eIF3m promotes fowl adenovirus serotype 4 replication via interacting with ORF1B protein.

Eukaryotic initiation factors (eIFs), a bunch of proteins that deeply involved in translation initiation of mRNA in eukaryotes, are closely associated with physiological and pathological processes. eIF3m, a core subunit in eIF3 complex, also played critical roles in virus infection. In this study, a co-immunoprecipitation coupled with mass spectrometry (Co-IP/MS) was performed to identify host factors that interacted with ORF1B, a unique non-structural protein of fowl adenovirus serotype 4 (FAdV-4). Among 2502 cellular proteins, eIF3m, especially its C-terminal part, was verified to interact w

Factors influencing Anti-SARS-CoV-2 IgG levels after vaccination in breast cancer patients.

BACKGROUND/OBJECTIVES: Breast cancer patients may exhibit impaired vaccine-induced immunity due to disease progression and treatment-related immunosuppression. This study examined clinical factors associated with anti-SARS-CoV-2 IgG responses post-COVID-19 vaccination, with a focus on disease stage and treatment timing. METHODS: We analyzed 63 breast cancer patients vaccinated between 2021 and 2022. Clinical data—including tumor size, stage, lymph node involvement, estrogen receptor (ER) status, and treatments (chemotherapy, radiation)—were collected. Serum anti-SARS-CoV-2 IgG levels were asse

Fowl adenovirus infections: A comprehensive review of prevalence, pathogenesis, diagnosis, control, and economic impact.

Fowl adenoviruses (FAdVs), used to be recognized as non-malignant pathogens, have now evolved into notable pathogens threatening global poultry productions. This review synthesizes current knowledge on the escalating challenges posed by FAdVs. There is already a wide variety of genetic types or strains within the virus population of the genus Aviadenovirus, FAdVs, where distinct serotypes are already present as the causative agent of Inclusion Body Hepatitis (IBH) and Hydropericardium Hepatitis Syndrome (HHS). These viruses are widely distributed throughout the globe, hypervirulent strains are

Advances in our understanding of anti-PF4 related immunothrombosis.

This article focuses on the central role of antibodies against platelet factor 4 (PF4) in mediating immunothrombosis, from classical heparin-induced thrombocytopenia (HIT) to vaccine-induced immune thrombocytopenia and thrombosis (VITT). The latter condition gained international attention during the rollout of vaccines against SARS-CoV-2. Since then, an increased awareness for anti-PF4 mediated disorders arose and patients were recognized with anti-PF4 disorders occurring without prior heparin or adenoviral vector vaccine exposure. These disorders include various acute and chronic VITT-like co

Mucosal immunization with an adenoviral vector expressing a prefusogenic F protein protects the upper and lower respiratory tracts of cotton rats against live respiratory syncytial virus challenge.

Respiratory syncytial virus (RSV) is a major cause of severe respiratory disease in infants, young children, and the elderly. Current licensed prefusogenic F-based RSV vaccines induce systemic immune responses through intramuscular injection. However, mucosal immunization will be required to elicit local sterilizing immunity to prevent virus replication in the nasopharynx and the lungs as well as virus shedding and transmission. Adenovirus vectors are a promising platform to develop RSV vaccines, but further evaluation of mucosal adenovirus-based prefusion F (preF) vaccines is still needed. In

A recombinant adenoviral vector vaccine expressing the ORF2 capsid protein confers robust protection against chicken astrovirus infection.

Chicken astrovirus (CAstV), which causes enteritis, nephritis, and growth retardation syndrome, including white chicken syndrome (WCS), represents a significant global economic burden, with no available vaccine. Its fecal-oral transmission route underscores the need for mucosal immunity at intestinal entry sites, while human adenovirus type 5 (HAdV-5) vectors are highly efficient at targeting mucosal tissues. To develop and evaluate a novel recombinant adenovirus-vectored vaccine against CAstV, we used the HAdV-5 vector (AdMax system) to construct rAd5-CAstV-ORF2, which expresses the immunodom

Multivalent fowl Adenovirus-Newcastle disease vaccine: comprehensive evaluation in SPF and commercial broiler breeders.

