(Sero)prevalence of (re)emerging major arbovirus infections in Africa: a systematic review and meta-analysis protocol.

BACKGROUND: Arbovirus infection outbreaks are becoming more widely reported in Africa. However, it is still difficult and crucial to better understand arbovirus transmission patterns, disease trends, and burdens. The epidemiology of these infections-dengue virus (DENV), Zika virus (ZIKV), chikungunya virus (CHIKV), West Nile virus (WNV), Rift Valley fever virus (RVFV), and yellow fever virus (YFV)-is unfortunately not well understood. This review provides an epidemiological inventory of DENV, ZIKV, CHIKV, WNV, RVFV, and YFV infections in Africa, with helpful results for risk mapping and upcomi

Spatiotemporal patterns of Rift Valley fever virus in Africa: a retrospective genomic epidemiology and phylodynamic modelling study.

BACKGROUND: Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic pathogen causing outbreaks in humans and ruminants across Africa and the Arabian Peninsula. Originally restricted to the Great Rift Valley, RVFV has expanded geographically, prompting its classification by WHO as a pathogen of pandemic potential. We investigated the evolutionary and spatial dynamics of RVFV across Africa. METHODS: We used genomic data generated at the International Livestock Research Institute Nairobi genomic laboratory (BioProject PRJNA1106221) and combined with publicly available datasets retrieved from

A systematic review and meta-analysis of mosquito arboviral infections detected through xenosurveillance in Africa: A focus on West Nile, Rift Valley fever, and chikungunya virus infections.

This systematic review and meta-analysis comprehensively evaluated the mosquito arboviral infections with West Nile virus (WNV), Rift Valley fever virus (RVFV), and chikungunya virus (CHIV), detected through xenosurveillance, reported in mosquito vectors across Africa in the last 25 years (2000-2024). The study analysed xenosurveillance data from 45 studies conducted with over 115,000 mosquito specimens, and incorporated moderator analyses to assess the influence of trap type, seasonality, regional location, settlement type, and screening methods on virus detection rates. The estimated pooled

A Cross-Sectional Study of Rift Valley Fever Exposure in Humans and Livestock in Southwestern Uganda Using a One Health Approach: Evidence of Elevated Seroprevalence Outside Recognized Outbreak Periods.

Rift Valley fever (RVF) transmission has intensified in southwestern Uganda since 2016. To quantify human and livestock exposure and associated risks, we conducted a cross-sectional serosurvey in Isingiro, Kabale and Rubanda districts between October and November 2023. A total of 766 humans and 2383 livestock were sampled and tested for RVF antibodies using ELISA, with structured questionnaires capturing demographic, behavioral and environmental data. Human seroprevalence was 11.5% (88/766), varying by district (13.8% Isingiro, 11.8% Rubanda, 6.8% Kabale; p = 0.04). Independent predictors from

Rift Valley fever virus dynamics in a transhumant cattle system in The Gambia.

Rift Valley fever (RVF) is a zoonotic disease of global concern, driven by environmental conditions, vector activity, and livestock mobility. Although RVF has been reported in The Gambia, its epidemiology remains poorly understood. This study developed a compartmental model to study RVF dynamics in the cattle population of the country. The model incorporated seasonally dynamic transmission parameters reflecting transhumant movement and ecological differences between two distinct ecoclimatic regions: the Sahelian area and the Gambia river. Parameterised using serological data linked to househol

Transboundary Animal Diseases Associated With Cross-Border Camel Movement. A Systematic Review and Meta-Analysis.

Transboundary animal diseases (TADs) are contagious diseases that significantly impact livestock health, public health and economic stability. In regions with frequent cross-border trade and transhumance involving camels, particularly the Middle East and Africa, TADs pose a heightened One Health risk due to their zoonotic potential and capacity to spread rapidly across national boundaries. This review synthesises current knowledge on TADs, with a focus on zoonotic pathogens affecting camels involved in cross-border movement, identifies geographical trends, and highlights research gaps to infor

Magneto-Plasmonics-Enhanced Colorimetric Lateral Flow Immunoassay Using Magnetic-Gold Nanostars.

Colorimetric lateral flow immunoassays (LFIA) have revolutionized point-of-care testing (POCT) methods by providing simple, rapid, accessible, and instrumentation-free detection of infectious diseases. However, conventional LFIAs relying on gold nanospheres (GNPs) can suffer from limited sensitivity due to weak colorimetric signal intensity at the test line, restricting their diagnostic potential. To overcome this limitation, we developed a magneto-plasmonics enhanced colorimetric LFIA (mpLFIA)─a breakthrough platform that integrates a novel hybrid nanoparticle system─magnetic gold nanostars (

Induction of a Th1-Type Polyfunctional T-Cell Response by the Four-segmented Rift Valley Fever Candidate Vaccine in Humans.

