Hypercementosis Associated with ENPP1 Mutations and GACI.

Mineralization of bones and teeth is tightly regulated by levels of extracellular inorganic phosphate (Pi) and pyrophosphate (PPi). Three regulators that control pericellular concentrations of Pi and PPi include tissue-nonspecific alkaline phosphatase (TNAP), progressive ankylosis protein (ANK), and ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1). Inactivation of these factors results in mineralization disorders affecting teeth and their supporting structures. This study for the first time analyzed the effect of decreased PPi on dental development in individuals with generalized art

Osteopontin regulates dentin and alveolar bone development and mineralization.

The periodontal complex is essential for tooth attachment and function and includes the mineralized tissues, cementum and alveolar bone, separated by the unmineralized periodontal ligament (PDL). To gain insights into factors regulating cementum-PDL and bone-PDL borders and protecting against ectopic calcification within the PDL, we employed a proteomic approach to analyze PDL tissue from progressive ankylosis knock-out (Ank-/-) mice, featuring reduced PPi, rapid cementogenesis, and excessive acellular cementum. Using this approach, we identified the matrix protein osteopontin (Spp1/OPN) as an

Comparative analysis of dentognathic pathologies in the Dmanisi mandibles.

OBJECTIVES: Due to the scarcity of the fossil record, in vivo changes in the dentognathic system of early Homo are typically documented at the level of individual fossil specimens, and it remains difficult to draw population-level inferences about dietary habits, diet-related activities and lifestyle from individual patterns of dentognathic alterations. The Plio-Pleistocene hominin sample from Dmanisi (Georgia), dated to 1.77 million years ago, offers a unique opportunity to study in vivo changes in the dentognathic system of individuals belonging to a single paleodeme of early Homo. Oral and maxillofacial considerations in Gardner's syndrome: a report of two cases.

Gardner's syndrome (GS) is a genetic disorder characterised by intestinal polyps, multiple osteomas, and soft-tissue tumours. Dentists play an important role in the syndrome diagnosis considering that craniomaxillofacial osteomas are a major criteria for Gardner's syndrome diagnosis. This study aimed to describe the main stomatological manifestation of GS and the importance of dentists in its diagnosis. Two patients presenting GS were evaluated. The first one had two osteomas in the mandible and GS was suspected. The colonoscopy confirmed the presence of polyposis and a prophylactic proctocole

Radiographic evaluation in clinical practice of the types and stage of incisor tooth resorption and hypercementosis in horses.

BACKGROUND: There are several reports of incisor tooth resorption and hypercementosis in horses but, to date, studies have been limited in case numbers and to advanced lesions. Tooth resorption in other species is a radiographic diagnosis of types of resorption that are often identified before clinical signs. Our goal was to evaluate radiographically incisor tooth resorption in a large population of horses, utilising interpretation criteria from canine and human dentistry. OBJECTIVE: To document and classify incisor tooth resorption and hypercementosis. STUDY DESIGN: Retrospective descriptiv

Malignant mesothelioma incidence by nation-wide cancer registry: a population-based study.

BACKGROUND: Most recently we reported that RHB‑104 triple antibiotics combination in culture is bactericidal and should be effective for treatment of Crohn's disease (CD)-associated with Mycobacterium avium subspecies paratuberculosis (MAP) (Alcedo et al. in Gut Pathog 14:32, 2016). The combination exhibited unique synergistic antimicrobial growth activity. The proprietary RHB-104 capsule formulation contains active ingredients (63.3 % Clarithromycin (CLA), 6.7 % Clofazimine (CLO) and 30 % Rifabutin (RIF)). In our earlier study, we could not dissolve the proprietary RHB-104 capsule formulation

Equine Odontoclastic Tooth Resorption and Hypercementosis: Histopathologic Features.

Equine odontoclastic tooth resorption and hypercementosis (EOTRH) is a painful progressive condition of older horses that involves multiple teeth, including canines and incisors. EOTRH is uncommonly recognized by veterinary pathologists and in some cases may be misdiagnosed as cementoblastoma. The cause is unknown. The goals of this study were to describe the histopathologic features of EOTRH in 17 affected horses from the United States and to increase awareness of this condition. Samples ranged from affected tooth to the entire rostral mandible and maxilla. Affected teeth exhibited cemental h

Histological evaluation of mandibular third molar roots retrieved after coronectomy.

There is a resurgence of interest in coronectomy for the management of mandibular third molars because it has a low risk of injury to the inferior dental nerve. However, there is concern that the root that is left in place will eventually become a source of infection. We describe the histological evaluation of 26 consecutive symptomatic coronectomy roots in 21 patients. All roots had vital tissue in the pulp chamber and there was no evidence of periradicular inflammation. Persistent postoperative symptoms related predominantly to inflammation of the soft tissue, which was caused by partially e

Irregular periapical radiopacity in mandibular premolars and molars.

