Reassessing Regional Tick-Borne Encephalitis Endemicity in Poland Through Seroprevalence Analysis in Blood Donors, 2021-2022.

TBEV is a major cause of viral central nervous system infections in Europe, with heterogeneous geographical distribution and substantial underdiagnosis in low-incidence regions. This study aimed to evaluate the validity of regional TBE risk classification in Poland by combining surveillance-based incidence data with serological markers of TBEV exposure. Plasma samples from 5541 blood donors residing in nine regions were tested by anti-TBEV IgG ELISA, followed by confirmatory VNT, IFA, and anti-NS1 IgG ELISA to differentiate infection-induced from vaccine-induced antibodies. Regions were classi

DNA Vaccine Encoding the prM-E Polyprotein of the Tick-Borne Encephalitis Virus.

We evaluated immunogenicity of the experimental DNA-vaccine construct pVAX-prM-E encoding the polyprotein prM-E of the tick-borne encephalitis virus (strain Sofjin). It was shown that pVAX-prM-E administered to mice using a needle-free jet injector induced the production of high levels of virus-neutralizing antibodies. The level of humoral response to pVAX-prM-E was comparable to that induced by administration of the commercial Tick-E-Vac vaccine. ELISpot analysis of the T-cell response showed that the obtained construct induces high levels of IFNγ in response to stimulation with a pool of spe

A replication-defective tick-borne encephalitis virus generated by NS1 trans-complementation confers robust protective efficacy.

Tick-borne encephalitis virus (TBEV) poses a severe threat to public health, causing neurological disorders with high morbidity and mortality in endemic regions. Therefore, developing safe and effective strategies for TBEV vaccines has long been a focus of attention. Herein, leveraging our well-established NS1 trans-complementation platform, we successfully constructed a high-titer replication-defective TBEV (TBEV-ΔNS1) using the BHK-21 cell line stably expressing Omsk hemorrhagic fever virus (OHFV) NS1 (designated BHKNS1). Retention of the NS1 deletion in TBEV-ΔNS1 was confirmed by continuous

Nuclear activities and interactome of the NS5 protein of tick-borne encephalitis virus.

Orthoflaviviruses are RNA viruses responsible for significant diseases in humans, domesticated animals, and wildlife. Their NS5 protein is central in viral replication, functioning both as an RNA-dependent RNA polymerase and a methyltransferase, while also modulating cellular processes, including the interferon response. Although viral replication is cytoplasmic, the NS5 protein of several mosquito-borne orthoflaviviruses cycles between the cytoplasm and the nucleus of infected human cells. However, the nuclear localization and function of NS5 of tick-borne orthoflaviviruses, such as tick-born

Trends in Tick-Borne Virus Ecology: Vector Incrimination, Focal Distribution, and Reservoir Associations.

Ticks are prominent hosts for many viruses that are pathogenic to humans on almost every continent. Despite the documented historical association of ticks with viral disease in humans, clinical case reports are low, likely due to under-recognition of these pathogens. A key feature of understanding tick-borne virus transmission to humans lies in the natural invertebrate and vertebrate hosts supporting ecological maintenance. As humans are often infected as incidental hosts during the tick-virus life cycle, determining relevant ticks and mammals involved in maintenance can inform prevention stra

Long-term patient-reported neurocognitive and functional outcomes after tick-borne encephalitis: a prospective controlled multicenter cohort study.

OBJECTIVE: To characterize cognitive symptoms the first year after tick-borne encephalitis (TBE). METHODS: We conducted a controlled, multicenter cohort study of 109 hospitalized patients ≥16 years with confirmed TBE in Norway. At 3 and 12 months after admission, patients completed BRIEF-A (Global Executive Composite (GEC) and subscales), Everyday Memory Questionnaire-Revised (EMQ-R), and Rivermead Post-Concussion Symptoms Questionnaire (RPQ), assessing executive functioning, memory, and post-TBE symptoms (higher scores indicate more difficulties). The primary outcomes were within-patient cha

Surveillance of tick-borne encephalitis virus and Tribeč Virus in Ixodes ricinus ticks from Slovakia.

Tick-borne viruses represent a significant and growing public health concern in Europe. This study investigated the minimum infection rates (MIRs) of tick-borne encephalitis virus (TBEV) and Tribeč virus (TRBV) in Ixodes ricinus ticks collected in Slovakia during 2016-2017 and 2023-2024. A total of 3,686 ticks (3,262 nymphs in 359 pools and 424 adults in 99 pools) from 13 sampling sites were screened for viral RNA. The overall MIRs were 0.38% for TBEV and 0.22% for TRBV. All positive detections occurred in nymphal pools. TBEV was detected at 8 of 13 sampling sites, with local prevalence rangin

Evidence for Powassan virus deletions and defective RNA in field-collected ticks.

