Alterations in the feline oral microbiome in common oral diseases - A comprehensive review.

The most prevalent diagnostic conditions in domestic cats (Felis catus) are oral diseases, affecting up to 90% of older cats. Periodontal disease (PD), feline chronic gingivostomatitis (FCGS), and tooth resorption (TR) are the principal clinically relevant entities, each with distinct histopathological and microbiological features. Certain molecular techniques, including 16S rRNA sequencing, shotgun metagenomics, and metatranscriptomics, have substantially advanced our understanding of the feline oral microbiome alterations. This review summarizes the findings of the healthy oral microbiome an

Impact of Austere Hot and Humid Conditions on the Physical and Biological Properties of Endodontic Sealers in Military Use.

INTRODUCTION: Nearly 15 million root canals are performed annually for the treatment of diseased and infected tooth pulp, allowing teeth to be functionally and esthetically maintained. Non-surgical root canal therapy ties directly into dental readiness and the maintenance of US posture by preventing and successfully treating endodontic disease. However, this therapy has a 5-20% failure rate with approximately 60% of these failures attributed to incomplete obturation of the root canal. The composition and performance of endodontic sealers are vital for successful root canal treatments and may b

Functional impact of pathogenic mutations in the runt homology domain of mouse Runx2 on skeletal and dental phenotypes in cleidocranial dysplasia.

Runt-related transcription factor 2 (RUNX2) is essential for skeletogenesis, and mutations in its gene cause cleidocranial dysplasia (CCD), an autosomal dominant skeletal disorder. The evolutionarily conserved 128-amino acid Runt homology domain (RHD) of human RUNX2 is essential for DNA binding and heterodimerization, and serves as a mutation hotspot associated with severe CCD phenotypes. To elucidate the functional impact of pathogenic RHD mutations in vivo, we generated 2 novel mouse lines: one carrying a missense mutation, c.695G>A, p.Arg232Gln (p.R232Q), corresponding to the human RUNX2 c.

Dental mesenchymal stem cell-derived exosomes for oral diseases: Classification, functionalization and clinical prospects.

The conventional therapeutic approaches for oral diseases primarily encompass dental restoration, pulp therapy, and maxillofacial surgical interventions. However, these methods are difficult to promote tissue regeneration and immune regulation. Mesenchymal stem cell-derived exosomes (MSC-Exos), mainly derived from tissues such as bone marrow, umbilical cord blood, umbilical cord, placenta, and fat, have been used to study the treatment of oral diseases. Conventional MSCs-Exos have relatively low immunogenicity and ethical issues, but exosomes derived from dental mesenchymal stem cells (DSCs) n

[Role and research progress of dental medicine in physical anthropology studies].

Physical anthropology is a discipline that studies human physical characteristics and their evolutionary patterns through the analysis of human biological remains. Oral tissues such as teeth and jawbones, serving as crucial or even exclusive materials among human biological remains, establish oral medicine's central role in this field. This paper reviewed the advances in oral medicine's research on key physical anthropology topics and methodologies, including human origins and evolution, paleopathology, paleodietary reconstruction, and paleomicrobiology. In human origins and evolution research

Oral hygiene and cancer risk: emerging evidence and public health perspectives.

Oral hygiene, traditionally regarded as a pillar of dental care, is increasingly recognized for its broader relevance to systemic health, including cancer prevention. Accumulating evidence suggests that poor oral hygiene may contribute to carcinogenesis through pathways involving chronic inflammation, microbial dysbiosis, and immune dysregulation. This review synthesizes epidemiological studies linking indicators of poor oral health—such as periodontal disease and tooth loss—to elevated risks of head and neck, gastrointestinal, and other cancers. It also explores biological mechanisms that pla

The role of Alanyl-tRNA synthetase 1 lactylation in tumors and other diseases.

Tumorigenesis and progression are driven by dysregulated genes and signaling pathways, with cellular metabolic reprogramming being a hallmark that supports neoplastic growth. Alanyl-tRNA Synthetase 1 (AARS1), a key enzyme with dual roles in metabolism and gene expression regulation, has emerged as a critical focus in cancer and disease research. As a member of the aminoacyl-tRNA synthetase (AARS) family, AARS1 canonically catalyzes the attachment of alanine to its cognate transfer RNA (tRNA) to ensure fidelity in protein synthesis. Emerging evidence reveals non-canonical roles of AARS1 in tumo

MECP2 Insufficiency Attenuates RUNX2-Dependent Osteoblast Differentiation via miR-126-3p/DKK1-Mediated Canonical Wnt Signaling Inhibition in Rett Syndrome.

Rett syndrome (RTT) is a rare neurodevelopmental disorder caused by loss-of-function mutations in the gene encoding methyl-CpG-binding protein 2 (MECP2) that is located on the X chromosome. Affected individuals also exhibit a variety of non-neurological symptoms such as kyphoscoliosis and osteoporosis. Thus, MECP2 may play a functional role in bone remodeling and osteoblast differentiation. This study aimed to clarify the molecular mechanisms underlying the deregulation of bone remodeling in RTT. Human deciduous tooth-derived mesenchymal stem cells that exhibit osteoblast plasticity were used

[A stratified framework for normal control tissue selection in ameloblastoma research].

Ameloblastoma is a locally invasive benign tumor of odontogenic epithelial origin. Research into its molecular mechanisms is hindered by the lack of unified norms for normal control tissue selection, which undermines data comparability. Current studies employ highly heterogeneous control tissues, including tooth germs, pericoronal tissue, normal oral mucosa and even commercial samples, thereby complicating data integration and potentially affecting the accurate interpretation of disease mechanisms. Based on principles of tooth development and the concept of "developmental arrest" in ameloblast

Discoidin Domain Receptor 2 Is Required for Tooth Extraction Socket Healing.

