Cytomegalovirus (CMV) Central Nervous System Infection and Recurrent Posterior Circulation Ischemic Strokes in an Immunocompetent Patient.

Cytomegalovirus (CMV) infection in an immunocompetent adult is rarely symptomatic. Neurological manifestations are uncommon and are predominantly characterized by meningitis, myelitis, and acute polyradiculoneuritis. We report a rare case of an immunocompetent patient who presented with prolonged headaches complicated by recurrent posterior circulation ischemic strokes associated with intracranial stenoses of the basilar trunk and posterior cerebral arteries. Analysis of his cerebrospinal fluid (CSF) revealed lymphocytic meningitis with moderate elevation of the CSF protein level, normal CSF g

Echovirus 19 Molecular Epidemiology in Russia and Some Central Asian Countries in 1999-2024.

Echovirus 19 (E19) is a rarely reported non-polio enterovirus (NPEV), although it has been associated with a spectrum of severe syndromes, including encephalitis, meningitis, and multisystem hemorrhagic disease in infants. Moreover, Echovirus 19 has been detected in patients with acute flaccid paralysis (AFP), suggesting a potential association between viral infection and CNS damage. Here we describe molecular epidemiology of Echovirus 19 in Russia and Central Asia from 1999 to 2024 using data on poliomyelitis surveillance. Echovirus 19 accounted for no more than 4% of annual NPEV isolates, ho

Enterovirus 2B protein: From viroporin to membrane stress sensor, mechanistic principles of multi-pathway regulation.

Enterovirus 2B is a highly conserved, functionally versatile, nonstructural protein. The classic viroporin model, which restricts its role to ion channel formation, cannot account for its interactions with host factors such as Bcl-2-associated X protein, interleukin enhancer-binding factor 2, and karyopherin subunit alpha 1, nor its functional divergence among enteroviruses. We propose a novel, testable membrane stress sensor hypothesis: the transmembrane helices of 2B sense lipid composition and pH changes, and through conformational dynamics, spatiotemporally coordinate calcium signaling, ce

Post-pandemic rebound of rotavirus and adenovirus in pediatric gastroenteritis: a 6-year antigen-based surveillance study from Turkey.

This report presents a 6-year antigen-based surveillance of pediatric viral gastroenteritis in Turkey, revealing substantial epidemiological shifts during and after the coronavirus disease 2019 (COVID-19) pandemic. Adenovirus circulation was strongly affected by COVID-19-related public health restrictions, showing a marked decline during restriction periods. However, rotavirus exhibited a relative increase in 2021 despite ongoing restrictions, followed by a more pronounced rise after the relaxation of measures, aligning with the "immunity debt" hypothesis associated with altered early-life vir

Molecular Characterization of Human Adenoviruses Before and After a Severe HAdV-7 Military Outbreak in Finland in 2024.

BACKGROUND: In 2024, a severe outbreak caused by human adenovirus type 7d (HAdV-7) occurred in Finnish military garrisons. Molecular typing was implemented to support outbreak control. The aim of this study was to determine adenovirus types circulating in Finnish garrisons during 2013-2023 by retrospective surveillance and to review earlier findings to better understand factors contributing to the 2024 outbreak. METHODS: Adenovirus-positive respiratory samples collected through routine surveillance (2013-2023) and enhanced surveillance (2024-2025) were typed using type-specific PCR assays tar

Detection of intranuclear inclusion bodies in hepatocytes of the poultry infected with fowl adenoviruses (Adenoviridae: Aviadenovirus).

INTRODUCTION: Fowl adenoviruses (FAdV) are infectious pathogens causing significant economic losses in poultry industry. Molecular, pathoanatomic, and pathomorphological methods are used for laboratory diagnosis of FAdV infection. The clinical picture and postmortem lesions are not sufficient to diagnose, unlike histological analysis. Histological examination reveals basophilic intranuclear inclusion bodies considered a pathognomonic sign of FAdV infection. Despite the informative value of the histological method, it has a limitation - the long duration of the procedure, which ranges from 5 to

Eugenol attenuates ferroptosis, mitochondrial damage, and inflammation in cardiac hypertrophy via the 14-3-3γ/AMPKα2 pathway.

BACKGROUND: Chronic stress-induced cardiac hypertrophy is often accompanied by energy metabolism within the mitochondria, iron homeostasis, and inflammation. Eugenol (Eug), as an antioxidant, scavenges reactive oxygen species (ROS) in the body and stabilizes the internal cellular environment. However, the mechanism of this process has not been fully elucidated in existing studies. METHODS: In this experiment, hypertrophy of H9c2 cells was induced by angiotensin II (Ang Ⅱ), and cardiac hypertrophy was induced by transverse aortic constriction (TAC) in Sprague-Dawley (SD) rats. Subsequently, th

Generation of DFNA5-deficient chickens reveals a species-specific role of pyroptosis in avian antiviral immunity.

The global burden of viral diseases in both humans and agricultural animals is exacerbated by dysregulated inflammatory cell death and cytokine storms. Pyroptosis, executed by gasdermin family proteins, is a key driver of this pathology, yet its functional conservation and therapeutic targeting in avian species remain critically unexplored. Here, we establish the first germline-edited heterozygous DFNA5-mutant chicken model using primordial germ cell mediated genome editing. We demonstrate that genetic disruption of DFNA5 markedly attenuates Raptinal-induced pyroptosis and reduces IL-1β and IL

Occurrence of Sensorineural Hearing Loss in Viral Meningitis and Encephalitis.

