AlphaFold-driven structure-guided identification of NS5 RdRp-targeting antiviral leads against Kyasanur Forest Disease Virus.

With no approved antiviral treatments currently available, Kyasanur Forest Disease Virus (KFDV) remains a serious and neglected tick-borne flavivirus posing a significant public health concern endemic to India. To address this therapeutic gap, we employed AlphaFold-based structural modelling integrated with structure-based lead identification targeting the RNA-dependent RNA polymerase (RdRp) domain of the viral NS5 protein. The predicted NS5 model exhibited high structural confidence, with pLDDT scores approaching 90 and a pTM score of 0.8, and was further validated using Verify3D, PROCHECK, a

A non-canonical JAK/STAT pathway promotes viral replication through the lipoprotein receptor-related protein in ticks.

Ticks transmit numerous viruses that pose significant threats to human and animal health, however, the molecular interactions between ticks and viruses remain poorly understood. Using Langat virus (LGTV)-a surrogate for tick-borne encephalitis virus (TBEV), we show that the JAK/STAT pathway promotes viral infection in Haemaphysalis longicornis. Rather than directly interacting with viral proteins, this proviral effect is mediated by a low-density lipoprotein receptor-related protein (LRP), whose expression is regulated by STAT. Silencing LRP in H. longicornis reduced LGTV infection, while ecto

Case 346: γ-Aminobutyric Acid A Receptor Encephalitis.

A 66-year-old man with a history of hypertension presented with left-sided jaw jerking, repetitive movements, aphasia, and dysarthria. He was afebrile and in no acute distress, with mild hypertension (153/81 mm Hg). Routine laboratory studies were unremarkable. Brain MRI showed multiple nonenhancing supratentorial cortical and subcortical hyperintensities on T2-weighted fluid-attenuated inversion recovery (FLAIR) images, which were nonspecific for infectious, inflammatory, vascular, or neoplastic etiologies. CT of the chest, abdomen, and pelvis and MRI of the cervical and thoracic spine were u

Screening Ticks for Crimean-Congo Hemorrhagic Fever Virus and Aigai Virus in Greece.

Ixodid ticks are vectors for a plethora of pathogens, including the Crimean-Congo hemorrhagic fever virus (CCHFV), which causes severe disease in humans. Two autochthonous CCHF human cases were reported in 2025 in Greece. The aim of the present study was to gain a better insight into the geographic distribution and prevalence of CCHFV and the related Aigai virus (AIGV) in ticks in Greece. Therefore, 680 ticks (135 Hyalomma and 545 Rhipicephalus ticks) collected during 2024 from livestock (sheep, goats, cattle) and from the environment were tested for CCHFV and AIGV. AIGV was detected in 12 adu

Langat Virus Infection Can Be Demonstrated in Both Tick Salivary Glands and Midgut Within 24 Hours of Blood Feeding.

The detailed mechanism and sequence by which tick-borne flaviviruses (TBFVs), such as Langat virus (LGTV), infect and disseminate in arthropod hosts remain undefined. To begin characterizing these processes, we used artificial membrane feeding chambers to feed adult Ixodes scapularis ticks with blood containing LGTV. At 24, 48, 72, and 96 hours (h) after attachment, we removed and dissected the partially fed ticks to obtain the midgut and salivary glands. Histology confirmed infection in cells of the digestive epithelium lineage; infection was noted in midgut generative cells and the more diff

Seroprevalence of Orthoflaviviruses in horses in 3 different Algerian climatic zones between 2021 and 2023.

Orthoflaviviruses are etiological agents of neurological diseases in equines. Horses are susceptible to equine Tick-borne encephalitis virus (TBEV), Usutu virus (USUV), and West Nile virus (WNV), although mostly WNV has been associated with clinical disease. The aim of the present study was to estimate the prevalence of TBEV, USUV and WNV in 289 healthy horses from nine provinces of Algeria. This study was conducted between March 2021 and December 2023. Sera that tested positive, doubtful, or negative close to the doubtful threshold in the Orthoflavivirus-ELISA were further tested by the virus

Clinical Characteristics and Neutralizing Antibody Levels in a Japanese Patient with Tick-borne Encephalitis: A Case Report.

Tick-borne encephalitis (TBE) is a severe neurological disease caused by infection with the TBE virus (TBEV), primarily transmitted to humans through tick bites. As there is no effective treatment, prevention through vaccination and avoidance of tick bites is essential. In Japan, only five cases had been reported up to 2023, all from Hokkaido, the northernmost main island. Here, we report the sixth case of TBE in Japan. The patient was a 58-year-old male, in whom magnetic resonance imaging revealed enhancing lesions in the brain and spinal cord, accompanied by severe neurological symptoms. Des

Powassan Virus Seroprevalence in a U.S. Servicemember Population at High Risk for Tick Exposure.

Powassan virus (POWV) is an emerging tick-borne flavivirus that can cause severe encephalitis in humans. Currently no vaccines or therapeutics are approved to treat POWV. POWV is spread by the deer tick, Ixodes scapularis, which is ubiquitous across the Northeastern United States. To better understand POWV prevalence in high-risk populations, we examined POWV seroprevalence in Cadets at United States Military Academy (USMA) in West Point, New York. Cadets at USMA, located in a heavily wooded area, are at high risk for tick exposure during outdoor military training. 1,051 serum samples from the

Immunopharmacological and structural design of a polyprotein based multi epitope vaccine against Powassan virus.

