Orthoflaviviruses are RNA viruses responsible for significant diseases in humans, domesticated animals and wildlife. Their NS5 protein is central in viral replication, functioning both as an RNA-dependent RNA polymerase and a methyltransferase, while also modulating cellular processes, including the interferon response. Although viral replication is cytoplasmic, the NS5 protein of several mosquito-borne orthoflaviviruses cycles between the cytoplasm and the nucleus of infected human cells. However, the nuclear localization and function of NS5 of tick-borne orthoflaviviruses, such as tick-borne

Ticks are highly competent arthropod vectors, capable of transmitting a great variety of viral, bacterial and protozoan pathogens. The continuous identification of tick-borne viruses (TBVs), especially those pathogenic to humans and animals, poses an increasing threat to public and veterinary health worldwide. Following an incubation phase, the clinical course of human TBV infections often starts with non-specific symptoms (such as fever) that can be followed by distinct clinical manifestations (such as haemorrhage, encephalitis or multi-organ failure). Importantly, a lack of specific diagnost

INTRODUCTION: Tacheng tick virus 1 (TcTV-1) is an emerging tick-borne nairovirus associated with human febrile illness. To date, no reliable detection method for TcTV-1 has been established. In this study, we developed and evaluated a rapid loop-mediated isothermal amplification (LAMP) assay for the detection of TcTV-1.
METHODS: The primers were designed based on the nucleocapsid protein (NP) gene of TcTV-1. Sensitivity was assessed using ten-fold serial dilutions of recombinant plasmids containing the target sequence. Specificity was evaluated using cDNA from Songling virus (SGLV), Yezo viru

Kyasanur Forest Disease (KFD), commonly known as 'monkey fever,' is a highly neglected tropical disease caused by the Kyasanur Forest Disease Virus (KFDV). KFD is endemic to the Western Ghats of Karnataka, India, with seasonal outbreaks during December to June every year. As there is no standard treatment regime, KFD can be fatal with a mortality rate of 2%-10%. Currently, KFD is detected through a non-specific IgM-ELISA followed by RT-PCR, which often delays diagnosis, leading to increased disease severity and even death. To address this, we focused on developing a specific antigen-based KFD

BACKGROUND: Tick-borne encephalitis (TBE), caused by tick-borne encephalitis virus (TBEV), is a zoonotic disease that can lead to severe neurological symptoms. Given the increasing number of reported human TBE cases in Europe, we developed a spatio-temporal predictive model to infer the year-to-year probability of human TBE occurrence across Europe at the regional and municipal administrative levels.
METHODS: We derived the distribution of human TBE cases at the regional level during 2017-2022 by using data provided by the European Centre for Disease Prevention and Control (ECDC), and at the

A 66-year-old man with a history of hypertension presented with left-sided jaw jerking, repetitive movements, aphasia, and dysarthria. He was afebrile and in no acute distress, with mild hypertension (153/81 mm Hg). Routine laboratory studies were unremarkable. Brain MRI showed multiple nonenhancing supratentorial, cortical, and subcortical hyperintensities on T2-weighted fluid-attenuated inversion recovery images, which were nonspecific for infectious, inflammatory, vascular, or neoplastic etiologies. CT of the chest, abdomen, and pelvis and MRI of the cervical and thoracic spine were unremar

Background/Objectives: While tick-borne encephalitis virus (TBEV) is genetically relatively conserved, the significant antigenic divergence between its main circulating subtypes hinders the development of broadly effective antiviral treatments and vaccines. Current inactivated TBEV vaccines offer limited cross-protection against heterologous strains, as evidenced by cases among vaccinated individuals in endemic regions. The aim of this study was to design a candidate mRNA vaccine and evaluate the breadth of protective immunity it elicits. Methods: Ten candidate mRNA-PrM/E-LNP vaccines were com