Hearing loss in viral neuroinfections is poorly investigated. The present study aimed to clarify if hearing loss can develop in viral meningitis and encephalitis. An OAE (oto-acoustic emission) was measured in adult patients with viral meningitis and encephalitis on the day of admission and 30-60 days after discharge. Frequencies were categorized as Low (1, 1.5, 2 kHz), Mid (3, 4, 5 kHz), Mid-high (6, 7, 8 kHz), and High (9, 10 kHz). An OAE was measured in 41 patients with viral meningitis and 8 patients with encephalitis. Six patients with viral meningitis (15%) experienced a decrease of > 5

UNC5A IgG in a Previously Reported CASPR2/LGI1-Negative VGKC-Associated Peripheral Nerve Hyperexcitability Syndrome: Long-Term Clinical and Electrophysiological Follow-Up.

Autoimmune peripheral nerve hyperexcitability (PNH) syndromes are most commonly associated with antibodies against CASPR2 or LGI1. However, a subset of patients with clinically and electrophysiologically confirmed PNH remains negative for these established neuronal surface antibodies despite persistent VGKC antibody positivity. This report presents the long-term follow-up of a patient previously diagnosed with a Morvan-like autoimmune PNH syndrome following enteroviral infection. Serial neurophysiological studies demonstrated persistent motor axonal hyperexcitability, while treatment with intr

Application of procalcitonin in pediatric emergency and critical care settings.

This expert consensus on the clinical application of procalcitonin (PCT) in pediatric emergency and critical care was convened by the Taiwan Society of Pediatric Emergency Medicine (TSPEM) and formulated by a committee of 14 leading experts. In pediatric emergency and critical care medicine, distinguishing between bacterial and viral etiologies remains a significant challenge, as clinical symptoms often lack specificity. While traditional biomarkers such as white blood cell count (WBC) and Creactive protein (CRP) provide reference values, they possess limitations regarding diagnostic timelines

Oncolytic virus encoding decorin and the CD40 ligand boosts T-cell response and achieves a durable antitumor response during breast cancer treatment.

Triple-negative breast cancer (TNBC) remains an aggressive disease with limited treatment options. Oncolytic adenoviruses represent a promising platform, but their efficacy as single agents is often insufficient. This study examined whether an oncolytic adenovirus co-delivering decorin (DCN) and CD40 ligand (CD40L), designated rAd.DCN.CD40L, could enhance antitumor activity and immune activation against TNBC. Recombinant adenoviruses expressing DCN (rAd.DCN), CD40L (rAd.CD40L), or both proteins were constructed and evaluated in breast cancer cells using cell viability, apoptosis, migration, in

The Golgi-resident protein ACBD3 facilitates the interaction between enteroviral 3A proteins and Exportin-1.

Enteroviruses are among the most common viral pathogens in humans, causing illnesses ranging from the common cold to severe and sometimes life-threatening diseases such as acute haemorrhagic conjunctivitis or poliomyelitis. To replicate efficiently within host cells, enteroviruses hijack several host factors, including the lipid kinase PI4KB and the adaptor protein ACBD3. During infection, ACBD3 is recruited to viral replication sites through a direct interaction between its C-terminal GOLD domain and viral non-structural 3A proteins. Here, we identify exportin 1 (XPO1) as a host factor involv

In vitro inhibition of STAT5 signaling limits CD8⁺ T cell exhaustion and improves outcomes in adoptive T-cell therapy.

T cell exhaustion remains a barrier to adoptive cell therapies. Although γc cytokines are used to expand T cells, their impact on T cell states remains incompletely defined. Using an in vitro model of repetitive TCR stimulation, we found that high-dose IL-2 or IL-15 promoted exhausted CD8⁺ T cell (TEX) differentiation, with increased inhibitory receptor expression and loss of stem-like features. During chronic LCMV Clone 13 infection, high IL2Rβ expression marked virus-specific CD8⁺ T cells with features of terminally exhausted TEX cells, whereas IL2Rβ deficiency favored progenitor-like popula

Going with the flow: peptide affinity membranes for high-throughput, high-yield purification of adenovirus and lentiviral vectors.

Membrane chromatography offers a compelling alternative to packed-bed resins for viral vector purification, as its open porosity and convective mass transport eliminate diffusive mass transfer limitations, enabling processing at substantially shorter residence times. Yet, the translation of affinity ligands from resins to membranes is challenged by the low density of reactive groups, which constrains functional ligand loading and limits binding capacity. Seeking to overcome this limitation, this study presents a method for functionalizing cellulose membranes with peptide affinity ligands and d

A multi-antigen chimeric PvCSP-MSP119 vaccine induces robust humoral responses against distinct stages of Plasmodium vivax.

Plasmodium vivax is the leading cause of malaria outside Africa, highlighting the need for an effective vaccine. However, the development of vaccines against this parasite remains challenging because of its complex life cycle and antigenic diversity. In this study, we evaluated vaccine formulations targeting both the pre-erythrocytic and erythrocytic stages of P. vivax using either individual antigens or a novel chimeric protein. The chimeric construct combined the C-terminal region and variant repeat domains of the P. vivax circumsporozoite protein (PvCSP) with the 19-kDa C-terminal fragment