Powassan virus (POWV) is a tick-borne flavivirus endemic to the United States, Canada, and parts of Russia. POWV remains an under-studied pathogen, despite the potential for serious and life-threatening neurologic complications following infection. While prior studies have characterized viral diversity due to single nucleotide polymorphisms, little is known about POWV recombination, defective RNAs (D-RNAs), and functional structural variants (SVs). Understanding POWV recombination in its natural vector can provide important insights into its replication and evolution. We analyzed POWV sequence

Divergent roles of Hsp70 chaperones in orthoflavivirus protein secretion and virion formation.

Orthoflaviviruses, such as tick-borne encephalitis virus (TBEV) and West Nile virus (WNV), can cause severe neurological disease and remain without specific antiviral treatments. We found that orthoflavivirus envelope (E) and non-structural protein 1 (NS1) interact with heat shock protein 70 (Hsp70) chaperones, key regulators of protein homeostasis and existing cancer drug targets. We examined how Hsp70 and endoplasmic reticulum-resident BiP contribute to viral protein secretion and infectivity of tick and mosquito-borne orthoflaviviruses. Targeting the Hsp70 nucleotide-binding domain with sma

The Toxoplasma gondii prevalence and its impact on tick-borne encephalitis virus infection among humans in Heilongjiang province of China.

Toxoplasmosis and tick-borne encephalitis (TBE) are endemic in Heilongjiang province, China, posing a significant threat to public health. Although some research has indicated that Toxoplasma gondii infection may suppress the host’s immune response and increase susceptibility to other pathogens, no studies have been conducted to explore the association between these two diseases. From 2020 to 2023, a total of 415 serum samples were collected and tested for both T. gondii and tick-borne encephalitis virus (TBEV) infections using enzyme-linked immunosorbent assay (ELISA) by us. The positive rate

Complications and long-term sequelae of tick-borne encephalitis: association with age and other predictors - a multicenter observational study.

Tick-borne encephalitis (TBE) is the most common non-purulent neuroinfection in Central Europe and a major public health concern in the Czech Republic, where vaccination coverage remains low. Even after recovery from the acute disease, many patients experience long-term sequelae known as postencephalitic syndrome (PES). This prospective multicentre study examined predictors of acute disease severity and long-term outcomes, including motor deficits and other forms of PES. A total of 209 patients with laboratory-confirmed TBE were enrolled at four Czech centres in 2023 and 2024; 187 completed si

Evolutionary history of Jamestown Canyon virus reveals complex multi-vector ecology.

Jamestown Canyon virus (JCV) is a historically understudied mosquito-borne virus of increasing concern in North America. We generated 658 whole-genome JCV sequences from northeast United States, including 84% (500/597) of all JCV-positive mosquitoes detected in Connecticut from 1997 to 2022. Then, we applied phylodynamic methods to demonstrate how mosquito phenology structures the maintenance and evolution of JCV. Our phylogenetic analyses estimate that JCV was introduced in the Northeast by at least the early 1700s, and the primary introductions of lineages A and B into Connecticut occurred d

Two Coding-Complete Genomes of Tick-Borne Encephalitis Virus Sequenced from Ixodes persulcatus Collected in Bulgan, Mongolia.

Tick-borne encephalitis virus (TBEV) is primarily transmitted by Ixodes spp. and poses significant health risks, leading to morbidity and mortality in humans. Two of the five subtypes, Siberian and Far Eastern are known to circulate in Mongolia. In 2021, Ixodes persulcatus ticks were collected from Bulgan aimag (province) using flagging and dragging methods and subsequently screened for TBEV using PCR. Positive samples underwent sequencing using an Oxford Nanopore Technologies-based hybrid capture approach, resulting in two coding-complete TBEV genomes from separate tick pools. Phylogenetic an

Cognitive outcome in relation to cerebrospinal fluid markers of neuronal and glial cell damage: A prospective study of tick-borne encephalitis.

OBJECTIVES: Tick-borne encephalitis (TBE) often results in long-term cognitive sequelae. While biomarkers of neuronal and glial injury are established in other neurological conditions, their role in predicting cognitive outcomes in TBE remains unclear. This study investigated whether acute-phase levels of neurofilament light (NfL), glial fibrillary acidic protein (GFAP), and S100B (calcium-binding protein B) in both cerebrospinal fluid (CSF) and serum, and tau and chitinase-3-like protein 1 (YKL-40) in CSF only, were associated with cognitive functioning at follow-up.
