Factors contributing to mosquito-borne disease: A systematic review in the Middle East and North Africa (MENA) region.

Mosquitoes can carry and spread many diseases caused by bacteria, viruses, or parasites. All these mosquito-borne diseases (MBD) represent a significant global burden of infectious diseases, including morbidity and mortality. This systematic review delves into the multifaceted factors contributing to the spread of MBD in the Middle East and North Africa (MENA) region. Following PRISMA guidelines, we reviewed peer-reviewed English-language observational and experimental studies published between May 1990 and January 2023, focusing on the interaction between population, environmental, disease, a

Localized Rift Valley fever virus persistence explains epidemic and interepidemic dynamics and guides control strategies.

Rift Valley fever (RVF) is an emerging disease with devastating impacts on livestock health and livelihoods. The risk of RVF virus (RVFV) emergence in new regions and the effectiveness of a strategy for preventing establishment are impacted by how infection persists at local scales. Multiple mechanisms have been proposed for its persistence in regions prone to epidemics, including maintenance via transovarial transmission (TOT) but whether and how TOT can support local persistence is not well understood. Through the development of host- and multi-vector climate-driven simulation models to recr

Research trends in Rift Valley fever virus: a bibliometric analysis from 1936 to 2024.

BACKGROUND: Rift Valley fever (RVF), first identified in Kenya in 1930, is a viral zoonosis transmitted by arthropods and poses significant risks to both public and animal health. Despite the wealth of published studies on the disease, the number of bibliometric analysis of Rift Valley fever virus (RVFV) is limited. This study utilized bibliometric analysis to identify research hotspots and emerging trends in RVFV studies, aiming to offer new insights and strategic references for future research directions and prevention strategies. METHODS: The study utilized Scopus to collect global publica

Seroprevalence of Rift Valley and Crimean-Congo Hemorrhagic Fever Viruses, Benin, 2022-2023.

We screened 650 febrile patients from Benin for Rift Valley fever and Crimean-Congo hemorrhagic fever viruses during 2022-2023. None were positive by reverse transcription PCR; 1.1% and 0.3%, respectively, had virus-specific IgG. False-positive results from malaria-associated antibodies likely reacting with histidine-tagged viral antigens mandate careful validation of serologic tests in malaria-endemic regions.

Maternal vaccination with live-attenuated Rift Valley fever virus protects offspring via immune transfer.

Rift Valley fever virus (RVFV) causes high rates of spontaneous abortions and neonatal mortality in ruminants resulting in severe socioeconomic and public health consequences. Maternal vaccination may protect pregnant animals, fetuses, and neonates via transfer of maternal antibodies; however, currently available live-attenuated RVFV vaccines are generally unsafe for use during pregnancy. RVFV-delNSs/NSm is a live attenuated strain that has demonstrated favorable safety and efficacy in pregnant livestock, yet studies investigating maternal vaccination as a strategy to protect neonates from RVF

Effectiveness and equity of vaccination strategies against Rift Valley fever in a heterogeneous landscape.

Spatio-temporal variations in environmental and socio-agricultural factors create heterogeneity in livestock disease transmission risk, raising challenges in identifying populations most at risk and how this risk changes over time. Consequently, effective vaccination strategies require careful planning to achieve optimal or equitable outcomes across regions. We developed a metapopulation model for Rift Valley fever transmission in livestock across the Comoros archipelago which incorporates livestock vaccination in addition to heterogeneity in viral transmission rates and animal movements. We u

Detection and Molecular Characterization of Rift Valley Fever Virus in Apparently Healthy Cattle in Uganda.

Rift Valley fever (RVF) is a zoonotic disease caused by the Rift Valley fever virus (RVFV), affecting humans, livestock, and wild ruminants. This study aimed to characterize and assess the genetic diversity of RVFV strains circulating among livestock in Uganda. Blood samples were collected between January 2021 and May 2024 from apparently healthy cattle, goats, and sheep in four districts. The samples were first screened for RVFV antibodies using ELISA; antibody-positive samples were subsequently tested for viral RNA using reverse transcriptase quantitative PCR (RT-qPCR). The PCR-positive samp

Identification and Characterization of a Metal Binding-Independent Rift Valley Fever Virus Endonuclease Inhibitor.

The viral cap-snatching mechanism, facilitated by the endonuclease (EndoN) domain of viral polymerases, is critical for viral gene transcription and translation and is therefore an attractive target for antiviral development. The successful development of EndoN inhibitor XOFLUZA (Baloxavir marboxil) has opened the possibility that the EndoN domain of negative-sense RNA viruses (NSVs) can be targeted in a similar fashion. Rift Valley Fever Virus (RVFV) belongs to the Bunyaviricetes class, which includes other pathogens with pandemic potential, such as severe fever with thrombocytopenia syndrome

Baculovirus-mediated production and purification of ferritin nanoparticles for rift valley fever vaccine development.

