Accelerating the clinical development of treatments for dengue: WHO dengue therapeutics landscape analysis and target product profiles.

Dengue, an Aedes mosquito-borne viral infection, is on the rise with climate and demographic change. In 2023, WHO declared dengue its highest grade of emergency, following the largest number of cases and deaths in recorded history. This emergency highlighted the absence of safe and effective dengue-specific treatments as a gap in the product landscape for care for people with dengue. To accelerate development of dengue-specific treatments for wide implementation and access in all dengue-endemic countries, WHO conducted a landscape analysis of the dengue therapeutics pipeline and convened a glo

Analysis of Cell Populations from Arbovirus-Infected Tissues by Flow Cytometry.

Arboviruses still represent a major challenge to public health in many parts of the world. Despite shared epidemiological characteristics, each arboviral disorder displays distinct cellular tropism and activation patterns, progressing to immunopathological outcomes that remain not fully elucidated. A deeper understanding of these processes requires methodologies capable of characterizing the often-complex cellular responses. In this context, flow cytometry emerges as a valuable tool, enabling the characterization of heterogeneous cell populations, the assessment of activation and proliferation

Assessing the Vector Competence for Chikungunya Virus in Mosquitoes.

Chikungunya virus (CHIKV) is a human pathogenic, mosquito-borne virus (arbovirus), which is causing epidemic outbreaks among human populations in Africa, Asia, Europe, and South- and Central America including the Caribbean. The development of novel approaches to prevent mosquito transmission of the virus in the field requires a detailed study of CHIKV interactions with its mosquito vectors in the laboratory. In this chapter, we describe how to prepare Aedes aegypti mosquitoes for the infection with freshly cultivated CHIKV. We also describe quantitative and qualitative viral detection assays i

A global database of West Nile virus host prevalence and competence.

West Nile virus (WNV) is one of the most widespread arboviruses globally and is maintained primarily through a bird-mosquito-bird transmission cycle, while other vertebrates play more limited roles. Host contributions to transmission depend on both infection evidence in natural populations (reflecting exposure and susceptibility) and reservoir competence, determined by the magnitude and duration of viraemia sufficient to infect mosquitoes. Despite extensive surveillance and experimental research, no comprehensive, standardised resource has integrated evidence on host exposure and infection in

Genomic characterization of highly pathogenic avian influenza A(H5N1) clade 2.3.4.4b into wild seabirds from coastal Ecuador.

INTRODUCTION: Highly pathogenic avian influenza A(H5N1) clade 2.3.4.4b has spread widely across South America, but its genomic diversity and patterns of introduction into Ecuador remain poorly characterized. METHODS: In this study, we report and characterize four genomes of A(H5N1) viruses detected in four individuals of two Ecuadorian seabird species: two magnificent frigatebirds (Fregata magnificens) and two blue-footed boobies (Sula nebouxii) sampled in 2023. RESULTS: These genomes represent confirmed detections of avian influenza in wild birds in the country. The viruses were assigned to

Swab-based viral RNA detection in hospitalized patients with Crimean-Congo hemorrhagic fever.

Data on Crimean-Congo haemorrhagic fever virus (CCHFV) RNA detection in non-blood specimens remain limited. We evaluated CCHFV RNA detection in nasopharyngeal (NP), oropharyngeal (OP), and sweat swab samples from hospitalized patients with CCHF. Among 50 patients with confirmed CCHF, CCHFV RNA was detected in at least one NP or OP swab in 91.5% of patients. Sweat swabs were positive in 34.9% of cases and sweat RNA detection was associated with greater disease severity. Among patients with paired admission and discharge samples 75% became swab PCR-negative at discharge. Upper respiratory swabs

Wesselsbron virus at the livestock-human interface: A practical guide to disease recognition and diagnosis.

Wesselsbron virus (WSLV) is a mosquito-borne Orthoflavivirus of the yellow fever virus (YFV) serocomplex, widely distributed across sub-Saharan Africa. It is neglected, and its true impact is likely underestimated. WSLV causes abortion, congenital malformations, and neonatal mortality in sheep, the primary recognized vertebrate host, but infections have also been documented in goats, cattle, camels, wildlife, and humans. The virus is maintained in complex cycles predominantly involving Aedes mosquitoes and incompletely defined domestic and wild vertebrate hosts. Direct infection via the placen

Kyasanur Forest Disease: feasible strategies for vaccine development and evaluation.

Kyasanur Forest Disease (KFD) is a tick-borne zoonotic infection endemic to certain parts of India, with expanding geographic distribution and recurrent focal outbreaks among forest-dependent populations in Western Ghats. The discontinuation of the formalin-inactivated KFD vaccine in 2022 has created a critical gap in prevention. New vaccine candidates are under development, but conventional Phase 3 efficacy trials are difficult to implement because of low incidence of disease, and clustered and unpredictable nature of outbreaks. KFD vaccine evaluation may require alternate regulatory approach

Regional and Temporal Dynamics of Dengue in Mexico (2020-2024): A Comprehensive Epidemiological Analysis.

