Whole-body irradiation causes long-term impairments in the maintenance and function of naïve CD4 T cells specific for self- and non-self-antigens.

Whole-body irradiation (WBI) consistently induces radiation-associated lymphopenia, a complication linked to poor prognosis and reduced overall survival. Understanding the quantitative and qualitative changes in naïve CD4 T cells after WBI is critical because these cells play key roles in both host defense and the propagation of autoimmunity. Here, we show that WBI triggers a rapid decline in the naïve CD4 T-cell pool, followed by thymus-dependent numerical recovery. Shortly after WBI, mice exhibit a reduced frequency of lymphocytic choriomeningitis virus (LCMV) epitope GP66-77-specific naïve

Metabolomics of in vivo inflammation identifies soluble sialic acid as a conserved myeloid-cell metabolite.

During an immune response, metabolism changes dramatically. Metabolites are oxidized to power immune cell functions, serve as building blocks for proliferation, and act as effectors to regulate pathogen or host cells. Though metabolic changes in cultured cells have been studied extensively, metabolism changes in vivo are less understood. Here, we measured metabolomic changes across six mouse tissues in three models of immune activation: CpG-DNA cytokine storm, lymphocytic choriomeningitis virus infection, and polyI:C viral mimetic injection; and carried out metabolomics in cultured macrophages

Dynamic expression of IL-21 in effector CD8 T cells helps control chronic infections.

CD8 T cells are critical players in immune responses against pathogens. Interleukin-21 (IL-21) is predominantly produced by CD4 T cells and exerts multifaceted effects on CD8 T cell regulation and function. Using Il21-reporter and Il21-fate mapping mice, we report that a subpopulation of activated CD8 T cells also produces IL-21 in the context of lymphocytic choriomeningitis virus (LCMV) infection. During the early effector phase of both acute and chronic infections, IL-21-expressing CD8 T cells exhibit substantial proliferative and cytotoxic capacities, with higher levels of interferon-γ (IFN

Sirtuin 1 Is Required for Optimal Mammarenavirus Multiplication.

Mammarenaviruses (MaAv) cause persistent infections in diverse rodent reservoirs worldwide and several are zoonotic pathogens with an important public-health burden in their endemic regions. Moreover, the globally distributed MaAv lymphocytic choriomeningitis virus (LCMV) is an underrecognized pathogen of clinical significance in congenital infections and immunocompromised individuals. The lack of FDA-approved vaccines or antivirals for MaAv infections underscores the urgent need for novel anti-MaAv therapeutic strategies. Neutral sphingomyelinase 2 (nSMase2) was recently identified as a host

LRRK2 G2019S reprograms innate and adaptive immunity to drive context-dependent host defense outcomes.

INTRODUCTION: Parkinson's disease (PD) is increasingly recognized as a neurological disorder characterized not only by neurodegeneration but also by chronic immune dysregulation across the lifespan. Although the initiating events underlying PD remain unclear, accumulating evidence suggests that inflammatory processes may contribute to disease susceptibility and progression. Mutations in leucine-rich repeat kinase 2 (LRRK2), particularly the gain-of-function G2019S variant, represent the common genetic cause of familial PD and have been implicated in immune regulation and infection susceptibili

Virus-specific memory CD4 T cells sustain long-term numerical and functional impairment following whole-body irradiation.

The increasing demand for nuclear power and radiotherapy has expanded potential avenues for human exposure to whole-body irradiation (WBI), underscoring the need to understand its impact on immune cells. While prior studies have explored radiation-induced disruptions in immune homeostasis, the direct effect of WBI on pre-existing memory CD4 T cells remains elusive. Using T cell receptor transgenic CD4 T cell chimeric mice infected with lymphocytic choriomeningitis virus-Armstrong, we demonstrate that a sublethal WBI dose (5 Gy) causes a significant and persistent reduction in the numbers of me

Innate immune activation profiles are associated with polarization of the T cell response to recombinant arenaviral vectors.

