Broad-spectrum molecular detection of zoonotic agents in wild micromammals: a nationwide surveillance study in the Republic of Korea.

BACKGROUND: Wild micromammals serve as key hosts for diverse zoonotic pathogens, maintaining these agents in natural ecosystems and facilitating potential spillover to humans. Assessing pathogen prevalence in micromammals is essential for evaluating zoonotic risk and informing public health interventions. METHODS: In this study, we applied the TaqMan Array Card (TAC) platform, a high-throughput molecular diagnostic tool, to micromammal-borne zoonotic pathogen surveillance. From October to November 2022, a total of 160 wild micromammals, including Apodemus agrarius (Striped field mouse) (90.6,

In vitro inhibition of STAT5 signaling limits CD8⁺ T cell exhaustion and improves outcomes in adoptive T-cell therapy.

T cell exhaustion remains a barrier to adoptive cell therapies. Although γc cytokines are used to expand T cells, their impact on T cell states remains incompletely defined. Using an in vitro model of repetitive TCR stimulation, we found that high-dose IL-2 or IL-15 promoted exhausted CD8⁺ T cell (TEX) differentiation, with increased inhibitory receptor expression and loss of stem-like features. During chronic LCMV Clone 13 infection, high IL2Rβ expression marked virus-specific CD8⁺ T cells with features of terminally exhausted TEX cells, whereas IL2Rβ deficiency favored progenitor-like popula

Investigating CD8 T Cell Exhaustion in Chronic LCMV Infection by Flow Cytometry.

CD8 T cells and the process of T cell exhaustion are central topics in both basic immunology and translational research, including clinical applications. Here, we provide a comprehensive, step-by-step guide to investigate CD8 T cells in murine in vivo lymphocytic choriomeningitis virus (LCMV) infection models. We outline viral stock preparation, infection, and viral titer evaluation. We put an emphasis on flow cytometry as a method to investigate phenotypical and functional characteristics of CD8 T cell exhaustion, but the protocols provided will also facilitate imaging or transcriptomic analy

Advances in neoadjuvant therapy for locally advanced head and neck squamous cell carcinoma (Review).

PURPOSE: Identifying factors that affect recovery or restoration of neurological function is a key goal of rehabilitation in neurology and ophthalmology. One such factor can be prolonged mental stress, which may be not only the consequence of nervous system damage but also a major risk factor, or cause, of neural inactivation. Using the visual system as a model of neural injury, we wished to study how patients' stress and personality profiles correlate with vision recovery as induced by therapy with alternating current stimulation (ACS) in patients with optic nerve damage. METHODS: Personalit

Upregulation of TNFR2 Precedes TOX Expression by Exhausted T cells and Restricts Antitumor and Antiviral Immunity.

PURPOSE: Exhaustion represents a collection of programmed T-cell differentiation states and an important mode of T-cell dysfunction. T-cell progression from progenitor to terminal exhaustion is associated with upregulation of the transcription factor thymocyte selection-associated HMG box (TOX). Our understanding of factors regulating TOX expression and the transition from progenitor to terminal exhaustion, however, remains incomplete. EXPERIMENTAL DESIGN: Single-cell RNA sequencing was used to evaluate the expression of TNF receptors on human and murine tumor-infiltrating CD8+ T cells. Flow

Metabolomic profiling reveals the potential of fatty acids as regulators of exhausted CD8 T cells during chronic viral infection.

Chronic infections induce CD8 T cell exhaustion, marked by impaired effector function. While intrinsic drivers are well studied, the role of the surrounding metabolic environment in shaping exhausted CD8 T cells (Tex) is less understood. Using untargeted metabolomics and the murine lymphocytic choriomeningitis virus infection model, we investigated systemic metabolite changes following acute vs. chronic viral infections. We identified distinct short-term and persistent metabolite shifts, with the most significant differences occurring transiently during the early phase of the sustained infecti

Efficiency of Lentiviral Vectors Pseudotyped with LCMV-G in Gene Transfer to Ldlr-/-ApoB100/100 Mice.

Background/Objectives: Lentiviral vectors (LVs) are most commonly pseudotyped with the vesicular stomatitis virus glycoprotein (VSV-G), which lends LVs a wide tropism as it uses the low-density lipoprotein receptor (LDLR) as the main receptor for cell entry. In some gene therapy and research applications, however, alternative pseudotypes can be useful. In this work, we characterized LVs pseudotyped with lymphocytic choriomeningitis virus (LCMV) glycoprotein, particularly in gene transfer to an LDLR-deficient mouse strain used to model cardiovascular disease, Ldlr-/-ApoB100/100. Methods: LCMV-L

Screening of the Pandemic Response Box Library Identified CRM1/XPO1 as an Anti-Mammarenavirus Druggable Target.

Mammarenaviruses (MaAv) cause persistent infection in their natural rodent hosts across the world and, via zoonotic events, can cause severe disease in humans. Thus, the MaAv Lassa virus (LASV) in Western Africa and the Junin virus (JUNV) in the Argentinean Pampas cause hemorrhagic fever diseases with significant case fatality rates in their endemic regions. In addition, the globally distributed MaAv lymphocytic choriomeningitis virus (LCMV) is an underrecognized human pathogen of clinical significance capable of causing devastating infections in neonates and immunocompromised individuals. Stepwise differentiation from precursor intermediates and distinct Th1 checkpoints promote CD4 Th1 cell differentiation during chronic viral infection.