BACKGROUND: Rift Valley fever (RVF) is a WHO-prioritized zoonotic, vector-borne disease with no licensed prophylaxis available for humans, highlighting the need for effective vaccine strategies. Nanoparticle-based platforms for antigen presentation offer a promising approach for vaccine development. RESULTS: In this work, we engineered ferritin (Ft) nanoparticles to display the immunogenic Gn domain of RVF virus (GnFt) and systematically assessed the production, purification, and physico-chemical properties of the purified nanoparticles. Baculovirus-based expression systems were evaluated in

Molecular detection of Ehrlichia ruminantium in ticks from ruminants during the 2021 Rift Valley fever outbreak in Mananjary, Madagascar.

Ehrlichia ruminantium, the causative agent of heartwater, is a tick-borne pathogen affecting livestock in Africa and the Caribbean. This disease is transmitted primarily by Amblyomma variegatum ticks and poses a significant threat to animal health. In Madagascar, the prevalence of E. ruminantium remains poorly documented. During a Rift Valley fever (RVF) outbreak in Mananjary, Madagascar (April-May 2021), we conducted a field study to assess the circulation of vector-borne pathogens in ticks collected from ruminants. Ticks were morphologically identified, and DNA was extracted for quantitative

Inter-epidemic seroprevalence of Rift Valley fever virus and associated risk factors in humans in Eastern Rwanda.

BACKGROUND: Rift Valley fever (RVF) is a mosquito-borne zoonosis that causes periodic and explosive epizootics/epidemics in Africa and the Arabian Peninsula. In Rwanda, RVF virus (RVFV) circulation has resulted into two major outbreaks in 2018 and 2022, both of which involving humans. Information on the magnitude of human exposure to RVFV in the country is scarce. This cross-sectional study aimed to investigate the seroprevalence of RVFV and associated risk factors in humans in the Eastern province of Rwanda, 3 years after the end of the 2018 outbreak. METHODOLOGY: A total of 552 outpatients

Rift Valley fever outbreak among animal handlers in Kinyogoga Village, Nakaseke District, Central Uganda: A case study.

Rift Valley fever (RVF) is a viral zoonosis which occurs sporadically in Uganda. On July 24, 2023, a 24-year-old male animal husbandry officer from Nakaseke District presented to a hospital in Kampala District with history of intermittent nosebleeds. He tested positive for Rift Valley fever virus (RVFV) by reverse transcriptase polymerase chain reaction (RT-PCR). We investigated to identify the source, estimate the magnitude, and determine the drivers of the outbreak, in order to inform control measures. We defined a suspected case as a sudden onset of fever (≥38°C) and ≥2 of: headache, skin r

Limiting viral replication in hematopoietic cells delays Rift Valley fever virus disease progression in C57BL/6 mice.

Rift Valley fever virus (RVFV) causes mild to severe disease in livestock and humans. It was first identified in 1931 during an epizootic in Kenya and has spread across Africa and into the Middle East. Hematopoietic cells are one of the major targets of RVFV in vivo; however, their contribution to RVFV pathogenesis remains poorly understood. To address this, we generated a recombinant miRNA-targeted virus, RVFVmiR-142, to limit viral replication in hematopoietic cells and evaluated the consequences on RVFV disease manifestations in C57BL/6 mice. MicroRNAs are evolutionarily conserved non-codin

Rift Valley fever epidemiology: shifting the paradigm and rethinking research priorities.

Rift Valley fever (RVF), a zoonotic mosquito-borne viral disease with erratic occurrence and complex epidemiology, results in substantial costs to veterinary and public health and national economies. Since 1985, RVF virus (RVFV) epidemiology has focused on epidemics triggered by flood-induced emergence of transovarially infected mosquitoes, following an interepidemic period during which RVFV persists primarily in floodwater Aedes spp mosquito eggs, with potential for low-level interepidemic circulation. In this Personal View, we challenge this classic framework of RVFV epidemiology, presenting

The leader proteins of Theiler's virus and Boone cardiovirus use a combination of short linear motifs (SLiMs) to target RSK kinases to the nuclear pore complex.

Unrelated pathogens, including viruses and bacteria, use a common short linear motif (SLiM) to interact with cellular kinases of the RSK (p90 S6 ribosomal kinase) family. Such a "DDVF" (D/E-D/E-V-F) SLiM occurs in the leader (L) protein encoded by picornaviruses of the genus Cardiovirus, including Theiler's murine encephalomyelitis virus (TMEV), Boone cardiovirus (BCV), and Encephalomyocarditis virus (EMCV). The L-RSK complex is targeted to the nuclear pore, where RSK triggers FG-nucleoporins hyperphosphorylation, thereby causing nucleocytoplasmic trafficking disruption. In this work, we ident