Arenavirus-based vectors are uniquely positioned to generate durable T cell responses against tumors, which remains a central challenge in cancer immunotherapy. Two members of the arenavirus family, Lymphocytic choriomeningitis virus and Pichinde virus, were genetically engineered to encode a non-oncogenic fusion protein, originating from human papillomavirus 16 E6 and E7. Hereafter, these vectors are referred to as HB-201 and HB-202, respectively. HB-201 alone or in combination with HB-202 elicited strong and durable E6/E7-specific CD8+ T cell responses in HPV 16+ cancer patients. Despite the

Extended IL-4 priming of macrophages induces an antiviral state that restricts lymphocytic choriomeningitis virus replication.

Viruses such as lymphocytic choriomeningitis virus (LCMV) and Lassa virus can directly suppress macrophage (Mϕ) activation, leading to viral chronicity and severe disease. Recent studies have shown that prolonged IL-4 stimulation induces a proinflammatory M2a phenotype, which upon subsequent TLR stimulation exhibits unique immune parameters. However, the innate immune response of such M2a Mϕ to virus infections remains unknown. Here, we investigated the response of IL-4-activated spleen-derived Mϕ (SpM) to LCMV infection to understand how IL-4 induced polarization influences the Mϕ - virus int

Lymphocytic Choriomeningitis Virus Bearing GPC Mutations for Enhanced Tumor Tropism and Strong Anti-Tumor Activity.

The development of effective viroimmunotherapies for cancer relies on the ability of replicating viruses to infect the entire tumor tissue and induce comprehensive inflammation within the tumor microenvironment (TME). In this study, we investigate the impact of mutations in the glycoprotein (GP) of lymphocytic choriomeningitis virus (LCMV) on tumor cell infectivity, replication, and the immune response. Specifically, we focus on two key positions in GP1 (181 and 185) that significantly enhance tumor cell infectivity and improve CD8+ T cell function in the GP-mutated LCMV strain LCMV P52. Our f

Mutations at Positions 181 and 185 Within the LCMV GP Protein Increase Tumor Cell Infectivity, Limit Induction of Type I Interferon, and Accelerate T Cell Activation.

The lymphocytic choriomeningitis virus (LCMV) represents a strong T cell activating virus with anti-tumoral properties. In recent work, we developed an attenuated cancer cell-adapted reassortant LCMV strain carrying tumor-tropic mutations. The immunological mechanisms underlying these mutations remained unknown. Here, we passaged wild-type LCMV-WE in human lung cancer H1975 cells for 52 passages. After 29 passages, the virus acquired the GP1 mutations I181M and R185W, involved in viral receptor interactions, which remained stable throughout the subsequent 33 passages. Compared with wild-type L

Propagation of side-stream partial nitritation-anammox disturbance to mainstream wastewater greenhouse gas emissions.

INTRODUCTION: The orientation of collagen fibers in native tissues plays an important role in cell signaling and mediates the progression of tumor cells in breast cancer by a contact guidance mechanism. Understanding how migration of epithelial cells is directed by the alignment of collagen fibers requires in vitro assays with standardized orientations of collagen fibers. METHODS: To address this issue, we produced micro-stripes with aligned collagen fibers using an easy-to-use and versatile approach based on the aspiration of a collagen solution within a microchannel. CD8+ T cell responses recognizing immunodominant Chlamydia antigens fail to protect against infection.

Chlamydia trachomatis (C.t.) causes the most common bacterial sexually transmitted infection, yet no licensed vaccine exists. CD4+ T cells are known to protect, while the role of CD8+ T cells is unclear. This study examined whether CD8+ T cells targeting five immunodominant C.t. proteins could protect against infection. Using an in silico approach, we identified six novel CD8+ T cell epitopes from ArtJ, GroES, IncA, OmpH, and major outer membrane protein (MOMP). These epitopes elicited C.t.-specific CD8+ T cells when delivered with the cationic adjuvant formulation 09b (CAF09b) liposomal adjuv

Common and Rare Emerging and Re-Emerging Infectious Diseases in Pediatric Transplant Recipients: A Comprehensive Review.

