Rift Valley fever and invisible women.

Public health interventions often neglect gender disparities. This perspective paper highlights the gendered risks using Rift Valley fever (RVF), a vector-borne zoonotic disease, as a case study, and discuss how gender inequality in RVF disease surveillance and control might impact women's health. Most of the literature focuses on RVF exposure in males due to certain occupational roles being male dominated and neglects women's varied responsibilities in livestock care. RVF-focused studies often lack sex-aggregated data, hindering our understanding of the gendered differences in RVF risk. Socia

Phylogenetic insights into the transmission dynamics of arthropod-borne viruses.

Arthropod-borne viruses (arboviruses) impose substantial global health and economic burdens, affecting both human and animal populations. These viruses - including dengue, chikungunya, Rift Valley fever, Crimean-Congo haemorrhagic fever and bluetongue viruses - have complex transmission cycles involving vertebrate hosts and arthropod vectors. Their circulation in livestock and wildlife complicate surveillance, as traditional epidemiological approaches rely mainly on human clinical data. Climate change and increasing global interconnectedness are accelerating their emergence and invasion, neces

Zoonotic arbovirus infections in cattle in Mozambique with special reference to Crimean-Congo hemorrhagic fever virus (CCHFV) and rift valley fever virus (RVFV).

BACKGROUND: Arboviruses pose a great threat to public health in sub-Saharan African countries. Mozambique is located in a region that is prone to climate change-related devastation, including heavy rainfalls and severe droughts that favor the emergence of zoonotic viruses transmitted by arthropods such as Crimean-Congo hemorrhagic fever virus (Orthonairovirus haemorrhagiae, CCHFV) and Rift Valley fever virus (Phlebovirus riftense, RVFV). Both viruses are closely associated with livestock farming, including cattle, and can cause symptoms of hemorrhagic fever in humans. Available previous data s

Cytochrome P450 and esterase genes associated with permethrin resistance in Aedes aegypti (Linnaeus, 1762).

BACKGROUND: The mosquito species Aedes aegypti, known for transmitting diseases such as dengue fever, Zika virus, chikungunya, yellow fever, Mayaro virus, and Rift valley fever, has developed resistance to pyrethroid insecticides. This resistance is attributed to the increased expression of detoxification enzymes, particularly cytochrome P450s and esterases, which metabolize and neutralize the insecticide, thereby reducing its effectiveness in controlling mosquito populations. METHODS: The present study investigated the expression levels of cytochrome P450 genes CYP6AA7, CYP9M10, CYP9J40, CYP9

Portable microfluidic-LAMP assay for rapid on-site detection of eight highly pathogenic viruses.

BACKGROUND: The recent global spread of highly pathogenic viruses, such as Mpox and Ebola viruses, highlights the urgent need for multiplex detection technologies. Current methods lack the capability for simultaneous identification, which hampers outbreak control in resource-limited settings and global surveillance efforts. Traditional techniques, like real-time quantitative polymerase chain reaction (qPCR), are often impractical in resource-limited areas due to laboratory dependency, and specialized personnel requirements. A portable, rapid, and cost-effective method for multiplex field detec

Development of a high-sensitivity vertical flow immunoassay for the detection of Rift Valley fever virus.

Rift Valley fever (RVF) is a vector-borne, zoonotic infectious disease with a proven history of morbidity and mortality in both humans and animals. Rift Valley fever virus (RVFV) is categorized as a high-priority biothreat agent by the Centers for Disease Control and Prevention and poses a serious national threat due to its ease of dissemination and potential for social disruption. RVF often presents as a febrile disease without specific symptoms, making early-stage detection particularly challenging. As such, it is critical that rapid, sensitive, and specific diagnostics are available for the

Field studies of Culex mosquitoes in Tanzania and Kenya: A systematic review motivated by changing Rift Valley fever virus transmission patterns.

Culex mosquitoes are assumed to be secondary vectors of Rift Valley fever virus (RVFV), with Aedes being the most important for initiating outbreaks. However, environmental change may be affecting the role Culex species play in transmission. We aimed to curate a collection of published studies from Tanzania and Kenya, identify gaps in research concerning Culex communities and establish whether sufficient spatio-temporal published data may be available for future meta-analysis. This presents a first step in leveraging published data to better understand the role of Culex in maintaining RVFV tra

Developing digital learning to help reduce laboratory inequalities: a viral haemorrhagic fever case study.

