Accelerating the clinical development of treatments for dengue: WHO dengue therapeutics landscape analysis and target product profiles.

Dengue, an Aedes mosquito-borne viral infection, is on the rise with climate and demographic change. In 2023, WHO declared dengue its highest grade of emergency, following the largest number of cases and deaths in recorded history. This emergency highlighted the absence of safe and effective dengue-specific treatments as a gap in the product landscape for care for people with dengue. To accelerate development of dengue-specific treatments for wide implementation and access in all dengue-endemic countries, WHO conducted a landscape analysis of the dengue therapeutics pipeline and convened a glo

Analysis of Cell Populations from Arbovirus-Infected Tissues by Flow Cytometry.

Arboviruses still represent a major challenge to public health in many parts of the world. Despite shared epidemiological characteristics, each arboviral disorder displays distinct cellular tropism and activation patterns, progressing to immunopathological outcomes that remain not fully elucidated. A deeper understanding of these processes requires methodologies capable of characterizing the often-complex cellular responses. In this context, flow cytometry emerges as a valuable tool, enabling the characterization of heterogeneous cell populations, the assessment of activation and proliferation

Assessing the Vector Competence for Chikungunya Virus in Mosquitoes.

Chikungunya virus (CHIKV) is a human pathogenic, mosquito-borne virus (arbovirus), which is causing epidemic outbreaks among human populations in Africa, Asia, Europe, and South- and Central America including the Caribbean. The development of novel approaches to prevent mosquito transmission of the virus in the field requires a detailed study of CHIKV interactions with its mosquito vectors in the laboratory. In this chapter, we describe how to prepare Aedes aegypti mosquitoes for the infection with freshly cultivated CHIKV. We also describe quantitative and qualitative viral detection assays i

Prevalence and factors associated with dengue fever serotypes in Eastern Sudan: a hospital-based cross-sectional study.

BACKGROUND: Dengue is a critical public health threat in Sudan, exacerbated by rapid urbanization and climatic factors. The 2024 outbreak in Kassala State occurred amidst a violent internal conflict and massive population displacement, synergistically accelerating transmission and complicating clinical management. This study aimed to determine the prevalence of dengue virus (DENV) serotypes and identify sociodemographic and clinical factors associated with specific serotypes among residents and internally displaced persons (IDPs) in Kassala during the 2024 epidemic. METHODS: A hospital-based

No-known-vector flaviviruses exhibit diverse replication and virulence phenotypes in mice.

The Orthoflavivirus genus (flaviviruses) includes globally significant arboviruses, which cycle between arthropod vectors (mosquitoes and ticks) and vertebrate hosts. In contrast, no-known-vector flaviviruses (NKVFVs) have been isolated from rodents and bats, but not arthropods, so they are thought to spread by vector-independent routes. However, little is known about the host range and pathogenic mechanisms of these viruses. To evaluate NKVFV pathogenesis, we infected wild-type, Ifnar1-/-, and Ifnar1-/- Ifngr1-/- mice by footpad inoculation with 12 NKVFVs: Entebbe bat virus (ENTV), Sokuluk vi

Detection, characterization, and evolutionary inferences regarding a novel virus identified in Amblyomma dissimile Koch, 1844 (Acari: Ixodidae) from the Amazon, Pará, Brazil.

Ticks are among the primary vectors of infectious agents of medical and veterinary importance, second only to mosquitoes in the number of pathogens transmitted to humans. These arthropods are capable of transmitting a wide diversity of microorganisms, including protozoa, bacteria, and viruses. Notable among tick-borne viruses are those associated with viral encephalitis, which cause severe acute and chronic neurological infections in humans. This study aimed to characterize the RNA virome of an Amblyomma dissimile specimen collected from a Boa constrictor rescued in the municipality of Ananind

Portable Duplex Diagnostic Platform for Point-of-Care Differentiation of Ebola and Lassa Virus.

Ebola virus (EBOV) and Lassa virus (LASV) infections present with indistinguishable early symptoms, creating a persistent diagnostic gap in coendemic regions. This challenge is compounded in resource-limited settings, where existing lateral flow assays are single-target, qualitative, and reliant on subjective interpretation. We developed a duplex colorimetric lateral flow immunoassay (LFIA) for simultaneous detection of EBOV VP40 and LASV nucleoprotein (NP), integrated with the Portable Rapid Onsite Versatile Image-based Detection System (PROVIDS), a compact optical reader that automates line

Design of a tetravalent dengue vaccine incorporating T-cell epitopes from NS3 and NS5 proteins and B-cell epitopes from the envelope protein using an immunoinformatics approach.

