Evaluation of methods to increase the expression of cytokine-induced killer cell chemoattractant cytokines in pancreatic cancer.

Adoptive Cell Therapies based on cytokine-induced killer cells (CIKs) can address the heterogeneity of solid tumors due to their multiple mechanisms of cancer cell recognition. However, tumor trafficking is one of the main limitations. In this work, we describe that a high proportion of CIKs obtained from pancreatic ductal adenocarcinoma patients express the CXCR3 and CCR5 receptors, and they migrate towards their corresponding chemokines CXCL10 and CCL5 in vitro. Using an immune competent orthotopic PDAC mouse model, we have investigated the ability of different clinically compatible interven

Replication-incompetent viral vaccine vectors ChAdOx1 and MVA as tools for evaluating T-cell responses to naturally processed antigens in vitro.

Assessing T-cell responses is critical for vaccine development. In vitro methods using SARS-CoV-2 or recombinant vaccinia virus with B cells effectively activate T-cells but require stringent biosafety conditions. As an alternative, we explored attenuated, replication-incompetent viral vectors, such as modified vaccinia Ankara (MVA) and chimpanzee adenoviral vectors (ChAdOx1 and ChAdOx2). These vectors successfully transduced B cells, as confirmed by GFP expression. B cells transduced with ChAdOx1 nCoV-19 (encoding SARS-CoV-2 Spike) activated autologous CD8⁺ and CD4⁺ T-cells. Similarly, B cell

Protocol for the injection of adenoviruses into brown adipose tissue in mice.

Brown adipose tissue (BAT) represents a specialized form of adipose tissue that has attracted much attention in the search for therapeutic interventions to treat obesity and its related metabolic disorders. Here, we present a protocol to modulate BAT metabolism in vivo in mice via the direct administration of adenovirus (Ad). We also describe steps for the delivery of postoperative care, including the administration of analgesics and the application of antiseptics to the surgical site to prevent potential infections. For complete details on the use and execution of this protocol, please refer

Laminin N-terminus α31 regulates corneal epithelial cell adhesion and migration through modifying the organization and proteolytic processing of laminin 332.

Laminin N-terminus α31 (LaNt α31) is a netrin-like protein generated by alternative splicing of the laminin α3 gene. While previously shown to regulate vascular permeability in vivo, its role in epithelial tissues remains less defined. Here, we demonstrate that LaNt α31 modulates epithelial cell behavior by altering laminin 332 (LM332) organization and hemidesmosome (HD) maturation. Adenoviral driven overexpression of LaNt α31 in corneal epithelial cells led to premature HD assembly, marked by enhanced recruitment of collagen XVII and BPAG1e to β4 integrin, and reduced cell migration. LaNt α31

Transient adenovirus-Cre infection causes long-lasting remodeling of the mammary gland immune landscape.

Understanding how immune cells respond to early oncogenic events is essential for designing immune-based strategies to intercept breast cancer. Mouse models that induce mammary tumorigenesis through Cre-mediated genetic manipulations can be used to study these early events. However, the immune effects of different induction methods remain unclear. Here, we compare adenovirus-delivered Cre with tamoxifen-inducible CreER systems in models targeting luminal mammary epithelial cells for p53-loss. We find that transient intraductal adenoviral infection produces not only an acute immune response but

Comprehensive surveillance of fowl adenovirus in the Americas reveals the circulation of multiple serotypes and evidence of recombination.

Fowl adenoviruses (FAdVs) comprise five species and twelve serotypes that infect chickens, several of which are associated with disease and significant economic losses in poultry farming. Although they are globally distributed, molecular characterization studies remain limited in many regions. This study aimed to elucidate the frequency and circulation of FAdV serotypes in South America, Central America, and the Caribbean. FAdV detection was conducted using qPCR and hexon gene sequencing on samples from chicken farms in Brazil, Peru, Guyana, Colombia, and El Salvador. In Brazil, FAdV was detec

Molecular detection and pathological evaluation of canine adenovirus 1 in Indian bear species.

In this study, we investigated infectious canine hepatitis (ICH), caused by canine adenovirus 1 (CAdV-1), in 125 bear carcasses submitted for examination. CAdV-1 is known to cause severe disease in domestic dogs and in wild canids and bears. Using molecular and/or histopathological methods, CAdV-1 infection was confirmed in 12 cases: 11 in sloth bears (Melursus ursinus) and one in an Asiatic black bear (Ursus thibetanus). Affected captive bears exhibited acute onset of high fever, jaundice, bloody vomitus, and foul-smelling diarrhoea, followed by death. Necropsy revealed widespread congestion

TGFβ pathway represses hepatic ribosome biogenesis and protein synthesis by regulating p70S6K-S6RP proteins.

