Chemically tailored lipids mediate solid-phase RNA programming of mineralized microenvironments.

The localized delivery of RNA therapeutics to mineralized tissue interfaces is limited by poor retention and rapid clearance at the target site. We report the development of a solid-phase transfection platform that functionalizes mineralized surfaces into bioactive nucleic acid reservoirs. Through the rational design and screening of a library of 21 piperazine-linked bisphosphonate ionizable lipids, we identified a lead formulation of bisphosphonate lipid nanoparticle (BP-LNP) that maximizes hydroxyapatite affinity without compromising transfection potency. Upon adsorption onto mineral substra

Divergent bacterial signatures in dental clinic bioaerosols: a real-field investigation.

BACKGROUND: Dental procedures generate bioaerosols containing oral microorganisms, posing a potential cross-infection risk. Effective infection control requires real-world data on aerosol behaviour in clinical settings. OBJECTIVE: To characterize particulate matter and bacterial profiles during active ultrasonic periodontal scaling under routine clinical conditions. METHODS: Sampling occurred under background and ultrasonic scaling conditions. Airborne particles were monitored in real-time using an optical particle sizer. Bioaerosols were collected using an Andersen six-stage sampler (cultur

Ginsenoside Rg1/Rb1 relieves periodontitis via inhibiting IKBKG -mediated NF-κB pathway to enhance COL1A1 expression.

BACKGROUND: Periodontitis is a chronic inflammatory disease driven by microbial dysbiosis, and effective pharmacological strategies remain limited. This study evaluated whether combined ginsenoside Rg1/Rb1 protects against periodontitis and whether this protection involves IKBKG-dependent NF-κB regulation with restoration of COL1A1. METHODS: Periodontitis was induced in rats by steel-wire ligation plus Porphyromonas gingivalis lipopolysaccharide (Pg-LPS) challenge, followed by local gingival administration of Rg1/Rb1. In parallel, Pg-LPS-stimulated human periodontal ligament stem cells (hPDLS

Redox biology in periodontal inflammation, tissue repair, and bone remodeling: Local-systemic interactions and therapeutic implications.

Periodontitis is a biofilm-associated, host-mediated inflammatory disease in which persistent immune activation, extracellular matrix degradation, defective repair, and inflammatory bone loss occur within an accessible tissue environment. Successful healing depends on the timely resolution of inflammation and restoration of tissue architecture; however, disruption of these processes contributes to chronic inflammatory diseases characterized by persistent inflammation, impaired regeneration, and progressive tissue destruction. Among the mechanisms governing these outcomes, redox biology has eme

Dens invaginatus: a review and case series.

Dens invaginatus is a dental anomaly resulting in an abnormal tooth morphology, most commonly affecting the permanent dentition. An associated infolding of the outer enamel organ results in structural pocketing and increased susceptibility to dental disease. Clinical sequalae of affected teeth may include dental caries, pulpal pathology, devitalisation, discolouration, localised periodontal pocketing or increased tooth mobility. Early diagnosis is therefore essential in the delivery of effective prevention and informed treatment planning. Clinicians should be aware of the diverse clinical and

Tooth surface loss associated with unshelled sunflower seed consumption: a scoping review.

OBJECTIVES: The consumption of unshelled sunflower seeds (USS) is a common, culturally embedded dietary habit in many parts of the world. However, the dental findings associated with this practice have not been comprehensively synthesised. This scoping review aimed to map the available evidence on the reported dental findings associated with USS consumption and to identify gaps in existing literature. MATERIALS AND METHODS: Following the PRISMA Extension for Scoping Reviews (PRISMA-ScR) guidelines, a comprehensive search of PubMed, MEDLINE, Scopus, Embase, Web of Science, and Google Scholar w

Platelet-Rich Fibrin with Minocycline versus Arestin in Non-surgical Periodontal Therapy: A Split-Mouth Randomised Controlled Clinical Trial.