Increased deposition of cementum is observed in a wide number of both benign and malignant conditions. Many cases are often diagnosed during routine examination as an incidental finding. Diagnosing correctly without confusing it with other similarly appearing lesions, thus avoiding subjecting the patient to unnecessary investigations and stress, is of prime importance. We report one such case, where the patient presented with the routine complaint of a painful tooth, during the investigation of which he was also diagnosed with hypercementosis affecting the mandibular second premolars and molar

Concrescence: assessment of case by periapical radiography, cone beam computed tomography and micro-computed tomography.

The aim of this article was to describe imaging aspects of concrescence analyzed by three imaging modalities. A second molar joined together with a third molar was imaged using digital periapical radiography, cone beam computed tomography (CBCT) and micro-computed tomography (Micro-CT). On periapical radiograph, the mesial root of the third molar is superimposed on the distal root of the second molar. On CBCT images, a large cementum union between bulbous roots was detected, confirming the diagnosis of concrescence. On micro-CT images, the cementum union appeared limited to the apical third of

Tooth alterations in areas of bisphosphonate-induced osteonecrosis.

OBJECTIVE: Osteonecrosis of the jaw is a potential side effect when using bisphosphonates. Most studies on the effects of bisphosphonates on teeth have been conducted in vitro or in animal models of tooth development. Therefore, the aim of this study was to describe alterations found in human teeth extracted from areas of bisphosphonate-induced osteonecrosis. MATERIALS AND METHODS: Using a retrospective study design, 16 teeth from 13 patients were extracted from areas of bisphosphonate-induced osteonecrosis during surgical debridement. The specimens were decalcified and embedded in paraffin.

Hypercementosis and concrescence of maxillary second molar with third molar: a case report and review of literature.

Concrescence signifies a rare developmental alteration, most commonly in maxillary molars, where two fully formed teeth with separate root canal structures are fused with cementum. Clinical detection is difficult due to lack of enamel involvement and radiographic detection may be challenging, as two dimensional images may be misdiagnosed as overlap, superimposition or close proximity of roots. Such a developmental anomaly can influence treatment outcomes for extraction, endodontic, periodontic, prosthodontic and orthodontic treatment. Legal complications could arise due to unexpected difficulti

Localized multiple cemental excrescences: a rare presentation of hypercementosis.

Hypercementosis is an abnormal thickening of cementum which may be generalized or localized. Localized hypercementosis is usually characterized by nodular enlargement of apical third of root. Hypercementosis in the form of multiple cemental spikes distributed throughout the entire length of root is a very rare finding. In this article, we report such a rare manifestation of localized hypercementosis affecting right maxillary third molar in a 24-year-old male. This finding was an accidental observation following a difficult extraction procedure as the spikes were not evident in the orthopantamo

Hypercementosis: a rare finding in a patient with systemic lupus erythematosus.

Hypercementosis is excessive deposition of non-neoplastic cementum over normal root cementum, which alters root morphology. This cementum may be either hypocellular or cellular in nature. The aetiopathogenesis of hypercementosis is ambiguous. Although most of the cases are idiopathic, several local and systemic factors are also linked to this condition, such as Paget's disease, acromegaly, vitamin A deficiency, etc. We report two rare cases of hypercementosis associated with systemic lupus erythematosus, not previously described in the literature, and also discuss the possible aetiopathogenesi

The oral pathological conditions of the Broken Hill (Kabwe) 1 cranium.

The Broken Hill (Kabwe) 1 cranium exhibits dental caries on ten teeth, multiple periapical lesions, periodontal disease, severe anterior dental wear, and hypercementosis, conditions all little-studied in Middle Pleistocene humans and making Broken Hill 1 of great value to discussions of the antiquity of oral pathological conditions. These individual pathological conditions, however, have never previously been described in detail; the focus has been on diagnosing an overarching syndrome connecting the temporal and oral lesions. This case study examines the individual dentoalveolar pathological

Determination and validation of criteria to define hypercementosis in two medieval samples from France (Sains-en-Gohelle, AD 7th-17th century; Jau-Dignac-et-Loirac, AD 7th-8th century).

OBJECTIVE: The main aim of this article was to develop different visual criteria allowing for an objective definition of hypercementosis (cementum hyperplasia). This preliminary study must notably show how to better understand at a later stage the significance of its frequency as well as its aetiologies, especially in past populations. MATERIALS AND METHODS: we set up a study protocol (macroscopic and photographic observations) on material consisting of 2 medieval samples from France (1) Sains-en-Gohelle sample-SG (AD 7th-17th century; 407 individuals; 5756 teeth observed, 319 with hypercemen
