Pathogenicity and virulence of Kyasanur Forest disease: A comprehensive review of an expanding zoonotic threat in southwestern India.

Kyasanur Forest Disease (KFD) is a tick-borne viral hemorrhagic fever caused by the KFD virus (KFDV). First identified in 1957 in Karnataka, India, KFD primarily affects humans and non-human primates. The disease has a broad clinical manifestation, ranging from asymptomatic infections to severe hemorrhagic complications, with a mortality rate of 3-10%. Over time, KFD has shown a gradual geographical expansion, with cases now reported beyond Karnataka. KFD has been a public health challenge for nearly seven decades, as no effective vaccine or antivirals are available. Although humoral immunity

Design and implementation of blood donor sample bioarchives to enhance preparedness for emerging and pandemic pathogens in England.

New and emerging infections with blood-borne pathogens pose an ongoing threat to the safety of blood transfusions and transplants. Bioarchives of stored blood donor samples represent a valuable pathogen screening resource for both ensuring safety of blood transfusions and for wider public health infectious disease surveillance. Large scale testing of donors enables early detection of pathogen spread and extent of population exposure. We have implemented two complementary systems for the bioarchiving of blood donor samples in England for these purposes. The CODONET bioarchive collects samples f

In search for tick-borne encephalitis virus and Rickettsia in Belgian Ixodes ricinus ticks: Evidence of rickettsia felis.

This study investigated the presence of tick-borne encephalitis virus (TBEV) in ticks collected in Belgium between 2018 and 2024, as well as the occurrence of Rickettsia spp. in ticks sampled in 2024. A total of 3739 ticks from 17 locations, of which 16 in Flanders and 1 in Wallonia, were collected from vegetation and tested in 510 pools for TBEV by RT-PCR. Sampling locations were selected based on potential exposure risk, including proximity to human TBE cases, evidence of TBEV antibodies in animals, or near the border between Belgium and the Netherlands. No TBEV RNA was detected in this coll

Seroprevalence of seven climate-sensitive zoonoses in Greenland and northern Sweden (1998-2017): High antibody prevalence against Rickettsia and Leptospira, with Leptospira possibly linked to global warming.

BACKGROUND: Climate change may alter zoonotic disease patterns in the Arctic, yet knowledge remains limited. DESIGN: Antibodies to seven zoonotic pathogens were analyzed in 660 unselected human sera drawn from serum banks from Greenland (n = 460) and Northern Sweden (n = 200) (1998-2017), frequency-matched with respect to sex, age, ethnicity and place of living. Greenlandic samples were tested for Francisella tularensis, Brucella melitensis, Brucella abortus, Coxiella burnetii, Rickettsia spp., and Leptospira spp., while Swedish samples also included Borrelia burgdorferi sensu lato (Bbsl) and

Comparison of the epidemiological and clinical fingerprints of Human Granulocytotropic Anaplasmosis and Human Monocytotropic Ehrlichiosis in the United States.

BACKGROUND: Human granulocytotropic anaplasmosis (HGA), caused by Anaplasma phagocytophilum, and human monocytotropic ehrlichiosis (HME), caused by Ehrlichia chaffeensis, are tick-borne zoonoses. The vast majority of Anaplasma phagocytophilum infections reported worldwide are from the United States. Ehrlichia chaffeensis infections are reported exclusively from North America, with the majority reported from the United States. We have recently summarized the available data on HGA and HME in the form of two systematic reviews. We compared data from these two systematic reviews to objectify diffe

RNA virome of ticks and fleas infesting Marmota himalayana from the Qinghai-Tibet plateau.

BACKGROUND: Ticks and fleas are important vectors and hosts for various emergent human pathogens, such as severe fever with thrombocytopenia syndrome virus, Crimean-Congo hemorrhagic fever virus, and tick-borne encephalitis virus. In the Qinghai-Tibet plateau of China, Marmota himalayana (marmots) serves as the primary reservoir for Yersinia pestis, primarily transmitted by fleas. Due to their wide distribution and large population, marmots serve as important host animals for ticks and fleas to survive and reproduce on the Qinghai-Tibet Plateau. However, the spectrum of RNA viruses found in ti

Rickettsia spp. in finnish ixodid ticks.

BACKGROUND: Rickettsia spp. are intracellular, arthropod-borne bacteria pathogenic to humans and animals. The clinical manifestations of rickettsial infections range from mild to severe, but diagnosis is sometimes missed owing to mild symptoms or empirical antibiotic treatment for suspected tick-borne diseases. This study aimed to determine the prevalence of Rickettsia spp. in ticks across Finland, explore possible regional variations, identify the infecting Rickettsia species, and assess rickettsial exposure of certain human patients. METHODS: We analysed 5101 ticks from 20 locations, collec

Construction of a CMV promoter-driven Langat virus cDNA clone and reporter viruses.