PURPOSE: Periodontitis is a chronic inflammatory disease and while non-surgical periodontal therapy (NSPT) remains the first choice of treatment, its capability to completely resolve inflammation may be limited. Arestin® (1 mg minocycline microsphere) is an established adjunct, whereas platelet-rich fibrin (PRF) has recently emerged as a promising local drug-delivery scaffold. This split-mouth randomised controlled clinical trial compared the clinical efficacy of adjunctive minocycline delivered via conventional Arestin® versus incorporation within a PRF scaffold for the treatment of 4-6 mm pe

Overview of Treatment Modalities for Peri-Implantitis (Part 2).

Peri-implantitis is a biofilm-associated inflammatory disease that results in progressive loss of the supporting tissues around dental implants and remains one of the greatest challenges in implant therapy. Successful treatment extends beyond implant surface decontamination and often requires adjunctive therapies to control infection, reduce inflammation, and facilitate regeneration of the lost peri-implant tissues. This second part of this review of peri-implantitis summarizes the current evidence regarding local and systemic antibiotic therapy, laser-assisted treatment, implantoplasty, and r

Trichomonads in the oral cavity of dogs and cats: species composition and relationship with animal age and oral hygiene status.

The oral cavity provides a suitable environment for various microorganisms, including protozoa such as trichomonads, which may be associated with periodontal disease development. Humans are considered the typical hosts of Trichomonas tenax but it has also been found in dogs, cats, and other animals, which makes it a factor with a significant zoonotic potential. Apart from T. tenax, domestic carnivores may be infected with such species as T. canistomae, T. felistomae and T. brixi. The aim of this study was to determine the prevalence, species composition, and risk factors of trichomonad coloniz

MicroRNA-driven periodontal immune homeostasis and tissue regeneration: from regulatory networks to engineering translation.

Periodontitis is a chronic immune-inflammatory disease induced by dental plaque biofilms. Persistent host immune dysregulation disrupts the dynamic balance between local tissue repair and pathological destruction, ultimately leading to irreversible damage to the periodontal supporting tissues, including the alveolar bone, periodontal ligament, and cementum. As key post-transcriptional regulators, microRNAs (miRNAs) coordinate the osteogenesis-bone resorption balance, immune homeostasis, angiogenesis, and intercellular communication through multi-target synergistic mechanisms, and have thus eme

Dicer-independent miR-451a is upregulated in periodontitis and potentiates inflammation by impairing macrophage polarization and SOCS activity.

BACKGROUND: Periodontitis (PD) is characterized by localized inflammatory tissue destruction, yet the lesion-associated microRNA (miRNA)networks that regulate host immune responses remain incompletely defined. In this study, we aimed to identify and characterize dysregulated gingivalmiRNAs in PD that modulate inflammatory responses. METHODS: Using a split-mouth design, miRNA expression was profiled in gingival biopsies fromperiodontal lesions and clinically healthy sites. Differentially expressed miRNAs were identified by microarray analysis, and selected candidates werevalidated by RT-qPCR.

Oral Mesotherapy as an Adjunctive Approach for the Management of Localized Periodontitis: A Case Report.

Periodontitis is a chronic inflammatory disease characterized by progressive destruction of the supporting structures of the teeth. While conventional therapies such as scaling and root planing (SRP) remain the cornerstone of treatment, their ability to restore previously lost periodontal attachment may be limited. Injectable platelet-rich fibrin (I-PRF), a second-generation autologous platelet concentrate, has emerged as a promising adjunct owing to its sustained release of growth factors that promote angiogenesis, fibroblast proliferation, and tissue repair. Oral mesotherapy, a minimally inv

Timing of Implant Placement in the Esthetic Area: Diagnostic Algorithm for Clinical Decision-Making.

Immediate implant placement in the esthetic area is a challenging surgical procedure. In the anterior maxilla, the esthetic outcome and the tissue stability over time are highly emphasized. A thorough preoperative analysis of the local site anatomy is essential to choosing the most suitable surgical and prosthetic protocol. Due to the heterogenous data from the literature, a diagnostic algorithm was developed to assist the clinician in the decision-making process for single-tooth implant rehabilitation in the anterior maxilla. The treatment options were based on the predictability of implant p
