In Vitro and In Vivo Models for Drug Development Against Two Hemorrhagic Hareavirales: Rift Valley Fever and Crimean Congo Hemorrhagic Fever Viruses.

Rift Valley fever virus (RVFV) and Crimean-Congo hemorrhagic fever virus (CCHFV) are designated by the World Health Organization as priority pathogens due to their epidemic potential, zoonotic transmission, and the absence of licensed therapeutics or vaccines. The development of effective antivirals critically relies on robust in vitro and in vivo models; however, progress is limited by the requirement for high-containment facilities. In this review, we provide a comprehensive overview of the experimental models currently available for RVFV and CCHFV, ranging from cell-based assays to animal m

Evaluation of Rift Valley fever vaccine candidates in pregnant rodent models.

BACKGROUND: Rift Valley fever virus (RVFV) causes significant disease in humans and livestock. Immunogenicity of candidate vaccines rMP-12 and RVax-1 show promise, but their placental tropism and potential effects on fetal outcomes remain incompletely understood, particularly across different animal models. Understanding species-specific placental replication is essential to optimize vaccine safety in pregnant populations. OBJECTIVE: To evaluate the placental tropism and fetal outcomes of RVFV candidate vaccines rMP-12 and RVax-1 in pregnant Sprague-Dawley (SD) rats and C57BL/6 mice. METHODS

Continental assessment of Rift Valley fever in dromedary camels: Systematic review and Meta-analysis, 1977-2025.

Rift Valley Fever (RVF) is a viral, mosquito-borne disease affecting livestock, including camels, and humans. The virus, belonging to the genus Phlebovirus, is primarily transmitted by mosquitoes of the genera Aedes and Culex, which act as both reservoirs and vectors, facilitating viral spread during outbreaks. This systematic review and meta-analysis aimed to provide a comprehensive synthesis of RVF epidemiology in camels across Africa from 1977 to 2025, estimate the true seroprevalence, and assess potential risk factors associated with infection. Following PRISMA guidelines, an exhaustive se

Pathogenicity of Rift Valley fever virus: Organ-specific clinical outcomes and hidden drivers of virulence - a narrative review.

Rift Valley fever virus (RVFV) is a mosquito-borne phlebovirus posing major threats to human and animal health. Despite decades of research, the determinants governing diverse clinical manifestations remain incompletely understood. This narrative review examines four central facets: i) exposure route impacts on disease outcome; ii) field strain diversity and organ-specific virulence; iii) genome reassortment driving viral evolution; and iv) organ-specific pathology including the overlooked placental/retinal infections. Differences among infection routes, viral strains, and host species complic

[Recombinant expression of rift valley fever virus nucleocapsid protein and generation of rabbit polyclonal antibodies].

Objective Prokaryotic expression and purification of the Rift Valley fever virus (RVFV) nucleocapsid protein (NP) were performed to establish a basis for serum NP antibody detection and to produce specific polyclonal antibodies against NP. Methods The constructed pET28a-ZH501-NP prokaryotic expression plasmid was transformed into BL21(DE3) competent cells, and the expression of the recombinant protein NP was induced by isopropyl-β-D-thiogalactoside (IPTG). The recombinant protein NP was purified using HisTrapTM HP and mixed with Freund's complete and incomplete adjuvants to immunize New Zealan

Preclinical efficacy of a favipiravir and nitazoxanide combination against Rift Valley Fever Virus.

Emerging and re-emerging viruses represent a major global health challenge. To address this, the WHO encourages the development of broad-spectrum medical countermeasures. This study evaluated the efficacy of repurposed broad-spectrum molecules against Bunyaviricetes viruses and more specifically Rift Valley fever virus (RVFV). In vitro, favipiravir and nitazoxanide showed low effective doses with an additive effect in combination against several viruses, including RVFV, Crimean-Congo hemorrhagic fever virus and Hantaan virus. In a murine model of severe RVFV infection, combination treatment re

The RNA helicase eIF4A as a novel target in insect cells to combat arboviral infections.

Arthropod-borne viruses cause major global health burdens, yet few vaccines or antivirals exist. Targeting host factors required for viral replication offers a promising approach. The DEAD-box RNA helicase eIF4A, a core component of the translation initiation complex eIF4F, unwinds structured 5' UTRs and is therefore critical also for the translation of many viral RNAs. The compound classes rocaglates and pateamines are potent eIF4A inhibitors in mammalian cells. Here we show that the natural rocaglate silvestrol strongly inhibits Rift Valley fever virus replication (RVFV) in human cells witho

Epidemiological and molecular characterization of Rift Valley fever outbreak in livestock in Burundi, May - November 2022.

An outbreak of Rift Valley fever (RVF) was officially reported for the first time in Burundi on 10th May 2022. The outbreak originated in the northern provinces and progressively spread to other regions of the country. This study presents (i) epidemiological investigations that were carried out through a countrywide emergency response and (ii) the characterization of the genotype of the RVF virus that caused the outbreak through phylogenetic analyses. Field teams visited each affected farm, collected data on observed syndromes, species and number of animals affected, farm's locations, and herd

Sensitivity analysis of factors influencing the ecology of mosquitoes involved in the transmission of Rift Valley fever virus.

