Overexpression of the transcriptional corepressor Tle1 enhances effector proliferation of CD8+ T cells during acute and chronic viral infections.

The transcriptional corepressor Tle (transducin-like enhancer of split) proteins interact with transcription factors such as TCF-1 and Runx3 to regulate transcriptional programmes during cellular development and differentiation; however, their roles in CD8+ T cell differentiation, particularly under conditions of chronic antigen stimulation, remain poorly defined. Here, we demonstrated that overexpression of Tle1, Tle3, and Tle4 improves the proliferation of antigen-specific effector CD8+ T cells during both acute and chronic viral infections. Notably, overexpression of Tle3 and Tle4, but not

Transient loss of KLF2 expression is essential for CD8 effector T cell expansion.

How T cell expansion, migration, and differentiation are coordinately controlled remains incompletely understood. The KLF2 transcription factor is critical for expression of cell surface molecules, which control lymphocyte traffic, and has been postulated to function as a mediator of T cell quiescence. KLF2 expression is rapidly and transiently silenced in activated T (and other) cells via proteolytic degradation and transcriptional silencing, but why recently activated T cells need to turn off KLF2 expression has remained unknown. Here, functions of transient KLF2 loss have been investigated

An autoimmunity-associated allele of PTPN22 enhances innate antiviral immunity to protect against acute coronavirus infection.

Allelic variation can impact viral clearance and disease severity. Still, our understanding of the effect of the autoimmunity-associated allelic variant of Ptpn22 (PEP-R619W) on antiviral immunity remains incomplete, as previous reports have only focused on chronic lymphocytic choriomeningitis virus infection. This research defines how the loss of Ptpn22 (PEP-null) and PEP-R619W changes antiviral immunity during an acute coronavirus infection. We address the hypothesis that CRISPR/Cas9-generated PEP-null and PEP-R619W mice have enhanced antiviral immunity over PEP-wild-type (WT) mice during co

Lymphocytic Choriomeningitis Virus Seroprevalence among Urban Pregnant Women and Newborns, Philadelphia, Pennsylvania, USA, 2021.

Lymphocytic choriomeningitis virus (LCMV) is a globally distributed rodentborne pathogen that can cause severe congenital infections. We conducted a retrospective cross-sectional seroepidemiologic study using remnant serum samples from pregnant women and newborns at 2 hospitals in Philadelphia, Pennsylvania, USA. We tested samples for LCMV IgG and IgM in 3 phases: a high-risk group determined by neighborhood deprivation index scores, a random sample of all birthing women, and a group with prenatally diagnosed neurologic malformations. We found LCMV IgG seroprevalence was 2.4% among 700 high-ri

Exposed phosphatidylserine is an inhibitory molecule in T cell exhaustion.

In cancer and chronic infection, CD8 T cell exhaustion is hallmarked by expression of inhibitory receptors such as PD1, TIM3, LAG3 and others1-3. Thus, inhibitory molecule focus has been limited to cell-surface proteins. Here we evaluate the surface lipid metabolite phosphatidylserine (PS) as a regulator of exhaustion. PS primarily localizes to the inner plasma membrane of live cells but is well known to be externalized to the outer membrane during cell death. The role of exposed PS on live immune cells is less clear. We show that viable, antigen-specific CD8 T cells externalize PS during lymp

ZFP148 is a transcriptional repressor of cytolytic effector CD8+ T cell differentiation.

Progenitor CD8+ T cells differentiate into effector and exhausted progenies during chronic antigen stimulation; however, mechanisms that restrain exhaustion and sustain effector differentiation remain incompletely defined. Here we identified the transcription factor ZFP148 as a repressor of CD8+ T cell effector differentiation. ZFP148-deficient CD8+ T cells displayed increased frequency of cytolytic effector cells and reduced frequency of exhausted cells compared with Zfp148fl/fl controls during chronic viral infection. Mechanistically, ZFP148 limited the chromatin accessibility of effector-dr

Irgm1 Restrains CD8+ T Cell Cytokine Production and Apoptosis via Cell-Extrinsic Regulation of Type I Interferon Signaling.