BACKGROUND: Inclusion body hepatitis (IBH) and Newcastle disease (ND) impose substantial economic burdens on global poultry production, with combined annual losses exceeding $3 billion worldwide. Current vaccination strategies requiring separate immunizations increase handling stress and operational costs. OBJECTIVE: To evaluate immunogenicity, cross-neutralization patterns, protective efficacy against virulent challenge, maternal antibody transfer, and commercial field performance of a novel multivalent inactivated vaccine containing fowl adenovirus (FAdV) serotypes 2, 8a, 8b, and 11 combine

A replication-defective bivalent adenovirus-vectored vaccine provides robust and durable protection against both canine distemper virus and canine parvovirus.

Canine distemper virus (CDV) and canine parvovirus (CPV) are highly contagious and often fatal pathogens in dogs. Current live-attenuated vaccines, while effective, carry the risk of virulence reversion. This study aimed to develop a safer, replication-defective bivalent vaccine using an adenovirus vector to simultaneously protect against both CDV and CPV. The recombinant vaccine, designated Ad5-(VP2 +H), was engineered to coexpress the CDV hemagglutinin (H) protein (CDV-H) and the CPV Viral Protein 2 (CPV-VP2). Its immunogenicity and protective efficacy were evaluated in mouse and canine mode

Intravenous infusion of engineered megakaryocytes to produce oncolytic platelets in vivo for enhanced cancer immunotherapy.

Tumor-targeted delivery of oncolytic viruses (OVs) via systemic administration could not only expand virotherapy beyond primary tumors to widespread metastases, but also improve clinical adherence and convenience. We here engineer megakaryocytes encapsulating oncolytic adenovirus type 5 (M-Ad5) to produce oncolytic platelets in vivo by leveraging the thrombopoiesis process. Upon intravenous administration, M-Ad5 travels through the lungs, where it can release OVs-harbored therapeutic platelets into circulation under pulmonary turbulence microenvironments. Under the shelter of platelets, OVs re

Adenoviral Inciting Antigen and Somatic Hypermutation in VITT.

BACKGROUND: Vaccine-induced immune thrombocytopenia and thrombosis (VITT) is a rare prothrombotic complication that occurs after adenoviral vector-based vaccination against coronavirus disease 2019; in rare cases, it can also occur after natural adenovirus infection. VITT is mediated by platelet-activating antibodies against the highly cationic protein platelet factor 4 (PF4). The underlying inciting antigen trigger and immunopathogenesis remain unknown. METHODS: We used antibody proteomics to determine the amino acid sequences of anti-PF4 antibodies from 21 patients with VITT and sequenced t

Novel, Stable, and Well-Secreted Single-Chain Fc Forms of Programmed Death-1 Immune Checkpoint Inhibitor Efficiently Block PD-1/PD-L1 Interaction.

Immune checkpoint inhibitors (ICI), such as antibodies against Programmed Death 1 (PD-1), are widely used to treat different types of cancer. Unfortunately, only a small subset of patients benefits from such treatments, and the systemic administration of ICIs can cause severe immune-related adverse events. To overcome these hurdles, we performed an in vitro proof-of-concept investigation of recombinant adenoviral vectors encoding different variants of secreted PD-1 (sPD-1), differing in size, cysteine mutations, oligomerization, and effector functions. First, eight sPD-1 variants were screened

Desmoglein 2 (DSG2)-knockout human respiratory epithelial cell model to study species B adenovirus receptor usage.

