tick-borne diseases. Among ticks collected using the flagging method in Osaka Prefecture, south-western Japan, we found a single I. pavlovskyi male that was identified based on its morphological features and the mitochondrial 16S rDNA sequence. To the

Orthoflavivirus omskense NS1 Protein Induces Microvascular Endothelial Permeability In Vitro.

Orthoflavivirus omskense (Omsk hemorrhagic fever virus, OHFV) is a tick-borne flavivirus that causes Omsk hemorrhagic fever (OHF), a severe zoonotic disease endemic to Western Siberia. Despite the fact that the role of NS1 proteins of various mosquito-borne flaviviruses in pathogenesis was investigated and their ability to affect human endothelial permeability was shown, the role of the NS1 protein of OHFV in pathogenesis is unstudied. In this work, the ability of OHFV NS1 to induce human endothelial permeability was investigated for the first time. It was shown that recombinant OHFV NS1 produ

Pharmacological Agent GW4869 Inhibits Tick-Borne Langat Virus Replication to Affect Extracellular Vesicles Secretion.

GW4869, a cell-permeable, selective inhibitor of neutral sphingomyelinase is a pharmacological agent that blocks the production and release of extracellular vesicles (EVs). Our previous studies have shown that GW4869 inhibits flaviviral loads in tick, mosquito and mammalian cells, including murine cortical neurons. Yet the mechanism(s) of GW4869 inhibitor upon viral infections were not addressed. In the current study, we focused on how GW4869 interferes with Langat Virus (LGTV, a tick-borne flavivirus) replication in ISE6 tick cells. First, we found that GW4869 is neither cytotoxic at tested d

Irreversible furin cleavage site exposure renders immature tick-borne flaviviruses fully infectious.

Flavivirus assembly is driven by the envelope glycoproteins pre-membrane (prM) and envelope (E) in the neutral pH environment of the endoplasmic reticulum. Newly budded, spiky particles are exported through the Golgi apparatus, where mildly acidic pH induces a major surface rearrangement. The glycoproteins reorganize into (prM/E)\₂ complexes at the surface of smooth particles, with prM trapped at the E dimer interface, thereby exposing a furin cleavage site (FCS) for proteolytic maturation into infectious virions. Here, we show that in the absence of furin, immature tick-borne flavivirus parti

Metagenomic evidence of viral secretion from tick salivary glands to saliva: implications for potential horizontal transmission.

Ticks transmit diverse viral pathogens to hosts during blood-feeding via saliva secretion. This study characterized viral compositions in salivary glands and saliva from adults of four tick species (Ixodes persulcatus, Rhipicephalus microplus, Haemaphysalis longicornis, and Haemaphysalis concinna) collected in China. Meta-transcriptomic analysis revealed distinct viromes across species, with Flaviviridae dominant in R. microplus, Nairoviridae in H. concinna and I. persulcatus, and Phenuiviridae in H. longicornis and I. persulcatus. Among 27 viruses detected in salivary glands, 14 were identifi

Tick-Borne Encephalitis (TBE) Vaccine in the National Immunisation Programme-For Whom, When and Where?

The incidence of Tick-borne encephalitis (TBE) cases has increased. The presumed location of transmission of Tick-borne encephalitis virus (TBEV) has been expanding increasingly in the western parts of Europe during the past decade. There has also been an increased incidence of surveillance-reported TBE cases in southern Sweden and southern Norway. Additionally, the warmer climate has enabled ticks to be present to a greater extent in the northern part of Scandinavia. In several reports, pre-school children tend to have a lower TBE incidence rate compared to teenagers, whereas both younger chi

A Deep Learning Approach for Classifying Developmental Stages of Ixodes ricinus Ticks on Images Captured Using a Microscope's High-Resolution CMOS Sensor.

This article presents a deep learning approach for classifying the developmental stages (larvae, nymphs, adult females, and adult males) of Ixodes ricinus ticks, the most common tick species in Europe and a major vector of tick-borne pathogens, including Borrelia burgdorferi, Anaplasma phagocytophilum, and tick-borne encephalitis virus (TBEV). Each developmental stage plays a different role in disease transmission, with nymphs considered the most epidemiologically relevant stage due to their small size and high prevalence. We developed a convolutional neural network (CNN) model trained on a da
