Cellular Responses to Flavivirus Infections: Stress Signaling at the Crossroads of Host Defense and Virus Infection.

Flaviviruses, encompassing notable pathogens, like Dengue, Zika, West Nile, and tick-borne encephalitis viruses, elicit complex cellular stress responses, involving pathways such as the unfolded protein response (UPR), integrated stress response (ISR), apoptosis, autophagy, and the antiviral immune response. These pathways regulate cell fate by either promoting survival to counteract virus-induced damage or triggering cell death programs under prolonged and irreparable stress. Therefore, the primary aim of flavivirus-induced cellular responses is to protect cells and hinder viral propagation.

Fluorescent Powassan Reporter Viruses Infect Neuron, Astrocyte and Microglial Cell Lines Independent of Attenuating D308N Envelope Protein Modification.

The Powassan virus (POWV) is a neurovirulent tick-borne virus that causes age-associated lethality and long-term neurologic sequelae in 50% of survivors. In aged mice the POWV strain LI9 mirrors human lethality and neuropathology; however, an avirulent POWV mutant, LI9-D308N, fails to enter the CNS or cause lethal disease. The D308N mutation is present in an envelope protein domain associated with cell attachment, yet the role of D308N mutations in cell tropism and neuroinvasion remains to be resolved. Here, we engineered fluorescent mScarlet3 and mNeonGreen reporter genes into WT LI9, and avi

Serological Evidence of Tick-Borne Encephalitis Virus (TBEV) Exposure Among Hospitalized Neurological Patients in Israel, 2010-2019.

BACKGROUND: Tick-borne encephalitis virus (TBEV) is an important cause of neuroinvasive infection in Europe and Asia. Israel is considered nonendemic, yet travel-associated cases have been reported. METHODS: Sera from hospitalized neurological patients in Israel (2010-2019) were screened for TBEV IgG antibodies. Positive/borderline results were confirmed by virus neutralization testing (VNT) against both TBEV and West Nile Virus (WNV). Confirmed TBEV-neutralizing sera were tested for anti-TBEV IgG and IgM by immunofluorescence assay. RESULTS: Of 12,651 suspected meningitis/encephalitis cases

A newly identified three-domain C-type lectin associated with blood feeding in the tick Ixodes ricinus.

Ixodes ricinus ticks are widely distributed throughout Europe and represent major vectors of tick-borne encephalitis virus and the Lyme borreliosis agent Borrelia burgdorferi sensu lato. In invertebrates, C-type lectins are commonly associated with innate immune functions, and several such lectins have been predicted in I. ricinus. Given the limited knowledge of lectin function in ticks, we characterized three carbohydrate-recognition domains (CRDs) of a novel C-type lectin identified in the I. ricinus transcriptome (IrCLec). The tertiary structures of CRD1, CRD2, and CRD3, predicted using the

A narrative review of climate- and environment-dependent dynamics of vector-borne diseases in the Nordic region: evidence from Finland.

INTRODUCTION: Vector-borne diseases (VBDs) pose a growing threat to human and animal health, driven by complex interactions among and between vectors, hosts, pathogens, and the environment. The Nordic region is warming at twice the global average rate, creating novel ecological conditions that reshape VBD transmission dynamics. OBJECTIVES AND METHODS: This narrative review synthesises current knowledge regarding major tick- and mosquito-borne diseases in the Nordic region, with a focus on Finland, based primarily on outputs from the VECLIMIT project (2020-2023), supported by other Nordic stud

A review of ticks and tick-borne diseases in Southeast Asia: identifying priorities, hurdles, and opportunities for health and knowledge gain.

Ticks and tick-borne diseases (TBDs) in Southeast Asia are poorly understood, confounding surveillance, and mitigation opportunities. Herein, we conducted a survey of experts (n = 25) to identify tick species and TBDs of greatest concern to Southeast Asia, 'drivers' which may alter their impacts, and research hurdles. These experts identified Dermacentor auratus, Haemaphysalis longicornis, Rhipicephalus linnaei, Amblyomma testudinarium sensu lato, Ixodes granulatus, Ixodes persulcatus, and Rhipicephalus microplus as ticks of greatest concern; and rickettsioses, severe fever with thrombocytopen

Field-isolate recombinant tick-borne encephalitis viruses define reporter-stability guidelines for antiviral testing in flaviviruses.

