A Long-Term (41 Years) Radiographic Follow-Up Study of the Onset and Development of a Jawbone Lesion in a Patient With Gnathodiaphyseal Dysplasia.

Gnathodiaphyseal dysplasia (GDD) is an autosomal dominant syndrome characterized by bone fragility, sclerosis of tubular bones, and cemento-osseous lesions of the jawbones. This report describes the long-term radiographic follow-up study of jaw lesions in a GDD-affected woman of >40 years of age from the age of one onward. At three years of age, widening of the diaphyseal cortices in the femur and radius was observed, although no symptoms were observed in the jawbones. At nine years of age, we observed a slightly sclerotic appearance in the alveolar and jawbones adjacent to the permanent tooth

A reliable micro-CT-based method reveals dynamic changes to alveolar bone and tooth root following ligature-induced periodontal injury in the mouse.

This study presents a method development and evaluation framework for assessing longitudinally the dynamic alveolar bone changes in a murine periodontal injury model using 3D Slicer software. Accurate and reproducible measurement of bone loss is crucial for periodontal research, yet traditional two-dimensional (2D) histological approaches lack the ability to capture three-dimensional (3D) alterations, while inconsistencies in image alignment, region of interest (ROI) selection, and segmentation have limited the widespread adoption of 3D micro-CT analysis in small animal models. Here, we presen

Trigeminal influences and Locus Coeruleus: Tips for countermeasures, therapies and precocious diagnosis in dementia.

Masticatory dysfunction is a risk factor for Alzheimer 's disease (AD), likely due to the trigeminal influences on the Locus Coeruleus (LC), a structure implicated in neurodegenerative processes and whose activity level is reflected by pupil size. Chewing activity acutely stimulates cognitive performance by increasing LC activation during task, as reflected by an increased pupil dilatation (mydriasis). The presence of a trigeminal sensorimotor imbalance is associated to an asymmetry in LC activity (reflected by an asymmetry in pupil size) and is detrimental for performance. These observations

Exploring the Role of Oral Microbiota in the Pathophysiology and Treatment of Bruxism.

Bruxism is an involuntary condition involving grinding and clenching of the teeth, occurring during both wakefulness and sleep. This behavior can lead to various detrimental effects on oral health, including significant tooth wear and damage, temporomandibular disorders (TMD), tooth sensitivity, gum recession, and persistent headaches along with ear pain or tinnitus. The underlying causes of bruxism have long been debated, with the consensus suggesting that psychological, genetic, and environmental factors contribute to its development. Traditionally, the etiology of bruxism has been linked to

Identification of cross-talk pathways and PANoptosis-related genes in periodontitis and atherosclerosis by bioinformatics analysis and machine learning.

BACKGROUND AND OBJECTIVES: Periodontitis(PD) is a chronic inflammatory disease that poses a serious threat to oral health and is one of the risk factors for atherosclerosis(AS). A growing body of evidence suggests that the two diseases are closely related. However, current studies have yet to fully understand the common genes and common mechanisms between PD and AS. This study aimed to screen the tandem genes of PD and AS and the potential relationship between the tandem genes and pan-apoptosis-related genes. By analyzing the relationship between the core genes and immune cells, it will provid

AI-assisted phenotyping in a zebrafish hypophosphatasia model enables early and precise detection of skeletal alterations.

Hypophosphatasia (HPP) is a rare genetic disorder mainly affecting bone and tooth mineralization in patients due to ALPL gene mutations. Understanding genotype-phenotype correlations in HPP remains challenging due to different severities and the disease's heterogeneity. To address this, we established a novel zebrafish animal model (alplwue7), which mimics severe HPP disease forms. To bypass limitations in human-based phenotypic classification of skeletal alterations in this transgenic line, we developed and trained an artificial intelligence (AI) model capable of image-based classification wi

Tooth loss progression and mortality among older adults: results from the Chinese longitudinal healthy longevity survey (CLHLS).

BACKGROUND: Previous studies on the association between tooth loss and mortality typically relied on a single assessment of baseline tooth count. Therefore, we aimed to examine whether tooth loss progression was associated with mortality independently of baseline tooth count among older adults, using data from the Chinese Longitudinal Healthy Longevity Survey. METHODS: A total of 8073 older participants were included (median age: 83.0, interquartile range: 73.0–91.0; men: 46.6%). The exposure was tooth loss progression, which was assessed by annual tooth loss and categorized into four groups:

Carnosic acid alleviated periodontitis by inhibiting ferroptosis via the Nrf2/GPX4 pathway.

BACKGROUND: Ferroptosis, caused by abnormal iron metabolism and lipid peroxidation, has been linked to pathogenic processes in several disorders. Its function and regulating mechanisms in periodontitis are still unclear, nevertheless. As a naturally derived phenolic diterpenoid molecule, carnosic acid (CA) serves multiple biological roles, including antioxidant, anti-inflammatory, and cytoprotective properties. Its potential for intervention in periodontitis and ferroptosis warrants further exploration. METHODS: In this investigation, we combined network pharmacology analysis with in vitro an

Genotypes and Phenotypes of Patients With TSPEAR-Related Disorder: Evidence of a Predominant Dental Phenotype.

TSPEAR (chr. 21q22.3) encodes a protein involved in tooth development and is predominantly expressed in the enamel knot. Biallelic loss of function variants in TSPEAR cause ectodermal dysplasia, tooth agenesis and sensorineural hearing loss. However, the role of TSPEAR in auditory processes is unclear. This study aimed at better delineating the allelic and clinical spectrum of TSPEAR-associated disorders. We identified homozygous and compound heterozygous causative variants in TSPEAR [NM_144991.3] in 11 patients from seven families. Abnormalities in tooth number and shape (conical teeth and to

[Charcot-Marie-Tooth Disease: Historical Evolution and Present Understanding].

Charcot-Marie-Tooth disease is a hereditary motor and sensory neuropathy characterized by progressive distal muscle weakness, atrophy, and sensory impairment. In this article, we review the original reports published in 1886 by Charcot, Marie, and Tooth, analyzing the clinical features they used to distinguish Charcot-Marie-Tooth disease from other forms of muscle atrophy. We then trace the historical development of the disease concept and examine evolving classification systems based on clinical, electrophysiological, and genetic findings. The identification of more than 140 causative genes h

Recent Advances of Electrochemical Sensors Based on Carbon-Based Nanoparticles for Dental Infection and Periodontitis Detection.

Effective treatment, lower long-term expenses, and the avoidance of significant consequences, such as tooth loss, depend on the early identification of periodontitis (PD) and other dental diseases. Dentists can significantly enhance patient outcomes by implementing preventive measures and individualized treatment regimens through early diagnosis and treatment. Novel biosensors are crucial because they identify specific biomarkers in biofluids, including saliva and gingival crevicular fluid (GCF), enabling early, precise, and noninvasive diagnosis. Compared to traditional approaches, our method

Mandibular Lateral Displacement in Growing Patients: Etiopathogenesis, Prophylaxis, and Early Treatment-A Literature Review.

Background: Mandibular lateral displacement is a common functional occlusal disorder that may occur independently or in conjunction with other types of malocclusion. Substantial evidence indicates that in growing patients, mandibular lateral displacement can have a significant negative impact on the development of both the morphology and function of the stomatognathic system. Early diagnosis and intervention are therefore crucial to preventing a range of developmental abnormalities, including skeletal and soft tissue asymmetries, temporomandibular joint dysfunction, and pathological tooth wear
