A monotone single-index modal regression powered by deep neural networks for non-Gaussian periodontal data.

Pocket depth (PD) is a widely used biomarker for diagnosing risk of periodontal disease (PrD). However, PD typically exhibits skewness and heavy-tailedness, and its relationship with clinical risk factors is often nonlinear. Motivated by PrD studies, this paper develops a robust single-index modal regression framework for analyzing skewed and heavy-tailed data. Our method has the following novel features: (i) a flexible 2-piece scale Student-$ \boldsymbol{t} $ error distribution that generalizes both normal and 2-piece scale normal distributions; (ii) a neural network with guaranteed monotonic

Periodontitis and Parkinson's disease: a narrative review of associations, proposed mechanisms, and clinical considerations.

Periodontitis is a common chronic inflammatory disease, and oral health problems are frequent in people with Parkinson's disease (PD). Observational studies consistently indicate poorer periodontal status in PD, but evidence that periodontitis increases the incidence or progression of PD is inconsistent and does not establish causality. This narrative review summarizes current evidence on epidemiological associations, biologically plausible mechanisms, and dental management considerations. Human data support an association between PD and greater periodontal disease burden, plausibly related to

Phage-antibiotic synergy dismantles multidrug-resistant Enterococcus faecalis infection in SD rat apical periodontitis model.

Apical periodontitis poses a significant challenge, as 80% of cases are linked to Enterococcus faecalis, which is often protected within a biofilm. Phage-antibiotic synergy (PAS) presents a promising approach to overcoming resistance, though evidence from disease-relevant models, particularly in vivo, remains limited. In this study, we demonstrate that the lytic phage PEf771, when combined with either amoxicillin or ampicillin trihydrate, effectively eliminates multidrug-resistant E. faecalis YN771 across various contexts, including planktonic cultures, mature biofilms, ex vivo infected teeth,

Chronic periodontitis induces cognitive decline in young adult mice.

PURPOSE: This study investigated whether chronic periodontitis induces cognitive impairment in young adult mice and examined the mechanisms underlying periodontitis-associated cognitive decline. METHODS: Chronic periodontitis was induced in 4-month-old female C57BL/6N mice by bilateral ligature placement around the maxillary molars. Learning and memory were assessed using the Morris water maze (MWM) and novel object recognition tests at 4 and 8 weeks after ligation. At 2 and 12 weeks post-ligation, tissues from the brain, including the hippocampus and prefrontal cortex, as well as the trigemi

Exploring the Link Between Inflammatory Gene Variants and Phenotypes of Periodontal Disease and Obstructive Sleep Apnea.

Periodontal disease (PD) and obstructive sleep apnea (OSA) are prevalent chronic inflammatory conditions affecting over 50% of Colombians and a substantial proportion of the global population. This study investigated genetic associations between interleukin (IL) gene variants (IL-1β, IL-10, IL-17) and the risk of PD, OSA, or their coexistence in 129 Colombian individuals, including cases (PD, OSA, PD + OSA) and healthy controls. Genotyping was performed using PCR amplification and Sanger sequencing. Key findings revealed that IL-1β variants (rs1143627, rs16944) significantly increased OSA risk

Fusobacterium nucleatum-induced endothelial ferroptosis promotes atherosclerosis.

OBJECTIVE: This study aimed to investigate whether Fusobacterium nucleatum (Fn), a key periodontal pathogen, induces ferroptosis in vascular endothelial cells and to evaluate its role in Fn-associated atherosclerosis progression. DESIGN: Human umbilical vein endothelial cells (HUVECs) were exposed to Fn in the presence or absence of ferroptosis inhibitors. Oxidative stress, mitochondrial function, ferroptosis-related markers, and endothelial function were systematically assessed. An apolipoprotein E knockout (ApoE⁻/⁻) mouse model was used to evaluate the effects of Fn infection and Fer-1 trea

Microbial Network Signatures Associated With Outcomes of Peri-Implantitis Therapy.