Dengue virus remains a major global health threat due to its four antigenically diverse serotypes and limited treatment and vaccine options. This study aimed to design and evaluate a rational tetravalent multi-epitope vaccine capable of eliciting broad cross-serotype immune responses against dengue virus. An immunoinformatics-driven strategy was employed to identify CTL, HTL, and B-cell epitopes from the NS3, NS5, and envelope domain III proteins of DENV-1 to DENV-4 based on antigenicity, allergenicity, and predicted immunogenic potential, followed by assessment of HLA population coverage. Immunologic and placental responses to the VSV-EBOV vaccine in a pregnant mouse model.

Ebola virus disease is a severe viral hemorrhagic illness with high maternal and perinatal mortality. Although the replication-competent vesicular stomatitis virus-based Ebola vaccine (VSV-EBOV) has been deployed successfully in outbreak settings, pregnant women were excluded from clinical trials, leaving critical safety gaps. To address this, we developed a murine model to assess pregnancy outcomes following VSV-EBOV exposure at different gestational stages. Pregnant C57BL/6 mice vaccinated at embryonic day 7 (E7), E10, or E15 mounted robust anti-Ebola virus (anti-EBOV) antibody responses com

Knowledge, attitudes and practices about dengue vaccines in Latin America and the Caribbean: a scoping review.

Dengue remains a major public health concern in Latin America and the Caribbean, where vaccination has emerged as a key strategy to reduce severe disease. However, its successful implementation depends on population knowledge, attitudes, and practices (KAP). This study aimed to synthesize available evidence on KAP related to dengue vaccination in the region. A scoping review was conducted following JBI guidelines. Searches were performed in international and regional databases, including MEDLINE/PubMed, Scopus, Embase, Web of Science, SciELO, and the Virtual Health Library. Original studies we

Transmission Dynamics of Dengue Virus, Ceará State, Brazil, 2010-2025.

We evaluated the transmission dynamics of dengue virus (DENV) during 2010-2025 in Ceará State, Brazil. Ceará has been a dengue hyperendemic region, where all 4 DENV serotypes co-circulate, predominantly DENV-1 and DENV-2, and where DENV-3 was recently reintroduced. Our findings provide an epidemiologic baseline for postlicensure dengue vaccine evaluation.

Immunoinformatics and reverse vaccinology design of self-amplifying RNA vaccine against marburg virus with structural dynamics and immune simulation analysis.

Marburg virus disease (MVD), a deadly zoonotic illness in humans, is caused by the Marburg virus (MARV), a single-stranded RNA virus belonging to the Orthomarburgvirus genus of the Filoviridae family. There are currently no approved vaccines or efficient antiviral therapies for MARV, despite high morbidity and fatality rates of the virus, particularly in Africa. Recognizing its epidemic potential and its inclusion on the WHO list of blueprint priority pathogens, an immunoinformatics approach was used to design novel multi-epitope saRNA vaccine candidates (MB-saVaxI and MB-saVaxII) targeting th

Predicting Clinical Outcome in Dengue Fever: A Retrospective Cohort Modeling Study.

BACKGROUND: Dengue fever demonstrates a broad spectrum of clinical severity, ranging from self-limiting illness to life-threatening complications. Early identification of patients at risk of progression remains a clinical challenge, particularly in high-burden settings. Although platelet count and World Health Organization (WHO) dengue classification are routinely used in clinical practice, reliance on individual parameters may not adequately capture the multifactorial nature of disease severity. Development of multivariable predictive models integrating clinical and laboratory markers availab

Integrated immunoreactivity analysis of MHC-I epitopes derived from the Marburg virus glycoprotein.

INTRODUCTION: Marburg virus (MARV) is a hemorrhagic fever virus, and its glycoprotein (GP) represents an important target for vaccine research. MHC class I (MHC-I)-restricted T cell epitopes are critical for inducing cytotoxic CD8+ T cell responses. In this study, we aimed to identify MHC-I-restricted T cell epitopes derived from the MARV Angola strain GP through an integrated bioinformatics and experimental approach. METHODS: We retrieved the MARV Angola strain GP sequence and divided it into 673 overlapping 9-mer peptides. By integrating five computational prediction algorithms, immunogenic

Yellow Fever Outbreaks Are Not Associated With Long-Term Negative Human Perceptions of Wild Primates in Brazil.

Human perceptions of wildlife are fundamental to guiding effective strategies for conserving biodiversity and public health. Zoonotic diseases affecting non-human primates, such as yellow fever (YF), may trigger negative human behaviors toward these species. We conducted 474 semi-structured interviews across seven municipalities in Minas Gerais (n = 324 respondents) and Bahia (n = 150), Brazil, and used mixed-effects models to evaluate whether human perceptions of marmosets (Callithrix sp.) and howler monkeys (Alouatta sp.) differed according to previous YF outbreaks. YF occurrence was not con
