A guanosine metabolism-bioenergetics intersection drives Parkinson's disease.

Several Parkinson's Disease (PD) linked mutations are known to drive deficits in a pathway that relies on an adequate supply of guanosine nucleotide triphosphate (GTP) to drive cellular neuronal processes. Similarly, there is strong evidence that a deficit in bioenergetic support for neuron function is also a major genetic driver of PD. We show here that the reliance on these two purine-based metabolites intersect at another PD susceptibility gene that encodes nucleoside diphosphate kinase (NDK) which converts ATP into GTP. We show that overexpression of NDK is strongly protective both in-vivo

Gut community-level analysis reveals an altered balance between Phocaeicola vulgatus and Bacteroides fragilis in Alzheimer's disease.

Gut microbiome differences in Alzheimer's disease (AD) are typically cataloged taxon by taxon, yet bacterial competition and cross-feeding make species' roles dependent on the entire community. We analyzed 274 stool metagenomes from 119 older adults (18 with AD) as communities, retaining 22 recurring across 1,000 runs. Using our AI framework, we identified 15 species differing in abundance in AD, particularly the commensal Phocaeicola vulgatus (Cohen's d -0.91, 95% CI [-1.23, -0.59]), a finding robust to repeated sampling. It correlated negatively with its sister species, Phocaeicola dorei ( r

Integrated multi-omic analysis of pediatric metastatic osteosarcoma reveals endothelial cell plasticity and lineage infidelity.

While recent research has increasingly focused on the role of fibroblasts and macrophages in osteosarcoma, the tumor vasculature remains poorly understood, particularly in metastatic disease. To address this gap, we performed single-nuclei multi-ome (RNA+ATAC) sequencing on 24 human metastatic osteosarcoma specimens. We found that endothelial cells adopt a hybrid endothelial-mesenchymal state resembling endothelial-to-mesenchymal transition (EndMT) and that a subset of diploid endothelial cells expresses osteoblastic transcriptional profiles and gene regulatory networks (GRN). Joint copy-numbe

Structural basis for the selective inhibition of the PI3KC3-C2 complex by Rubicon in endolysosome maturation and mitophagy.

UNLABELLED: Rubicon is a negative regulator of autophagy and the endolysosomal network (ELN) and an antagonist of the class III phosphatidylinositol 3-kinase complex II (PI3KC3-C2). Inhibition of Rubicon is considered a potential means to therapeutically upregulate autophagy and the ELN to treat Parkinson's disease and other conditions characterized by autophagic and ELN dysfunction. Rubicon is specific for the UVRAG-containing PI3KC3-C2 over the purely autophagic ATG14- containing PI3KC3-C1 complex. Here, we determined the high-resolution cryo-electron microscopy structure of PI3KC3-C2 in com

Metabolomic, lipidomic, and N-glycomic analyses of a human cell model of Krabbe disease reveal treatable deficits in glycosylation and serine-ceramide metabolism.

Krabbe disease is a rare autosomal recessive lysosomal disease caused by deficiency of galactocerebrosidase (GALC), leading to accumulation of galactosylceramide and formation of the toxic metabolite galactosylsphingosine (psychosine). While psychosine accumulation is well-established as a primary pathogenic mechanism, the broader metabolic consequences of GALC deficiency remain incompletely understood. In this study, we used stable isotope tracing to comprehensively characterize metabolic perturbations in a human oligodendrocellular Krabbe disease model. This approach revealed elevated de nov

The Non-Human Primate Sample Size Calculator: a design tool for longitudinal research projects.

Nonhuman primates (NHP) are crucial models of human health and disease and offer the opportunity to bridge the gap between basic research and the clinic. Given the ethical considerations, high costs, and logistical constraints associated with NHP studies, careful planning is essential to maximize scientific rigor while minimizing animal use. Statistical power calculations, to determine optimal sample size, require knowing the expected number of animals at each timepoint, a challenge for longitudinal studies that must account for natural deaths or disease during a study. Here, we leveraged data

Cellular senescence is associated with age-related loss of liver zonation and hepatocyte function.