As arthropod-borne viruses continue to threaten populations globally, there is a pressing need for experimental systems that enable rapid antiviral discovery. Reverse-genetics platforms producing recombinant reporter orthoflaviviruses have been developed to address this gap. Here, we present two new recombinant tick-borne encephalitis viruses (TBEVrec) generated on a European-subtype Haselmühl Tiho1 isolate backbone. A reporter gene, either eGFP or Nluc, was inserted in the capsid-coding region of the genome downstream of the capsid RNA regulatory signal and separated from the complete viral p

Under-ascertainment of medically attended tick-borne encephalitis virus infections by tick-borne encephalitis surveillance in selected areas of Austria, Denmark, Finland, Germany, Latvia, Sweden, and Switzerland.

OBJECTIVES: Antibodies against tick-borne encephalitis (TBE) virus (TBEV) nonstructural protein 1 (NS1) immunoglobulin G are elevated in TBEV-infected individuals, but not in TBE vaccinated individuals, enabling estimation of TBEV infection prevalence in countries where TBE vaccines are used. METHODS: We used data from NS1 seroprevalence studies to estimate the incidence of medically attended TBEV infections and then compared this incidence with the incidence of surveillance-reported TBE cases to estimate the under-ascertainment multiplier for medically attended TBEV infections identified by

Tick-Borne Encephalitis in the 21st Century: Epidemiology, Clinical Challenges, and Prevention Strategies.

Tick-borne encephalitis (TBE) is an emerging viral infection in Europe and Asia. Climate and environmental changes are expanding the distribution and activity of Ixodes ticks, increasing human exposure. Most infections are asymptomatic, but symptomatic cases may present with a biphasic illness that can progress to meningitis or encephalitis, sometimes leading to long-term neurological sequelae, particularly in the elderly population. Currently, treatment is limited to supportive care, as no specific antiviral therapy is available. Effective vaccines exist and represent the main preventive stra

Internal poly(A) replacement strategy within 3' UTR for live-attenuated tick-borne encephalitis virus.

Tick-borne encephalitis virus (TBEV), a prominent orthoflavivirus transmitted by ticks, poses a risk of potentially fatal encephalitis in humans. Currently, the only available inactivated vaccines have limited usage, underscoring the need for developing novel vaccine strategies. The 3' untranslated region (UTR) of TBEV can be divided into variable and conserved regions, where structured RNA elements play crucial roles in viral replication and virulence. In this study, we found that deleting sequences in both the variable region and most of the conserved region within the 3'UTR eliminated viral

Rocky Mountain Spotted Fever Presenting as an Ischemic Stroke: A Rare Manifestation.

Rocky Mountain spotted fever (RMSF) is among the most severe tick-borne rickettsial infections encountered in the United States. Its presentation ranges from the classic triad of fever, headache, and rash to central nervous system (CNS) conditions, including meningitis, encephalitis, and seizures. Notably, RMSF presenting as an ischemic stroke due to CNS vasculitis is rare, which can prove fatal if recognition is delayed. We present the case of a woman in her 50s with a history of well-controlled diabetes mellitus type 2 and hypertension, who presented with fever, chills, body aches, and rash.

First report on the successful in vitro feeding of adult Ixodeshexagonus.

Artificial feeding systems have become essential tools for studying tick physiology, pathogen acquisition, and transmission without relying on vertebrate hosts. While in vitro feeding has been successfully established for several ixodid species, including Ixodes ricinus, Dermacentor spp., and Rhipicephalus spp., no system has yet supported complete engorgement and reproduction of Ixodes hexagonus, a widespread European tick and vector of Borrelia spp. and the tick-borne encephalitis virus. In this study, we report the first successful in vitro feeding of adult I. hexagonus using a silicone-mem

Prevalence of antibodies against Tick-Borne Encephalitis Virus in blood donors in the states of Saxony and Berlin/Brandenburg, eastern Germany.