Discoidin Domain Receptor 2 (DDR2) is a collagen-activated tyrosine kinase required for bone and tooth development as well as regeneration of calvarial and long bones. Although DDR2 is expressed in alveolar bone, its functions in this site have not been previously examined. The present study used global and conditional knockout approaches and lineage tracing to examine the functions of DDR2 in alveolar bone formation during tooth socket healing. First molars were extracted from the maxillae of wild-type and globally Ddr2-deficient (Ddr2LacZ/LacZ) mice, and socket healing was measured after 1,

Co-Existing Charcot-Marie-Tooth Disease Type II and Parkinson's Disease Linked to a Novel DNAjB2 Pathogenic Variant.

BACKGROUND: The DNAjB2 gene encodes a co-chaperone protein that interacts with the heat shock protein (HSP) family to maintain protein quality control and preserve neuronal integrity. Variants in this gene have been associated with the axonal form of Charcot-Marie-Tooth Disease (CMT2). Recent literature has also suggested an association between DNAjB2 variants and neurodegenerative disorders such as Parkinson's disease (PD). DESIGN/METHODS: Case Report. CASE DESCRIPTION: We present a 36-year-old female patient initially diagnosed with CMT2 at the age of 28, who later developed symptoms of PD

Lentivirus-based HPSC therapy provides effective and long-term treatment in hypophosphatasia mouse model.

Hypophosphatasia (HPP) is a multi-system metabolic disorder characterized by skeletal hypomineralization and perinatal lethality in severe cases and rickets, muscle weakness, and premature tooth loss in milder forms. It is caused by loss-of-function mutations in the ALPL gene encoding the tissue-nonspecific isozyme of alkaline phosphatase (TNALP). Currently, enzyme replacement therapy with asfotase alfa (STRENSIQ) is the only treatment approved for the most severe forms of the disease. It enhances survival and improves bone mineralization, but it is burdensome for the patients who must undergo

Resolution of Chronic Diarrhoea Following Treatment of Periodontal Disease in a Cat.

An association between periodontal disease and comorbid disorders, including gastrointestinal signs, has been reported in cats and investigated in humans, where a bidirectional relationship between oral and gastrointestinal microbiomes exists. A 5-year-old neutered female domestic shorthair cat was presented with a 2-year history of small-bowel diarrhoea. The cat's appetite was reduced, and coat quality had deteriorated. On examination, the cat was found to have periodontal disease affecting multiple teeth and a matted coat. Biochemistry revealed mild hyperproteinaemia, haematology was unremar

Nanozymes with superoxide dismutase activity: Mechanisms, classification, and biomedical applications.

Nanozymes with superoxide dismutase (SOD) activity represent a class of artificial enzymes that mimic the catalytic function of natural SOD. This review systematically summarizes the recent advancements in SOD-like nanozymes, focusing on their catalytic mechanisms, material classifications, and biomedical applications. It begins by elucidating the enzymatic mechanisms of native SOD isoforms dependent on their metal cofactors (Cu/Zn, Mn, Fe, Ni). The article then classifies and discusses various synthetic nanozymes, including those based on metals, metal oxides, metal-organic frameworks (MOFs),

Endosomes as central hubs of interorganellar communication and cellular homeostasis.

Endocytosis mediates the internalization of extracellular cargo via vesicular trafficking, enabling targeted delivery to specific organelles and maintaining cellular homeostasis. Endosomes function as dynamic hubs that orchestrate intracellular communication. While they primarily internalize material from the cell surface, their role extends far beyond passive transport. Through continuous cycles of sorting, fusion, and fission, endosomes engage in extensive crosstalk with other organelles via three fundamental modes: vesicle-mediated cargo transport, membrane contact site-mediated non-vesicul

Clinical studies on the effect of type 1 diabetes on oral health in children and adolescents aged 6-17 years. A literature review.

BACKGROUND AND AIMS: Type 1 diabetes mellitus is a chronic metabolic disease of autoimmune origin with early manifestations, which appear predominantly in childhood. Its incidence has increased in most European countries mainly due to refined diets and increased consumption of sweets. Diabetes is known as a predisposing factor for the development of oral diseases; therefore, prevention at an early age is essential. Genetic factors and environmental factors contribute to the risk of disease, indicating a multifactorial etiology. Along with chronic periodontitis and other oral manifestations, su

[Oral homeostasis imbalance under hypoxia].

Hypoxia disrupts oral homeostasis through multiple interconnected pathways, including interference with tooth germ development, impairment of salivary gland function and salivary buffering capacity, compromise of the oral mucosal barrier, imbalance in jawbone and alveolar bone remodeling, and alterations in the diversity and functionality of the oral microbiome. These disturbances collectively contribute to the onset and progression of oral diseases. Moreover, disruption of oral homeostasis may, in turn, affect systemic homeostasis, increasing the risk of disorders in other organ systems throu

Safety, efficacy, and distal nerve Schwann cell biodistribution in mice and NHPs to support translation of AAV9 RNAi therapy for CMT1A.

Charcot-Marie-Tooth (CMT) type 1A, the most common inherited demyelinating peripheral neuropathy, is caused by PMP22 gene duplication, leading to overproduction of PMP22 protein in Schwann cells. To treat CMT1A, we developed a PMP22 gene silencing therapy using adeno-associated viral vectors (AAV9) to deliver a therapeutic miRNA expression cassette (U6.miR871) via lumbar intrathecal administration. A single injection produced long-term miR871 expression, triggered selective RNA interference against the PMP22 mRNA, and subsequently lowered protein levels and improved disease manifestations in a
