Functional gastrointestinal (GI) tract disorders affect a substantial proportion of the global population and are often preceded by intestinal infections that cause injury to enteric neurons and glia through unrestrained immune responses. However, the mechanisms that limit infection-induced inflammation and protect the enteric nervous system remain poorly understood. Here, we defined such neuron-glia-macrophage interactions after West Nile virus (WNV) infection; this model neurotropic virus causes GI tract dysmotility in mice via injury of enteric neurons through a T cell-mediated cytolytic me

BACKGROUND: From January to May of 2024, 27 cases of neuroinvasive disease due to West Nile Virus (WNV) were confirmed from Kerala. The number of cases were more than those seen over past three years combined. Cases were not clustered geographically and were primarily reported from four districts, viz, Kozhikode, Thrissur, Malappuram and Ernakulum. To understand the circulation of WNV, a focal serosurvey was conducted in the regions from where cases were reported.
METHODS: A cross-sectional study was done in four districts of Kerala. Patients, family members and immediate neighbours were recr

West Nile virus (WNV) is an emerging infectious disease causing neuroinvasive diseases, including meningitis and encephalitis. Although infection has been documented in birds in Bangladesh, WNV has not been detected in humans. Since 2007, the Institute of Epidemiology, Disease Control and Research in Bangladesh has conducted hospital-based acute encephalitis syndrome (AES) surveillance to monitor Japanese encephalitis (JE), which is typically detected in 2% to 3%. Our study aimed to determine the presence of WNV, dengue virus (DENV), and chikungunya virus (CHIKV) among persons enrolled in AES

The yellow fever vaccine containing the live attenuated 17D strain is one of the most effective methods for preventing yellow fever infection and has long been considered safe. However, rare but serious adverse events such as yellow fever vaccine-associated viscerotropic disease (YEL-AVD) have been reported. YELAVD is a life-threatening clinical condition that can mimic wild-type yellow fever infection, is associated with a high risk of mortality, and may pose diagnostic challenges, particularly in non-endemic regions. A 57-year-old male patient with no underlying comorbidities, history of imm

West Nile virus (WNV) is a zoonotic Orthoflavivirus transmitted by mosquitoes that is responsible for outbreaks of meningitis and encephalitis worldwide. Driven by climate change, WNV has expanded as a global public health concern, particularly in temperate regions. However, there are still no specific approved therapies, reinforcing the need for antiviral development. Previous works have documented that WNV multiplication strictly depends on certain cellular lipids. To identify novel lipid-related therapeutic targets, we analyzed the infection-driven alterations in the CNS lipidome, the prima

Studies focused on the epidemiological surveillance of arboviruses that cause potentially zoonotic diseases, such as dengue, Zika, or emerging viruses like West Nile virus (WNV), are critical due to their significant impact on public health. Although research on these infectious agents is increasing in Colombia, regions remain where the presence of zoonotic agents is still unknown. To address this knowledge gap, the present study aimed to investigate the current status of WNV circulation in wildlife in two municipalities of the department of Casanare (El Yopal and Paz de Ariporo) from the Colo

Guillain-Barré syndrome (GBS) and GBS-like syndromes secondary to West Nile virus (WNV) are rare and poorly characterized. GBS is an immune-mediated disease that affects peripheral nerves, classically in a symmetrical and ascending pattern. It is generally associated with infectious microbes, but not classically with WNV. WNV, although a relatively new pathogen in the United States, has become a leading mosquito-borne viral infection in the country. We present a case of a patient who came to the hospital in Delaware, USA, with fever and confusion that rapidly progressed to worsening encephalop

INTRODUCTION: Since the rise of molecular high-throughput technologies, many diseases are now studied on multiple omics layers in parallel. Understanding the interplay between microRNAs (miRNA) and their target mRNAs is important to understand the molecular level of diseases. While much public data from mRNA experiments are available for many diseases, few paired datasets with both miRNA and mRNA expression profiles are available. This study aimed to assess the possibility of predicting miRNA expression data based on mRNA expression data, serving as a proof of principle that such cross-omics p

West Nile virus (WNV) causes human disease with variable severity. Toll-like receptor 3 (TLR3) is essential for innate immune responses to viral infections, including WNV. The non-synonymous single-nucleotide polymorphism (SNP) rs3775291 (C/T), which alters TLR3 function, has been linked to susceptibility to several viral pathogens, but its role in WNV infection is unclear. This study investigates the association between rs3775291 and WNV infection in hospitalized patients. A case-control study was conducted in Cyprus, including 20 hospitalized WNV patients and 22 healthy controls (HCs). Genot

West Nile virus (WNV) is a rare trigger of acute inflammatory demyelinating polyneuropathy (AIDP), more commonly associated with meningoencephalitis. This case highlights an atypical post-infectious presentation of WNV-associated AIDP following recent viral illness and explores the associated diagnostic and therapeutic challenges. A 36-year-old male patient with heavy alcohol use, chronic tobacco exposure, and a recent upper respiratory infection developed rapidly progressive bilateral weakness, numbness, and paresthesias over several days, ultimately becoming unable to walk. Examination revea

Background/Objectives: Flaviviruses, including West Nile virus (WNV), pose global health challenges due to their worldwide distribution, pathogenicity, and lack of effective treatments or vaccines. Today, WNV is considered the most important causative agent of viral encephalitis worldwide. This study investigated the different forms of the main WNV antigen-the preM/E protein-in the context of its immunogenic and protective properties. Methods: The recombinant adenovirus type 2 (rAd2) vectors expressing different forms of the WNV preM/E genes were obtained using standard molecular biology techn

The mosquito midgut functions as a key interface between pathogen and vector. However, studies of midgut physiology and virus infection dynamics are scarce, and in Culex tarsalis-an extremely efficient vector of West Nile virus (WNV)-nonexistent. We performed single-cell RNA sequencing on Cx. tarsalis midguts, defined multiple cell types, and determined whether specific cell types are more permissive to WNV infection. We identified 20 cell states comprising 8 distinct cell types, consistent with existing descriptions of Drosophila and Aedes aegypti midgut physiology. Most midgut cell populatio

For many RNA viruses, immunity is triggered when RIG-I-like receptors (RLRs) detect viral RNA. However, only a minority of infected cells undergo innate immune activation. By examining these "first-responder" cells during West Nile virus infection, we found that specific accumulation of antigenomic negative-sense viral RNA (-vRNA) underlies innate immune activation and that RIG-I preferentially interacts with -vRNA. However, flaviviruses sequester -vRNA into membrane-bound replication compartments away from cytosolic sensors. We found that single-stranded -vRNA accumulates outside of replicati

West Nile virus (WNV) is a neurotropic mosquito-borne orthoflavivirus, representing a relevant public health threat. Identification of biomarkers that would predict the course of WNV infection is of interest for the early identification of patients at risk and for supporting decisions on therapeutic interventions. In this study, serum levels of glial fibrillary acidic protein (sGFAP) and neurofilament light chain (sNfL), which are markers of brain tissue damage and inflammation, were analysed in 103 subjects with laboratory-confirmed WNV infection, comprising 13 asymptomatic blood donors, 23 w

This study introduces a novel forecasting tool for West Nile Virus (WNV) risk at the municipal level, based on a previously developed climate-dependent epidemiological model coupled with a data-driven model and field data in an ensemble framework. The modelling system was evaluated at the municipal level in the regions of Central Macedonia (RCM) and Thessaly (RTH) in Greece for the period 2020-2024 and classifies the municipalities according to the risk level of occurrence of a WNV human case (WNVhc) within each forecast season. The modelling system produces seasonal forecasts updated monthly
