Tick-borne encephalitis vaccination coverage among outdoor workers in Sweden: a nationwide survey.

Tick-borne encephalitis (TBE) is an emerging public health concern in Sweden, with a steadily increasing incidence over the past two decades. Climate change, expanding tick populations, and increased human exposure contribute to the growing burden of disease. Although TBE vaccination is highly effective, it is not included in Sweden's national immunization program, and data on vaccination coverage among high-risk occupational groups remain unknown. We conducted a nationwide cross-sectional Knowledge, Attitude, and Practice (KAP) survey in 2024 to assess TBE vaccination coverage among major out

Climate drivers of tick-borne encephalitis in the Veneto Region, Italy, 2007-2024.

Tick-borne encephalitis (TBE) is an arboviral disease in Europe, with increasing incidence in northeastern Italy. Transmission of the virus via Ixodes ticks is influenced by seasonal dynamics and environmental factors. This study aimed to describe TBE epidemiology in the Veneto Region, assess spatiotemporal patterns, and examine associations with environmental determinants, including possible impacts of climate change. We conducted a population-based study using hospital discharge records to identify TBE cases from 2007 to 2024. Temporal trends and spatial patterns of age-standardized incidenc

Co-release of tick-borne encephalitis virus RNA and structural proteins via virus-like particles and extracellular vesicles.

Tick-borne encephalitis virus (TBEV) is a medically important flavivirus that causes severe neurological diseases in humans. The assembly of flaviviruses is initiated by the interaction between capsid (C) proteins and viral genomic RNA, yet the molecular determinants that govern RNA encapsidation remain unclear. In this study, we established a TBEV virus-like particle (VLP) system to analyse viral factors that influence viral RNA incorporation independently of productive infection. Using a reporter-containing TBEV minigenome, we investigated the contribution of UTRs to the incorporation of RNA

Protective human antibodies against Powassan virus.

UNLABELLED: Powassan virus (POWV) is a tick-borne orthoflavivirus that can cause severe neuroinvasive disease. There are no approved vaccines or therapeutics, and the incidence of POWV infection in humans is rising. Here, we isolated and characterized a panel of human monoclonal antibodies (mAbs) from a convalescent donor using single B-cell sorting with POWV EDIII and full-length E ectodomain as antigen bait. Binding and neutralization assays with both POWV reporter virus particles and an authentic strain identified multiple neutralizing mAbs that target distinct epitopes across the E glycopr

Evidence of Heartland and Bourbon Viruses Transmitted by Ticks in Suffolk County, New York.

A cohort of 107 subjects (2019-2024) between the ages of 20 and 93 years old with an acute febrile illness after a recent tick bite in Suffolk County, New York, was assessed for infection with Powassan, Heartland, and Bourbon viruses. The available blood samples collected from these subjects were sent to the New York State Department of Health for screening using a plaque-reduction neutralization test. Of the 107 subjects analyzed, two had neutralizing antibodies to Bourbon virus, and one had neutralizing antibodies to Heartland virus. A fourfold rise in neutralizing antibody titer in two of t

Seasonality and environmental drivers of tick-borne encephalitis virus prevalence in Ixodes ricinus ticks from Southern Norway.

The tick-borne encephalitis virus (TBEV) is the causative agent of neurological disease in humans, and it is mainly transmitted by the tick Ixodes ricinus in western Europe. In Norway, previous surveillance studies have detected the virus along the coast from Nordland County in the north to the Swedish border in the southeast. However, all human cases of tick-borne encephalitis are restricted to the southernmost part of the country, presenting a significant increase in the incidence in the last decades. The incidence and prevalence of TBEV is consistently linked to climatic factors. We evaluat

Targeted monitoring informed by mapping the ongoing spread of tick-borne encephalitis virus, the Netherlands.

BACKGROUNDTick-borne encephalitis virus (TBEV) was detected in the Netherlands in 2015. Current monitoring through serology in deer, tick collection and reported human cases, may underestimate areas of virus circulation. Including estimates of suitability of the habitat for TBEV can guide risk-based monitoring.AIMWe aimed to improve the accuracy of the TBEV distribution map by adding estimates of TBEV habitat suitability to guide targeted tick monitoring in newly identified risk regions.METHODSHabitat suitability for TBEV was assessed by calculating the basic reproduction number (R0) for 1 km2

Bemnifosbuvir and remdesivir inhibit tick-borne encephalitis virus infection in complementary in vitro and ex vivo disease models.

The geographical distribution and incidence of tick-borne encephalitis (TBE) have steeply increased over the past decades, raising to represent a major health concern in Asia and Europe. Symptoms of TBE, caused by infections with tick-borne encephalitis virus (TBEV), range from mild flu-like symptoms to severe neurological disease, often accompanied by long-term sequelae persisting for several years following pathogen encounter. While effective vaccines against TBEV are available, no antiviral drugs are currently approved and therapeutic options for patients suffering from TBE are limited to s

Tick-Borne Encephalitis Vaccine Effectiveness and Impact in Slovenia, 2019-2024.

Slovenia has a high tick-borne encephalitis (TBE) incidence and TBE vaccination is recommended for all residents ≥1 year-of-age. Despite widespread TBE vaccine use in Slovenia, TBE vaccine effectiveness (VE) has not been reported for Slovenia. We defined individuals who received ≥3 TBE vaccine doses in accordance with the recommended vaccination schedule as fully vaccinated. Using the screening method, TBE VE was determined by comparing the proportions of TBE cases and survey respondents who were fully vaccinated. TBE incidence and VE were used to estimate the number of TBE cases prevented by

Current Knowledge on Tick-Borne Encephalitis Virus Interaction with Ticks: Acquisition, Dissemination, and Persistence.

