Overview of Delivery Methods for Gene Editing.

The clinical success of CRISPR-based interventions depends primarily on the efficient delivery of editing components into target cells. While base and prime editing have refined genomic precision, achieving therapeutic efficacy requires specialized vehicles that can navigate systemic circulation, escape endosomes, and ensure cell-specific entry. Delivery platforms are traditionally categorized into viral and nonviral systems. Viral vectors, namely adeno-associated vectors (AAV), lentiviral vectors, and adenoviral vectors, employ evolved mechanisms to achieve high transduction efficiency and pr

Production and Storage of High-Capacity Adenoviral Vectors for the Delivery of Advanced CRISPR Systems.

Genome editing based on engineered CRISPR systems is advancing rapidly, with the field increasingly moving toward approaches that avoid the induction of mutagenic double-stranded DNA breaks (e.g., RNA-programmable base editing, prime editing, and donor DNA transposition). These nuclease-free strategies often rely on large or multi-component molecular assemblies that can include gene-sized donor DNA substrates. There is, nonetheless, a paucity of vehicles capable of delivering such large and complex genome-editing components effectively and, ideally, in defined stoichiometric ratios. High-capac

Completed clinical trials of p53-targeted therapeutic strategies in cancer: A cross-sectional ClinicalTrials.gov registry analysis.

The tumor suppressor gene TP53 is one of the most frequently altered genes in human cancers and plays a central role in regulating cell cycle arrest, apoptosis, and genomic stability. Therapeutic strategies targeting the p53 pathway have been investigated to restore or exploit p53 function in human malignancies. However, the characteristics, reporting completeness, and clinical development patterns of completed trials evaluating p53-targeted therapeutic strategies have not been comprehensively summarized. This study aims to characterize the design, therapeutic strategies, reporting completenes

Host life history traits as drivers of viral infection in European bats.

Bats are recognized as natural reservoirs of a wide range of viruses. Understanding the factors that determine patterns of viral occurrence in reservoir species, particularly viral richness and viral detection frequency, is essential for explaining host-pathogen dynamics and anticipating public health risks. In this study, we evaluated the influence of life-history traits on viral frequency (defined as the proportion of positive detections across studies) in European bats for six important viral groups (Adenoviridae, Herpesviridae, Papillomaviridae, Filoviridae, Coronaviridae, and the genus Ly

Engineering human endogenous retrovirus-K for improving immunogenicity and broad cancer targeting.

BACKGROUND: HERV-K (HML-2) is the most studied transcriptionally active endogenous retrovirus in human cancers, yet immune tolerance and incomplete mechanistic understanding have impeded its clinical use as an immunotherapy target. We hypothesized that rational engineering of the HERV-K envelope putative immunosuppressive domain (ISD) could improve immunogenicity. METHODS: We performed a mutational screen across the ISD region, evaluated NF-κB activation and then analyzed antigen expression in human monocyte-derived dendritic cells (moDCs) using flow cytometry, RNA-seq, and TEM. Human PBMCs w

Safety and immunogenicity of an adenoviral-vectored vaccine against Crimean-Congo haemorrhagic fever in the UK (CCHF01): an open-label, first-in-human, non-randomised, phase 1 clinical trial.

BACKGROUND: There are no approved vaccines for Crimean-Congo haemorrhagic fever virus (CCHFV), which remains a WHO priority pathogen for vaccine development. We evaluated the safety and immunogenicity of ChAdOx2 Crimean-Congo haemorrhagic fever (CCHF), a novel, non-replicating adenoviral-vectored vaccine, in a first-in-human phase 1 clinical trial. METHODS: CCHF01 was an open-label, first-in-human, non-randomised, phase 1 trial conducted at a single centre, the University of Oxford Centre for Clinical Vaccinology and Tropical Medicine in Oxford, UK. Healthy adults aged 18-55 years, with no co

In silico and in vitro immunogenicity risk assessment of process- and product-related protein impurities reported in adenoviral-vectored COVID-19 vaccines.

Process-related protein impurities (PRPIs) such as host cell proteins (HCPs) in biologic products have been associated with unexpected immune responses, adverse events, and the discontinuation of clinical programs. While an HCP threshold of 100 ng/mg (PPM) is considered to be 'acceptable', that threshold may be too high if individual impurities have strong intrinsic immunogenicity or, in contrast, may be overly cautious if many of the HCP impurities have low immunogenicity. In silico methods for predicting the immunogenicity of PRI and HCP may help triage higher risk from lower risk entities.

High Exposure of Intravenous Alemtuzumab is Associated with Increased Risk of Adenovirus reactivation in Patients with Non-Malignant Disease Undergoing Allogeneic HCT.

INTRODUCTION: Alemtuzumab, a recombinant humanized immunoglobulin G1 monoclonal antibody directed against human CD52, is a widely used serotherapy strategy in pediatric allogeneic hematopoietic stem cell transplantation (HCT) in non-malignant diseases (NMD) to help reduce the risk of graft rejection (GR) and graft versus host disease (GVHD). Excessive exposure to alemtuzumab potentially delays immune reconstitution and increases the risk of infections. METHOD: We conducted a retrospective analysis at a single institution of 95 pediatric patients with NMD who underwent HCT and received intrave

Molecular surveillance of adenovirus and coronavirus in Procellariiformes, Brazil: Detection of new divergent adenoviruses.