BACKGROUND: Transforming growth factor-beta (TGFβ)-superfamily signaling has been implicated in the regulation of hepatocyte growth and regeneration after acute or chronic liver injury. However, the precise mechanisms underlying TGFβ signaling in the distinct hepatic cell types during the progression of liver fibrosis remain largely unknown. We aim to identify the downstream molecular mechanisms of TGFβ-signaling modulation on hepatocytes. METHODS: To modulate TGFβ-superfamily signaling in vivo, Smad3 or Smad7 were adenovirally overexpressed in mouse liver. Parallelly, hepatosphere cultures w

Medical Gas Plasma Inactivates Adenoviruses via Capsid Oxidation.

Pandemics threaten societies, driving the need for new therapeutics. Medical gas plasma, a partially ionized gas, has been proposed as an antiviral agent, but its mechanisms of action are elusive. We employed a certified argon plasma jet and dynamic infection monitoring. Gas plasma exposure reduced adenovirus infectivity in lung cells in treatment time-dependently, with 90s yielding infectivity losses of 96%. The antioxidants N‑acetylcysteine and tyrosine, and the nitric oxide scavenger carboxy‑PTIO, abrogated gas plasma-mediated virus inactivation, implicating hydroxyl radicals and peroxynitr

Establishment and evaluation of the detection method for human adenovirus and its subtypes based on RAA-LFD technology.

In this research, we developed a rapid assay for detecting human adenovirus (HAdV) and its prevalent subtypes (including HAdV3, HAdV4, HAdV7, HAdV14, HAdV11, and HAdV55) based on recombinase-mediated isothermal amplification combined with lateral flow chromatography (RAA-LFD). We designed specific primers and probes targeting conserved regions of the Hexon genes of HAdV and its common subtypes, and established the recombinase-aided amplification (RAA) reaction system. The optimal primer and probe combinations were identified utilizing real-time fluorescence RAA. Subsequently, the RAA-lateral f

Short-Term Change in IgG Antibody Elicited by Omicron BA.5 Infection and Inhaled Ad5-nCoV Vaccine Among Healthcare Workers.

BACKGROUND: Little is known about the immune response induced by the SARS-CoV-2 Omicron BA.5 variants or the inhaled adenovirus vector vaccine. In this study, we investigated the short-term profile of antireceptor-binding-domain IgG antibody responses elicited by either the virus or the vaccine. METHODS: A cohort of healthcare workers who were infected with Omicron BA.5 or who received the inhaled adenovirus vector vaccine was identified between August and December 2022. Blood samples were collected twice to detect IgG antibodies against the receptor-binding domain of the SARS-CoV-2 spike pro

A Tumor Cell Membrane-Coated CD40 Ligand-Encoding Oncolytic Adenovirus Serves as a Potent Nano-Vaccine for Ovarian Cancer Immunotherapy.

Oncolytic virus-based vaccines for advanced ovarian cancer have limitations owing to their receptor-dependent tropism, host immune clearance, and ascites-mediated viral neutralization. To mitigate these drawbacks, a novel nano-vaccine platform was developed by incorporating immunostimulatory oncolytic adenoviruses within tumor cell membranes. This biomimetic system enabled sustained antigen release and tumor-localized delivery of co-stimulatory molecules, creating immunological hotspots through in situ viral replication and PANoptosis. The nano-vaccine achieved selective viral tropism with a 3

Molecular epidemiology and clinical characteristics of human adenovirus in children in Hangzhou (2022-2023).

Human adenovirus (HAdV), a leading cause of pediatric acute respiratory infections (ARIs), exhibited altered transmission patterns during the period of time in which COVID-19 nonpharmaceutical interventions (NPIs) were being applied. In this study, we investigated the molecular and clinical characteristics of pediatric HAdV infections in Hangzhou across two epidemiological phases: the 2022 group (March 2022-February 2023), representing a period of strict NPIs with initial post-policy adjustment, and the 2023 group (March 2023-February 2024), reflecting resumption of normal social activities. HSP90α promotes fowl adenovirus serotype 4 replication in LMH cells via interacting with hexon.

Heat shock protein 90 alpha (HSP90α), a conservative chaperone protein, is closely involved in signal transduction and virus proliferation, but how it engages the lifecycle of fowl adenovirus serotype 4 (FAdV-4) remains unknown. Here, we demonstrated that the expression of HSP90α was significantly upregulated in Leghorn male hepatoma (LMH) cells infected with FAdV-4. Functional study revealed that overexpression of HSP90α promoted FAdV-4 replication in vitro, whereas knockdown of HSP90α exerted the opposite effect. Subsequently, co-immunoprecipitation (Co-IP) assay showed that HSP90α, particul

Stimulation of Human Adenovirus Infection Modulated by Emerging Micropollutants.

Contaminants of emergent concern (CECs), such as pharmaceuticals and microplastics, are increasingly found in aquatic environments, yet their interactions with viral pathogens remain underexplored. This study evaluated the effects of antibiotics, antidepressants, microfibers, and nanoplastics on human adenovirus type 5 (HAdV-5) replication in A549 cells. A series of in vitro assays simulating distinct exposure scenarios across the viral replication cycle was conducted. Results showed that individual pre- or post-infection exposure to CECs did not significantly impact HAdV-5 replication. Howeve
