Now you see me, now you don't: What and how viral RNAs are detected by cytoplasmic pattern-recognition receptors.

In a recent issue of Molecular Cell, Genoyer et al.1 show that during West Nile virus infection, escaped, cytoplasmic, anti-genomic negative-sense viral RNA triggers an antiviral response via RIG-I activation. Flaviviruses typically hide -vRNA in replication compartments to avoid detection, but some -vRNA leaks, possibly aided by viral capsid proteins.

West Nile Virus Chorioretinitis: First Case with Ocular Involvement in Türkiye.

A 59-year-old man who experienced severe visual loss in the right eye for two days following a febrile illness (high fever lasting for 15 days) presented to our center for a second opinion. On examination, his Snellen best corrected visual acuity (BCVA) was 1/10 in the right eye and 9/10 in the left eye. On fundoscopy, we observed a few track-like, cream-colored linear lesions in the superior fundus of the left eye and a small whitish foveal discoloration together with a temporally pallid disc in the right eye. On autofluorescence imaging, there were some scattered hyperautofluorescent patchy

An integrated proteomics approach identifies phosphorylation sites on viral and host proteins that regulate West Nile virus infection.

Upon infection, viruses alter the proteome, creating a hospitable environment for infection. Cells respond to limit viral replication, including through protein regulation by post-translational modifications. We use mass spectrometry to define proteome alterations during West Nile virus (WNV) infection. Our studies identify upregulation of HERPUD1, which restricts WNV replication through a mechanism independent of its role in endoplasmic reticulum (ER)-associated degradation (ERAD). We also identify modifications on viral proteins, including a WNV NS3 phosphorylation site that impacts viral re

Identification of West Nile virus infection by anti-premembrane antibodies in Nicaraguan children prior to 2007-2009.

UNLABELLED: Since its introduction to the Western Hemisphere in 1999 in New York City, West Nile virus (WNV) has spread throughout the continental USA and moved into Canada, Mexico, Caribbean, and Central and South Americas. While WNV has caused ~7 million human infections and >59,000 cases in the USA and >6,000 cases in Canada, only few human cases have been reported in Latin America. Due to the cross-reactivity of anti-envelope antibodies, the detection of WNV infection by serology to explore its epidemiology in Latin America, where multiple flaviviruses co-circulate, remains a challenge. Long-Term Humoral Immune Response After West Nile Virus Convalescence in Horses in a Geographic Area of Multiple Orthoflavivirus Co-Circulation.

BACKGROUND: In the last three decades, West Nile virus (WNV, Flaviviridae, Orthoflavivirus genus) has become one of the most important encephalitic agents worldwide, causing substantial numbers of cases in humans and horses every year by re-emerging in endemic areas and emerging in new territories. It is considered that after natural WNV infection, humans and birds develop long-term immunoprotection, but data on immunoprotection in horses is scarce. HYPOTHESIS: West Nile virus infection provides long-term humoral immunity in subclinically infected horses. ANIMALS: Client-owned, naturally WNV

Reprogramming of liver metabolism during West Nile virus infection unveils novel aspects of disease pathophysiology.

BACKGROUND: West Nile virus (WNV) is a neurotropic mosquito-borne flavivirus responsible for outbreaks of encephalitis and meningitis worldwide. About 20% of infected patients exhibit abnormal liver function tests, although the participation of this organ in the pathophysiology of the disease remains unclear. To fill this gap, this study explores changes in liver metabolism during WNV infection. METHODS: Given the relevance of the liver as a major immune and metabolic organ, the changes in response to WNV infection were analyzed in the mouse model combining transcriptomics, lipidomics and his

Mosquito abundance, blood feeding, and West Nile virus infection in association with woody plant encroachment in the U.S. Great Plains.

Knowledge of the utilization by mosquito species of different habitats is critical for determining risk for the transmission of mosquito-borne pathogens. The Great Plains region of the United States is experiencing a biome-level change as grasslands are being converted to forests through the process of woody plant encroachment by eastern redcedar (ERC). There is a need to evaluate how mosquitoes respond to fine scale habitat characteristics within areas experiencing ERC encroachment. This study examined the influence of fine-scale vegetation differences on mosquito abundance, bloodmeal hosts,

Tissue tropism, pathology, and pathogenesis of West Nile virus infection in saltwater crocodile (Crocodylus porosus).

West Nile virus (WNV) is one of the leading causes of economic losses to the saltwater crocodile farming industry due to skin lesions, known as "pix", induced by the infection. Our previous study suggested a possible immunopathological pathway causing these lesions. We therefore resolved to investigate the kinetics of WNV-infection and the elicited immune responses in experimentally challenged saltwater crocodile hatchlings. Employing virus isolation, quantitation of viral genome loads in tissues by RT-qPCR and immunohistochemistry, we demonstrated that upon infection, the virus replicates in

The paradoxical impact of drought on West Nile virus risk: insights from long-term ecological data.

Mosquito-borne diseases are deeply embedded within ecological communities, with environmental changes-particularly climate change-shaping their dynamics. Increasingly intense droughts across the globe have profound implications for the transmission of these diseases, as drought conditions can alter mosquito breeding habitats, host-seeking behaviours and mosquito-host contact rates. To quantify the effect of drought on disease transmission, we use West Nile virus as a model system and leverage a robust mosquito and virus dataset consisting of over 500 000 trap nights collected from 2010 to 2023

Adaptive immune response to West Nile virus infection in the Collaborative Cross mouse model: A database of cellular phenotypes and Quantitative Trait Loci.

West Nile virus (WNV) infection can lead to a wide range of clinical outcomes, from asymptomatic to self-limiting febrile and serious neuro-invasive disease. Knowledge of the genetic factors contributing to the heterogeneity of this disease can inform pathogenic mechanisms, help guidethe development of cell based therapeutics, vaccines and inform lifestyle choices. Yet this knowledge is incomplete in humans. Here we present data from a large-scale experiment aimed at identifying quantitative trait loci (QTL) associated with adaptive immune cellular phenotypes observed in response to infection

Emergence of West Nile virus infection in Kerala, South India: A case of concern.

West Nile virus (WNV) infection is emerging as a disease of public health concern in Kerala state, India with recurring outbreaks since 2011. With its tropical climate, biodiversity hot spots of Western ghats, forest cover, plenty of water bodies and bird sanctuaries, Kerala provides an ideal ecological niche for vector breeding and transmission of WNV. In this work, we reflect on the peculiar features of the outbreaks of WNV fever in Kerala and highlight the knowledge gaps, research priorities and the need for effective control measures. Information on transmission patterns of WNV, its circul
