Rapid Construction and Characterization of Infectious cDNA Clones and Reporter Viruses of Enteroviruses, Including Enterovirus A71 and Coxsackievirus B5, with Systematic Identification of Critical Determinants for Successful Reporter Virus Generation.

Enteroviruses are positive-sense single-stranded RNA viruses and common pathogens that are responsible for diverse public health diseases. To facilitate the study of the virus biology and pathogenesis of enterovirus, we developed a rapid method for construction of the enteroviral cDNA clones including enterovirus A71 (EV-A71) and coxsackievirus B5 (CVB5). As described for EV-A71, the full-length cDNA of CVB5 was amplified by long-distance PCR and cloned into a T7 promoter-containing plasmid using directional seamless cloning technology. The virus was successfully rescued by single transfection

An IRES-like cis-acting element located within the EV-A71 coding region drives translation independent of the 5'-IRES and modulates viral fitness through regulated binding of viral RNA to 3D polymerase.

Enterovirus A71 (EV-A71) is a leading cause of hand, foot, and mouth disease and is occasionally associated with severe neurological complications in young children. Previous studies of EV-A71 and related enteroviruses suggested that the virus possibly contains additional cis-acting elements (CAEs) within the major open reading frame (ORF) regulating viral protein translation. To systematically explore CAEs in the EV-A71 genome and characterize their functions, we utilized bicistronic reporter plasmids to screen the entire genome, identifying internal ribosome entry site (IRES)-like CAEs withi

Modelling the impacts of meteorological factors on herpangina: a Bayesian spatiotemporal analysis.

BACKGROUND: Herpangina is a common enteroviral illness in young children with recurrent seasonal surges. Although meteorological conditions influence transmission, evidence capturing non-linear and delayed effects across space remains limited. We aimed to quantify the exposure-lag-response associations of temperature, precipitation, and relative humidity with herpangina risk in Japan using a Bayesian spatiotemporal model. METHODS: Weekly sentinel surveillance herpangina cases and meteorological variables were obtained for the 47 prefectures from January 2001 to December 2024. We applied a Bay

Coxsackievirus A6 on the rise: epidemiology, pathogenicity, evolutionary dynamics, and antiviral strategy.

SUMMARYIn recent years, coxsackievirus A6 (CVA6) has become a predominant cause of hand, foot, and mouth disease (HFMD) worldwide, surpassing enterovirus A71 (EV-A71) and CVA16. The rise of CVA6 is of particular public health concern due to its association with atypical and severe clinical presentations, including extensive vesiculobullous eruptions and neurological complications. These diverse and often non-classical manifestations, which also occur in adults, complicate clinical diagnosis and highlight the need for enhanced molecular surveillance. Furthermore, the potential impact of enterov

Infection-specific long-chain fatty acid metabolism as a broad anti-enterovirus target.

Enteroviruses are arguably the most numerous group of viruses infecting humans. While most enterovirus infections are benign and self-resolving, their sheer number inevitably increases the chances of multiple complications. The diversity of enteroviruses means that the development of vaccines is only economically feasible against a select few, and no direct-acting or host-targeted anti-virals are approved to treat enteroviral infections, largely due to the rapid development of resistance against all experimental drugs. Here, we explored a universal property of enterovirus infection - a massive

Gasdermin D-dependent neutrophil pyroptosis exacerbates Coxsackievirus A6-induced neuroinflammation through NET formation.

BACKGROUND: Coxsackievirus A6 (CVA6) is a nonenveloped, single-stranded RNA virus linked to neurological complications. Emerging evidence suggests neutrophil pyroptosis drives inflammation. However, the role of neutrophil pyroptosis in CVA6 pathogenesis remains unexplored. METHODS: Ten-day-old wild-type (WT), Caspase-1 KO, and GSDMD KO mice were infected with a lethal dose of CVA6. For in vivo and in vitro studies, we used the caspase-1 inhibitor belnacasan, disulfiram, and anti-Ly6G antibody. We also generated neutrophil-specific PAD4-knockout mice (PAD4 Ne-KO) by deleting Padi4 under the S1

Discordant trends in enteroviral infections and meningitis in the post-COVID-19 era: Contributions of type distribution and type-specific risk.

BACKGROUND: After the COVID-19 pandemic, enterovirus circulation rebounded, but enteroviral meningitis remained low in South Korea, suggesting that viral activity alone may not reflect disease burden. METHODS: We analyzed hospital-based and national surveillance data from 2015 to 2025 across the pre-pandemic period (2015-2019), pandemic period (2020-2021), and post-pandemic period (2022-2025). A Kitagawa-type decomposition was applied to quantify the relative contributions of changes in circulating type distribution and type-specific risk. RESULTS: In the post-pandemic period, despite partia

Cross-System Transcriptomics Suggests Enterovirus A71 Exploits Wound Healing Programs to Target Neural Progenitors in Human Spinal Cord Organoids.

INTRODUCTION: The mechanisms of EV-A71 immune evasion and neuroinvasion are poorly understood. Inactivated vaccines provide protective immunity without the dysregulated inflammation and viral persistence caused by active infection. In this bioinformatics-driven study, our objective was to utilize computational transcriptomics to investigate this dichotomy and determine the specific neural cell populations susceptible to enteroviral infection. METHODS: We compared bulk RNA-seq data from THP-1 macrophages infected with active versus inactivated EV-A71 to identify viral hijacking programs. We th

Discovery of non-nucleoside inhibitors of the enterovirus D68 3D polymerase through crystallographic fragment and high-throughput biochemical screening.