BACKGROUND: Rift Valley fever virus (RVFV) is a mosquito-borne virus that affects livestock and humans. The 4-segmented live-attenuated human vaccine candidate hRVFV-4s has shown a strong safety profile and excellent tolerability in healthy adults during a first-in-human clinical trial, while also eliciting both neutralizing antibody and T-cell responses. Recognizing the critical role of cellular immunity in vaccine-induced protection and immune durability, this study aimed to comprehensively characterize the cytokine secretion profile, the antigen-specific breadth of RVFV-specific T-cell resp

Understanding the Rift Valley fever exposure risk: A comparative perspective from a multi-country study in East and Central Africa, 2021-24.

Rift Valley fever (RVF) is a concern in East and Central Africa, particularly following periods of heavy rainfall and flooding. However no human outbreaks have been reported in the Democratic Republic of the Congo (DRC). To assess whether this reflects a true absence of virus circulation, we estimated RVF seroprevalence in Goma (eastern DRC) and examined context-specific risk factors, comparing the findings with data from outbreak-prone countries. A two-year longitudinal study, across six health facilities in DRC, Kenya and Uganda, enrolled febrile subjects aged ≥10 years. Human serum samples

Development of reference material from Rift Valley fever virus RNA.

BACKGROUND: Rift Valley fever virus (RVFV) has mainly been reported in African countries, and molecular diagnosis is performed using nucleic acid amplification tests. Nucleic acid reference materials (RMs) are necessary for the performance evaluation and quality assessment of molecular diagnosis of Rift Valley fever. However, metrological certified RMs for RVFV nucleic acids are currently unavailable. METHODS: In this study, RVFV RNA RMs (KRISS 111-10-537, 538, and 539) were developed from in vitro transcribed RNA containing a three-segment genome, including the L, M, and S segments. The RNA

Live-cell 3D-SIM of Rift Valley fever virus NSs filaments reveals a polygon web architecture.

A defining feature of Rift Valley fever virus (RVFV) is the incorporation of the NSs protein into large filamentous assemblies inside infected nuclei [R. Swanepoel, N. K. Blackburn, J. Gen. Virol. 34, 557-561 (1977).], as judged from fixed specimens. To gain insight into the 3D structure of NSs filaments within live-cell nuclei, we used genetic-code expansion (GCE) to incorporate trans-cyclooct-2-en-L-lysine into the protein. This enabled site-specific fluorescent labeling with tetrazine dyes for live-cell structured illumination microscopy (SIM). Our superresolved images revealed the complete

Unreported Rift Valley fever virus circulation during 2023-2024 El Niño event detected by slaughterhouse-based surveillance in southern Kenya.

Rift Valley fever (RVF) is a zoonotic arbovirus, and livestock cases are often underreported in endemic countries due to reliance on passive clinical surveillance. During the 2023-2024 El Niño event, Kenya experienced widespread flooding, but no RVF outbreaks were reported in the southern regions. We implemented slaughterhouse-based surveillance in southern Kenya from May 2023-June 2024, using five consecutive cross-sectional surveys. Cattle, sheep, and goats were tested for anti-RVFV IgG and IgM antibodies, with concurrent recording of post-mortem lesions. Using age estimates from dentition,

Ferritin nanoparticles displaying rift valley fever virus glycoprotein elicit potent dendritic cell activation in vitro.

Rift Valley fever (RVF) is a mosquito-borne zoonosis of major concern for human and animal health, yet no licensed human vaccine exists. Here, we engineered a self-assembling nanoparticle vaccine candidate by genetically fusing the RVF virus glycoprotein Gn to the N-terminus of a hybrid bacterial ferritin, generating nanoparticles that display 24 copies of Gn on their surface. Cryo-electron microscopy at 6 Å resolution confirmed ordered and symmetric presentation of the antigens, consistent with the structural models of ferritin and Gn. When incubated with human monocyte-derived dendritic cell

Rift Valley fever: An emerging zoonotic threat to global health and agriculture.

Rift Valley fever (RVF) is an acute, mosquito-borne zoonotic disease caused by the RVF virus (RVFV), which belongs to the Phlebovirus genus of the Bunyaviridae family. The infection mainly targets domestic ruminants such as sheep, goats, cattle, and camels, with sheep showing the highest vulnerability. The virus is spread predominantly through the bites of Aedes and Culex mosquitoes, although humans may also become infected by direct exposure to blood, organs, or secretions of diseased animals. In livestock, the illness is marked by sudden fever, ocular and nasal discharges, vomiting, and swol

A Species-Independent Lateral Flow Test to Detect Rift Valley Fever Virus Antibodies Using a Double Antigen Approach.

Rift Valley fever virus (RVFV) is a re-emerging, vector-borne pathogen endemic to Africa and the Arabian Peninsula, posing an increasing threat to human and animal health. Outbreaks have severe economic and social impacts on farmers, communities, and governments. Current diagnostic methods rely on PCR and ELISA; however, rapid pen-side tests would enable faster, cost-effective monitoring and outbreak control. Here, a species- and immunoglobulin class-independent capillary flow immunodiagnostic assay (lateral flow test; LFT) for detecting RVFV-specific antibodies is described. The assay uses a