With an increasing number of human adenoviruses identified, the selection of potential therapeutic vectors broadens. For safety reasons, achieving cell-specific gene delivery is crucial to minimize off-target effects. Therefore, it is essential to gain a systematic understanding of adenovirus receptor-usage. Our aim is to establish a human-originated in vitro model for comparative analysis of human adenoviruses receptor usage. Based on our previous work of human CD46 and coxsackievirus and adenovirus receptor (CAR) knockout cell lines, we generated desmoglein 2 (DSG2) knockout cell lines using

Netrin-5 Preserves Blood-Brain Barrier Integrity via Wnt3a/β-Catenin Pathway Activation in Murine Cerebral Ischemia.

Blood-brain barrier compromise represents a pivotal pathological mechanism in ischemic stroke, driving neurological deterioration. Netrin-5, an axon guidance protein family member, demonstrates regulatory potential for BBB integrity. Employing middle cerebral artery occlusion (MCAO) mice and oxygen-glucose deprivation/reperfusion (OGD/R) in human brain microvascular endothelial cells (HBMVECs), we found that Netrin-5 was significantly downregulated in the murine cortex post-MCAO and was also downregulated in HBMVECs upon OGD/R exposure. Adenoviral Netrin-5 delivery in MCAO mice attenuated cere

Pharmacological Vitamin C Plus Oncolytic Adenoviruses Orchestrate Immunogenic Tumor Ferroptosis.

High-dose Vitamin C (VitC) substantially boosts the anti-tumor effect of oncolytic adenoviruses (oAds), but optimizing its therapeutic potential remains to be fully explored. This study aims to investigate the synergistic effects of VitC and oAds on tumor cell viability and the underlying mechanisms. The CCK-8 assay and Flow cytometry were employed to detect the viability and apoptosis of tumor cells treated with VitC, oAds and VitC plus oAds. The combination therapy increased the oncolytic effect by 25-fold in CT26 cells and 10-fold in 4T1 cells, highlighting that VitC could enhance the oncol

Adenovirus, herpesvirus, and parvovirus survey in Iberian wolves (Canis lupus signatus) of Cantabria, Spain.

The study of infectious diseases in wild carnivores is essential to their conservation. Adenovirus, herpesvirus and parvovirus are DNA viruses able to cause severe disease in carnivores; however, data on their occurrence in Iberian wolves (Canis lupus signatus) are limited. Herein, 130 spleen samples from Iberian wolves in Cantabria, northern Spain, were tested for these viruses using broad-range nested PCRs and real time PCR. All positive amplicons were confirmed by direct Sanger sequencing in both directions. Additionally, exposure to adenovirus and Carnivore protoparvovirus 1 (CCPV1) was ev

High-capacity adenoviral vector-mediated expression of an LDLR/transferrin chimeric protein in muscle reduces atherosclerosis in Ldlr-/- mice.

Familial hypercholesterolemia (FH) is a genetic disorder caused by mutations in the low-density lipoprotein (LDL) receptor, leading to impaired uptake of LDL and its accumulation in arterial walls and other tissues. This accumulation results in cardiovascular disease and early mortality. Treatments, including statins, ezetimibe, PCSK9 inhibitors, and bempedoic acid, are often insufficient in homozygous FH patients, particularly those with null mutations in the LDL receptor (LDLR). To address this unmet need, we have developed two helper-dependent adenoviral (HD-Ad) vectors for the expression o

An adenovirus-vectored strategy expressing IFN-λ3 and IL-22 protects neonatal piglets from porcine epidemic diarrhea virus.

Porcine epidemic diarrhea virus (PEDV) causes devastating enteric disease in neonatal piglets, often resulting in nearly 100 % mortality. Frequent genetic variation further undermines the effectiveness of existing vaccines and control strategies, highlighting the urgent need for novel antiviral interventions. In this study, we developed a recombinant adenovirus-vectored strategy (rAd5-IFNλ3-IL22) co-expressing interferon lambda 3 (IFN-λ3) and interleukin-22 (IL-22) to concurrently target viral replication and attenuate epithelial injury. In vitro, rAd5-IFNλ3-IL22 expressed and secreted IFN-λ3