The liver is organized into tightly regulated zones with distinct metabolic functions but zonation erodes with age. Cellular senescence contributes to aging and liver diseases, however, its impact on aging biology is ill-defined. As part of The Cellular Senescence Network Consortium, we used multiple spatial transcriptomics approaches (GeoMx, Visium, CosMx) with snRNA-seq to profile senescence signatures, zonation markers, and metabolic pathways in livers from wild-type (WT) mice of multiple ages. We observed a loss of canonical zone signatures in aged mouse livers characterized by "expansion"

A clinical care intensity atlas of 505 diseases from 90 million people.

No single population-derived measure ranks the entire diagnosed phenome by clinical care intensity on one scale. From insurance claims contributed by 90 million US enrollees, we derived a care-intensity score that ranks 505 diseases by one uniform scoring rule applied identically to every diagnosis, with no disease-specific clinical input. It tracks Global Burden of Disease disability weights (Spearman ρ = 0.53, n = 130), a pharmacy-only signal recovers much the same order (ρ = 0.71), and a related utilization summary predicts one-year in-hospital death close to a validated comorbidity index.

Asymmetric dimethylarginine (ADMA) modulates frequency-dependent muscarinic antagonism of β-adrenergic regulation of rat papillary muscle contractility.

Accumulating evidence implicates the endogenous nitric oxide synthase inhibitor asymmetric dimethylarginine (ADMA) in cardiovascular impairments. Although the detrimental cardiovascular effects of ADMA have largely been attributed to endothelial dysfunction, recent studies suggest that ADMA also directly affects cardiomyocyte function. However, the action of ADMA on myocardial contractility remains poorly understood. Here, we tested the hypothesis that continuous exposure to 10 µM exogenous ADMA alters the contractile responsiveness of rat right ventricular papillary muscles to sympathetic and

Vascularized Brain Organoid: A Versatile Platform Models Brain Cancer and Traumatic Brain Injury.

Human iPSC-derived brain organoids are revolutionizing tools to study layers biology, synergize disease modeling, and accelerate therapeutic discoveries that overcome obstacles in monolayer cell culture or animal models. The neurovascular unit including vasculature and microglia is critical for brain development, maintenance of synaptic plasticity and neural activity, and the high metabolic demands of long-term culture. We present a methodology to incorporate these important components during organoid generation and discuss potential approach, aiming consistent production of vascularized organ

Tandem repeat expansions in DAPK1 , ANK3 , and RPL14 are associated with diverse neurodegenerative diseases.

Tandem repeat expansions (TREs) cause over 50 neurological conditions, yet their contribution to neurodegenerative disease risk at a population scale remains incompletely characterized. We performed a TRE association study across 6,539 short tandem repeat loci in 276,411 individuals from the UK Biobank and 44,370 individuals from the All of Us Research Program, using two composite neurodegenerative phenotypes to increase statistical power and capture pleiotropic effects. Meta-analysis across the two cohorts identified associations at eight established pathogenic TRE loci, including C9orf72 , D

Improved diagnostic identification of urothelial carcinoma through solid-state nanopore determination of urinary hyaluronan size distribution.

Urothelial carcinoma (UC) is among the most common malignancies worldwide and is known to exhibit a high recurrence rate. The relative lack of validated, non-invasive biomarkers for the disease challenges early detection and negatively impacts patient outcomes. The linear polysaccharide hyaluronan (HA) has been recognized as a potential source of diagnostic information for UC, with its urinary concentration shown to be predictive of disease severity. Here, we use solid-state nanopore (SSNP) sensing to investigate the value of urinary HA size distribution as an independent and complementary pre

NeuroMorph: A Unified Morphological Reference Space for Cross-Disease Brain Profiling.

