Rift Valley Fever at Europe's Risk Frontier: Climate, Vectors, and One Health Preparedness.

Background: Rift Valley fever (RVF) is a livestock-amplified, mosquito-borne zoonosis caused by Rift Valley fever virus (RVFV). Continental Europe has no established RVFV transmission, but a rare introduction could have major consequences if susceptible ruminants amplify infection before detection. Methodology: This critical narrative review synthesizes 69 scientific and institutional sources identified through structured PubMed/MEDLINE and Europe PMC searches, citation verification, and qualitative appraisal. Findings: European preparedness should distinguish probability from impact and weigh

Entomo-virological investigation in urban forest fragments and intradomiciles during a dengue outbreak in Salinas, MG, Brazil.

BACKGROUND: Mosquitoes (Diptera: Culicidae) are among the most important disease vectors worldwide. Several species exhibit high levels of anthropophily and are frequently found in human dwellings and forest fragments near urban areas. OBJECTIVES: In this integrative study combining mosquito collection, viral detection, and ecological analyses, the assemblage of diurnal mosquitoes was investigated across three distinct environments - intradomiciles, and two distinct urban forest fragments (UFFs) - during a dengue outbreak in the city of Salinas, Minas Gerais, Brazil. METHODS: Sampled mosquit

Impact of metabolic comorbidities on the clinical course of West Nile virus infection.

BACKGROUND: Italy has experienced a marked increase in the incidence of human infections caused by West Nile virus (WNV). Metabolic and other comorbidities, particularly hyperglycemia, are associated with unfavorable outcomes in infectious diseases. This study aimed to investigate the association between metabolic comorbidities, with a specific focus on blood glucose (BG) levels at hospital admission, and the clinical course of WNV infection. METHODS: 34 patients (22 males, 12 females, mean age 69 ± 14 years) with confirmed WNV infection were included. Clinical history, metabolic comorbiditie

Utility of blood biomarkers in inpatients with West Nile virus infection: an exploratory study.

BACKGROUND: West Nile virus (WNV) infection is an emerging public health concern in Southern Europe and may present with heterogeneous clinical phenotypes, ranging from predominantly neurologic manifestations to systemic inflammatory and septic presentations. While lymphocytopenia and immune dysregulation are well-recognized features of WNV infection, data on routine hematological parameters and composite inflammatory indices, and their potential association with different clinical presentations, remain limited. METHODS: We retrospectively analyzed a multicenter Italian cohort of 30 consecuti

Diagnosis and Surveillance of West Nile Virus Infection in Horses: Current Methods, Challenges, and Future Directions.

West Nile virus (WNV) is a mosquito-borne flavivirus of growing importance for both human and equine health in Europe. Horses are highly susceptible to neurological disease and, because they share ecological exposure with humans, they represent valuable sentinels for detecting local viral circulation within a One Health framework. However, diagnosis of WNV infection in equines is complicated by the short and low-level viraemia, which limits the sensitivity of molecular assays, and by serological cross-reactivity with related flaviviruses and the confounding effects of vaccination. In this narr

Disseminated Intracranial and Spinal Dysembryoplastic Neuroepithelial Tumor: A Case Report with a Systematic Review.

Background: Disseminated intracranial and spinal dysembryoplastic neuroepithelial tumors (DNETs) are exceptionally rare, with only five prior cases reported. Methods: This study presents a case of a 47-year-old woman with DNETs in the right hippocampus and lumbar spine, treated surgically, and systematically reviews the existing literature (total n = 6). Results: While primary tumors occurred in both locations, secondary lesions were predominantly spinal (83%). The key features included frequent obstructive hydrocephalus (67%, often requiring shunting) but rare epilepsy (n = 1, 17%). Two fatal

Acute Flaccid Paralysis Followed by Guillain-Barré Syndrome in West Nile Virus Infection: A Case Report.

Neuroinvasive West Nile virus infection can present with a broad spectrum of neurologic manifestations, creating diagnostic and management challenges. We report a case that initially presented with acute flaccid paralysis and subsequently evolved to include features consistent with Guillain-Barré syndrome during hospitalization. This case highlights the dynamic nature of neuromuscular involvement in neuroinvasive West Nile virus infection and the importance of ongoing clinical reassessment as neurologic findings evolve.

Quantitative analysis of chikungunya virus with special emphasis on biosensors: A review.

Chikungunya is a viral fever caused by a single stranded RNA virus of the genus alpha virus of the family Togaviridae and transmitted to human by the bite of Aedes aegypti mosquito. The incubation time frame of this virus is short, enduring about 2-4 days. Chikungunya virus disease is typically self-constrained, non-lethal, with fever settling inside a couple of days. A number of conventional methods such as ELISA, RT-PCR and PCR are available for determination of chikungunya virus. Besides the advantages of being highly sensitive and selective, these methods have some drawbacks like time-cons

Epidemiological and genomic features of chikungunya virus disease in travellers returning from Cuba, September 2025 to January 2026: a GeoSentinel analysis.

