Exposed yet unmapped: evidence of differential flood exposure in deprived urban areas using citizen science.

BACKGROUND: Due to their immature immune system, neonates are at high risk of infection. This vulnerability when combined with limited resources and health education in developing countries can lead to sepsis, resulting in high global neonatal mortality rates. Many of these deaths are preventable. The BabyGel pilot trial tested the feasibility of conducting the main randomised trial, with the provision of alcohol handgel to postpartum mothers for prevention of neonatal infective morbidity in the rural community. This secondary analysis sought to evaluate the methods of detecting infections in

African-specific genetic loci determine iron status and risk of severe malaria and bacteremia in African children.

Iron is essential for both humans and pathogens, yet its genetic regulation remains understudied in African populations. Here, we report genome-wide association studies of six iron-related biomarkers in 3928 children from five sites across Africa, with replication in 2868 African American adults and investigate associations with severe malaria and bacteremia. We identify previously unreported loci at genome-wide significance, for transferrin at GTF3C5, and for hepcidin at CHCHD7/SDR16C5. Variants tagging the DUP4 haplotype, encoding the Dantu blood group (rs552439837) are associated with solub

From Plato to Pareto: Defining the shape of infection's disease space.

If the immune system is an interconnected network, then the evolution of an archetype that is ideal for fighting one pathogen should result in trade-offs decreasing its ability to fight others. How many archetypes are there in an immune system? We infected diverse mice with Plasmodium chabaudi and identified five distinct archetypes of responses on the basis of the host's position in microbial load, immune activity, and host damage space. To better understand the nature of these archetypes, we developed a mathematical model of a generalized host-pathogen system. This model explains the number

Species-specific iNOS expression distinguishes epithelial and myeloid IFNγ responses in tuberculosis.

RATIONALE: Tuberculosis vaccines aim to elicit protective immunity, with IFNγ-producing CD4⁺ T cells considered central mediators of host defense. In murine models, IFNγ activates macrophages to induce inducible nitric oxide synthase (iNOS), resulting in nitric oxide-dependent control of Mycobacterium tuberculosis (Mtb). OBJECTIVE: To define the relevance of IFNγ-induced iNOS activity in humans and other natural host of virulent mycobacterial species. METHODS: We systematically compared IFNγ-induced effector responses in Mtb-infected myeloid cells across species. Using bulk RNA sequencing, f

Exploiting autophagy-targeting natural compounds for potential antimicrobial actions.

Natural products are biologically active compounds used for therapeutic interventions for various diseases, particularly infections. Autophagy is an intracellular catabolic pathway involving lysosomal degradation and is closely associated with immunological pathways, effectively combating bacterial, viral, fungal, and parasitic infections. Accumulating evidence suggests that autophagy activation or inhibition by natural products promotes antimicrobial responses against various pathogens. Numerous natural products can modulate autophagy through diverse signaling pathways, suggesting their poten

Profiling complement proteins in pediatric tuberculosis children with or without serological evidence of prior SARS-CoV-2 exposure.

INTRODUCTION: Coinfection with Mycobacterium tuberculosis (TB) and SARS-CoV-2 poses unique challenges in pediatric population due to overlapping immune pathways and potential immune dysregulation. The complement system, a key arm of innate immunity, plays a dual role in pathogen clearance and inflammation. METHODS: This study aimed to investigate complement activation in TB in children with or without serological evidence of prior SARS-CoV-2 exposure. Children aged 2-17 years with pulmonary or extrapulmonary TB were recruited from two tertiary hospitals in South India. Participants were group

From pandemic influenza to novel coronaviruses: emerging infectious diseases of the 21st century.

Emerging infectious diseases have risen significantly in the twenty-first century as ecological disruption, climate change, expanding human-animal interfaces, and global mobility intensify opportunities for pathogen transmission. This review synthesizes historical and contemporary evidence across viral, bacterial, fungal, and parasitic threats to characterize how diverse pathogens emerge and spread. Foundational events such as the 1918 influenza pandemic, mid-century influenza pandemics, the emergence of HIV/AIDS, and the eradication of smallpox provide context for understanding modern disease

Adapting the Daily Inventory of Stressful Events for SMS Delivery: Recommendations for Research.

Vernal keratoconjunctivitis (VKC) is a chronic and potentially sight-threatening disease. Topical corticosteroids (Cs) seem to be the only effective treatment for this condition, although severe side effects may occur owing to their prolonged use. More recently, cyclosporine (Cyc) eye drops have been reported as a valid alternative, but so far such treatment has only been successfully experimented for a short time and in small numbers of patients. The aim of our study is to evaluate the long term safety and efficacy of topical cyclosporine eye drops in children suffering from VKC. Over a perio

Projecting long-term excess risks of major infectious diseases associated with future extreme weather events in Thailand.