Vector-borne diseases are a major global health concern, with Rift Valley fever (RVF) serving as a key example due to its impact on both human and animal health. Predicting and controlling such diseases requires understanding how environmental factors shape mosquito ecology. Due to mosquito abundance, distribution, and behavior being influenced by ecological conditions, identifying the drivers of these dynamics is essential for anticipating transmission risk. This study aims to assess the sensitivity of ecological factors governing the ecology of mosquitoes, using a deterministic, compartmenta

Infectious Agents of Non-Malarial Febrile Illnesses in 20 African Countries, 2019-2024: A Systematic Review and Meta-Analysis.

BACKGROUND: The growing attention to non-malarial febrile illnesses (NMFIs), driven by the decline in malaria cases across the African continent, has revealed pathogens previously overshadowed by the burden of malaria. Recent outbreaks, the resurgence of diseases and the rising antimicrobial resistance burden, compounded with data gaps in previous studies, have underscored the need to closely examine this area in recent times. METHODS: We searched for articles through PubMed, Scopus, ScienceDirect and Web of Science databases to identify the common causative agents of NMFIs and estimate their

Systematic review and meta-analysis for the occurrence of Crimean-Congo hemorrhagic fever, Tularemia, and Rift Valley fever in pastoralist systems in Africa.

INTRODUCTION: Pastoralism is a livestock production system practiced in areas with harsh environmental conditions and is characterized by low investment in animal health, increasing the risk of animal disease outbreaks. Zoonotic diseases such as Crimean-Congo hemorrhagic fever (CCHF), Rift Valley fever (RVF), and tularemia may spread through livestock product value chains. METHODS: Published studies on the three diseases were systematically searched in PubMed, Web of Science, Scopus, and Google Scholar using predetermined search terms. Early outbreak detection in endemic settings using a novel method applied to sparse Rift Valley fever incidence data.

Rift Valley Fever (RVF) is a mosquito-borne zoonotic disease often linked to climatic factors. However, identifying the onset and duration of outbreaks can be hindered by high numbers of zero counts in sparse surveillance data. We analyzed monthly RVF data from Kenya (2015-2022), compared the performance of widely used outbreak-detection methods, and developed Negative Binomial models augmented with a hidden Markov process and zero-inflation to address data sparsity. Our proposed framework estimates monthly probabilities of transitioning between endemic and epidemic phases, improving outbreak

Dissecting placental host-pathogen interactions: Rift Valley fever virus infection in early human trophoblast stem cells.

Rift Valley fever virus (RVFV) is a mosquito-borne Phlebovirus and zoonotic pathogen affecting maternal-fetal health. Vertical transmission is linked to miscarriage and severe fetal outcomes, but mechanisms of placental pathogenesis remain unclear. We used first-trimester human trophoblast stem cells (hTSCs) to model infection at the maternal-fetal interface. Immunofluorescence, qRT-PCR, western blotting, and single-cell transcriptomics showed that hTSCs are highly susceptible to RVFV. Strand-specific viral transcriptomics confirmed the ambisense S segment and revealed preferential transcripti

A Newly Established ELISA for the Surveillance of Rift Valley Fever in Dromedary Camels and Their Owners, Kenya 2018.

In 2024 Kenya had a population of 4.78 million camels that contributed to the livelihoods of pastoralist communities in northern Kenya. Previous studies in Kenya, Saudi Arabia and eastern Africa demonstrated high seroprevalence of Middle East respiratory syndrome coronavirus (MERS-CoV)-specific antibodies in dromedary camels, as well as sporadic transmission of MERS-CoV from camels to humans. Based on the MERS-CoV data and the very close contact between owners and their camels in northern Kenya, we speculated that camels may also transmit other zoonotic viruses, such as Rift Valley fever virus

Determining factors of the blood feeding behaviour of mosquito vectors of West Nile and Rift Valley fever viruses in Madagascar.

UNLABELLED: In Madagascar, the health burdens caused by West Nile virus (WNV) and Rift Valley Fever virus (RVFV) vary across regions, likely resulting from the variations of their vector capacity. This study aimed to assess the degree of vector-hots contact and to develop statistical models to identify factors influencing host preferences. Bimonthly mosquito collections were performed in 25 sites of Madagascar from February to October 2019. Engorged mosquitoes were collected from indoors and outdoors resting locations within each site. Blood meal identification was performed using host-specifi

Computer-Aided Discovery of Small-Molecule Inhibitors of Pathogenic New World Arenavirus Entry and Replication.

Pathogenic New World arenaviruses (NWAs), including Junín (JUNV) and Machupo (MACV) viruses, rely on host-virus entry processes that represent attractive points for antiviral intervention. Guided by the known use of human transferrin receptor 1 (hTfR1) by several NWAs for cell entry, we conducted a structure-based virtual screening campaign targeting the MACV GP1-hTfR1 interaction interface to identify small molecules capable of inhibiting early infection. From an in silico screen of commercially available drug-like compounds, 25 candidates were selected and tested in cell-based assays, yieldi

Cross-Sectional Analysis of Serologic Response to Arthropod-Borne and Hemorrhagic Fever Viruses in Ghanaian Livestock Herders.

Zoonotic diseases account for more than 60% of emerging infectious diseases, and they are the leading cause of pandemics. As humans and livestock become increasingly transient and the environment and climate change, disease vectors expand into previously untouched geographical regions, spreading pathogens. In this study, we assessed the seroprevalence of arthropod-borne and hemorrhagic fever viruses of similar clinical presentation and endemicity in high-risk populations in Ghana (animal handlers). Using a microneutralization assay, we compared total IgG prevalence with live virus neutralizing
