Dengue infection remains a major global health burden, ranging from asymptomatic infection to life-threatening severe dengue, which is characterized by increased vascular permeability and leakage of plasma from the intravascular compartment into the extravascular space. Plasma leakage during the critical phase of dengue illness, which follows the febrile phase, is a major contributor to disease severity and adverse clinical outcomes. Risk stratification remains challenging because clinical warning signs and routine laboratory abnormalities may become apparent only after vascular leakage has be

Serological diagnosis of neglected arboviruses remains hindered by extensive intra-genus cross-reactivity, compromising both assay specificity and epidemiological accuracy. In this study, we focused on yellow fever virus (YFV) as a model system to design and validate a generalizable de novo protein design framework integrating evolutionary genomics with structural modeling to rationally engineer diagnostic antigens with reduced cross-reactivity and improved cost-effectiveness. Through comprehensive evolutionary mapping over time of the YFV polyprotein, we identified conserved yet discriminator

Dengue virus (DENV), an enveloped mosquito-borne flavivirus, is the causative agent of dengue and prevalent in tropical and subtropical regions. Severe cases can progress to dengue hemorrhagic fever or shock syndrome, posing significant public health challenges. In the absence of specific antiviral drugs, antimicrobial peptides (AMPs) have emerged as promising therapeutic agents against DENV due to their broad-spectrum antibacterial and antiviral properties. This study investigated the synthetic AMP A4S7, which effectively inhibited DENV-2 RNA and protein expression and reduced viral progeny p

Bundibugyo virus disease (BVD), an Ebola virus species with no licensed vaccine or therapeutic, reemerged in May 2026 as a cross-border outbreak in Uganda and the Democratic Republic of the Congo. During a 2-day workshop, July 8-9, 2026, we conducted an early action review of the outbreak response using the 7-1-7 framework (7 days to detect, 1 day to notify, 7 days to complete early response actions) to assess timeliness and identify bottlenecks and enablers across 9 response pillars. Uganda declared its outbreak on May 15, 2026; by July 8, the country had recorded 20 confirmed cases (15 impor

BACKGROUND: Arboviral surveillance in Africa is limited by fragmented diagnostic capacity and insufficient integration of molecular detection with genomic epidemiology, particularly in settings where dengue (DENV), Zika (ZIKV), and chikungunya (CHIKV) viruses co-circulate and present with overlapping clinical syndromes.
METHODS: We conducted a molecular surveillance study across multiple regions in Senegal, including samples collected from 367 individuals with febrile or nonfebrile illness. A tiered workflow was implemented using multiplex reverse transcription quantitative polymerase chain r

Hepacivirus bovis (BovHepV) has been detected globally since its initial description in 2015, but its transmission routes remain largely unknown. Arthropod-borne or vertical transmission is a common route for several livestock viruses, including Rift Valley fever virus and bluetongue virus. Moreover, many members of the genus Orthoflavivirus (family Flaviviridae) are transmitted by hematophagous arthropods, raising the possibility that hepaciviruses such as BovHepV, which also belongs to the Flaviviridae family, could share similar transmission routes. In this study, we examined possible trans

BACKGROUND: Dengue virus (DENV) is considered an emerging threat to blood safety. While transfusion-transmitted DENV (TT-DENV) cases have been documented, TT-DENV has never been reported in Chinese mainland. In this study, a retrospective investigation was conducted after two patients developed dengue fever (DF) following apheresis platelet (PLT) transfusion from the same healthy donor.
METHODS: Blood samples from the recipients and the donor were tested for DENV RNA by nucleic acid testing (NAT). Nonstructural protein 1 (NS1) antigen and anti-DENV immunoglobulin M/G (IgM/IgG) antibodies were