Structural MRI is routinely acquired in clinical practice, yet quantitative morphometry has had limited impact on clinical decision-making. Overlapping symptoms, trajectories and comorbidities remain difficult to interpret within disease-specific frameworks, leaving it unclear how individual patients relate to the broader organization of brain disease. Here, we construct a cross-disease morphological reference space from 110,591 T1w MRI scans of 78,794 participants, spanning four disease families, 19 diagnoses, and seven subtypes. To construct this space, we developed NeuroMorph, an AI framewo

One Health tick bite exposure biomarkers against Ixodes ricinus salivary peptides.

The changing distributions and activity patterns of tick populations underscore the urgent need for enhanced surveillance and risk assessment strategies for ticks and tick-borne diseases (TBDs) affecting humans and animals. Biomarkers of exposure to tick salivary peptides can be used to identify tick bite history and to monitor tick species distributions. Here, we derived peptides from previously identified immunogenic proteins, namely IrCalreticulin, IrLip1, and IrSPI. Bioinformatic predictive analysis of 3D structure and computed docking strength were used to identify candidate peptides base

The Urine Ammonium-pH Index is a Measure of Kidney Tubular Function.

Dysfunction of the tubulointerstitial compartment is a key driver of chronic kidney disease (CKD) progression. However, tubular function remains largely unaddressed by routine clinical assessment. Here, we show that the urine ammonium-pH index (uAPI), a composite of urinary ammonium and pH, reflects kidney tubular function and predicts kidney function decline. Using acid/base, dietary, and potassium perturbations, segmental disruption of tubular ammonium handling, mathematical modelling, and data from patients with renal tubular acidosis, we identified defective ammoniagenesis as the main uAPI

A small molecule inhibitor of CD28 costimulation restrains pathogenic T-cell responses in inflammatory bowel disease.

UNLABELLED: CD28 costimulation contributes to pathogenic T cell responses in inflammatory bowel disease (IBD), but current B7-directed blockade also limits CTLA-4 signaling. Using a sensitive NanoBiT split-luciferase screening platform, we identified and optimized CA-23, a small molecule antagonist that directly binds human and mouse CD28 without measurable binding to CD80, CD86, or CTLA-4. CA-23 inhibited CD28-B7 engagement and CD28-dependent T cell activation without agonist activity in human whole blood and peripheral blood mononuclear cells. CA-23 achieved exposure in the colon and mesente

Off-Target MAP2K7 Inhibition Drives the Antileukemic Activity of the PD-1/PD-L1 inhibitor ACS-69 in Pediatric T-ALL.

T-cell acute lymphoblastic leukemia (T-ALL) remains a high-risk pediatric malignancy characterized by aggressive disease behavior and poor outcomes after relapse, underscoring the need for novel targeted therapies. Our group reported that aberrant activation of the MAP2K7-JNK signaling axis is implicated in T-ALL pathogenesis, promoting leukemic cell survival, proliferation, and treatment resistance. In this study, we evaluated the anti-leukemic properties of the compound ASC69 in T-ALL cell lines and patient-derived xenograft cells using cell viability, apoptosis, and cell cycle analyses. Pat

Fractal-fractional model for analysis of tuberculosis infection using Ethiopia incidence data.

In this study, a fractional-order S E I u I T R model was proposed to examine the transmission of tuberculosis (TB) in the community. The proposed model was rigorously examined for well-posedness. The basic reproduction number was also derived, and the disease-free equilibrium was obtained. The local and global stabilities of the proposed model were also examined. The proposed model was fitted with the actual TB incidence in Ethiopia, while numerical simulations were carried out in MATLAB to validate the results obtained in this study. Local and global sensitivity analyses were carried out

Validated Synthetic Patient Generation for Small Longitudinal Cohorts: Coagulation Dynamics Across Pregnancy.

Small longitudinal cohorts, common in maternal health, rare diseases, and early-phase trials, limit computational modeling because enrollment is slow and the data are too sparse to train reliable models. We present multiplicity-weighted Stochastic Attention (SA), a generative framework based on modern Hopfield networks. Stochastic attention stores real patient profiles as memory patterns in a continuous energy landscape. The resulting distribution is a finite mixture with one component centered on each stored profile. We generated the reported cohort with Langevin dynamics; direct sampling fro