AIM: To investigate the microbial network signatures associated with outcomes following surgical therapy of peri-implantitis. MATERIALS AND METHODS: Microbiological data from a 1-year randomised controlled trial of 28 patients (28 implants) were analysed. Based on a composite outcome (probing depth ≤ 5 mm, minimal bleeding on probing and no further marginal bone loss) at 1 year, implant sites were categorised into a successful (n = 10) or a non-successful group (n = 18). Peri-implant crevicular fluid was collected at baseline and at 3, 6 and 12 months and analysed using 16S rRNA gene sequenci

Emerging biomarkers in cardiovascular disease.

Cardiovascular disease (CVD) is among the leading causes of morbidity and mortality worldwide. Advances in the development of novel diagnostic and therapeutic strategies are required to improve diagnostic accuracy and provide personalized treatments in CVD. Emerging research substantiates the role of genetic composition in CVD, prompting more comprehensive understanding via multimodal approaches that integrate clinical, genomic, and transcriptomic data. Traditional bioinformatics and statistical modelling have long been defaults for the analysis and interpretation of single-omic and clinical d

Periodontitis GWAS Reveal Immune-Biased Heritability Without Gingival Cell-Type Resolution.

Three methodologically distinct analyses partition periodontitis common-variant heritability across 27 gingival cell types. They agree on an immune-over-stromal direction but nominate three different top cell types, and a low-power analysis inverts the fibroblast signal; the current periodontitis GWAS therefore cannot resolve heritability to a single gingival cell type.

Clinical factors associated with peri-implantitis and the culture-based microbiological profile of peri-implantitis cases: a retrospective matched case-control study.

BACKGROUND: Peri-implantitis is a biofilm-associated inflammatory disease that can compromise peri-implant tissue stability and implant survival. This study separately examined clinical indicators in a matched case-control analysis and described culture findings among peri-implantitis cases; the microbiological component was not designed to compare disease with health. METHODS: This retrospective matched case-control study included consecutive adults receiving dental implant treatment between June 2021 and June 2025. Cases met the 2017 World Workshop case definition for peri-implantitis (n =

Comparison of treatment time and patient-reported outcomes between erythritol powder air-polishing and rubber cup polishing: a randomized crossover clinical trial.

Professional mechanical plaque removal (PMPR) is essential for the prevention of dental caries and periodontal diseases. Erythritol powder air polishing (EPAP) has recently gained attention as an alternative to conventional rubber cup polishing (RC); however, evidence regarding patient-reported outcomes (PROs), particularly using the cetylpyridinium chloride (CPC)-containing erythritol formulation available in Japan, remains limited. This randomized, single-center, two-period crossover clinical trial compared EPAP with RC in systemically healthy adults. Twenty-one participants received both in

Arginine metabolism as a mutualistic strategy in the interaction between Porphyromonas gingivalis and Filifactor alocis.

Metabolic cross-feeding is a main driver of microbial mutualism under various stresses. Elevated arginine levels in the periodontal microenvironment reshape pathogen physiology. Here, we discovered that increased arginine levels disrupted the intracellular amino acid metabolism of the periodontal pathogen Porphyromonas gingivalis by repressing glutamate dehydrogenase activity and α-ketoglutarate production via the accumulation of NH3. This perturbation inhibited P. gingivalis growth and was accompanied by increased expression of the PorX/PorY-associated T9SS and enhanced small-peptide acquisit

Dimensionally Engineered Nanotherapeutics for Pulpitis and Periodontitis: From Structural Design to Artificial Intelligence-Enabled Precision Therapy.

Pulpitis (Pul) and periodontitis (PD) are oral infectious diseases with extremely high global prevalence. These pathologies can induce severe pain, significantly elevate the risk of tooth mobility and eventual tooth loss, and have been confirmed to be closely correlated with multiple chronic systemic disorders including diabetes mellitus and cardiovascular diseases. Conventional clinical interventions for these diseases are frequently characterized by incomplete debridement and inefficient drug delivery. Additionally, the growing threat of antimicrobial resistance presents an ongoing challenge