Immunity-related GTPases (IRGs) are a family of proteins that maintain cellular homeostasis by promoting autophagy and mitophagy. Mutations in human IRGM and genetic deletion of mouse Irgm1 have been linked to increased severity of inflammatory bowel disease, cancer, sepsis, and various infections. While IRGM/Irgm1 are known cell-intrinsic regulators of inflammation, their roles in T cell function remain poorly understood. We previously demonstrated that Irgm1 deficiency leads to increased production of proinflammatory mediators, including Granzyme B and interferon-γ, and increased apoptosis i

Role of Src family kinases Fyn and Lyn in arenavirus infection.

Arenaviruses encompass a diverse group of infectious agents, some of which, like Lassa virus (LASV), pose significant threats to global public health due to their acute virulence, while others, such as lymphocytic choriomeningitis virus (LCMV), are less pathogenic but still clinically relevant. Both Old World arenaviruses LASV and LCMV exploit host tyrosine kinase signaling to establish infection, though the molecular mechanisms remain incompletely understood. In this study, phosphorylated receptor tyrosine kinase antibody array screening revealed that recombinant LASV (rLCMV-LASV GP) infectio

Chronic infection perturbs the affinity hierarchy of antiviral B cells.

A key function of the germinal center (GC) reaction consists in the preferential expansion and enrichment of high-affinity B cell clones. Whether and how persistent viral infection thwarts this purpose remains ill-defined. Here, we transferred monoclonal lymphocytic choriomeningitis virus-specific B cells into persistently infected hosts. High-affinity B cells expanded vigorously, forming GCs and abundant antibody-secreting cells. When failing to clear viremia or to drive viral mutational escape, the expanded B cell population contracted, ending in its quasi-complete disappearance from the spl

Perinatal infection elicits clonally restricted T follicular helper cell responses that drive antibody-mediated viral control.

Perinatal infection often results in viral persistence, as observed for hepatitis B virus in humans. Commonly held concepts suggest that persistence is associated with immunotolerance, but this fails to predict the emergence of antiviral antibodies and eventual viral load control. Studying perinatal lymphocytic choriomeningitis virus (LCMV) infection, the prototypic mouse model of neonatal antiviral immunotolerance, we observe antibody-mediated viral load suppression and canonical germinal center (GC) reactions. This neonatal response is driven by antiviral CD4+ T follicular helper (Tfh) cell

B cell immunity to the Lassa virus glycoprotein is a correlate of vaccination-induced virus control in mice.

Lassa fever, a viral hemorrhagic fever caused by the arenavirus Lassa virus (LASV), affects thousands of individuals annually, highlighting the need for a vaccine. Yet, immunological correlates of viral load control remain poorly defined. Here we study vaccination-induced immunity in a surrogate LASV challenge model in mice. We find that LASV-cross-reactive B cell immunity induced by the glycoprotein of distantly related arenaviruses, such as lymphocytic choriomeningitis virus (LCMV), provides significant viral load control. Counter to common concepts, suppression of viremia is observed in the

Selective depletion of virus-specific CD8 T cells from the liver after PD-1 therapy with Fc-intact antibody during chronic infection.

Anti-programmed cell death 1 (PD-1) antibody therapy is now widely used in various cancers. However, the role of the antibody Fc region in PD-1 directed immunotherapy is not well understood. Preclinical studies commonly use species-mismatched rat anti-mouse antibodies, which may not accurately reflect antibody-Fc gamma receptor (FcγR) interactions. Here, we used mouse anti-mouse PD-1 antibodies to investigate how the Fc region influences therapeutic efficacy for enhancing CD8 T cell responses using mouse models of chronic lymphocytic choriomeningitis virus infection and CT26 tumors. Increased Constitutive Interferon-β Levels and Altered CD4 T Cell Homeostasis Induced by Expression of a Viral Glycoprotein.