Adenoviruses are widespread DNA viruses that infect a broad range of vertebrate hosts and are recognized for their long-term host coevolution, remarkable genetic diversity, and broad pathogenic potential. Coronaviruses circulate among wild birds due to their evolutionary potential and are relevant to animal and public health. However, the diversity and ecological role of these viruses in pelagic seabirds remain poorly understood. This study provides the first molecular survey of adenoviruses and coronaviruses in Procellariiformes from the South Atlantic. Fifty stranded individuals representing

Immunogenicity and protective efficacy of single-dose adenoviral vaccines expressing Crimean-Congo haemorrhagic fever virus GP38, Gc, or nucleoprotein antigens in mice: a preclinical study.

BACKGROUND: Crimean-Congo haemorrhagic fever virus (CCHFV) is a highly pathogenic, tick-borne orthonairovirus responsible for severe human disease, yet no approved vaccines or therapeutics exist. The glycoprotein precursor (GPC), including its GP38, Gn, and Gc components, and the nucleoprotein (NP) have been explored as vaccine targets with varying success. A chimpanzee adenoviral vector (ChAdOx) vaccine encoding full-length GPC previously conferred complete protection in mice, supporting Phase I evaluation. However, the essential protective components and mechanisms of vaccine-induced immunit

Intramuscular self-amplifying RNA and adenoviral RSV vaccines program distinct systemic and respiratory mucosal immune architectures.

Respiratory syncytial virus (RSV) remains a major cause of severe lower respiratory tract disease in infants, older adults, and immunocompromised individuals, underscoring the need for vaccine strategies that balance systemic protection with frontline respiratory immunity. Although adenoviral and RNA-based RSV are both promising platforms, how the platform itself shapes systemic versus respiratory mucosal immunity under the same antigenic context remains insufficiently defined. We conducted a head-to-head comparison of an LNP-delivered saRNA vaccine (LNP-RSV-preF saRNA) and a recombinant Ad5-v

Viral metagenomic characterization of HAdV-B-related signals in bronchoalveolar lavage fluid from pediatric and adult clinical cohorts.

Human adenovirus (HAdV) is an important respiratory virus in children, but sequence-level evidence for HAdV-related signals in lower respiratory tract samples remains limited. In this study, we analyzed metagenomic next-generation sequencing data from 189 bronchoalveolar lavage fluid (BALF) libraries from pediatric and adult clinical cohorts, including 30 pediatric and 159 adult patients, together with laboratory environmental controls. MEGAN read-level classification was used to screen for Adenoviridae-related signals. HAdV-related contigs were further reviewed, and a BALF-derived HAdV-B-rela

Polyclonal breast milk-derived secretory IgA confers broad antiviral protection against multiple respiratory viruses.

Passive immunization is an important tool for protecting individuals with primary immune deficiencies from infectious diseases and offers a rapid response strategy for protecting vulnerable populations, particularly during the interval between pathogen emergence and vaccine deployment. Current approaches rely on intravenous delivery of IgG-enriched plasma or monoclonal antibodies, which limits accessibility, scalability, and effectiveness at mucosal surfaces, the primary entry point for respiratory viruses. Here, we tested LCTG-001, a polyclonal secretory IgA preparation derived from pooled hu

Hemagglutinin and fusion proteins precede measles virus ribonucleoproteins and marshal cell-to-cell spread in airway epithelium.

Measles virus (MeV) is the most contagious human respiratory pathogen. To understand how it spreads in the airways, we infected well-differentiated primary cultures of human airway epithelial cells (HAE). We discovered that the MeV attachment protein (hemagglutinin, H), the fusion (F) protein, and the assembly organizer protein (matrix, M) reach newly infected cells ahead of, and independently from, the replicative complex proteins. To characterize the mechanisms of MeV spread, we generated a replication-deficient adenoviral vector expressing the MeV M, F, and H proteins in a doxycycline-induc

Highly neutralizing anti-IFN-ω autoantibodies underlie severe pediatric adenovirus infection.

Neutralizing autoantibodies (auto-Abs) against type I IFNs (IFN-α and IFN-ω) have emerged as major acquired risk factors for severe viral infections. Human adenovirus (HAdV) is highly prevalent in children and can cause severe pneumonia, but the host determinants of disease severity remain poorly understood. We describe here a 5-year-old child with life-threatening HAdV-B7 pneumonia requiring intensive care who carried high-titer anti-IFN-ω auto-Abs of the IgG1 and IgG3 subclasses. Peripheral blood mononuclear cells produced normal amounts of IFN-α and IFN-ω ex vivo; however, IFN-ω was undetec

Layer-by-layer assembly of bioactive nanofilms for in situ gene transfection of a LAMA3 mutant on hydrogenated TiO₂ nanotubes: in vitro evaluation of epithelial adhesion and hemidesmosome formation.

BACKGROUND: Reliable peri-implant epithelial sealing is paramount for the prevention of peri-implantitis. Remarkably, both hemidesmosomes (HDs) and the internal basal lamina, which notably includes laminin332-essential for transmucosal epithelial sealing-are significantly diminished compared to their natural tooth counterparts. In situ gene therapy has emerged as a promising approach for the spatial regulation of HD formation. METHODS: In this study, we aim to develop a bioactive coating with an exceptional capacity for the in situ transfection of a recombinant adenovirus carrying a mutant of

HtpG-High-Expressing Escherichia coli Promotes Fibrosis in Chronic Pancreatitis.

BACKGROUND & AIMS: Chronic pancreatitis (CP) is a progressive fibroinflammatory disease with rising global incidence and no curative therapy. Gut dysbiosis has been associated with CP, but its causal contribution to disease pathogenesis remains unclear. We intended to identify specific pathogenic microbial species that promote CP and to delineate the underlying pathogenic mechanisms. METHODS: Multi-omics analyses were used to identify key microbial signatures in CP patients and mice. Novel pathogenic Escherichia coli strains were isolated and bioengineered for functional studies.