Anaplasma capra is a recently discovered bacterial tickborne pathogen that was first identified in goats in 2012 and later in humans in China in 2015. We detected and genetically characterized A. capra in adult patients with suspected Crimean-Congo hemorrhagic fever (CCHF) in Turkey during the 2022 outbreak. We screened all patients for A. capra by using nested PCR targeting the gltA and groEL genes. We confirmed CCHF by molecular and serological tests. Of 263 patients, 6 (2.28%) were positive for A. capra. Sequence analysis revealed 84.16%-100% gltA gene similarity and 90.24%-100% groEL gene

Dengue continues to pose a major global threat, infecting nearly 390 million people annually. Recognizing the importance of studying vector biology and therein vector competence ( v c ), recent research focuses on mosquito parameters to inform transmission modeling and vector control strategies. This study models relationship between Aedes vectors and dengue pathogens, highlighting v c as a key driver of within-vector infection dynamics and endemic persistence. Using a predator-prey framework, we show that endemic conditions emerge naturally from the biological interplay between vector's str

Pathogen genomic analysis is central to tracking, understanding and containing outbreaks1-13, but the complexity and cost of state-of-the-art phylogenetic tools limit global access and impact. Here we introduce Delphy, an exact reformulation of Bayesian phylogenetics14-17 designed to transform its speed, scalability and accessibility while retaining Bayesian state-of-the-art accuracy. Delphy's central data structure, an explicit mutation-annotated tree, takes advantage of the high sequence similarity of large-scale epidemic datasets18-20 for efficient tree exploration and convergence. BACKGROUND: In Türkiye, where both Crimean-Congo Hemorrhagic Fever (CCHF) and brucellosis are endemic, these infections share common presenting symptoms, including fever and musculoskeletal complaints, as well as overlapping laboratory findings such as thrombocytopenia and elevated liver enzymes, making early differential diagnosis challenging. Given their distinct infection control and management requirements, this study aimed to evaluate the diagnostic performance of routine hematological and biochemical parameters for differentiating these infections before confirmatory test results become

West Nile virus (WNV) is an emerging/re-emerging flavivirus representing one of the major global causes of mosquito-borne encephalitis, yet no approved therapeutics or vaccines currently exist. To characterize the kinetics of WNV-specific immunity, we evaluated the humoral response in a longitudinal cohort of 55 WNV lineage 1-infected blood donors up to 1 year. We demonstrate that WNV infection elicits robust and durable binding and neutralizing antibodies. Using a biphasic decay model, we estimated the half-life of neutralizing antibodies to be 37 days during the rapid decay phase and 472 day

INTRODUCTION: Toscana virus (TOSV) and Leishmania species are sand fly-borne pathogens that pose a significant public health threat and an increasing risk of exposure in Europe. The objective of this study was to identify the phleboviruses circulating in an area endemic for sand fly-borne pathogens and to estimate infection prevalence in wild sand fly populations. We also aimed to analyse co-infections with L. infantum and the symbiotic bacterium Wolbachia.
METHODS: The study was conducted in southern Spain. Sand flies were collected using CDC light traps, morphologically identified and DNA/R

The ongoing Orthoebolavirus bundibugyoense-Bundibugyo virus (BDBV)-epidemic is the worst Ebolavirus outbreak in a decade and is rapidly expanding, with >4600 suspected cases in the Democratic Republic of Congo (DRC) alone. It has caused 2186 deaths and spread to neighboring countries. As BDBV has no approved vaccine, timely response will require adapted use of existing vaccines approved for other ebolaviruses and expedited development of novel vaccines targeted toward BDBV. Access to vaccines is fundamental to public health emergency response and generally depends on two expert bodies in each

BACKGROUND: West Nile virus (WNV) is an emerging pathogen in Europe with an unprecedented northward expansion. In Romania, the virus has been endemic for at least three decades, and human infection cases are registered yearly, sometimes culminating in outbreak events. Bucharest, the capital city of Romania, is the largest urban area of the country, encompasses a multitude of mosquito and bird habitats, and has been continuously reported as an active hotspot for WNV transmission. The circulation of lineage 2 has been documented in Bucharest since 2010 in mosquito and human populations. Our stud
