The southern region of Poland, rich in natural and tourist attractions, encourages outdoor recreation and spending time in nature. Ongoing climate change, including global warming, has contributed to an increased abundance of ticks and an extended period of their seasonal activity. Consequently, the number of tick-borne disease cases continues to rise, and their diagnosis and treatment are often challenging and prolonged. The aim of this study is to summarize current knowledge on the occurrence of ticks in tourist areas of southern Poland and to raise public health awareness among individuals

UNLABELLED: Powassan Viruses (POWV) are tick-borne Flaviviruses that cause lethal encephalitis and long-term neurologic sequelae. POWVs are divided into two lineages that share 86% genetic identity. POWV-LB (Lin I) was isolated from a fatal case of encephalitis, while POWV-LI9 (Lin II) is a tick-derived isolate that causes age-dependent lethality in mice. Domain III of Flavivirus envelope proteins (EDIII) directs cell attachment, and determines murine neuropathology and lethality. Here we developed POWV LB reverse genetics, analyzed the CNS of LB and recLB mutant infected mice, and assessed th

Deer tick virus (DTV), or lineage II Powassan virus, is an emergent tick-borne encephalitis virus in North America. Survivors frequently sustain neurologic sequelae. Nationally reported cases have been increasing. DTV is thought to be maintained in nature by multiple modes including horizontal transmission (from viremic host to tick), cofeeding transmission (between ticks feeding nearby) and by transovarial transmission (female to progeny). Analysis of the relative importance of each mode has been hindered by low enzootic transmission. In 2021, Martha's Vineyard, Massachusetts experienced an e

Tick-borne diseases (TBDs) have proliferated in Germany. The two most prevalent TBDs, Lyme-borreliosis and tick-borne encephalitis, can present with nonspecific symptoms and lead to serious neurological complications. To date, a review synthesising the risk factors of acquiring a TBD in Germany is missing. Such a review could mitigate infection risk and support early protection. A scoping review was conducted using the databases MEDLINE/PubMed and Web of Science. Studies on the risk of acquiring TBDs through tick bites in Germany, written in German or English, were included in this review. Two

Wild bird species contribute significantly to the rapid geographic dissemination of tick-borne encephalitis viruses (TBEV) and West Nile virus (WNV), facilitating the establishment of new natural foci of these orthoflaviviruses. However, the impact of TBEV and WNV population variability on shaping these foci, as well as the potential emergence of new human-pathogenic viral variants, remain underexplored. This study aimed to assess the genetic heterogeneity of TBEV (Siberian and Far Eastern genotypes) and WNV, isolated simultaneously from the tissues of a single garden reed warbler (Acrocephalu

OBJECTIVES: To examine the incidence, clinical presentation, and prognosis of infectious encephalitis among adults in Denmark.
METHODS: Prospective, population-based cohort study using the Danish Study group for Infections of the Brain database to identify all Danish residents ≥18 years of age hospitalized with infectious encephalitis from 2015 to 2023. Cases were defined in alignment with the International Encephalitis Consortium criteria. Multivariable modified Poisson regression was used to compute adjusted relative risks (adj. RRs) with 95% CIs for intensive care unit admission and 6-mont

UNLABELLED: Tick-borne orthoflaviviruses (TBOVs) are a growing global health concern. Several members of this viral group cause fatal disease in humans with increasing case numbers throughout the last few decades. The innate immune response, especially interferon (IFN)-dependent signaling, is an essential part of the vertebrate defense system that counteracts infection with TBOVs and other viruses. Although they activate the same signaling cascade, IFNs belonging to the type I and III families trigger differing gene expression patterns. Which genes the two IFN families induce to restrict infec

Kyasanur Forest Disease Virus (KFDV) is a zoonotic tick-borne flavivirus that causes Kyasanur Forest Disease (KFD), a disease primarily found in India. KFD historically been confined to Karnataka state of southern India, however, in recent years, it has spread beyond its original endemic region, raising significant public health concerns. There are currently no KFD specific treatments or vaccines available. Rapid development of effective vaccine against KFD is urgently needed. We developed and evaluated a whole virion inactivated KFD virus vaccine candidate, adjuvanted with aluminum hydroxide

TRIpartite Motif (TRIM) protein 5 alpha (TRIM5α) is a well characterized cellular inhibitor of lentivirus replication that limits transmission of related viruses between primates. We previously reported that TRIM5α derived from humans and rhesus macaques inhibits replication of orthoflaviviruses belonging to the tick-borne encephalitis virus (TBEV) serocomplex, including TBEV, Kyasanur forest disease virus and Langat virus (LGTV), but interestingly not the tick-borne Powassan virus (POWV). To further characterize the primate TRIM5α and orthoflavivirus interface, we screened TRIM5α variants fro

