Exploring the Role of Cerebrospinal Fluid and Serum Mid-Regional Pro-Adrenomedullin in Tick-Borne Encephalitis: A Pilot Study.

Background: Adrenomedullin (ADM) is a multifunctional peptide with vasoregulatory, antimicrobial, and anti-inflammatory properties. Its stable fragment, mid-regional pro-adrenomedullin (MR-proADM), is a validated biomarker in sepsis and systemic infections, but its role in viral neuroinfections remains unexplored. Tick-borne encephalitis (TBE), caused by the tick-borne encephalitis virus (TBEV), is a major viral infection of the central nervous system (CNS) associated with long-term neurological sequelae. This study aimed to assess MR-proADM levels in cerebrospinal fluid (CSF) and serum of pat

Neurological Manifestations and High Viral Load as Independent Predictors of Mortality in Severe Fever With Thrombocytopenia Syndrome.

BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS), an emerging tick-borne viral hemorrhagic fever, is characterized by high mortality rates. While neurological complications (eg, seizures, encephalitis) have been identified as adverse prognostic factors in severe cases, their association with viral replication, immune responses, and neuroinflammation remain poorly defined and urgently require systematic investigation. METHOD: A cohort of 277 patients with SFTS was included and stratified based on neurological symptoms. Clinical characteristics, laboratory results, and immune mark

Vector-Borne Infections in Romania: From Surveillance to Prediction.

Vector-borne infections are a growing public health concern in Romania, influenced by ecological diversity, climate change, and socioeconomic factors. West Nile virus, tick-borne encephalitis, and Lyme borreliosis represent the most significant threats, with additional risks posed by emerging pathogens, such as leishmaniasis, and the potential reintroduction of malaria. While surveillance systems exist for human cases and, to a lesser extent, vectors, these remain fragmented, underfunded, and limited in their integration across human, veterinary, and environmental health domains. By highlighti

Tick salivary cystatin Iristatin limits the virus replication in skin of tick-borne encephalitis virus-infected mice.

Tick-borne encephalitis virus (TBEV) is flavivirus transmitted to the host via tick saliva which contains various molecules with biological impacts. One of such molecules is Iristatin, a cysteine protease inhibitor from Ixodes ricinus that has been shown to have immunomodulatory properties. To characterize Iristatin in the relation to TBEV, we investigate whether this tick inhibitor has any capacity to influence TBEV infection. Mice were intradermally infected by TBEV with or without Iristatin and the viral multiplication was determined in skin and brain tissues by RT-PCR two and 5 days after

Tick-borne encephalitis vaccine uptake, effectiveness, and impact in Sweden from 2018 to 2022.

Tick-borne encephalitis (TBE) is an increasing health threat in Sweden and elsewhere in Europe. TBE vaccination is commonly recommended in Sweden, but limited data are available on uptake, effectiveness, and impact of TBE vaccination. General population surveys conducted in 2019-2022 were used to estimated TBE vaccine uptake. TBE vaccine effectiveness (VE) was estimated using the screening method utilizing the surveys and public health TBE surveillance data, which predominately includes hospitalized TBE cases, from 2018 to 2022. Impact of TBE vaccination was calculated based on disease inciden

Early transcriptomic changes at the skin interface during Powassan virus transmission by Ixodes scapularis ticks.

INTRODUCTION: Powassan virus (POWV), a vector-borne pathogen transmitted by Ixodes ticks in North America, is the causative agent of Powassan encephalitis. As obligate hematophagous organisms, ticks transmit pathogens like POWV at the tick bite site, specifically during the initial stages of feeding. Tick feeding and salivary factors modulate the host's immunological responses, facilitating blood feeding and pathogen transmission. However, the mechanisms of immunomodulation during POWV transmission remain inadequately understood. In this study, we investigated the global cutaneous transcriptom

Genotype-driven sensitivity of mice to tick-borne encephalitis virus correlates with differential host responses in peripheral macrophages and brain.

BACKGROUND: Tick-borne encephalitis (TBE) is the most common tick-borne viral infection in Eurasia. Outcomes range from asymptomatic infection to fatal encephalitis, with host genetics likely playing a role. BALB/c mice have intermediate susceptibility to TBE virus (TBEV) and STS mice are highly resistant, whereas the recombinant congenic strain CcS-11, which carries 12.5% of the STS genome on the BALB/c background, is more susceptible than BALB/c mice. In the present study, we employed these genetically distinct mouse models to investigate the host response to TBEV infection in both periphera

Dysregulation of the kynurenine pathway is related to persistent cognitive impairment in tick-borne encephalitis.

BACKGROUND: Tick-borne encephalitis (TBE) patients frequently suffer cognitive and neuropsychiatric sequelae, which may be severe and long-lasting. The underlying mechanisms remain poorly understood, but likely involve dysregulation of immunological and neuroinflammatory processes. Here, we investigated the activity of the kynurenine pathway (KP), an immunologically induced set of enzymes generating neuroactive metabolites via degradation of the amino acid tryptophan and their correlation with the cognitive performance in TBE patients. METHODS: KP metabolite concentrations were measured in se

Evolutionary Patterns and Genotype-Specific Amino Acid Mutations of Tick-Borne Encephalitis Virus.

Tick-borne encephalitis virus (TBEV) is a significant tick-borne flavivirus responsible for severe human diseases. Here, we analyzed the genetic diversity and evolutionary dynamics of TBEV using 263 genome sequences from the NCBI database and identified key amino acid mutations. TBEV sequences were classified into five genotypes-Baikalian, European, Far-Eastern, Himalaya, and Siberian-showing ORF nucleotide similarity of 81.5% to 88.0% and amino acid similarity of 93.0% to 96.4%. Extensive recombination between genotypes was not observed. Entropy analyses revealed highly variable sites distrib

Establishment of reverse genetics systems for Colorado tick fever virus.

The Colorado tick fever virus (CTFV), which has 12-segmented double-stranded RNA genomes, is a pathogenic arbovirus that causes severe diseases in humans. However, little progress has been made in the analysis of replication mechanisms and pathogenicity. This virological constraint is due to the absence of a reverse genetics system for CTFV; therefore, we aimed to establish the system. Initially, the efficacy of CTFV replication was investigated in various cell lines. CTFV was found to grow in many cell types derived from different hosts and organs. Subsequently, BHK-T7 cells stably expressing