The established pathogens, including cytomegalovirus, Epstein-Barr virus, and bacterial bloodstream infections, remain leading causes of morbidity and mortality in pediatric transplant recipients (PTR). However, over the past two decades, the landscape of infectious diseases in this population has evolved substantially, marked by an increasing burden of emerging and re-emerging pathogens. The global rise in multidrug-resistant bacteria and fungi has complicated antimicrobial management and limited therapeutic options. Concurrently, novel and re-emerging viral threats, most notably severe acute

A Bacterial Microbiome Is Dispensable for the Induction of CD8 T Cell Exhaustion.

Prolonged antigen exposure in chronic viral infections reduces the effector capacity of cytotoxic T cells-a phenomenon known as T cell exhaustion. Development of T cell exhaustion is driven by high viral titers, strong TCR stimulation, and high antigen concentrations associated with strong inflammatory signals. A largely unexplored factor has been the influence of the microbiome in these processes. Here, we report that T cell exhaustion progresses independently of the presence or absence of a microbiome in chronic lymphocytic choriomeningitis virus (LCMV) infections. Virus-specific CD8 T cells

A Clinical Review of Congenital and Perinatal Infections: From Womb to Follow-Up.

The acronym "TORCH" was originally introduced in 1974 by Dr Nahmias to collectively describe the most common pathogens responsible for congenital and perinatal infections, including Toxoplasma gondii, others, rubella virus, cytomegalovirus (CMV), and herpes simplex virus. With advances in diagnostic capabilities and expanded epidemiologic surveillance, the list of recognized congenital pathogens has grown to include syphilis, varicella-zoster virus, Zika virus, HIV, parvovirus B19, and lymphocytic choriomeningitis virus. While the TORCH acronym continues to be widely used in clinical and educa

Accumulation of free nuclei denotes defective phagocytic capacity of macrophages and occurs after infection with Listeria monocytogenes and lymphocytic choriomeningitis virus.

Efficient phagocytosis of pathogens is a key effector function of the innate immune system. Impaired phagocytic activity can result in uncontrolled pathogen proliferation and life-threatening infections. However, reliable methods to detect early dysfunction of the phagocytic system in vivo are limited. Here, we used a mouse model of Listeria monocytogenes infection to determine blood parameters which correlate with limited macrophage function. We found that lack of macrophages led to accumulation of nuclei in the blood. Further analysis of nuclei revealed that these nuclei were released from b

Prevalence of antibodies against hantavirus, Leptospira, lymphocytic choriomeningitis virus and Chlamydia psittaci in pest control workers in Germany - a cross-sectional study.

BACKGROUND: Pest control workers may be exposed to zoonotic pathogens during their work, yet data on the prevalence of zoonotic infections in this occupational group in Germany is scarce. This study investigated the seroprevalence of antibodies against four pathogens commonly transmitted by rodents and pigeons among pest control workers in Germany. METHODS: Blood samples of participants were analysed for antibodies against hantavirus, Leptospira, lymphocytic choriomeningitis virus (LCM virus) and Chlamydia psittaci. Additional data was collected using a questionnaire. RESULTS: Among the 154

PARP14 is an interferon (IFN)-induced host factor that promotes IFN production and affects the replication of multiple viruses.

PARP14 is a 203 kDa multi-domain protein that is primarily known as an ADP-ribosyltransferase, and is involved in a variety of cellular functions including DNA damage, microglial activation, inflammation, and cancer progression. In addition, PARP14 is upregulated by interferon (IFN), indicating a role in the antiviral response. Furthermore, PARP14 has evolved under positive selection, again indicating that it is involved in host-pathogen conflict. We found that PARP14 is required for increased IFN-I production in response to coronavirus infection lacking ADP-ribosylhydrolase (ARH) activity and

Protein Phosphatase 2A Promotes CD8+ T Cell Effector Function through the Augmentation of CD28 Costimulation.

Protein phosphatase 2A (PP2A) is one of the most abundant serine/threonine phosphatases and plays critical roles in regulating cell fate and function. We previously showed that PP2A regulates the differentiation of CD4+ T cells and the development of thymocytes. Nevertheless, its role in CD8+ T cells remains elusive. By ablating the catalytic subunit α (Cα) of PP2A in CD8+ T cells, we revealed the essential role of PP2A in promoting the effector functions of CD8+ T cells. Notably, PP2A Cα-deficient CD8+ T cells exhibit reduced proliferation and decreased cytokine production upon stimulation in