Dengue remains a major public health concern in Mexico, with incidence and clinical burden increasing substantially in recent years. Using national surveillance data from 2020 to 2024, this study provides a comprehensive overview of the changing epidemiological landscape, with an emphasis on geographic distribution, temporal trends, serotype dynamics, and disease severity. Over this period, Mexico saw an almost fivefold increase in dengue cases, reaching over 123,000 confirmed infections and 426 deaths in 2024, the highest annual number recorded. Seasonal patterns shifted, with sustained trans

Monocyte-associated C1q upregulation and complement activation contribute to vascular endothelial and multi-organ injury in severe dengue.

Dengue virus (DENV) infection causes a broad spectrum of clinical manifestations, ranging from self-limiting dengue fever (DF) to life-threatening severe dengue (SD). Increased vascular permeability and endothelial dysfunction are hallmarks of SD and contribute to plasma leakage, shock, and multi-organ injury, yet the underlying mechanisms remain incompletely understood. Here, we integrated dynamic bulk and single-cell RNA sequencing data from peripheral blood mononuclear cells (PBMCs) of dengue patients at different disease stages to investigate the mechanisms associated with disease progress

Kyasanur forest disease: A brief overview and relevance to public health in the Czech Republic.

Kyasanur forest disease is a tick-borne haemorrhagic disease endemic to southern India. The cases have recently been on the rise in India, and the area of occurrence is expanding. This is primarily due to climate change, deforestation, and increased human contact with forest ecosystems. The symptoms include fever, myalgia, haemorrhagic manifestations, and neurological complications. The case-fatality rate ranges from 3% to 10%. Kyasanur forest disease may be underdiagnosed, given its possible subclinical course. In India, an inactivated vaccine was available until 2022 but was withdrawn due to

New-onset refractory status epilepticus with a hyperferritinemic hyperinflammatory phenotype in Kyasanur Forest Disease.

BACKGROUND: Kyasanur Forest Disease (KFD) is a tick-borne flaviviral infection endemic to India. While neurological manifestations are recognized, new-onset refractory status epilepticus (NORSE) has not been previously reported. We describe NORSE in the setting of severe KFD-associated hyperinflammation and multiorgan dysfunction. CASE PRESENTATION: A 53-year-old previously healthy woman with virologically confirmed KFD developed generalized tonic-clonic seizures that progressed to NORSE during a severe systemic illness. She had profound cytopenias, severe hepatitis, coagulopathy, myocarditis

Interactions between Tick-Borne Encephalitis Virus Nonstructural Protein 1 and Blood-Brain Barrier Tight Junction Proteins: Potential Clues to Strain-Specific Neuropathogenicity.

Tick-borne encephalitis virus (TBEV) invades the central nervous system (CNS) through strain-specific mechanisms that remain poorly understood. For mosquito-borne orthoflaviviruses such as dengue and yellow fever viruses, the nonstructural protein 1 (NS1) has been shown to disrupt endothelial barrier integrity by targeting tight junction proteins (TJPs), facilitating viral neuroinvasion. However, comparable mechanisms in TBEVs remain largely unexplored. Here, we investigated the potential interaction of NS1 from high (Hypr)- and low (Vs)-pathogenic TBEV strains with different blood-brain barri

Early-warning surveillance of West Nile and Usutu viruses in wetland and larval habitat waters and in mosquito excreta using digital PCR.

West Nile virus (WNV) and Usutu virus (USUV) are mainly avian mosquito-borne pathogens, increasingly cause human and equine disease in temperate regions. Mostly asymptomatic, current surveillance in France is reactive, based on clinical and veterinary reports, delaying signals for vector control and preventative interventions. We implemented in Southern France, in 2024 and 2025, a proactive surveillance methodology combining molecular xenomonitoring of mosquito excreta and environmental water detection of viral RNA in wetlands and mosquito larval habitat, through a multiplex reverse transcript

Genomic Surveillance of Dengue Virus During the 2024 Outbreak in a Northeastern Brazilian State.

Dengue remains a major public health challenge in tropical regions, where viral genetic diversity, vector ecology and human mobility contribute to recurrent outbreaks. In 2024, Brazil experienced its largest recorded dengue epidemic, with intense transmission across the Northeast region. This study characterizes the epidemiological, genomic and phylogeographic features of dengue virus (DENV) circulation during the 2024 outbreak in Alagoas, northeastern Brazil. Of 6102 suspected cases analysed by molecular diagnostics, 1607 (26.3%) were confirmed as DENV infections. Epidemiological analyses rev

Human exposure to Crimean-Congo hemorrhagic fever virus is associated with livestock management and handling practices in southcentral Tanzania, 2023-2024.

BACKGROUND: Crimean-Congo hemorrhagic fever virus (CCHFV) is a tick-borne zoonotic pathogen widely distributed across Africa, Asia, and Europe. Human infection occurs through tick bites or contact with infected animal tissues. In Tanzania, evidence of human exposure exists, but data on population-level seroprevalence and associated behavioral risk factors remain limited. This study assessed human CCHFV seroprevalence and identified demographic, behavioral, and environmental factors associated with prior exposure in livestock-keeping communities in southcentral Tanzania. METHODS: A community-b
