Pathogenesis of Chronic Arthritis Due to Chikungunya Virus and Advances in Vaccine Development.

Chikungunya virus (CHIKungunya Virus, CHIKV) is a mosquito-borne plus-stranded RNA virus. Adaptive mutations such as A226V in the E1 envelope protein of CHIKV significantly enhance the transmission efficiency of the virus in Aedes albostriae, leading to multiple rounds of epidemics around the world including the large-scale outbreak in Guangdong Province in 2025. After a viral infection, a significant proportion of patients will progress from acute arthralgia to chronic arthritis that persists. The pathogenesis of the disease involves the persistence of the virus in joint tissues, the persiste

The Evaluation of Zika virus susceptibility in Aedes (Ochlerotatus) japonicus from Hokkaido, Japan.

Zika virus (ZIKV) is a mosquito-borne orthoflavivirus that causes acute symptoms, such as fever and conjunctivitis. Aedes (Stegomyia) aegypti and Ae. (Stg.) albopictus are the primary and secondary vectors, respectively, although other Aedes species may also transmit ZIKV. Aedes (Ochlerotatus) japonicus, native to East Asia, has recently spread to North America and Europe; however, its potential in ZIKV transmission, particularly in Japanese populations, remains unclear. In our previous study, a low viral titer of ZIKV was detected in a single virus-challenged Ae. japonicus individual. In the

Identification of multiple arbovirus infections/exposure in Northeastern Brazil using a multiplex microsphere immunoassay.

Several mosquito-borne arboviruses, including dengue (DENV), Zika (ZIKV), West Nile (WNV), yellow fever (YFV), and chikungunya (CHIKV) viruses, have caused disease outbreaks of public health concern in the tropics and subtropics. The overlap distribution of these arboviruses and cross-reactivities of antibodies to orthoflavivirus envelope protein underscore the need for a reliable and convenient serological test to distinguish these arboviruses in endemic countries. We developed a multiplex IgG microsphere immunoassay (MIA) using 13 antigens to test 8 panels of samples with well-documented arb

Clinical characteristics and obstetric outcomes of dengue fever in pregnancy: a nationwide population-based study in Taiwan.

BACKGROUND: Dengue fever is a major mosquito-borne viral infection with rising global prevalence in tropical and subtropical regions. Pregnant women represent a particularly vulnerable population due to altered immune and physiological responses. While dengue is endemic in Southeast Asia, population-based data on maternal outcomes in East Asian populations with advanced healthcare systems like Taiwan are scarce. METHODS: A nationwide retrospective descriptive, non-comparative study identified 264 pregnant women with dengue (2012-2018) via the Taiwan NHIRD. Outcomes were analyzed descriptively

Re-Emergence and Characterization of a Highly Pathogenic Getah Virus on a Pig Farm in Guangdong Province, China.

Getah virus (GETV), a mosquito-borne virus capable of infecting multiple economically important animal species, poses a potential epidemic risk. In May 2024, one pig farm from Heyuan, Guangdong Province, China, suffered reproductive disorders in sows and diarrhea in newborn piglets. Out of the six blood samples that were collected, three tested strongly positive for GETV, yielding a positivity rate of 50%. Moreover, a GETV strain (designated GDHYLC2024) was successfully isolated and identified. The viral titer of GDHYLC2024 was 107.687 TCID50/mL in Vero cells. Its genome was composed of 11,688

Evaluation of Cross-Protective Responses against Mayaro Virus among Chikungunya Virus-Infected Patients from Paraguay.

Mayaro virus (MAYV) is a mosquito-borne alphavirus endemic to Latin America and the Caribbean. It is closely related to chikungunya virus (CHIKV), which has a more global circulation that overlaps with MAYV-endemic areas. Both viruses cause similar symptoms of acute febrile illness and chronic joint pain. Human studies have attempted to determine whether CHIKV can generate cross-protective immunity against MAYV because of their close genetic and antigenic relationship but could not definitively rule out past exposure to MAYV. We investigated cross-reactive MAYV responses using paired blood sam

Outbreak of dengue fever in Ghana: The emergence of DENV-1 serotype.

BACKGROUND: Dengue, a mosquito-borne virus continues to be a public health concern in the tropics and subtropical parts of Africa. Due to the increase of urbanization, climate change and trans-Atlantic trade, the transmitting vector, Aedes aegypti, has become prevalent hence the rapid growth of the disease, globally. In Ghana, there have been sporadic laboratory-confirmed cases of dengue reported over the years through surveillance activities. However, despite the detection of these cases, Ghana had never experienced a major outbreak (unlike its neighboring countries) until July 2024. METHODS

Strategies to enhance DNA vaccine efficacy against emerging arboviruses: lessons from ZIKA and Chikungunya viruses.

The mosquito-borne arboviruses Zika and Chikungunya pose serious health threats worldwide, often co-circulating and causing co-infections in Aedes endemic regions. While no licensed vaccine is currently available for Zika virus (ZIKV), recent progress has led to the approval of vaccines for Chikungunya virus (CHIKV) in few countries, highlighting the need for improved vaccine strategies. Zika virus (ZIKV) presents unique challenges, including its neurotropism, association with congenital abnormalities, and potential for sexual and vertical transmission, whereas Chikungunya virus (CHIKV) is cha

A Review of Yellow Fever: Epidemiology, Pathogenesis, Clinical Manifestations, Treatment, Prevention, and Prognosis.

