Seroprevalence and Geographical Distribution of Rift Valley Fever in Small Ruminants in Mauritania: Evidence of Endemic Circulation and Regional Risk Hotspots.

Rift Valley fever (RVF) is a mosquito-borne viral zoonosis that causes severe illness in livestock and humans, with significant economic and health repercussions. Mauritania is considered an RVF focus in West Africa. A cross-sectional survey was conducted in 2023 in 12 of Mauritania's 15 provinces. Serum samples from 849 small ruminants (428 goats and 421 sheep) were analyzed by ELISA for IgG and IgM antibodies against RVF virus. Logistic regression analyses were performed to assess associations between seropositivity and species, age, sex, season, and geographic location. In total, 14.1% of t

NanoLuc-based microneutralization assay for high-throughput detection of neutralizing antibodies against rift valley fever virus.

Plaque reduction neutralization tests (PRNTs) are the gold standard for measuring neutralizing antibodies but are labor-intensive, low-throughput, and subject to variability from manual plaque counting. We developed a NanoLuc-based microneutralization (mNT) assay using rMP12-NanoLuc that quantifies luminescence in culture supernatants as a surrogate for infection. The assay runs in 96-well plates without overlay, fixation, or staining, using 35 focus-forming unit (FFU) per well and a 40 hour readout with back-titration to ensure accuracy. mNT titers from vaccinated animal sera showed strong co

Climate-resilient livestock systems in Sub-Saharan Africa: a one health systems framework integrating pastoralism, animal health, and sustainable development.

Pastoral livestock systems in Sub-Saharan Africa face accelerating climate threats that compromise food security, animal health, and human well-being across millions of rural households. Rising temperatures, recurrent droughts, erratic rainfall, and floods degrade rangeland productivity, restrict pastoral mobility, and reshape disease ecology, driving outbreaks of trypanosomiasis, tick-borne infections, and Rift Valley fever. These biophysical pressures interact with governance failures, farmer-herder conflict, and weak veterinary infrastructure to generate compound vulnerabilities that extend

Rift Valley fever virus infects trophoblast cell lines in an LRP1-dependent manner.

Rift Valley fever virus (RVFV) is a mosquito-borne phlebovirus that poses a growing global threat, causing severe disease in humans and livestock, including hepatitis, haemorrhage and encephalitis. Documented cases of vertical transmission in humans and livestock highlight risks during pregnancy. Although RVFV can directly infect the placenta, how it enters trophoblast cells remains unclear. Here, we investigated the role of low-density lipoprotein receptor-related protein 1 (LRP1), a known RVFV entry factor, in human trophoblast infection. Using JEG-3, JAR and HTR-8/SVneo trophoblast cell lin

Persistent circulation of Rift Valley fever virus lineage C in Rwanda, 2022-2025.

Rwanda has experienced recurrent Rift Valley fever virus outbreaks in the last decade. In this study, we investigated whether these outbreaks resulted from repeated introductions or sustained local circulation. We generated RVFV whole-genome sequences from livestock samples collected between 2022 and 2025 using Nanopore sequencing. Genomic analyses indicated the outbreaks resulted from sustained local circulation of lineage C rather than repeated introductions, suggesting ongoing transmission likely driven by sporadic spillover. This study underscores the importance of continuous genomic One H

Climate Change, Urbanization, and the Emerging Urban Threat of Rift Valley Fever in Tropical and Subtropical Cities: A Narrative Review.

Rift Valley fever (RVF) is a climate-sensitive mosquito-borne zoonosis long regarded as a rural, pastoral disease, yet accelerating tropical urbanization and intensifying climate variability may be reshaping its epidemiology at the urban-peri-urban interface. This narrative review examines how global climate change and urban-specific climatic conditions jointly shape RVF virus (RVFV) vector habitats, transmission, and burden in tropical and subtropical cities, synthesizing 51 of 412 English-language records identified by a structured, non-systematic search of PubMed, Scopus, Web of Science, an

Development of a Novel Immune Gene Array to Assess Mosquito Gene Expression During Arbovirus Infection.

Assessing mosquito vector competence for emerging arboviruses, including the underlying determinants of infection, is a foundational line of investigation towards understanding arbovirus transmission dynamics and vector-virus interactions. We developed a novel quantitative immune gene array that measures the relative expression of 22 immune genes with functional representation across five major immune pathways using quantitative reverse-transcriptase polymerase chain reaction. Homologous primers were developed for Culex tarsalis, Cx. quinquefasciatus, and Ae. aegypti mosquitoes. As an initial

Simulating the Transmission of Rift Valley Fever Virus via Raw Milk Consumption in Inter-Epidemic Periods.

Rift Valley fever virus (RVFV) is a zoonotic, vector-borne disease affecting livestock and humans. The consumption of raw unpasteurized milk has been epidemiologically linked to RVFV exposure in humans and recognized as a potentially important non-vector route for RVFV transmission. This study aims to investigate the potential of RVFV transmission to humans through the consumption of infected raw milk. For this purpose, we conducted agent-based stochastic simulations to assess the potential for RVFV transmission through ingestion of contaminated raw milk. We created synthetic populations of co

Cross-border introduction risk of Rift Valley fever virus: surveillance evidence from intercepted livestock in southwestern Saudi Arabia.

