Filovirus reactive antibodies in wildlife hunting communities suggest ongoing zoonotic spillover in forested Guinea.

Forested Guinea has been the site of multiple high-consequence viral spillover events, including the index case of the 2013-16 West African Ebola epidemic. Deforestation and wildlife hunting rapidly expand the human-animal interface, increasing opportunities for transmission. Here we integrate serologic, spatial and questionnaire data to characterize zoonotic filovirus spillover events and risk factors amongst wildlife hunter communities in Forested Guinea. Hunters and close contacts from 43 villages in 2017 (n = 511) and 2023 (n = 648) were screened using a multiplex microsphere-based immunoa

Interactions between Tick-Borne Encephalitis Virus Nonstructural Protein 1 and Blood-Brain Barrier Tight Junction Proteins: Potential Clues to Strain-Specific Neuropathogenicity.

Tick-borne encephalitis virus (TBEV) invades the central nervous system (CNS) through strain-specific mechanisms that remain poorly understood. For mosquito-borne orthoflaviviruses such as dengue and yellow fever viruses, the nonstructural protein 1 (NS1) has been shown to disrupt endothelial barrier integrity by targeting tight junction proteins (TJPs), facilitating viral neuroinvasion. However, comparable mechanisms in TBEVs remain largely unexplored. Here, we investigated the potential interaction of NS1 from high (Hypr)- and low (Vs)-pathogenic TBEV strains with different blood-brain barri

Unsupervised identification of low-frequency antigen-specific TCRs using distance-based anomaly scoring.

Identifying antigen-specific T cell receptors (TCRs) within the diverse human repertoire remains challenging, particularly for low-frequency clonotypes. Here, we present TCR-RADAR (TCR Rare Antigen-specific Detection by Anomaly Ranking), an unsupervised approach that detects low-frequency antigen-specific TCRs through distance-based anomaly detection in TCR sequence space. Using TCRdist3 to quantify sequence distances, we identify query TCRs that are anomalous relative to reference repertoires within their V-J gene combinations. We validated this approach across three immunological contexts: C

Zoonotic arboviruses and associated risk factors in donkeys from northeastern Algeria.

Rift Valley fever virus (RVFV), Crimean-Congo hemorrhagic fever virus (CCHFV), and several zoonotic flaviviruses represent an emerging threat to animal and human health. In Algeria, particularly in the northern regions, data on the circulation of these viruses remain limited, especially for RVFV and CCHFV. In this context, we aimed to assess the seroprevalence of these arboviruses among donkeys across three provinces in northeastern Algeria and to identify factors associated with seropositivity. Between 2019 and 2024, 183 serum samples were collected and analyzed by ELISA. Binary logistic regr

Wolbachia introgression in Rio de Janeiro remains at sub-optimal levels 30 months after its decline: challenges in the sustainability of wMel interventions for dengue control.

BACKGROUND: The deployment of the Wolbachia wMel strain is currently underway in multiple dengue-endemic municipalities across Brazil. The efficacy of this strategy in Rio de Janeiro remains uncertain, primarily due to the difficulty in sustaining high wMel prevalence in regions previously subjected to large-scale releases. A key contributing factor was the routine rotation of insecticides within the framework of Integrated Vector Management (IVM), which led to the use of the larvicide Spinosad for Aedes control in urban areas. This compound was associated with a precipitous decline in Aedes a

Household representatives' understanding of dengue solutions and larval indices surveillance systems in southern Thailand: A mixed methods study.

BACKGROUND: Dengue fever remains a major public health concern in tropical regions, particularly in Southeast Asia. Effective prevention depends on the community's understanding of both dengue control measures and vector surveillance systems. This study assessed and explored household representatives' understanding of dengue solutions (care and preventive measures) and the Aedes larval index surveillance system in southern Thailand. METHODS: A community-based convergent mixed-methods study was conducted among 824 household representatives selected through stratified random sampling. Quantitat

Facilitators and barriers to community-based livestock abortion reporting for Rift Valley Fever surveillance in Uganda: A COM-B analysis.

Rift Valley Fever Disease (RVFD) causes substantial economic losses via livestock abortion storms, yet community-based abortion reporting for early warning remains suboptimal in endemic settings. This study analysed the behavioural determinants of livestock abortion reporting to inform RVF surveillance interventions in Uganda. We conducted a cross-sectional study using qualitative data collection methods in Isingiro District, Uganda, employing the Capability, Opportunity, Motivation-Behaviour (COM-B) framework. We conducted 29 key informant interviews with national/district policymakers and te

Vector competence for dengue-2 virus of permethrin resistant/ susceptible Aedes aegypti sub-populations from selected sites in Lamu County, Kenya.