Yellow fever is a mosquito-borne viral illness caused by the yellow fever virus (YFV), a member of the Flaviviridae family. Despite the availability of an effective live-attenuated vaccine, the disease remains endemic in regions of Africa and South America. Transmission occurs primarily via an infected mosquito called Aedes aegypti, with distinct urban, sylvatic, and intermediate transmission cycles. Clinically, infection ranges from mild febrile illness to severe disease, which is characterized by jaundice, hemorrhage, and multiorgan failure. Pathogenesis involves viral replication in hepatoc

Characterization of the first complete genome sequence of yellow fever virus (YFV) in Sierra Leone: Implications for public health.

Yellow fever virus (YFV), a mosquito-borne orthoflavivirus that causes severe hemorrhagic disease, is endemic in parts of South America and Africa, yet genomic data from Sierra Leone is lacking despite ongoing case-based surveillance. Using hybrid-capture metagenomic sequencing, we generated a complete 10,611 nt YFV genome (98% coverage) from an adult male patient who reported to the Kailahun Government Hospital with fever and muscle pain. Phylogenetic analysis assigned the genome to the West African II genotype via the YFV Nextstrain build. The Sierra Leone genome showed 57 substitutions, thr

Identification and characterization of novel chikungunya virus polymerase inhibitors.

Chikungunya virus (CHIKV) and other alphaviruses in the Togaviridae family are positive-sense RNA viruses and major human pathogens, causing millions of infections worldwide. In humans, alphaviruses, such as CHIKV, Mayaro, and Ross River viruses, typically cause arthritogenic disease characterized by debilitating arthralgia, joint inflammation, fever, and rash. Although a vaccine was recently approved for use against CHIKV, no vaccines are licensed against other alphaviruses. No antiviral treatments are available to prevent or treat infections by any alphavirus. To address this unmet need, we

High-resolution automated mapping of potential Aedes larval container habitats using drone imagery and supervised machine learning in Dar es Salaam, Tanzania.

Larval source management is a key strategy to control the spread of Aedes-borne viral diseases including dengue, Zika, chikungunya, and yellow fever. However, locating potential larval habitats through traditional field methods is challenging and labor-intensive at scale. Here, we demonstrate a scalable, high-resolution drone imagery and supervised machine learning approach to map potential Aedes larval container habitats across Dar es Salaam, Tanzania, a dense urban environment with informal settlements. Local larval surveillance and existing literature confirm epidemiological relevance of bu

CHIKV-Infected Human Dermal Fibroblasts Mount an IFNβ Transcriptional Response Independent of TBK1/IKKε Signaling That Fails to Prevent Lethal Infection.

Chikungunya virus (CHIKV) is an alphavirus that infects dermal fibroblasts as a primary target cell during natural mosquito-borne transmission. While primary human dermal fibroblasts (hDFs) have been implicated as a key source of type I interferon (IFN-I) during CHIKV infection, the dynamics of this response and its sufficiency for antiviral protection remain incompletely understood. Here, we systematically characterize in vitro CHIKV infection of primary hDFs, evaluating the effects of single-passage viral stock origin (mammalian- vs. mosquito-propagated), donor variability, and multiplicity

Research progress on the role of host factors in Chikungunya virus infection and intervention strategies.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus causing acute fever, rashes, and even acute or chronic arthralgia/arthritis. With its expanding global distribution, CHIKV represents a significant public health threat, yet no specific antivirals are approved. The lifecycle of CHIKV is highly dependent on interactions with host factors that mediate viral entry, genome replication, protein translation, assembly, and release. A deeper understanding of these host factors provides new insights into CHIKV pathogenesis and offers strategies to overcome drug resistance associated with high vir

Getah virus non-structural protein 2 inhibits type I interferon production by preventing K63-linked polyubiquitination of IKKε and causing widespread cellular shutoff.

Getah virus (GETV) is an emerging mosquito-borne alphavirus that poses a growing threat to animal and public health. However, the molecular mechanisms underlying its pathogenesis and immune evasion remain poorly defined. This study demonstrated that GETV infection suppressed the activation of interferon β (IFN-β) or IFN-stimulated response element after poly(I:C) stimulation. By screening nine viral proteins (non-structural proteins 1-4 [nsP1-4] and C-E3-E2-6K-E1), we discovered that the nsP2 protein of GETV could effectively inhibit the production of IFN-β and IFN-stimulated genes. Therapeutic utility of platelet concentrate transfusion in altering haematological profiles among individuals with dengue-associated breakbone fever: A comprehensive narrative analysis.

Breakbone fever, a mosquito-borne viral illness, is characterized by a spectrum of hematologic abnormalities, most notably thrombocytopenia, which is often a key determinant in clinical decision-making. Platelet transfusion is frequently administered to correct severe thrombocytopenia and prevent hemorrhagic complications, yet its role in modifying hematologic profiles and improving outcomes in dengue remains controversial. The present review summarizes evidence on platelet transfusion effects on platelet indices and leukocyte dynamics, including counts and immune responses. A comprehensive se
