The taiga tick Ixodes persulcatus (Schulze, 1930) (Acari, Ixodidae), widespread in the northern Palearctic, is the vector of dangerous human pathogens including tick-borne encephalitis virus. Knowledge of its ecology is important for minimising tick bite exposure risk. We studied the diel activity of male and female I. persulcatus in the north of the range (Karelia, Russian Federation) and tested its relationship with environmental factors including temperature, relative humidity and light intensity. The field experiment was conducted in May-June 2024. Cages with unfed adult ticks (males and f

Ticks are vectors of a wide range of pathogens, relevant to both veterinary and public health. Although tick surveillance is frequently conducted in the Netherlands, data on tick-borne pathogens in free-living herbivores remains limited, leaving the impact of these pathogens on animal health poorly understood. This study aimed to characterise tick species and determine seroprevalence and infection prevalence of selected tick-borne pathogens in large herbivores at the Oostvaardersplassen nature reserve and questing ticks collected in the area. Between September 2023 and December 2024, blood was

Tick-borne encephalitis (TBE) remains a severe public health threat in affected areas with shifting geographic distribution linked to environmental change. However, systematic tick surveillance is limited due to the cost and effort of field monitoring. Here, we evaluate the potential of using national citizen science tick data reports for predicting tick-human interaction and TBE risk alongside socioeconomic and environmental data. We integrate citizen science observations with socioeconomic and environmental data, and apply statistical and machine learning models to identify key drivers and a

Background and Clinical Significance: When diverse clinical presentations coincide with complex laboratory findings, particularly in older adults, the diagnostic process can be especially challenging. Case Presentation: We report the case of a geriatric patient who was hospitalized with initial gastrointestinal and respiratory symptoms, followed by progressive chest pain and profound weakness, accompanied by elevated transaminases, troponin elevation, and hyponatremia, initially suggesting multiple competing diagnostic entities. During the clinical course, the patient developed neurological sy

Tick-borne diseases constitute a major group of emerging vector-borne infections and represent a growing public health concern in many European countries. In Europe, the most clinically significant agents transmitted by Ixodes ricinus are tick-borne encephalitis virus (TBEV) and Borrelia burgdorferi sensu lato, the causative agents of tick-borne encephalitis and Lyme borreliosis, respectively. Ixodes ricinus also serves as a vector for other pathogenic microorganisms, including louping-ill virus (LIV) and Anaplasma phagocytophilum. Various endosymbionts, such as Wolbachia pipientis and Midichl

Climate change is reshaping the epidemiology of vector-borne diseases in Europe by altering the ecological conditions that determine vector survival, seasonal activity and pathogen transmission. Rising temperatures, milder winters, prolonged warm seasons and changing precipitation patterns are increasing the suitability of parts of Europe for competent mosquito, tick and sandfly vectors. These changes, combined with human mobility and land-use change, increase the probability that imported pathogens encounter permissive conditions for local transmission. This Opinion article examines autochtho

Flaviviruses, encompassing notable pathogens, like Dengue, Zika, West Nile, and tick-borne encephalitis viruses, elicit complex cellular stress responses, involving pathways such as the unfolded protein response (UPR), integrated stress response (ISR), apoptosis, autophagy, and the antiviral immune response. These pathways regulate cell fate by either promoting survival to counteract virus-induced damage or triggering cell death programs under prolonged and irreparable stress. Therefore, the primary aim of flavivirus-induced cellular responses is to protect cells and hinder viral propagation.

The Powassan virus (POWV) is a neurovirulent tick-borne virus that causes age-associated lethality and long-term neurologic sequelae in 50% of survivors. In aged mice the POWV strain LI9 mirrors human lethality and neuropathology; however, an avirulent POWV mutant, LI9-D308N, fails to enter the CNS or cause lethal disease. The D308N mutation is present in an envelope protein domain associated with cell attachment, yet the role of D308N mutations in cell tropism and neuroinvasion remains to be resolved. Here, we engineered fluorescent mScarlet3 and mNeonGreen reporter genes into WT LI9, and avi

BACKGROUND: Tick-borne encephalitis virus (TBEV) is an important cause of neuroinvasive infection in Europe and Asia. Israel is considered nonendemic, yet travel-associated cases have been reported.
METHODS: Sera from hospitalized neurological patients in Israel (2010-2019) were screened for TBEV IgG antibodies. Positive/borderline results were confirmed by virus neutralization testing (VNT) against both TBEV and West Nile Virus (WNV). Confirmed TBEV-neutralizing sera were tested for anti-TBEV IgG and IgM by immunofluorescence assay.
RESULTS: Of 12,651 suspected meningitis/encephalitis cases

Ixodes ricinus ticks are widely distributed throughout Europe and represent major vectors of tick-borne encephalitis virus and the Lyme borreliosis agent Borrelia burgdorferi sensu lato. In invertebrates, C-type lectins are commonly associated with innate immune functions, and several such lectins have been predicted in I. ricinus. Given the limited knowledge of lectin function in ticks, we characterized three carbohydrate-recognition domains (CRDs) of a novel C-type lectin identified in the I. ricinus transcriptome (IrCLec). The tertiary structures of CRD1, CRD2, and CRD3, predicted using the

