Clinical Characteristics and Neutralizing Antibody Levels in a Japanese Patient with Tick-borne Encephalitis: A Case Report.

Tick-borne encephalitis (TBE) is a severe neurological disease caused by infection with the TBE virus (TBEV), primarily transmitted to humans through tick bites. As there is no effective treatment, prevention through vaccination and avoidance of tick bites is essential. In Japan, only five cases had been reported up to 2023, all from Hokkaido, the northernmost main island. Here, we report the sixth case of TBE in Japan. The patient was a 58-year-old male, in whom magnetic resonance imaging revealed enhancing lesions in the brain and spinal cord, accompanied by severe neurological symptoms. Des

Powassan Virus Seroprevalence in a U.S. Servicemember Population at High Risk for Tick Exposure.

Powassan virus (POWV) is an emerging tick-borne flavivirus that can cause severe encephalitis in humans. Currently no vaccines or therapeutics are approved to treat POWV. POWV is spread by the deer tick, Ixodes scapularis, which is ubiquitous across the Northeastern United States. To better understand POWV prevalence in high-risk populations, we examined POWV seroprevalence in Cadets at United States Military Academy (USMA) in West Point, New York. Cadets at USMA, located in a heavily wooded area, are at high risk for tick exposure during outdoor military training. 1,051 serum samples from the

Immunopharmacological and structural design of a polyprotein based multi epitope vaccine against Powassan virus.

Powassan virus (POWV) poses a growing neurotropic threat with limited immunopharmacological interventions, requiring the development of targeted vaccine strategies. The current study focuses on developing a specific vaccine using bioinformatics tools to target T- and B-cell epitopes from a genome polyprotein sequence of POWV. Four vaccine constructs with possible immunogenic qualities were assembled. Two specific epitopes were selected from B-cell, and T-cell for each construct. The heat-labile enterotoxin S63 adjuvant was attached to the N-terminal, while the PADRE order and the His-tag were

Conserved histidine residues and the control of tick-borne encephalitis virus maturation.

The maturation and infectivity of orthoflaviviruses is driven by interactions and structural changes of the two envelope proteins, prM and E, which are controlled by protonation and deprotonation events during the viral life cycle when the virus encounters acidic and neutral pH environments inside and outside the cell. Histidine residues act as sensors for these pH-induced conformational transitions, which differ between mosquito-borne and tick-borne orthoflaviviruses in some aspects. We identified six histidine residues that are conserved among the envelope proteins of mammalian tick-borne or

Kyasanur Forest Disease: Advances in Experimental Animal Models, Immune Assays, and Challenges in Vaccine Development.

Kyasanur Forest Disease (KFD) is a tick-borne viral haemorrhagic fever endemic to forested regions of South India, with case fatality rates ranging from 3% to 10% and seasonal outbreaks causing recurrent illness among at-risk populations such as forest workers and villagers. The historically used formalin-inactivated KFD vaccine showed limited effectiveness in field studies, providing only about 62% protection. This moderate level of protection highlights the need for improved vaccines that offer higher efficacy and longer-lasting immunity for populations living in endemic areas. A major chall

No-known-vector flaviviruses exhibit diverse replication and virulence phenotypes in mice.

UNLABELLED: The Orthoflavivirus genus (flaviviruses) includes globally significant arboviruses, which cycle between arthropod vectors (mosquitoes and ticks) and vertebrate hosts. In contrast, no-known-vector flaviviruses (NKVFVs) have been isolated from rodents and bats, but not arthropods, so are thought to spread by vector-independent routes. However, little is known about the host range and pathogenic mechanisms of these viruses. To evaluate NKVFV pathogenesis, we infected wild-type, Ifnar1 -/- , and Ifnar1 -/- Ifngr1 -/- mice by footpad inoculation with 12 NKVFVs: Entebbe bat virus (ENTV),

Development and evaluation of single-plex TaqMan real-time quantitative PCR assays for the detection of six tick-borne pathogenic viruses in northeastern China.

BACKGROUND: Northeastern China is a recognized hotspot for tick-borne viral diseases. However, there remains a lack of methods capable of simultaneously detecting these emerging and re-emerging tick-borne viruses. METHODS: Primer and probe sets were designed using Beacon Designer 8.0, targeting conserved regions of viral genomes from Alongshan virus (ALSV), Tick-borne encephalitis virus (TBEV), Severe fever with thrombocytopenia syndrome virus (SFTSV), Beiji nairovirus (BJNV), Yezo virus (YEZV), and Songling virus (SGLV). Recombinant plasmids were constructed to assess assay sensitivity, whil

Roles of ticks in the occurrence of emerging and re-emerging vector-borne viruses of zoonotic importance in West Africa.

Ticks are arthropod vectors that transmit a wide range of emerging and re-emerging vector-borne diseases (VBDs), many of which are of considerable zoonotic importance. Recently, tick-borne diseases (TBDs), particularly those caused by arboviruses, have emerged as a significant public health concern in West Africa. The complex socio-ecological landscape in West Africa contributes to the circulation of several tick-borne arboviruses (TBAs), including Crimean-Congo hemorrhagic fever virus (CCHFV) and Dugbe virus (DUGV), both members of the genus Orthonairovirus, as well as other tick-borne flaviv

Serious tick-borne infections - considering the strengths and weaknesses of currently used laboratory diagnostic methods.