Enterovirus D68 (EV-D68) is a non-polio picornavirus that has caused increasing rates of severe respiratory illness and acute flaccid myelitis in children worldwide this century. There are no approved vaccines or antivirals for EV-D68. Thus, we conducted a crystallographic fragment screening (CFS) and a high-throughput screening (HTS) biochemical assay against the EV-D68 RNA-dependent RNA polymerase 3D (3D pol ) to identify ligandable sites and non-nucleoside compounds that can spearhead anti-enteroviral drug discovery. The CFS, involving 650 fragments, identified 68 hit compounds (~10% hit ra

Fibrillarin-mediated 2'-O-methylation serves as a translation brake on uncapped enterovirus RNA.

2'-O-methylation (Nm) is a key epitranscriptomic mark across diverse RNAs, yet its role in antiviral defense remains largely unexplored. To systematically investigate the landscape and functional significance of internal Nm modifications on enterovirus RNAs, a sensitive sequencing method termed RACE-Nm was established, in combination with the machine learning-based predictor NanoNm, to map Nm sites on low-abundance RNAs at single-nucleotide resolution. A conserved Nm site was identified at nucleotide 307C within the viral internal ribosome entry site (IRES), which was catalyzed by the methyltr

Emergence of an echovirus 17 subtype causing a viral meningitis outbreak, Israel, 2025 to 2026.

An outbreak of enteroviral meningitis occurred in Israel between August 2025 and May 2026. Echovirus type 17 was identified in 72 hospitalised patients, most of them infants (n = 44). Many patients (n = 45) were from densely populated Haredi (strictly Orthodox) communities. Clinical course was mild. Phylogenetic analysis suggests extensive local transmission. Echovirus 17 infections declined abruptly following the implementation of community restrictions imposed during a military conflict. This outbreak highlights the value of routine molecular enterovirus surveillance.

Structure-based macrocyclization of α-ketoamides leads to potent inhibitors of coronaviral and enteroviral proteases.

Viral proteases represent validated targets for direct-acting antivirals and the treatment of associated infections. In co-crystal structures of Mpro of SARS-CoV-2 with peptidomimetic inhibitors, we noticed a spatial proximity of sidechains filling the S1' and S2 pockets, as well as those filling S3 and S1 pockets. To enhance molecular rigidity, the proximal residues were conformationally fixed by macrocyclization. We report the synthesis of two macrocyclic series, i.e. exocyclic nitriles with linked P3 and P1 residues and endocyclic α-ketoamides with linked P1' and P2 residues, and characteri

Enteroviruses antagonize the host restriction factor ABL2 via proteasomal degradation to facilitate viral replication.

Enteroviruses, including Coxsackievirus B3 (CVB3), are significant human pathogens that cause severe diseases, such as viral myocarditis, pancreatitis, and encephalitis. ABL proto-oncogene 2 (ABL2), a non-receptor tyrosine-protein kinase, regulates diverse physiological processes and participates in virus infection; however, its role in enterovirus infection remains uncharacterized. Here, we demonstrate a novel host-virus interaction: enteroviruses degrade ABL2 via the ubiquitin-proteasome system through their non-structural protein 2B. Furthermore, ABL2 functions as an antiviral restriction f

Molecular insights into NLRP1 Gly/N-degron recognition by CRL2ZER1.

The Gly/N-degron pathway is a branch of the proteasomal degradation pathway that specifically targets proteins initiated with an N-terminal glycine. The E3 ligase substrate adaptors ZYG11B and ZER1 have been identified as being responsible for recognizing the target proteins of the Gly/N-degron pathway. Previously, it has been shown that the Gly/N-degron pathway activates the human NLRP1 inflammasome by degrading the autoinhibitory N-terminal fragment of NLRP1 after cleavage by the enteroviral 3C protease. However, the recognition of the NLRP1 Gly/N-degron is not yet fully understood. Here, we

Clinical and Laboratory Profiles, Time to Presentation, and Antibacterial Therapy in Children with Aseptic Meningitis: A Comparison of Confirmed Enteroviral and Unidentified-Etiology Cases.

BACKGROUND/OBJECTIVES: Pediatric aseptic meningitis remains diagnostically challenging because early manifestations may be incomplete, cerebrospinal fluid (CSF) findings vary with disease stage, and etiologic confirmation is not always available. This study compared hospitalized children with PCR-confirmed enteroviral meningitis and meningitis of unidentified etiology, focusing on presentation timing, CSF patterns, and antibacterial therapy. METHODS: This single-center retrospective observational study included 87 pediatric patients hospitalized with aseptic meningitis or meningoencephalitis.

Development of a universal primer set for the identification of enterovirus serotypes using next-generation sequencing.

Enteroviruses have been recognized as major etiological agents of viral myocarditis, a significant contributor to sudden cardiac death. However, the causative virus genomes are rarely identified in forensic autopsy specimens, because extensive nucleic acid degradation compromises analysis. The aim of this study was to develop a next-generation sequencing (NGS)-based method for detecting and identifying the serotype of enteroviruses, which are one of the primary viral causes of myocarditis. We developed a primer set using sequences from 21 representative enterovirus serotypes. The sensitivity o

How to differentiate pediatric enteroviral meningoencephalitis from scrub typhus meningoencephalitis: A single-center retrospective study from Yunnan Province, China.

BACKGROUND: During the summer-autumn epidemic season, pediatric scrub typhus meningoencephalitis (ST) and enteroviral meningoencephalitis (EV) often present with overlapping clinical manifestations and routine cerebrospinal fluid (CSF) findings, making early differential diagnosis challenging and delaying appropriate treatment. Practical diagnostic strategies based on readily available parameters are lacking. OBJECTIVE: To evaluate whether common clinical and laboratory indicators can differentiate pediatric ST meningoencephalitis from EV meningoencephalitis. METHODS: This single-center retr
