Assessing the Vector Competence for Chikungunya Virus in Mosquitoes.

Chikungunya virus (CHIKV) is a human pathogenic, mosquito-borne virus (arbovirus), which is causing epidemic outbreaks among human populations in Africa, Asia, Europe, and South- and Central America including the Caribbean. The development of novel approaches to prevent mosquito transmission of the virus in the field requires a detailed study of CHIKV interactions with its mosquito vectors in the laboratory. In this chapter, we describe how to prepare Aedes aegypti mosquitoes for the infection with freshly cultivated CHIKV. We also describe quantitative and qualitative viral detection assays i

Analysis of Laboratory Testing for Imported Dengue Fever in the Republic of Korea, 2020-2025.

Objectives: Dengue fever is an acute, mosquito-borne disease that is widely endemic in tropical and subtropical regions. While no autochthonous cases have been reported in the Republic of Korea, imported cases continue to occur. This study analyzed laboratory testing data from 2020 to 2025 to support the surveillance of imported mosquito-borne infections. Methods: A total of 2,904 laboratory test requests for dengue fever submitted to the Korea Disease Control and Prevention Agency and 17 provincial Institutes of Health and Environment were reviewed. Laboratory diagnosis was performed using re

MicroRNA Responses Associated with Vector-Borne Diseases in Humans and Arthropod Vectors: A Systematic Review.

Vector-borne diseases (VBDs) represent a significant public health challenge. Early and accurate diagnosis is crucial for effective disease management and control, with host-encoded microRNAs (miRNAs) emerging as promising disease biomarkers. This systematic review surveys research conducted on miRNA responses in disease vectors and human hosts of VBDs. A systematic search of four databases and manual searching identified 93 qualifying articles. Numerous VBDs were described, with the most common focus on dengue (21%; n = 15) and Chagas disease (11%; n = 8). Twenty-eight in vivo human studies h

Rift Valley Fever at the Crossroads of Ecology Mobility and Fragmented Health Governance.

Rift Valley fever (RVF) remains one of the most recognisable yet persistently elusive zoonotic diseases in Africa, despite decades of scientific advances in virology, epidemiology, and early warning. While RVF dynamics are well characterised in eco-epidemiological terms, its continued emergence, amplification, and persistence suggest that biological complexity alone does not fully explain ongoing transmission patterns. This review adopts a socio-ecological One Health systems perspective to examine how interactions between eco-epidemiological processes, livestock mobility, and health governance

Seroepidemiological investigation and risk factors of Q fever and Rift valley fever in livestock in Bangladesh.

Rift Valley fever (RVF) is an emerging and re-emerging mosquito-borne zoonosis of major economic, public health, and veterinary importance, often causing great population-level impact. Q fever remains a neglected zoonotic disease of considerable public health and veterinary significance, responsible for notable economic losses for livestock farmers. Despite their significance, the epidemiology of RVFV and Coxiella burnetii (Q fever) remains poorly understood, with little evidence on their seroprevalence and associated risk factors. We conducted a cross-sectional study to estimate the seropreva

A global database of West Nile virus host prevalence and competence.

West Nile virus (WNV) is one of the most widespread arboviruses globally and is maintained primarily through a bird-mosquito-bird transmission cycle, while other vertebrates play more limited roles. Host contributions to transmission depend on both infection evidence in natural populations (reflecting exposure and susceptibility) and reservoir competence, determined by the magnitude and duration of viraemia sufficient to infect mosquitoes. Despite extensive surveillance and experimental research, no comprehensive, standardised resource has integrated evidence on host exposure and infection in

Wesselsbron virus at the livestock-human interface: A practical guide to disease recognition and diagnosis.

Wesselsbron virus (WSLV) is a mosquito-borne Orthoflavivirus of the yellow fever virus (YFV) serocomplex, widely distributed across sub-Saharan Africa. It is neglected, and its true impact is likely underestimated. WSLV causes abortion, congenital malformations, and neonatal mortality in sheep, the primary recognized vertebrate host, but infections have also been documented in goats, cattle, camels, wildlife, and humans. The virus is maintained in complex cycles predominantly involving Aedes mosquitoes and incompletely defined domestic and wild vertebrate hosts. Direct infection via the placen

SNX17 facilitates entry and infection of Rift Valley fever virus and related bunyaviruses.

Bunyaviruses are a group of multisegmented, negative-sense RNA viruses. Rift Valley fever virus (RVFV) is a representative member that can cause sporadic human outbreaks. We previously identified low-density lipoprotein receptor-related protein 1 (LRP1) as a host entry factor for RVFV and several bunyaviruses. This screen also identified additional factors as proviral host proteins, including sorting nexin 17 (SNX17). Here, we describe efforts to characterize SNX17 in bunyaviral infection. Our findings reveal that the removal of SNX17 results in a substantial reduction of RVFV infection in SNX

Interactions between Tick-Borne Encephalitis Virus Nonstructural Protein 1 and Blood-Brain Barrier Tight Junction Proteins: Potential Clues to Strain-Specific Neuropathogenicity.