The laboratory diagnosis of tick-borne infections is a major interdisciplinary issue, closely linked not only to advances in molecular biological methods but also to changes in ecosystems and biodiversity caused by climatic and anthropogenic factors. These factors significantly influence the epidemiological situation both globally and in the Czech Republic, where a marked increase in the incidence of serious tick-borne infections has been observed in recent years. Current diagnostic approaches combine indirect serological methods (e.g., ELISA, Western blot, and immunofluorescence assays) with

Structures of asymmetric particles of tick-borne encephalitis virus provide insight into flavivirus assembly and maturation.

Flaviviruses are globally distributed human pathogens. However, the mechanisms underlying flavivirus assembly and maturation remain poorly understood. Here, we show that many particles of tick-borne encephalitis virus (TBEV) are asymmetric and lack subsets of surface heterodimers. Immature particles of TBEV contain incomplete spikes, providing evidence that their coats assemble directly from heterodimers of premembrane (prM) and envelope (E) proteins. Exposure of TBEV particles to acidic pH in the Golgi complex promotes maturation. The spikes and herringbone regions in TBEV maturation intermed

Restraint of Powassan virus replication by TRIM5α facilitates viral avoidance of antiviral immunity.

TRIpartite Motif (TRIM) protein 5 alpha (TRIM5α) is a well-characterized cellular inhibitor of lentivirus replication that limits transmission of related viruses between primates. We previously reported that TRIM5α derived from humans and rhesus macaques inhibits replication of orthoflaviviruses belonging to the tick-borne encephalitis virus (TBEV) serocomplex, including TBEV, Kyasanur forest disease virus and Langat virus (LGTV), but interestingly not the tick-borne Powassan virus (POWV). To further characterize the primate TRIM5α and orthoflavivirus interface, we screened TRIM5α variants fro

First evidence of exposure to West Nile Virus in horses within Denmark, 2025.

A nationwide serological screening of 822 horses was conducted in Autumn 2025 to investigate potential exposure to West Nile Virus (WNV) within Denmark. Serum samples were tested for antibodies against WNV, tick-borne encephalitis virus (TBEV) and Usutu virus (USUV). Four horses without any history of travel outside of Denmark, or previous vaccination against WNV, tested positive for WNV neutralising antibodies. The results support local exposure to WNV indicating that virus infected mosquitoes have been present in Denmark and underline the usefulness of horses as sentinels for the risk of hum

Diel activity of adult taiga ticks Ixodes persulcatus in the north-western part of its range (Karelia, Russia).

The taiga tick Ixodes persulcatus (Schulze, 1930) (Acari, Ixodidae), widespread in the northern Palearctic, is the vector of dangerous human pathogens including tick-borne encephalitis virus. Knowledge of its ecology is important for minimising tick bite exposure risk. We studied the diel activity of male and female I. persulcatus in the north of the range (Karelia, Russian Federation) and tested its relationship with environmental factors including temperature, relative humidity and light intensity. The field experiment was conducted in May-June 2024. Cages with unfed adult ticks (males and f

Tick-borne pathogens in a nature reserve area in the Netherlands: serological and molecular assessments in large herbivores and Ixodes ricinus ticks.

Ticks are vectors of a wide range of pathogens, relevant to both veterinary and public health. Although tick surveillance is frequently conducted in the Netherlands, data on tick-borne pathogens in free-living herbivores remains limited, leaving the impact of these pathogens on animal health poorly understood. This study aimed to characterise tick species and determine seroprevalence and infection prevalence of selected tick-borne pathogens in large herbivores at the Oostvaardersplassen nature reserve and questing ticks collected in the area. Between September 2023 and December 2024, blood was

Exploring prediction of tick and tick-borne encephalitis cases in Sweden using citizen science data.

Tick-borne encephalitis (TBE) remains a severe public health threat in affected areas with shifting geographic distribution linked to environmental change. However, systematic tick surveillance is limited due to the cost and effort of field monitoring. Here, we evaluate the potential of using national citizen science tick data reports for predicting tick-human interaction and TBE risk alongside socioeconomic and environmental data. We integrate citizen science observations with socioeconomic and environmental data, and apply statistical and machine learning models to identify key drivers and a

Microorganisms in Ixodes ricinus ticks in southern and southwestern Norway.

Tick-borne diseases constitute a major group of emerging vector-borne infections and represent a growing public health concern in many European countries. In Europe, the most clinically significant agents transmitted by Ixodes ricinus are tick-borne encephalitis virus (TBEV) and Borrelia burgdorferi sensu lato, the causative agents of tick-borne encephalitis and Lyme borreliosis, respectively. Ixodes ricinus also serves as a vector for other pathogenic microorganisms, including louping-ill virus (LIV) and Anaplasma phagocytophilum. Various endosymbionts, such as Wolbachia pipientis and Midichl

Climate Change and Autochthonous Vector-Borne Disease Transmission in Europe: Dengue as a Sentinel Signal for Surveillance and Preparedness.

Climate change is reshaping the epidemiology of vector-borne diseases in Europe by altering the ecological conditions that determine vector survival, seasonal activity and pathogen transmission. Rising temperatures, milder winters, prolonged warm seasons and changing precipitation patterns are increasing the suitability of parts of Europe for competent mosquito, tick and sandfly vectors. These changes, combined with human mobility and land-use change, increase the probability that imported pathogens encounter permissive conditions for local transmission. This Opinion article examines autochtho