Powassan virus (POWV) poses a growing neurotropic threat with limited immunopharmacological interventions, requiring the development of targeted vaccine strategies. The current study focuses on developing a specific vaccine using bioinformatics tools to target T- and B-cell epitopes from a genome polyprotein sequence of POWV. Four vaccine constructs with possible immunogenic qualities were assembled. Two specific epitopes were selected from B-cell, and T-cell for each construct. The heat-labile enterotoxin S63 adjuvant was attached to the N-terminal, while the PADRE order and the His-tag were

Conserved histidine residues and the control of tick-borne encephalitis virus maturation.

The maturation and infectivity of orthoflaviviruses is driven by interactions and structural changes of the two envelope proteins, prM and E, which are controlled by protonation and deprotonation events during the viral life cycle when the virus encounters acidic and neutral pH environments inside and outside the cell. Histidine residues act as sensors for these pH-induced conformational transitions, which differ between mosquito-borne and tick-borne orthoflaviviruses in some aspects. We identified six histidine residues that are conserved among the envelope proteins of mammalian tick-borne or

Kyasanur Forest Disease: Advances in Experimental Animal Models, Immune Assays, and Challenges in Vaccine Development.

Kyasanur Forest Disease (KFD) is a tick-borne viral haemorrhagic fever endemic to forested regions of South India, with case fatality rates ranging from 3% to 10% and seasonal outbreaks causing recurrent illness among at-risk populations such as forest workers and villagers. The historically used formalin-inactivated KFD vaccine showed limited effectiveness in field studies, providing only about 62% protection. This moderate level of protection highlights the need for improved vaccines that offer higher efficacy and longer-lasting immunity for populations living in endemic areas. A major chall

No-known-vector flaviviruses exhibit diverse replication and virulence phenotypes in mice.

UNLABELLED: The Orthoflavivirus genus (flaviviruses) includes globally significant arboviruses, which cycle between arthropod vectors (mosquitoes and ticks) and vertebrate hosts. In contrast, no-known-vector flaviviruses (NKVFVs) have been isolated from rodents and bats, but not arthropods, so are thought to spread by vector-independent routes. However, little is known about the host range and pathogenic mechanisms of these viruses. To evaluate NKVFV pathogenesis, we infected wild-type, Ifnar1 -/- , and Ifnar1 -/- Ifngr1 -/- mice by footpad inoculation with 12 NKVFVs: Entebbe bat virus (ENTV),

Roles of ticks in the occurrence of emerging and re-emerging vector-borne viruses of zoonotic importance in West Africa.

Ticks are arthropod vectors that transmit a wide range of emerging and re-emerging vector-borne diseases (VBDs), many of which are of considerable zoonotic importance. Recently, tick-borne diseases (TBDs), particularly those caused by arboviruses, have emerged as a significant public health concern in West Africa. The complex socio-ecological landscape in West Africa contributes to the circulation of several tick-borne arboviruses (TBAs), including Crimean-Congo hemorrhagic fever virus (CCHFV) and Dugbe virus (DUGV), both members of the genus Orthonairovirus, as well as other tick-borne flaviv

Serious tick-borne infections - considering the strengths and weaknesses of currently used laboratory diagnostic methods.

The laboratory diagnosis of tick-borne infections is a major interdisciplinary issue, closely linked not only to advances in molecular biological methods but also to changes in ecosystems and biodiversity caused by climatic and anthropogenic factors. These factors significantly influence the epidemiological situation both globally and in the Czech Republic, where a marked increase in the incidence of serious tick-borne infections has been observed in recent years. Current diagnostic approaches combine indirect serological methods (e.g., ELISA, Western blot, and immunofluorescence assays) with

Structures of asymmetric particles of tick-borne encephalitis virus provide insight into flavivirus assembly and maturation.

Flaviviruses are globally distributed human pathogens. However, the mechanisms underlying flavivirus assembly and maturation remain poorly understood. Here, we show that many particles of tick-borne encephalitis virus (TBEV) are asymmetric and lack subsets of surface heterodimers. Immature particles of TBEV contain incomplete spikes, providing evidence that their coats assemble directly from heterodimers of premembrane (prM) and envelope (E) proteins. Exposure of TBEV particles to acidic pH in the Golgi complex promotes maturation. The spikes and herringbone regions in TBEV maturation intermed

Restraint of Powassan virus replication by TRIM5α facilitates viral avoidance of antiviral immunity.

TRIpartite Motif (TRIM) protein 5 alpha (TRIM5α) is a well-characterized cellular inhibitor of lentivirus replication that limits transmission of related viruses between primates. We previously reported that TRIM5α derived from humans and rhesus macaques inhibits replication of orthoflaviviruses belonging to the tick-borne encephalitis virus (TBEV) serocomplex, including TBEV, Kyasanur forest disease virus and Langat virus (LGTV), but interestingly not the tick-borne Powassan virus (POWV). To further characterize the primate TRIM5α and orthoflavivirus interface, we screened TRIM5α variants fro

First evidence of exposure to West Nile Virus in horses within Denmark, 2025.

A nationwide serological screening of 822 horses was conducted in Autumn 2025 to investigate potential exposure to West Nile Virus (WNV) within Denmark. Serum samples were tested for antibodies against WNV, tick-borne encephalitis virus (TBEV) and Usutu virus (USUV). Four horses without any history of travel outside of Denmark, or previous vaccination against WNV, tested positive for WNV neutralising antibodies. The results support local exposure to WNV indicating that virus infected mosquitoes have been present in Denmark and underline the usefulness of horses as sentinels for the risk of hum
