Surveillance-based insights into mosquito-borne disease trends: Implications for public health in Poland and Europe (2018-2024).

INTRODUCTION: Mosquito-borne diseases continue to pose a significant global health challenge, driven by environmental, climatic, and socio-demographic factors. This study investigates the epidemiological trends of malaria, dengue fever, chikungunya, and West Nile virus (WNV) in Poland and Europe, from 2018 to 2024, with a focus on regional differences, local transmission, and temporal dynamics. MATERIAL AND METHODS: Data were obtained from national and international surveillance systems. Annual case counts for each disease, including autochthonous cases, were analyzed using Poisson regression

MyD88 signaling in myeloid cells induces gastrointestinal tract injury and systemic inflammation after West Nile virus infection.

Humans with autoantibodies to type I interferons (IFNs) are at higher risk for neurological disease after West Nile virus (WNV) infection. IFN signaling deficient WNV-infected mice develop greater inflammation in the brain, which correlates with loss of gut barrier integrity. Here, we investigate the immune pathways that mediate this virus-gut-brain axis of inflammation. Myd88-/- mice administered anti-IFNAR1 antibody and infected with WNV exhibit reduced GI tract damage and inflammation. GI tract injury caused by WNV infection requires gasdermins or IL-1 receptor expression in hematopoietic c

Mapping the bird risk index for West Nile virus in Europe and its relationship with disease occurrence in humans.

West Nile virus (WNV) is a zoonotic mosquito-borne Flavivirus, with bird populations reservoirs. Although often asymptomatic, infection in humans can cause febrile symptoms and, more rarely, severe neurological symptoms. Previous studies assessed environmental drivers of WNV infections, but most overlooked areas with potential WNV circulation despite no reported human case, and mixed mechanisms affecting hosts vs. vectors. Our objective was to generate a WNV Bird Risk Index (BRI) mapping the potential of WNV circulation in bird communities across Europe. We first used a bird traits-based model

Rift Valley fever virus activates multiple cell death pathways in neurons.

Neuronal injury and death contribute to long-term impairments and lethality during viral encephalitis. Rift Valley fever virus (RVFV), an arbovirus with epidemic potential, can manifest as late-onset encephalitis in humans, yet this disease outcome remains understudied. A lethal rodent model of RVF encephalitis is characterized by a dysregulated immune response, neuronal necrosis, and blood-brain barrier breakdown that precedes lethality. In this study, we built upon this prior work using both in vivo and in vitro models to interrogate the mechanism of cell death in neurons during RVFV infecti

The Therapeutic Potential for Steroid Treatment Strategies in the Treatment of Murine Venezuelan Equine Encephalitis Virus (VEEV) Infection.

One highly consequential presentation of Venezuelan equine encephalitis virus (VEEV) infection is encephalitis. Here we considered anti-inflammatory interventions to limit the effects of this using a BALB/c subcutaneously challenged mouse model of disease. This disease model nearly ubiquitously presents with severe encephalitis, where viral neuroinvasion correlates with much of the outward clinical signs of disease. A selection of already licenced, commonly used anti-inflammatory drugs were tested in mice developing encephalitis (starting treatment at 24 h post challenge). Drug regimens were u

Ecologic Factors Contributing to West Nile Virus Hyperendemicity in Central South Carolina: An Integrated Vector-Human-Environmental Study.

West Nile virus (WNV) is an endemic arboviral infection in the United States that has undergone phylogenetic evolution since its introduction 25 years ago. An integrated vector-human-pathogen study was conducted in the summer of 2023 to unearth contemporary Culex quinquefasciatus habitat patterns and human transmission spillover foci in South Carolina, a state with historically little WNV data. A serosurvey revealed WNV seroprevalence 10 times the national average (22% versus 2%, respectively), with unusual epidemiologic risk factors. Female Culex quinquefasciatus WNV positivity was low (2.7%)

Donor-Derived West Nile Virus Infection in Kidney Transplant Recipients, France, 2025.

We report 2 cases of donor-derived West Nile virus infection in kidney transplant recipients in France. Both recipients had mild disease develop and recovered without sequelae. A more proactive screening strategy in France, particularly during periods of highest risk for West Nile virus circulation, would help reduce risk for donor-derived infections.

Introduction of a European Central-South-Eastern West Nile Virus Lineage 2 Strain in Italy in 2023: Evidence from the First Locally Acquired Neuroinvasive Case in the Calabria Region.

West Nile virus lineage 2 (WNV-2) is a growing public health concern in Europe causing West Nile fever or West Nile neuroinvasive disease (WNND) with substantial morbidity and mortality; however, genomic data from southern Italy are limited despite recent expansion of autochthonous transmission. The aim of the study was to characterize the phylogenetic and molecular features of the WNV-2 strain responsible for the first autochthonous human infection reported in Calabria (2023), and two more additional WNND cases detected in 2024. Full WNV-2 genomes were generated from the three cases. Phylogen

Durable protection against lethal Rift Valley fever hepatitis and encephalitis following low-dose ΔNSsΔNSm vaccination in mice.

Rift Valley fever virus (RVFV) is a zoonotic arbovirus that infects and causes disease in both humans and livestock. In humans, RVFV infection mostly results in febrile illness but has the potential to cause severe complications such as hepatitis and encephalitis. There are currently no licensed vaccines for human use; however, several candidates have been shown to be safe and immunogenic in pre-clinical studies. However, there remain gaps in our knowledge, including the duration of immunity and efficacy of protection from divergent disease manifestations over time. This pilot study investigat

WEST NILE VIRUS DISSEMINATION AMONG INDIVIDUAL WILD MOSQUITO VECTORS IN NORTHERN COLORADO, 2023.