OBJECTIVES: Tick-borne encephalitis virus (TBEV) is a major vector-borne pathogen in Europe, but its true infection burden remains unclear due to the high proportion of asymptomatic infections and the inability of conventional serology to distinguish vaccination- from infection-induced antibodies. We applied a non-structural protein 1 (NS1)-based serological approach to estimate TBEV seroprevalence, incidence, and manifestation index in eastern Germany (Saxony and Berlin/Brandenburg). METHODS: A total of 1,391 anonymized blood donor sera (18-72 years), collected between May and September 2021

Powassan Virus in the United States: An AI-Integrative One Health Scoping Review of Ecological Drivers, Zoonotic Interactions, Symptom Profiles, and Public Health Implications.

BACKGROUND: Powassan virus (POWV) is the only tick-borne encephalitis-group flavivirus endemic to the United States (U.S.) and is an increasingly serious public health threat. Reported U.S. cases remain relatively few but have grown steadily-from fewer than two annually before 2005 to a record 60 in 2024-and neuroinvasive disease carries a 10-15% case fatality with lasting neurological sequelae in roughly half of the survivors. Examining POWV through a One Health lens, we conducted a scoping review of its ecological and zoonotic drivers, its interactions with co-circulating tick-borne pathogen

Virome Diversity and Zoonotic Risks of Haemaphysalis concinna Ticks in Northeastern China.

Haemaphysalis concinna is a prevalent tick vector in Northeastern China, exhibiting a broad host range and the capacity to transmit over 40 human pathogens. Although numerous virome studies have investigated this tick species in Northeastern China, individual-level viral prevalence, coinfection dynamics, and the host spectra of the harbored pathogenic viruses remain poorly characterized. In this study, we conducted metatranscriptomic sequencing combined with TaqMan-based real-time quantitative PCR (RT-qPCR) validation on individual ticks to systematically profile the RNA virome of 112 adult H.

How Does Inter-Epitope Spacer Variation Within Artificial Immunogens Based on T-Cell Epitopes of Tick-Borne Encephalitis Virus Affect Immunogenicity?

Background/Objectives: In recent years, researchers have directed considerable attention toward the activation of the T-cell response in the development of vaccines against viral infections, including the tick-borne encephalitis virus (TBEV). A particularly promising approach to developing effective and safe T-cell vaccines involves the use of artificial multi-epitope immunogens. The selection of spacers that link epitopes within a construct can have a significant effect on the immunogenicity of multi-epitope constructs. The objective of this study was to design multi-epitope TBEV immunogens u

Milk Modulates the Gastrointestinal Stability of Tick-Borne Encephalitis Virus: Implications for Alimentary Transmission.

Tick-borne encephalitis virus (TBEV) can be transmitted alimentarily through contaminated dairy products, yet the mechanisms by which the virus survives the digestive tract remain poorly understood. In this study, we investigated the stability of TBEV in milk under simulated gastrointestinal conditions. While milk is known to preserve viral infectivity at low temperatures, our results demonstrate that in the gastric environment and at physiological temperature, it exerts a destabilizing effect, significantly reducing TBEV viability. All major milk fractions-whey, casein, and lipids-contribute

Comparative transcriptomic insights into ticks infected by alongshan and tick-borne encephalitis viruses.

Ticks are hematophagous ectoparasites and key vectors of a wide range of pathogens, including viruses, bacteria, and protozoa. Climate change and environmental shifts have contributed to the expansion of tick populations and the pathogens they harbor. Ticks rely solely on their innate immune system to combat infections, utilizing mechanisms such as RNA interference, phagocytosis, and antimicrobial peptide expression. However, pathogens have evolved strategies to evade or manipulate these defenses to facilitate their replication and transmission. This study compares the infection dynamics of tw