CD4+ helper T cells are essential for controlling viral infections. During chronic LCMV infection, CD4+ T cells differentiate into heterogeneous populations, including a TCF-1hi progenitor subset that serves as a reservoir to continuously replenish type 1 helper (Th1) and follicular helper (Tfh) T cells. The gradual loss of CD4+ Th1 cell responses impairs the immune system's ability to control viral replication and contributes to the development of CD8+ T cell exhaustion. However, the mechanisms directing Th1 differentiation and the factors underlying their progressive decline during chronic i

A Novel Murine Model of Hemophagocytic Lymphohistiocytosis-Like Inflammation in ZNFX1 Deficiency.

Hemophagocytic lymphohistiocytosis (HLH) is a severe inflammatory syndrome characterized by persistent activation of lymphocytes and macrophages. Recently, deleterious autosomal recessive mutations in ZNFX1 were reported to predispose pediatric patients to HLH-like disease upon viral trigger. The objective of this study was to assess the suitability of Znfx1-mutant (Znfx1mut) mice infected with lymphocytic choriomeningitis virus (LCMV) as a model of HLH-like inflammation observed in patients. Following LCMV infection, Znfx1mut mice were monitored for pathophysiological signs of HLH, and their

Sphingosine kinase 2 suppresses neutrophil responses to promote viral persistence while attenuating immune pathology.

Chronic viral infections often suppress immune cell functions, which helps restrict immune pathology but leads to viral persistence. However, the underlying mechanisms are not completely understood. We recently found that sphingosine kinase 2 (SphK2)-deficient (Sphk2 -/-) mice succumb to lymphocytic choriomeningitis virus (LCMV) infection due to immune pathology. In addition to heightened T cell immunity, a notable increase in neutrophil numbers was observed in LCMV-infected Sphk2 -/- mice. Depletion of neutrophils increased the viability of virus-infected Sphk2 - / - mice, supporting the role

WAS Protein Deficiency Disrupts Memory B Cell Formation During Acute LCMV Infection.

Wiskott-Aldrich syndrome (WAS) is a rare x-linked monogenic immunodeficiency disease, caused by the mutation of WAS gene encoding WAS protein (WASp). Previous findings in WAS patients show B cell perturbations in the periphery, characterized by diminished B-cell numbers and phenotype abnormalities, including reduced frequency of classical CD27+ memory B cells (MBCs), accompanied by an unusual expansion of atypical CD21low MBCs. The mechanism underlying these abnormalities in MBCs developmental pathway has not been completely dissected. In this study, WASp knock-out mice undergone with acute ly

Memory-like CD8+ T cells lacking PD-1 adapt to persistent stimulation by reducing TCR signal transduction rather than increasing exhaustion.

CD8+ T cells respond to persistent stimulation during chronic viral infection by stably expressing co-inhibitory receptors and other exhaustion-related molecules. Here we addressed how memory-like CD8+ T (TML) cells, which sustain the immune response to chronic infection thanks to their stem-like properties, adapt to chronic stimulation when they cannot express the co-inhibitory receptor PD-1. We found an increased initial generation and stable long-term persistence of TML cells lacking PD-1 during chronic viral infection. However, these cells had a reduced ability regenerate upon acute restim

Sulfated glycosaminoglycans inhibit LCMV entry and modulate antiviral immunity and pathology.

Viral infections remain a major challenge due to the limited availability and efficacy of current treatments. Existing antivirals primarily target viral replication but are often virus-specific and can lead to drug resistance. Sulfated glycosaminoglycans (GAGs) have emerged as promising broad-spectrum agents that block viral binding and entry into host cells. Here, we show that highly sulfated GAGs restrict the infectivity of both pathogenic and non-pathogenic Arenaviruses. Using the lymphocytic choriomeningitis virus (LCMV) model, we demonstrate that GAG exposure reduces viral entry and infec

Non-cytolytic re-engineering of a viral vaccine vector enables durable effector-memory T cell immunity by reinforcing type I IFN induction.

Replication-deficient viral vector systems hold promise for CD8 T cell-based vaccination, but the molecular mechanisms accounting for platform-specific differences in immunogenicity remain ill-defined. When comparing prototypic single-cycle vaccine delivery platforms, we found that lymphocytic choriomeningitis virus (rLCMV)-based vectors, which are non-cytolytic, elicited more durable and effector-memory-differentiated CD8 T cell responses than vectors based on cytolytic vesicular stomatitis virus (rVSV). Hence, we re-engineered rVSV to be non-cytolytic (rVSVMq). This vector induced more durab

Glutathione is critical for NK cell-mediated immunity.

Natural killer (NK) cells are essential for immune protection against tumors and viruses. Disease environments impose oxidative stress and impair immune cell functions. Glutathione (GSH) is a major cellular antioxidant and is critical for the immune response, but how it modulates NK cell function remains largely unknown. Using a mouse model with a specific deletion of the catalytic subunit of glutamate-cysteine ligase (Gclc) in NK cells, we demonstrate that GSH supports interleukin-15 (IL-15)-driven activation of NK cells. Gclc deficiency causes an intracellular accumulation of reactive oxygen

Oncolytic virotherapy potentiates chemo-PD-1 immunotherapy by engaging chemo-resistant bystander CD8+ T cells.

BACKGROUND: The efficacy of combined chemotherapy and programmed cell death protein-1 (PD-1) immune checkpoint blockade (ICB) is constrained by the collateral cytotoxicity of chemotherapy toward proliferating tumor-specific CD8+ T (TTST) cells, a population indispensable for antitumor immunity. This study aimed to overcome this limitation by targeting a potential chemotherapy-resistant immune cell reservoir. METHODS: By using the murine acutely resolved lymphocytic choriomeningitis virus (LCMV) infection and tumor models (colorectal cancer and melanoma), we characterized the susceptibility of