INTRODUCTION: Vector-borne diseases (VBDs) pose a growing threat to human and animal health, driven by complex interactions among and between vectors, hosts, pathogens, and the environment. The Nordic region is warming at twice the global average rate, creating novel ecological conditions that reshape VBD transmission dynamics.
OBJECTIVES AND METHODS: This narrative review synthesises current knowledge regarding major tick- and mosquito-borne diseases in the Nordic region, with a focus on Finland, based primarily on outputs from the VECLIMIT project (2020-2023), supported by other Nordic stud

BACKGROUND: Tick-borne viruses (TBVs) are emerging pathogens that affect human and animal health, yet their diversity, epidemiology, and clinical impact in Africa remain poorly synthesised. This systematic review aimed to map TBV diversity, quantify prevalence and seroprevalence, assess clinical outcomes, and evaluate the extent of One Health implementation in surveillance studies across Africa.
METHODS: We searched PubMed, Embase, Scopus, Web of Science, CAB Abstracts, African Index Medicus, and grey literature up to 9 April 2026. Independent reviewers screened records, extracted data, and a

Ticks and tick-borne diseases (TBDs) in Southeast Asia are poorly understood, confounding surveillance, and mitigation opportunities. Herein, we conducted a survey of experts (n = 25) to identify tick species and TBDs of greatest concern to Southeast Asia, 'drivers' which may alter their impacts, and research hurdles. These experts identified Dermacentor auratus, Haemaphysalis longicornis, Rhipicephalus linnaei, Amblyomma testudinarium sensu lato, Ixodes granulatus, Ixodes persulcatus, and Rhipicephalus microplus as ticks of greatest concern; and rickettsioses, severe fever with thrombocytopen

As arthropod-borne viruses continue to threaten populations globally, there is a pressing need for experimental systems that enable rapid antiviral discovery. Reverse-genetics platforms producing recombinant reporter orthoflaviviruses have been developed to address this gap. Here, we present two new recombinant tick-borne encephalitis viruses (TBEVrec) generated on a European-subtype Haselmühl Tiho1 isolate backbone. A reporter gene, either eGFP or Nluc, was inserted in the capsid-coding region of the genome downstream of the capsid RNA regulatory signal and separated from the complete viral p

OBJECTIVES: Antibodies against tick-borne encephalitis (TBE) virus (TBEV) nonstructural protein 1 (NS1) immunoglobulin G are elevated in TBEV-infected individuals, but not in TBE vaccinated individuals, enabling estimation of TBEV infection prevalence in countries where TBE vaccines are used.
METHODS: We used data from NS1 seroprevalence studies to estimate the incidence of medically attended TBEV infections and then compared this incidence with the incidence of surveillance-reported TBE cases to estimate the under-ascertainment multiplier for medically attended TBEV infections identified by

Tick-borne encephalitis (TBE) is an emerging viral infection in Europe and Asia. Climate and environmental changes are expanding the distribution and activity of Ixodes ticks, increasing human exposure. Most infections are asymptomatic, but symptomatic cases may present with a biphasic illness that can progress to meningitis or encephalitis, sometimes leading to long-term neurological sequelae, particularly in the elderly population. Currently, treatment is limited to supportive care, as no specific antiviral therapy is available. Effective vaccines exist and represent the main preventive stra

Tick-borne encephalitis virus (TBEV), a prominent orthoflavivirus transmitted by ticks, poses a risk of potentially fatal encephalitis in humans. Currently, the only available inactivated vaccines have limited usage, underscoring the need for developing novel vaccine strategies. The 3' untranslated region (UTR) of TBEV can be divided into variable and conserved regions, where structured RNA elements play crucial roles in viral replication and virulence. In this study, we found that deleting sequences in both the variable region and most of the conserved region within the 3'UTR eliminated viral

Rocky Mountain spotted fever (RMSF) is among the most severe tick-borne rickettsial infections encountered in the United States. Its presentation ranges from the classic triad of fever, headache, and rash to central nervous system (CNS) conditions, including meningitis, encephalitis, and seizures. Notably, RMSF presenting as an ischemic stroke due to CNS vasculitis is rare, which can prove fatal if recognition is delayed. We present the case of a woman in her 50s with a history of well-controlled diabetes mellitus type 2 and hypertension, who presented with fever, chills, body aches, and rash.

Artificial feeding systems have become essential tools for studying tick physiology, pathogen acquisition, and transmission without relying on vertebrate hosts. While in vitro feeding has been successfully established for several ixodid species, including Ixodes ricinus, Dermacentor spp., and Rhipicephalus spp., no system has yet supported complete engorgement and reproduction of Ixodes hexagonus, a widespread European tick and vector of Borrelia spp. and the tick-borne encephalitis virus. In this study, we report the first successful in vitro feeding of adult I. hexagonus using a silicone-mem

OBJECTIVES: Tick-borne encephalitis virus (TBEV) is a major vector-borne pathogen in Europe, but its true infection burden remains unclear due to the high proportion of asymptomatic infections and the inability of conventional serology to distinguish vaccination- from infection-induced antibodies. We applied a non-structural protein 1 (NS1)-based serological approach to estimate TBEV seroprevalence, incidence, and manifestation index in eastern Germany (Saxony and Berlin/Brandenburg).
METHODS: A total of 1,391 anonymized blood donor sera (18-72 years), collected between May and September 2021