Climate change is postulated to impact infectious disease transmission, yet few studies have characterised the excess risks of infectious diseases associated with extreme weather events. To address this, we conducted a study estimating and projecting the impacts of extreme heat and precipitation on the incidence of major infectious diseases in Thailand. We developed, fitted and validated an analytical framework to model province-level disease cases and their relationship with extreme weather indicators based on historical data. We used generalised additive models to delineate the relationship

Pathological findings in 61 free-ranging leopards (Panthera pardus) from the Greater Kruger National Park, South Africa. Part Two: non-parasitic conditions.

Few reports on diseases in free-ranging leopards (Panthera pardus) are available. A retrospective review of tissues from free-ranging leopards from the Greater Kruger National Park, South Africa, that were submitted for histological examination was conducted. Non-parasitic lesions were recorded in 61 free-ranging juvenile to elderly leopards. They had died or been euthanized between 1998 and 2023. Parasitic conditions in these leopards have been reported separately. Traumatic injury was common (41%). Respiratory tract conditions included pneumoconiosis (36%) and tuberculosis (23%), presumed to

Disseminated angiostrongylosis with involvement of the central nervous system as a cause of sudden death in a dog in Germany.

BACKGROUND: The metastrongyloid nematode Angiostrongylus vasorum, known as "French Heartworm", is an emerging concern in canine health in Europe. Foxes are considered the main reservoir hosts, and affected carnivores predominantly show cardiovascular and respiratory signs. However, a disseminated course of infection can have diverse and severe consequences, including fatal outcomes. CASE PRESENTATION: A six-year-old male Labrador Retriever was presented to the clinic with progressive episodes of stupor lasting a few seconds and dyspnea. Radiographic examination revealed multifocal pulmonary c

Integrated pathological and molecular characterisation of respiratory disease in extensively reared Sicilian Black pigs: detection of Mycoplasma hyopneumoniae, porcine circovirus type 2, and concurrent pulmonary silicosis.

The Sicilian Black pig is a well-adapted autochthonous breed, yet its susceptibility to respiratory disease remains poorly defined due to limited data on pathogen circulation and lesion patterns in extensive production systems. This study characterised the pathological features of Porcine Respiratory Disease Complex (PRDC) in this breed by examining lungs and tracheobronchial lymph nodes from 59 animals through pathological assessment, molecular detection of major respiratory pathogens, and evaluation of crystalline particulates using scanning electron microscopy coupled with energy-dispersive

Etiologies of community-acquired febrile illness identified by TaqMan Array Card qPCR on blood samples: a systematic review and meta-analysis.

UNLABELLED: Community-acquired febrile illness presents a significant diagnostic challenge. The TaqMan Array Card (TAC) enables simultaneous detection of many bacterial, viral, parasitic, and fungal pathogens. This systematic review aimed to characterize the etiologies of community-acquired fever diagnosed by TAC directly on blood specimens. A systematic search of PubMed, Scopus, Embase, and Web of Science (from inception to January 2026 in English) identified studies using TAC to detect pathogens directly in human blood to identify the etiology of community-acquired febrile illness. Fasciola hepatica in alpine dairy farming: prevalence trends, risk factors and associations with Salmonella Dublin seropositivity.

Fasciola hepatica impairs animal health and productivity in ruminants and has been associated with Salmonella Dublin infection, a zoonotic pathogen in cattle that causes diarrhea, abortions, and pneumonia. This work reassessed F. hepatica seroprevalence in Tyrolean dairy herds and investigated associations with alpine pasturing, the breed, S. Dublin occurrence and milk performance. In late autumn/early winter 2023, bulk tank milk samples from 3645 farms were tested using a commercial ELISA test (IDEXX Fasciolosis Verification Test, Montpellier, France). Global burden of enteric infectious diseases, diarrhoeal diseases, and corresponding aetiologies, 1990-2023: a systematic analysis for the Global Burden of Disease Study 2023.

BACKGROUND: Enteric infectious diseases claim more than 1 million lives annually and are among the top ten causes of death in children younger than 5 years. Remarkable global investment has been dedicated to enteric infectious disease prevention and control; however, the shifting global health landscape is testing the continuance of progress. To evaluate the current status and guide future interventions, we present the latest epidemiological estimates of enteric infectious diseases from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2023 and assess progress towards the G

Molecular characterization and antifungal susceptibility of Aspergillus spp. isolated from critically ill COVID-19 patients: implications for suspected COVID-19-associated pulmonary aspergillosis (CAPA) in a Brazilian tertiary referral hospital.

COVID-19-associated pulmonary aspergillosis (CAPA) is a severe complication in critically ill patients, but species identification and antifungal resistance data from middle-income countries remain limited. We analyzed 17 Aspergillus isolates from respiratory specimens of 15 critically ill patients with COVID-19 in São Paulo State, Brazil, during 2020-2021. Patients were retrospectively classified according to ECMM/ISHAM criteria as probable CAPA (n = 5), possible CAPA (n = 7), or non-CAPA (n = 3). Complementary reassessment using the 2024 FUNDICU definitions classified patients as probable in