Type I interferons (IFN-Is) are critical antiviral cytokines that restrict viral replication and limit viral disease. A remarkable recent discovery is that human autoantibodies (autoAbs) neutralizing the activities of IFN-Is phenocopy inborn errors of immunity and markedly exacerbate susceptibility to life-threatening infections. Development of these pathogenic autoAbs in humans is strongly linked to genetic and nongenetic factors affecting thymic function, and they are estimated to be present in >100 million people worldwide with a prevalence that increases with age. Here, we review major adv

Introduction. Alongshan virus (ALSV) is a tick-borne Flaviviridae. It has been detected in Ixodes ricinus and Ixodes persulcatus across China, Russia, Finland, Switzerland and Germany. Hypothesis/Gap Statement. However, the clinical relevance and the pathogenicity of ALSV in humans remain unclear. Sensitive and specific molecular tools are needed to support surveillance and to enable clinical investigations of ALSV in suspected cases of tick-borne meningoencephalitis. Aim. We aimed to develop, validate and integrate two ALSV-specific TaqMan real-time quantitative PCR (qPCR) assays on our open,

Group A Streptococcus (GAS) infections can lead to neuropsychiatric sequelae in children, yet the mechanisms driving post-infectious brain pathology remain poorly defined. In a mouse disease model, Th17 lymphocytes induce microglial activation, blood-brain barrier (BBB) dysfunction, and neural circuit impairment; however, the transcriptional programs underlying these effects, and the specific Th17-derived cytokines involved are unclear. Using mouse genetics, single-cell RNA sequencing, and spatial transcriptomics, we show that GAS infections induce inflammatory gene programs in microglia and b

Antibody-dependent enhancement (ADE) of infection is a phenomenon observed during secondary infection caused by some viruses (primarily, from the Flaviviridae family), including dengue virus (DENV), Zika virus (ZIKV), West Nile virus (WNV), tick-borne encephalitis virus, yellow fever virus, and some others. There are two types of ADE mediated by (i) Fc receptors and (ii) receptors for complement system components. The first type involves 4 types of Fc receptors for IgG molecules (FcRI, FcRIIa, FcRIIb, and FcRIIIa) which ensure the binding of the virus-antibody complex to the target cell and it

The prevalence, epidemiological history and disease risk posed by pathogenic tick-borne viruses (TBVs) remain poorly understood in Northeast Asia. Here, over 12,000 ticks were collected from diverse ecological environments in Northeast China, leading to the identification of 44 TBVs from which 1,221 viral sequences were assembled. This included six human/mammal-infecting TBVs: Alongshan virus, Beiji nairovirus, Mukawa virus, Nuomin virus, tick-borne encephalitis virus (TBEV), and Yezo virus. Aga, Izumo, and Nishinomiya in Japan were identified as tick diversity hotspots, while Shangzhi City, G

Tick-borne diseases represent a growing public health concern worldwide, yet the microbial factors that govern pathogen transmission remain incompletely understood. Over the past decade, high-throughput metagenomics and functional studies have revealed that two distinct microbial communities-the vertebrate host's skin microbiota and the tick's own microbiome-act synergistically as key modulators of pathogen acquisition, persistence within the vector, and successful transmission to the vertebrate host. At the feeding site, the skin microbiota orchestrates local cutaneous immunity, influences in

Prolonged tick attachment with deep subcutaneous embedding is uncommon in humans but may complicate tick removal in primary care. Firm fixation is mediated by the tick's barbed hypostome and the secretion of glycine-rich salivary cement, which together anchor the mouthparts within the dermis. Awareness of these mechanisms is important when standard extraction techniques fail. A patient presented with a live, engorged tick deeply embedded in the skin after several days of attachment. The tick's anterior body and mouthparts were firmly fixed within the dermis, and surrounding erythema migrans wa

Tick-borne encephalitis (TBE) is an emerging infectious disease with effective vaccines available. However, breakthrough TBE occurs in previously vaccinated patients, a condition that is difficult to diagnose. We report a case of a previously healthy teenager who fell severely ill and was hospitalized for three months despite full TBE vaccination. Initial fever and headache progressed during the first week to encephalitis and, since autoimmune encephalitis was suspected, treatment with immunoglobulins and methylprednisolone was given. This case demonstrates the delayed response of TBEV IgM, ty