During September 2025-January 2026, 111 travellers (61 female/50 male; median age:  53 years) who acquired chikungunya virus (CHIKV) in Cuba were reported to GeoSentinel. Upon return, 64.2% (70/109) were potentially viraemic. Only 8.2% (9/98) had received pre-travel consultations. The CHIKV was of East-Central-South Africa genotype, closely related to Brazilian strains. International travellers can serve as arboviral outbreak sentinels and, if viraemic, risk introducing CHIKV into areas with established Aedes spp. vectors. Their effective surveillance can trigger adequate public health respons

Intranasal infection of Rift Valley fever virus in the ferret model suggests multiple routes of neuroinvasion with consequential encephalitis and ophthalmitis.

Rift Valley fever virus (RVFV) causes haemorrhagic fever, ophthalmitis, and in severe cases, encephalitis in humans. Despite the well-recognised neurotropism, the mechanisms of central nervous system (CNS) invasion remain poorly understood. We investigated RVFV ingress into the CNS and resulting neuropathogenesis following intranasal inoculation in ferrets (Mustela putorius furo) (n = 15). At 3 DPI, before RVFV dissemination to the CNS, mild multifocal suppurative rhinitis and multifocal interstitial pneumonia were detected in the nasal cavity and lungs respectively, supported by detection of

From Hepatitis to Encephalitis: Neuroinvasion and Antiviral Development in Rift Valley Fever Virus Infection.

Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic pathogen that causes substantial livestock losses and a range of severe human illnesses, including hemorrhagic disease, hepatitis, retinitis, vision loss, and delayed encephalitis. Despite its public health and One Health importance, no approved RVFV-specific antiviral therapy or licensed human vaccine is available. Therapeutic development is challenged by the progression of RVFV disease from acute viremia and hepatic injury to delayed neurologic and ocular complications, highlighting the need for countermeasures that protect both sys

Rational Design of a Chimpanzee Adenoviral-Vector Vaccine Against Yellow Fever Through the Modification of Antigen Transmembrane Domains.

Background/Objectives: Chimpanzee adenoviral-vectored vaccines have proven to be both safe and effective, with a manufacturing and distribution pipeline capable of rapid global supply, as demonstrated during the COVID-19 pandemic. Yellow fever is a mosquito-borne viral hemorrhagic disease endemic in parts of Africa and Latin America, and although an effective live attenuated vaccine exists, its use is limited by safety and eligibility restrictions. Moreover, large outbreaks continue to expose critical challenges, such as an insufficient vaccine supply, reliance on fractional dosing, and slow a

Internal poly(A) replacement strategy within 3' UTR for live-attenuated tick-borne encephalitis virus.

Tick-borne encephalitis virus (TBEV), a prominent orthoflavivirus transmitted by ticks, poses a risk of potentially fatal encephalitis in humans. Currently, the only available inactivated vaccines have limited usage, underscoring the need for developing novel vaccine strategies. The 3' untranslated region (UTR) of TBEV can be divided into variable and conserved regions, where structured RNA elements play crucial roles in viral replication and virulence. In this study, we found that deleting sequences in both the variable region and most of the conserved region within the 3'UTR eliminated viral

Culex restuans (Diptera: Culicidae) in the upper Midwest: a review of ecology, biology, control, and contributions to human WNV in the region.

West Nile virus (WNV) is a mosquito-borne flavivirus endemic to the United States. Several regions, including parts of the Midwest, experience sustained WNV transmission with annual local and regional outbreaks. In the upper Midwest, Culex pipiens Linnaeus and Culex restuans Theobold, play key roles in WNV circulation, with Cx. pipiens considered the primary epidemic vector responsible for human cases and Cx. restuans linked to early-season enzootic amplification. These species share ecological niches, have similar host preferences, and are morphologically difficult to distinguish. Both are im

Land and climate suitability for West Nile virus in Atlantic archipelagos guided by historical data from Europe.

West Nile (WNV) is a zoonotic mosquito-borne virus with an expanding geographical and epidemic activity worldwide. Computational studies have contributed to the understanding of factors driving WNV occurrence, particularly in North America and Europe. Archipelagos have largely been overlooked, despite the risk to unique local avian species and human populations. In this study, we apply an ecological niche approach, trained on WNV occurrence and (a)biotic factors from European countries to project ecological suitability for WNV occurrence across several Atlantic archipelagos. The approach gives