INTRODUCTION: Rift Valley fever (RVF) is a mosquito-borne transboundary zoonotic disease with major implications for animal and public health. Following the 2000 outbreak in Saudi Arabia and Yemen, the risk of reintroduction through cross-border livestock movement remains a continuing concern. This study aimed to estimate the seroprevalence of Rift Valley fever virus (RVFV) among illegally introduced livestock intercepted in the Jazan region of southwestern Saudi Arabia and to assess the added value of combined serological and molecular testing for detecting recent infection under routine surv

Spatial heterogeneity of Rift Valley fever burden in Kenya: a 10-year One Health zDALY evaluation.

BACKGROUND: Rift Valley fever (RVF) is an endemic zoonotic disease in Kenya, with recurring outbreaks that affect both humans and livestock. Despite its impact, the full burden of RVF remains poorly quantified. This study presents the first integrated and spatially disaggregated estimates of RVF burden in Kenya using a One Health approach. METHODS: We quantified the national and subnational burden of RVF in Kenya from 2014 to 2024 using the zoonotic disability-adjusted life years (zDALYs) framework, which combines human health loss (disability-adjusted life years [DALYs]) and livestock-relate

BTX-A51 treatment modulates host innate immune responses via NF-κB and β-catenin signaling during Rift Valley fever virus infection in vitro.

Rift Valley fever virus (RVFV) is a zoonotic and mosquito-borne hemorrhagic pathogen of high pandemic potential, endemic to most countries in Africa. It most severely causes disease in small domestic ruminants and is transmitted to humans. RVFV has been declared a priority pathogen by the World Health Organization for vaccine and therapeutic development. Outbreaks of this virus are known to cause great disturbances to the economy, agriculture, food availability, and healthcare system of local communities. No U.S. FDA-approved vaccines or therapeutics exist for the prevention or treatment of RV

Discovering host-viral protein interactions in autophagy: A LIR discovery pipeline for identifying LC3-interacting region motifs in highly virulent viruses.

Hemorrhagic fever virus (HFV) infections are highly fatal, posing a significant global pandemic threat as they continue to emerge in new locations. HFVs and other highly virulent viruses (HVVs), such as Nipah virus, exploit host cell pathways, including the conserved host autophagy pathway, to promote viral replication. The Atg8/microtubule-associated protein 1 light chain 3 (LC3) proteins are necessary for autophagosome formation and maturation. Proteins interact with Atg8-family proteins through LC3-interacting region (LIR) motifs, which is a short linear motif (SLiM) found in intrinsically

Spatio-temporal dynamics of Rift Valley fever virus circulation in sentinel livestock herds (2021-2024) before the initial fatal human outbreaks in Senegal.

BACKGROUND: Multiple detections of Rift Valley fever virus (RVFV) infections in humans, animals and mosquito vectors suggest permanent circulation of RVFV in Senegal. This study investigated the spatial and temporal dynamics of RVFV circulation in central (Sahelian) and southeastern (Sudanian) Senegal from March 2021 to June 2024. METHODS: A total of 976 initially seronegative sheep and goats from sentinel herds in 10 districts and 2 areas were monitored for 40 months. Serum samples were collected quarterly and tested by ELISA, while mosquitoes were collected monthly and screened for RVFV RNA

Smartphone AI-Enabled Lateral Flow Immunoassay Platform Using Advanced Quantum Dots for Intelligent Quantitative Diagnostics.

Lateral flow immunoassays (LFIAs) are widely used in point-of-care diagnostics for their low cost and simplicity, but conventional formats rely on subjective visual readout and lack quantitative accuracy and sensitivity. Here, we present a smartphone-guided, artificial-intelligence-enhanced LFIA platform that unites a rationally engineered dual-modal nanoprobe with on-device machine learning, demonstrated for quantitative detection of Rift Valley fever virus (RVFV). The platform combines three synergistic principles: advanced magnetic quantum dot nanoparticles (AQDs) that integrate a magnetic

Rift Valley fever virus virulence factor NSs targets ATF4- and XBP1-driven unfolded protein response.

Rift Valley fever virus (RVFV) is a mosquito-borne arbovirus that infects humans and domestic ruminants, significantly impacting public health and the economy in African and Middle Eastern countries. RVFV non-structural protein NSs is a major viral virulence factor and possesses multiple functions to counteract host antiviral responses. One of its functions is to block host transcription by promoting degradation of subunits of the host transcription factor IIH (TFIIH). However, the biological significance of this function in the RVFV life cycle remains unclear. We used RVFV strain MP-12 and it

Establishment of an ex vivo vascular tissue model to investigate bunyavirus-triggered vascular endothelial injury.

UNLABELLED: Severe fever with thrombocytopenia syndrome, an emerging infectious disease caused by the tick-borne bunyavirus severe fever with thrombocytopenia syndrome virus (SFTSV), is associated with a high clinical fatality rate of 12%-50%. Vascular endothelial injury triggered by SFTSV infection has been linked to severe disease progression and mortality. Currently, a suitable model to investigate the underlying mechanisms of SFTSV-induced vascular damage is lacking. Here, we show that the ex vivo human vascular tissue can be used as a model for studying bunyavirus-triggered endothelial in