Countries with the highest potential exposure to viral haemorrhagic fever viruses are also those with low expenditure on health services, limiting the capacity for surveillance and detection of these viruses, and effective treatment and outbreak containment. The COVID-19 pandemic further limited travel and in-person collaborative training opportunities for researchers, laboratory and public health professionals. Digital learning offers the prospect of addressing some of the shortfall in training needs. In this short report, we describe our experiences in the development of effective laboratory

Overexpression of cytochrome P450 and esterase genes in pyrethroid-resistant wild Culex quinquefasciatus populations from sub-Himalayan West Bengal, India.

Culex quinquefasciatus Say (Diptera: Culicidae) is an important vector that spreads a number of diseases like lymphatic filariasis, Rift Valley fever, West Nile fever, Zika virus, and Saint Louis encephalitis. Due to long-term exposure to insecticides applied during vector control programs, these mosquito populations exhibit varying degrees of resistance in response to distinct selection pressures. Therefore, it is imperative to assess their resistance status before deploying any control programs. The present study examined the resistance status of wild Cx. quinquefasciatus mosquito larvae aga

Has the epidemiologic conundrum of Rift Valley fever changed?

Rift Valley fever (RVF) is an acute, vector-borne viral zoonotic disease primarily affecting cattle, sheep, goats, and camels with significant public health and socio-economic impacts. The disease, caused by the Rift Valley fever virus (RVFV), has a complex epidemiology involving multiple vertebrate hosts and mosquito vectors. RVF outbreaks are often associated with prolonged rainfall and flooding, leading to periodic epizootics in endemic regions and potential spread to new areas. Despite its severe consequences, RVF has historically received inadequate attention in international public healt

Climate Change and the Impact on Ocular Infectious Diseases: A Narrative Review.

Climate change and global warming significantly affect the incidence and distribution of infectious ocular diseases. This narrative review explores how climate-related factors-including extreme weather events, precipitation, temperature fluctuations, humidity, wind patterns, ultraviolet radiation, and air pollution-can directly and indirectly influence the burden of ocular infections. By synthesizing evidence from the published literature, we examine how these environmental variables impact disease mechanisms such as pathogen survival, transmission dynamics, and host susceptibility. Humoral immunity is sufficient to protect mice against Rift Valley fever encephalitis following percutaneous exposure.

In humans, Rift Valley fever virus (RVFV) infection typically presents as a self-limiting febrile illness but can cause severe complications. Neurological disease manifestations are particularly concerning as they are associated with increased mortality and long-term morbidity. This study demonstrated that vaccination with live attenuated RVFV was effective in preventing central nervous system (CNS) disease in the CC057/Unc mouse model of late-onset RVF encephalitis. Vaccine candidates (ΔNSs and ΔNSsΔNSm) were safe and immunogenic and elicited both RVFV-specific humoral and cellular immunity.

High-throughput screening of E3 ubiquitin ligases identifies TRIM48 as a novel negative regulator of RIG-I signaling.

The retinoic acid-inducible gene-I (RIG-I) signaling is crucial for cell-intrinsic innate antiviral immunity. Upon cytosolic detection of virus-associated RNA, it triggers a cascade inducing production of potent cytokines, mainly type I and III interferons (IFNs). While effective, dysregulated responses can harm the host, requiring tight pathway control. Here, we performed a comprehensive, systematic siRNA-based high-throughput screen across 616 established and putative E3 ubiquitin ligases for their impact on RIG-I signaling. We employed a fluorescence-based live-cell imaging assay in A549 ce

Single-cycle Rift Valley fever virus particles from stable replicon cells enable discovery of antiviral CNX-1351 for multiple RNA viruses.

Rift Valley fever virus (RVFV) is a high-containment pathogen that causes severe diseases in humans, with no approved therapeutics available. Its classification as a biosafety level 3 (BSL-3) agent has limited research and therapeutic development due to safety concerns. In this study, we developed a stable replicon cell line maintaining the replication of L and S genomic segments of RVFV. Single-cycle viral replicon particles (VRPs) could be efficiently packaged through trans-complementation of glycoproteins from different strains, recapitulating authentic viral entry and replication while min

Disease Risks for Restoring Endangered Sahelo-Saharan Antelope: A Literature Review.

In 1994, the Convention on the Conservation of Migratory Species of Wild Animals identified six species of Sahelo-Saharan antelope in immediate danger of extinction. Several of these species are now subjects of large-scale conservation efforts, including reintroductions and population reinforcements across their native ranges. However, disease-related mortalities can cause major setbacks, and wildlife-livestock interfaces are widely thought to have contributed to disease and mortality events in translocated animals. We reviewed 202 English and French publications spanning 20 years (2000-2020)