Langat virus (LGTV) is a tick-borne member of the Flaviviridae family and a biosafety level 2 surrogate for studying tick-borne encephalitis virus (TBEV) replication and pathogenesis. Here, we report the construction of a plasmid encoding a cytomegalovirus (CMV) promoter-driven LGTV cDNA that initiates infection following direct transfection of mammalian cells. Incorporation of three introns eliminated viral cDNA-associated toxicity in bacteria, enabling stable propagation of the full-length plasmid. Transfection of this construct resulted in high-level production of infectious LGTV, which exh

Integrated approach for sustainable management and control of Kyasanur forest disease in alignment with sustainable development goals.

Kyasanur forest disease virus (KFDV) is a highly pathogenic tick-borne flavivirus that is endemic to India, primarily affecting rural communities in forested regions. The virus is transmitted through the bite of infected Haemaphysalis spinigera ticks, leading to a severe febrile illness characterized by symptoms that can range from mild to life-threatening. In approximately 20 % of cases, patients exhibit biphasic symptoms, which can progress to serious complications, including encephalitis and hemorrhagic manifestations. This review aims to provide a comprehensive analysis of the clinical pre

Update on emerging and re-emerging vector-borne diseases.

Vector-borne diseases (VBDs) are spread by ticks and mosquitoes on every inhabitable continent and pose a persistent and escalating threat to global public health. The incidence of the vector-borne RNA viruses, which are the focus of this article, has increased in recent decades. This diverse group comprises multiple genera of viruses, ranging from dengue, the most common mosquito-borne virus in the world, to more obscure viruses like Oropouche virus, circulating in South America, and multiple orthonairoviruses recently identified in China. The transmission dynamics of these diseases are shape

Detection of Tick-borne Encephalitis Virus in Ixodes Ticks Collected Near Ulaanbaatar, Mongolia.

BACKGROUND: Tick-borne encephalitis virus (TBEV) is a flavivirus that causes a range of clinical presentations including severe neurological complications in humans. In Mongolia, TBEV is primarily transmitted by the Ixodes persulcatus tick, which serves as both a vector and reservoir. Historically, TBEV in Mongolian ticks has been predominantly described in the forested regions of the Bulgan and Selenge aimags (provinces). METHODS: In 2021 and 2024, I persulcatus ticks (n = 240) were collected from Bulgan and Tuv aimags in northern Mongolia, respectively, and pooled into 52 groups of 1 to 7 t

Multiplex serological screening of wild boar as sentinels of emerging zoonoses: HEV, WNV, and TBEV distribution in Saxony, Germany.

Targeted wildlife surveillance enables early detection of emerging infections and facilitates prevention of wildlife-human spillover events. The Eurasian wild boar (Sus scrofa) serves as a valuable indicator species for wildlife surveillance due to its broad European distribution and its ability to adapt to diverse habitats. Wild boar and domestic pigs are the main reservoir for hepatitis E virus (HEV) genotype 3, which can be transmitted to humans mainly through consumption of undercooked meat. Wild boar can also be infected by vector-borne pathogens like the mosquito-transmitted orthoflavivi

Genome-wide association study identifies ABCG1 as a susceptibility locus for tick-borne encephalitis.

Tick-borne encephalitis (TBE) is a viral infection of the central nervous system, caused by the tick-borne encephalitis virus (TBEV) presenting clinically as meningitis, meningoencephalitis, and meningoencephalomyelitis. To investigate genetic susceptibility to TBE, and its severe forms, we conducted a genome-wide association study in the European population comprising 1,600 TBE cases and 9,699 controls. We identified several suggestive (p < 1 × 10-5) intronic and exonic variants in ABCG1, the only gene significantly associated with TBE susceptibility. These variants were shown to influence AB

Assessment of human health risks to tick-borne infections in urban green spaces (UGS) - a study protocol.

BACKGROUND: As the world becomes increasingly urbanised, with projections of about two-thirds of a burgeoning population living in urban areas by 2050, several governments are continuously making efforts to create, maintain, and expand urban green spaces and enhance their ecological connectivity. However, the advantages of these green infrastructures are weighed down by their risk of higher human exposure to wildlife, zoonotic diseases, and wildlife-associated ectoparasites, leading to spillage of vector-borne illnesses. METHODS/DESIGN: The research study aims to examine the risk of acquiring

Evaluation of three virus RNA extraction platforms for the detection of human norovirus, hepatitis A virus, hepatitis E virus and tick-borne encephalitis virus in different foodstuffs.

In the investigation of foodborne outbreaks, molecular detection of foodborne viruses involves virus extraction, RNA purification and quantitative detection using RT-qPCR. The NucliSENS® easyMAG® platform, previously used for virus RNA extraction from various foods, has been discontinued and alternative methods are needed. Here, we compared eMAG® (fully automated) and eGENE-UP® (semi-manual) RNA extraction systems with the easyMAG® platform for detecting viruses from virus stocks and food matrices. Various foods (couscous vegetable mix, green beans, goat milk, bottled water and ham pâté) were