Besides the strong Type I interferon (IFN-I) response induced by infections, IFN-I is also produced constitutively at lower levels. Constitutive IFN-I production is regulated by resident microbiota and is essential for induction of efficient immune responses. Here, we demonstrate in transgenic (tg) mice that expression of a single viral envelope gene, the glycoprotein (GP) of lymphocytic choriomeningitis virus (LCMV), increased constitutive IFN-β levels and induced numerous IFN-stimulated genes (ISGs). In addition, self-antigen-driven CD44highCD62Llow memory-phenotype CD4 T cells and regulator

Mammarenaviruses depend on endogenous fatty acid synthesis in cell culture systems.

Lassa virus (LASV) and lymphocytic choriomeningitis virus (LCMV) are Old World mammarenaviruses that, like all viruses, rely on host-derived biological molecules to complete their replication cycle. Identifying host factors essential for mammarenavirus replication may reveal novel targets for antiviral intervention. To this end, we found that replication of recombinant tri-segmented r3LCMV and r3LCMV LASV chimera (r3LCMV-LASV) was sensitive to reductions in both exogenously supplied and endogenously synthesized lipids. Lipidomic analysis on mock-infected and virus-infected VeroS cells revealed

Development and validation of two economical and flexible immunoassays for detecting antibodies against LCMV in mouse serum.

Measurement of antibody responses to viral infection is essential for surveillance, diagnostics, epidemiological research, and natural history of infection studies. However, current methods to detect virus-specific antibodies are often resource-intensive and impractical for deployment in outbreak settings or in field-based studies. This manuscript presents two economical, high-throughput immunoassays-the cytoblot immunoassay (CBA) and strip immunoblot assay (SIA)-for detecting and quantifying anti-lymphocytic choriomeningitis mammarenavirus (LCMV) antibodies in mouse serum. To validate, we tes

PIM1 kinase-regulated cellular metabolism sustains differentiation and function of effector CD8+ T cells during chronic viral infection.

CD8+ T-cell differentiation during chronic viral infection is supported by metabolic reprogramming to meet distinct bioenergetic demands. Early effector CD8+ T-cell differentiation and function are supported by the PI3K-Akt-mTOR pathway, while the differentiation of late exhausted CD8+ T cells remains incompletely understood. We first characterized the metabolic heterogeneity of the progenitor, effector, and exhausted CD8+ T-cell subsets in chronic infection by utilizing the Compass algorithm, which provides metabolic state predictions based on single-cell RNA sequencing data and flux-based an

Nociceptor neurons shape antiviral immunity.

The nervous and immune systems cooperate to protect the host, yet the contribution of sensory neurons to antiviral immunity remains incompletely defined. Nociceptor neurons do more than relay pain: they detect viral products and inflammatory cues through pattern-recognition receptors (PRRs), including Toll-like receptors (TLRs) and RIG-I-like receptors (RLRs), and respond to mediators such as type I interferons (IFNs), tumor necrosis factor (TNF), and interleukin-1β (IL-1β). Upon activation, these fibres release neuropeptides and neurotransmitters, including calcitonin gene-related peptide (CG

Krüppel-like factor 2 programs early exhausted T cell states and restrains antiviral immunity.

A key challenge in improving T cell-mediated immunotherapies is defining the factors that regulate functional versus exhausted T cell fates. Through multi-round in vivo CRISPR screens in chronic lymphocytic choriomeningitis virus Clone 13 (LCMV Cl13) infection and transcription factor (TF) benchmarking, we identified Krüppel-like factor 2 (KLF2) as a top TF driving CX3CR1+ effector-like exhausted cell (Texeff-like) differentiation. Overexpression of KLF2 converted CX3CR1⁻ cells into Texeff-like cells by direct engagement of key loci. Conversely, loss of KLF2 increased inhibitory receptor expre