West Nile virus (WNV) transmission risk is typically estimated from pooled whole-mosquito infection data, which may overestimate the proportion of mosquitoes capable of transmission. To assess natural viral dissemination in field-collected Culex tarsalis, we tested infection rates in tissues of 1,793 individual mosquitoes collected from northern Colorado in August 2023. Abdomens were screened for WNV ribonucleic acid (RNA), and corresponding thorax and head tissues from positive mosquitoes were tested. Fifteen mosquitoes had detectable abdominal infections, but WNV RNA was detected in only 53%

Scenario-driven shifts in future Usutu and West Nile virus outbreak characteristics in the Netherlands.

Mosquito-borne virus transmission is shaped by its ecological context, including land use, climate, and population dynamics. Future changes in these factors may therefore affect the risk and intensity of Usutu virus (USUV) and West Nile virus (WNV) outbreaks in the Netherlands. We compared a reference scenario to four future Shared Socio-economic Pathway scenarios developed for the Netherlands, which differed in land use, host distribution, mosquito distribution, and temperature. Temperature (between + 1.0 °C and + 1.7 °C) and mosquito abundance (between + 5% and + 10%) were predicted to incre

Association of avian biodiversity and West Nile Virus circulation in Culex mosquitoes in Emilia-Romagna, Italy.

BACKGROUND: West Nile Virus (WNV) is a zoonotic arbovirus maintained in a transmission cycle between Culex mosquitoes and birds, occasionally spilling over into humans. The impact of avian biodiversity on WNV circulation remains debated, with studies reporting both negative and positive correlations (dilution and amplification effects respectively) across different settings. In Europe, this relationship remains largely unexplored, particularly in regions with high WNV transmission, such as Emilia-Romagna in Northern Italy. METHODS: We explored the association between avian biodiversity and WN

Evaluation of the safety and insecticidal efficacy of ivermectin-treated bird feed formulations in different avian species.

BACKGROUND: West Nile virus (WNV) is maintained in an enzootic cycle between reservoir host birds and Culex (Cx.) spp. mosquitoes. This relationship presents a potential target for vector control strategies. Ivermectin (IVM), an endectocidal drug that selectively affects invertebrates while remaining safe at high concentrations in mammals and birds, can be delivered to Culex tarsalis via blood meals from birds fed IVM-treated bird feed. In this study, we evaluated the safety, efficacy, and utility of IVM-treated bird feed as a novel vector control strategy by assessing its impact on multiple b

West Nile Virus in Europe: Epidemiology, Vector Ecology, Environmental Drivers, and the Role of Equine Sentinel Surveillance in a One Health Framework.

West Nile virus (WNV) is a mosquito-borne flavivirus that remains an important public and veterinary health concern across Europe. Periodic outbreaks affecting humans, horses, and wildlife highlight the complex ecological interactions underlying viral circulation. This narrative review aims to synthesize current knowledge regarding WNV epidemiology, transmission dynamics, and surveillance strategies in Europe, with particular attention to the Romanian context. Available surveillance data indicate recurrent seasonal transmission in several European regions; however, reported case numbers may be

Reframing West Nile Virus in Latin America: From Enzootic Evidence to Human Risk-Surveillance Gaps and One Health Actions.

West Nile virus (WNV) is a mosquito-borne flavivirus with one of the widest global distributions. Since its discovery in Uganda in 1937, it has become a major zoonotic pathogen, and after its introduction into the United States in 1999, it spread rapidly across the Americas, becoming the leading cause of neuroinvasive arboviral disease. Its expansion illustrates a remarkable ecological adaptability, further intensified by climate change. In Latin America and the Caribbean, WNV circulation has been consistently documented in birds, horses, and mosquitoes; however, confirmed human cases remain d

Venezuelan equine encephalitis virus infection causes chronic neurobehavioral outcomes, cellular remodeling, and hippocampal single-cell transcriptomic changes.

Venezuelan equine encephalitis virus (VEEV), a neuroinvasive alphavirus, can cause significant neurological deficits in humans. Viral infections, including VEEV, have been linked to neurological diseases such as Parkinson's and Alzheimer's, though mechanisms remain unclear. Currently, not only are there no therapeutic options for VEEV available, but there is also limited information on the host responses following infection that contribute to neurological sequelae. To fill this gap in knowledge, longitudinal neuropathological, behavioral, and single-cell transcriptomic changes were examined in

Hospitalizations associated with endemic and non-endemic mosquito-borne arboviruses in Canada, 2002-2023.

Mosquito-borne arboviruses pose a growing public health concern in Canada, particularly in the context of climate change and increased global travel. This study aimed to quantify the burden of endemic and non-endemic mosquito-transmitted arboviral diseases in Canada by examining hospitalization trends from 2002 to 2023. Using administrative hospital data from the Canadian Institute for Health Information (CIHI) and national West Nile virus (WNV) surveillance data, we analyzed patient demographics, temporal and spatial patterns, and disease classification. Hospitalizations were classified as en

Catch basin larvicide treatments impact adult mosquito West Nile virus vector species in metropolitan Milwaukee, WI, U.S.A.

Roadside stormwater catch basins are frequently treated with larvicides in metropolitan areas because they are ideal larval growth sites for West Nile virus (WNV) vector species Culex pipiens Linnaeus and Cx. restuans (Theobald). Despite the wide-spread implementation of catch basin treatments in integrated vector management programs, there is little entomological evidence to suggest catch basin larvicide treatments reduce adult mosquito abundance and thereby impact WNV risk. We evaluated the impact of treating catch basins at three of four sites (700-1000 basins each) with a formulation of Ly