Dengue virus, primarily transmitted by Aedes aegypti mosquitoes, is endemic in Lamu County. There is currently no definitive treatment for dengue; therefore, vector control remains the most effective method to curb transmission. Prolonged exposure of Ae. aegypti to insecticides may confer resistance. This study aimed to determine the levels of resistance to permethrin, a commonly used insecticide, and to compare the vectorial competence and genetic differences of resistant versus susceptible populations for dengue-2 virus. Ae. aegypti mosquitoes were collected from Lamu (Shela, Hindi, and Moko

From IL-6 production to IL-6R buffering: a unified IL-6-IL-6R framework for immune homeostasis during viral infection.

Interleukin-6 (IL-6) is a pleiotropic cytokine that coordinates innate and adaptive immunity and maintains tissue homeostasis during infection and inflammation. Although transient IL-6 production is essential for host defense, dysregulated or sustained IL-6 activity contributes to immunopathology across infectious and inflammatory diseases. Dengue virus (DENV) infection provides an important context in which this balance may become disrupted. Elevated circulating IL-6 levels are associated with endothelial dysfunction, vascular leakage, coagulopathy, and severe dengue; however, the mechanisms

Early-warning surveillance of West Nile and Usutu viruses in wetland and larval habitat waters and in mosquito excreta using digital PCR.

West Nile virus (WNV) and Usutu virus (USUV) are mainly avian mosquito-borne pathogens, increasingly cause human and equine disease in temperate regions. Mostly asymptomatic, current surveillance in France is reactive, based on clinical and veterinary reports, delaying signals for vector control and preventative interventions. We implemented in Southern France, in 2024 and 2025, a proactive surveillance methodology combining molecular xenomonitoring of mosquito excreta and environmental water detection of viral RNA in wetlands and mosquito larval habitat, through a multiplex reverse transcript

Genomic Surveillance of Dengue Virus During the 2024 Outbreak in a Northeastern Brazilian State.

Dengue remains a major public health challenge in tropical regions, where viral genetic diversity, vector ecology and human mobility contribute to recurrent outbreaks. In 2024, Brazil experienced its largest recorded dengue epidemic, with intense transmission across the Northeast region. This study characterizes the epidemiological, genomic and phylogeographic features of dengue virus (DENV) circulation during the 2024 outbreak in Alagoas, northeastern Brazil. Of 6102 suspected cases analysed by molecular diagnostics, 1607 (26.3%) were confirmed as DENV infections. Epidemiological analyses rev

Human exposure to Crimean-Congo hemorrhagic fever virus is associated with livestock management and handling practices in southcentral Tanzania, 2023-2024.

BACKGROUND: Crimean-Congo hemorrhagic fever virus (CCHFV) is a tick-borne zoonotic pathogen widely distributed across Africa, Asia, and Europe. Human infection occurs through tick bites or contact with infected animal tissues. In Tanzania, evidence of human exposure exists, but data on population-level seroprevalence and associated behavioral risk factors remain limited. This study assessed human CCHFV seroprevalence and identified demographic, behavioral, and environmental factors associated with prior exposure in livestock-keeping communities in southcentral Tanzania. METHODS: A community-b

The Mystery of Dürer's Fever.

Albrecht Dürer's illness has often been interpreted through a splenic and malarial framework, largely because of the famous self-portrait in which he points to a painful area on his left side and refers to a "yellow spot." Yet this reading may be too organ-centered. The image may not simply localize pain; it may depict a true cutaneous lesion. Once the malaria hypothesis is weakened, the conjunction of recurrent fever, constitutional decline, and a painful marked lesion becomes newly legible through the modern spectrum of late-onset autoinflammatory disease, especially VEXAS syndrome and relat

Introgression Shapes Genetic Variation Associated With Humid Environments in Aedes aegypti Mosquitoes From Saudi Arabia.

Introgression between genetically distinct populations generates variation that may promote adaptation to diverse environmental conditions. In the mosquito Aedes aegypti, the forest-associated subspecies Aedes aegypti formosus (Aaf) underwent a domestication process approximately 5000 years ago, giving rise to the globally invasive, human-associated arbovirus vector Aedes aegypti aegypti (Aaa). Despite extensive human-mediated transport, the two subspecies remain largely distinct, with admixture documented mainly in coastal Africa and Saudi Arabia. We examined Saudi Arabian Ae. aegypti across