Tick-borne encephalitis virus (TBEV) is a major arthropod-borne flavivirus responsible for severe neurological disease in humans across Europe and Asia. It is maintained in nature through complex interactions within ticks and between tick vectors, vertebrate hosts and environmental factors. This review summarizes current knowledge on TBEV-tick interactions, focusing on virus acquisition, dissemination, vector competence, and long-term persistence within tick vectors. TBEV is acquired by ticks during blood feeding on viremic hosts or through co-feeding transmission under experimental conditions

A Household Cluster of Tick-Borne Encephalitis in Belgium in 2025: Is the Epidemiology Evolving?

Despite serological evidence of tick-borne encephalitis virus (TBEV) circulation in Belgian animals since 2007, confirmed autochthonous human infection was only first documented in 2020. We review the current national epidemiologic situation and investigate a household cluster of confirmed autochthonous cases identified in 2025. A cohabiting couple experienced a near-simultaneous onset of meningoencephalitis and tested positive for TBEV-specific IgM and IgG, with confirmation by PRNT90. One patient reported a recent tick bite, and both patients reported consumption of unpasteurized milk and go

Fatal European subtype tick-borne encephalitis in a fully vaccinated immunocompetent child: a case report with viral sequencing.

BACKGROUND: Tick-borne encephalitis (TBE) is considered to have a more favourable outcome in children compared to adults, and death from TBE in children is infrequent. Severe cases are primarily seen in immunocompromised or unvaccinated individuals. We report a case of TBE with a fatal outcome in a previously healthy child. The case is unique in several aspects: an atypical clinical presentation without fever or biphasic illness, and a fulminant course despite full vaccination and absence of known predisposing risk factors for severe disease. In addition, sequencing revealed the TBE virus (TBE

RNF138-Mediated Ubiquitination and Degradation of NS5 Restricts Tick-Borne Encephalitis Virus Infection.

Tick-borne encephalitis virus (TBEV) causes severe neurological disease. However, whether host restricts its replication and the underlying mechanisms remain incompletely delineated. Here, we identify the E3 ubiquitin ligase RNF138 as an intrinsic restriction factor targeting the viral RNA-dependent RNA polymerase (RdRp) NS5. Mechanistically, RNF138 directly interacts with the RdRp domain of NS5 through its ubiquitin-interacting motif, catalyzes K48-linked polyubiquitination and induces degradation of TBEV NS5. Ubiquitin-remnant profiling and mutational analyses identify K372, K462, and K470 w

Molecular Mimicry by the Tick-Borne Encephalitis Virus E Protein: A Hidden Link to Autoimmunity.

In this study, we combined computational predictions with experimental validation as a hybrid strategy to explore whether the E protein of tick-borne encephalitis virus (TBEV) possesses autoimmune potential. Using in silico homology searches, we identified two viral epitopes (evglekl and vtgtqgt) within the TBEV E protein that share sequence identity with fragments of the human proteins DNAH7 and CSMD2. Antibodies against these epitopes were detected in the plasma of a subset of patients after natural TBEV infection. Notably, no such antibodies were found in recipients of the Tick-E-Vac vaccin

Economic burden estimation to inform prevention policies against emerging zoonotic diseases: The case of tick-borne encephalitis in France.

OBJECTIVES: Economic evidence on health losses can guide prioritisation of prevention policies for emerging zoonotic risks. Tick-borne encephalitis (TBE) is gaining attention in Western Europe, and notifications are rising in France. Our goal is to scope existing European burden estimates and to generate the first national figures for France using a standardized framework that can inform health policies. STUDY DESIGN: This study was designed as a nationwide model-based cost-of-illness analysis combining a systematic literature review with a decision-tree simulation to estimate the costs of TB

Tissue tropism and mRNA expression profiles of selected innate immunity-related genes following experimental tick-borne encephalitis virus and louping ill virus infection of sheep.

UNLABELLED: Tick-borne encephalitis virus (TBEV) and louping ill virus (LIV) are genetically and antigenically closely related tick-borne flaviviruses responsible for different disease outcomes in sheep. In order to determine whether key components of the innate immunity could contribute to the observed difference in virus dissemination and disease outcome, 8-month-old sheep were inoculated intradermally with TBEV Neudoerfl strain or LIV LI/31 strain. Only LIV-infected sheep developed fever and clinical signs. No TBEV RNA was detected in serum and examined tissues, except for low amounts in th

Circulating miRNAs as Biomarkers of Tick-Borne Encephalitis Severity: Association with Cytokine Profile in Febrile, Meningeal, and Encephalitic Forms.

Tick-borne encephalitis (TBE) is a neuroinvasive flavivirus infection with a wide spectrum of clinical manifestations whose molecular mechanisms remain insufficiently characterized. MiRNAs regulate pro-inflammatory cytokine production; therefore, changes in their profiles across different forms of TBE may determine the nature of the cytokine response. The aim of this work was to identify the specific features of the circulating miRNA and cytokine profiles in different clinical forms of TBE, as well as to analyze the correlations between them. The study included patients with febrile, meningeal