Tick-borne encephalitis virus (TBEV) poses an increasingly significant threat to public health. Building on our previous TBEV-host interactome study, we further refine the virus-host interaction network and show that multiple viral proteins, including prM, NS1, NS2B3, NS4A, and NS5, are associated with modulation of diverse host cellular pathways, including innate immune responses, autophagy, ribosomal biogenesis, homologous recombination, and cell cycle regulation. Notably, TBEV infection is associated with a modest but reproducible accumulation of host cells in the G0/G1 phase. Mechanistical

BACKGROUND: The greatest impact on the burden of CNS infections resulted from preventive measures, mainly vaccination programs, that have decreased the incidence of many CNS infections significantly. Here, we highlight the main cornerstones of vaccination programs on bacterial and viral CNS infections and point at future chances of upcoming vaccines.
MAIN BODY: Vaccination programs have significantly decreased the number of cases due to Haemophilus influenzae type B and Neisseria meningitidis. For pneumococcal meningitis, new vaccines that prevent neuroinvasive serotypes have the potential to

With no approved antiviral treatments currently available, Kyasanur Forest Disease Virus (KFDV) remains a serious and neglected tick-borne flavivirus posing a significant public health concern endemic to India. To address this therapeutic gap, we employed AlphaFold-based structural modelling integrated with structure-based lead identification targeting the RNA-dependent RNA polymerase (RdRp) domain of the viral NS5 protein. The predicted NS5 model exhibited high structural confidence, with pLDDT scores approaching 90 and a pTM score of 0.8, and was further validated using Verify3D, PROCHECK, a

Ticks transmit numerous viruses that pose significant threats to human and animal health, however, the molecular interactions between ticks and viruses remain poorly understood. Using Langat virus (LGTV)-a surrogate for tick-borne encephalitis virus (TBEV), we show that the JAK/STAT pathway promotes viral infection in Haemaphysalis longicornis. Rather than directly interacting with viral proteins, this proviral effect is mediated by a low-density lipoprotein receptor-related protein (LRP), whose expression is regulated by STAT. Silencing LRP in H. longicornis reduced LGTV infection, while ecto

A 66-year-old man with a history of hypertension presented with left-sided jaw jerking, repetitive movements, aphasia, and dysarthria. He was afebrile and in no acute distress, with mild hypertension (153/81 mm Hg). Routine laboratory studies were unremarkable. Brain MRI showed multiple nonenhancing supratentorial cortical and subcortical hyperintensities on T2-weighted fluid-attenuated inversion recovery (FLAIR) images, which were nonspecific for infectious, inflammatory, vascular, or neoplastic etiologies. CT of the chest, abdomen, and pelvis and MRI of the cervical and thoracic spine were u

Ixodid ticks are vectors for a plethora of pathogens, including the Crimean-Congo hemorrhagic fever virus (CCHFV), which causes severe disease in humans. Two autochthonous CCHF human cases were reported in 2025 in Greece. The aim of the present study was to gain a better insight into the geographic distribution and prevalence of CCHFV and the related Aigai virus (AIGV) in ticks in Greece. Therefore, 680 ticks (135 Hyalomma and 545 Rhipicephalus ticks) collected during 2024 from livestock (sheep, goats, cattle) and from the environment were tested for CCHFV and AIGV. AIGV was detected in 12 adu

The detailed mechanism and sequence by which tick-borne flaviviruses (TBFVs), such as Langat virus (LGTV), infect and disseminate in arthropod hosts remain undefined. To begin characterizing these processes, we used artificial membrane feeding chambers to feed adult Ixodes scapularis ticks with blood containing LGTV. At 24, 48, 72, and 96 hours (h) after attachment, we removed and dissected the partially fed ticks to obtain the midgut and salivary glands. Histology confirmed infection in cells of the digestive epithelium lineage; infection was noted in midgut generative cells and the more diff

Orthoflaviviruses are etiological agents of neurological diseases in equines. Horses are susceptible to equine Tick-borne encephalitis virus (TBEV), Usutu virus (USUV), and West Nile virus (WNV), although mostly WNV has been associated with clinical disease. The aim of the present study was to estimate the prevalence of TBEV, USUV and WNV in 289 healthy horses from nine provinces of Algeria. This study was conducted between March 2021 and December 2023. Sera that tested positive, doubtful, or negative close to the doubtful threshold in the Orthoflavivirus-ELISA were further tested by the virus