Tick-borne encephalitis virus (TBEV) invades the central nervous system (CNS) through strain-specific mechanisms that remain poorly understood. For mosquito-borne orthoflaviviruses such as dengue and yellow fever viruses, the nonstructural protein 1 (NS1) has been shown to disrupt endothelial barrier integrity by targeting tight junction proteins (TJPs), facilitating viral neuroinvasion. However, comparable mechanisms in TBEVs remain largely unexplored. Here, we investigated the potential interaction of NS1 from high (Hypr)- and low (Vs)-pathogenic TBEV strains with different blood-brain barri

Zoonotic arboviruses and associated risk factors in donkeys from northeastern Algeria.

Rift Valley fever virus (RVFV), Crimean-Congo hemorrhagic fever virus (CCHFV), and several zoonotic flaviviruses represent an emerging threat to animal and human health. In Algeria, particularly in the northern regions, data on the circulation of these viruses remain limited, especially for RVFV and CCHFV. In this context, we aimed to assess the seroprevalence of these arboviruses among donkeys across three provinces in northeastern Algeria and to identify factors associated with seropositivity. Between 2019 and 2024, 183 serum samples were collected and analyzed by ELISA. Binary logistic regr

Facilitators and barriers to community-based livestock abortion reporting for Rift Valley Fever surveillance in Uganda: A COM-B analysis.

Rift Valley Fever Disease (RVFD) causes substantial economic losses via livestock abortion storms, yet community-based abortion reporting for early warning remains suboptimal in endemic settings. This study analysed the behavioural determinants of livestock abortion reporting to inform RVF surveillance interventions in Uganda. We conducted a cross-sectional study using qualitative data collection methods in Isingiro District, Uganda, employing the Capability, Opportunity, Motivation-Behaviour (COM-B) framework. We conducted 29 key informant interviews with national/district policymakers and te

Early-warning surveillance of West Nile and Usutu viruses in wetland and larval habitat waters and in mosquito excreta using digital PCR.

West Nile virus (WNV) and Usutu virus (USUV) are mainly avian mosquito-borne pathogens, increasingly cause human and equine disease in temperate regions. Mostly asymptomatic, current surveillance in France is reactive, based on clinical and veterinary reports, delaying signals for vector control and preventative interventions. We implemented in Southern France, in 2024 and 2025, a proactive surveillance methodology combining molecular xenomonitoring of mosquito excreta and environmental water detection of viral RNA in wetlands and mosquito larval habitat, through a multiplex reverse transcript

Type I IFN coordinates resistance and tolerance against Zika virus disease by inducing NK cell recruitment and limiting neutrophil-driven neuropathology in the brain.

Zika virus (ZIKV) neurological disease arises from both direct viral cytopathic effects in the central nervous system (CNS) and host-driven neuroinflammation, with disease outcomes determined by the balance between antiviral resistance and disease tolerance. Type I interferons (IFN-I) are well-established antiviral mediators that also regulate neuroinflammation and preserve blood-brain barrier (BBB) integrity. Previous in vitro studies showed that the African-lineage ZIKV strain MR766 (ZIKVMR766) is more resistant to IFN-I-mediated antiviral effects than the Asian-lineage strain PE243 (ZIKVPE2

Evidence of West Nile Virus Infections in Wild Boars (Sus scrofa) in the Netherlands, 2018-2021.

INTRODUCTION: Orthoflaviviruses are emerging in Europe and have potential significant impact on animal and public health. Usutu virus (USUV) and West Nile virus (WNV) have been first detected in the Netherlands in 2016 and 2020, respectively. Wild boars (Sus scrofa) have been suggested as an interesting population to detect the circulation of orthoflaviviruses. We aim to provide epidemiological insights of USUV and WNV infections in wild boars in the Netherlands. We further explore the interpretation of serological results, taking into account cross-reactivity patterns. METHODS: A total of 15

Seroprevalence and durability of West Nile virus (WNV) neutralizing antibody responses in asymptomatic adults.

OBJECTIVE: West Nile virus (WNV) is a mosquito-borne pathogen of growing public health concern that is endemic in Israel and has been responsible for intermittent outbreaks. The seroprevalence and durability of functional antibody responses in asymptomatic WNV-infected populations remain unclear. METHODS: We screened a large cohort of asymptomatic healthcare workers (HCWs) for WNV IgG and neutralizing antibody titers. Predictors of seropositivity and neutralization levels were assessed using multivariable regression. Among seropositive individuals, longitudinal samples, collected up to 44 mon

Confidence Intervals for a Proportion Estimated From Pooled Samples Based on Firth's Corrected Score.

Interval estimation of a proportion estimated from testing of pooled samples has been studied by a variety of researchers, who have examined both exact and asymptotic methods. Recent work in point estimation has seen the use of Firth's correction to maximum likelihood estimation to reduce bias effectively. We consider the Firth-corrected score statistic, using either expected or observed information, as the first derivative function of a penalized likelihood and develop score-based confidence intervals wholly within this framework. We evaluate their performance, comparing them by information t
