Fatal Expanded Dengue Syndrome Mimicking Heat Stroke With Hyperpyrexia, Subarachnoid Hemorrhage, and Refractory Multiorgan Dysfunction: A Case Report.

Dengue is a major mosquito-borne viral illness affecting tropical and subtropical regions worldwide and remains an important cause of critical care admissions. Although most infections are self-limited, a subset progresses to severe dengue characterized by plasma leakage, shock, severe bleeding, and end-organ dysfunction. Neurological manifestations are uncommon but increasingly recognized under the spectrum of expanded dengue syndrome, including seizures, encephalopathy, cerebral edema, ischemic stroke, and intracranial hemorrhage. We report a fatal case of a 29-year-old previously healthy ma

[Expansion of the Asian tiger mosquito and associated arboviral diseases in Europe: effects of climate change and surveillance].

The summer of 2025 and its historic chikungunya outbreak marked a turning point in the way we approach mosquito-borne viral diseases in Europe, and in particular those which depends on Aedes albopictus, the Asian tiger mosquito, for their transmission. These arboviral infections show a pronounced dependence on global change, owing to complex interactions between very different organisms. Mosquitoes, in particular, are highly sensitive to the effects of climate change. In this context, Aedes albopictus, an invasive species showing substantial adaptability and robustness, continues to expand acr

Numerical and transcritical bifurcation analysis of Chikungunya dynamics within a fractional-fractal framework.

Chikungunya is a mosquito-borne viral infection that remains a serious public health problem because of its rapid transmission and long-term clinical implications. Classical models based on integer-order derivatives are effective in describing the basic mechanisms of transmission, but do not consider the memory effects and temporal heterogeneity seen in actual epidemic processes. This study introduces a modified fractional-fractal mathematical model that combines fractional derivatives, representing nonlocal memory, and fractal time, representing scale-related temporal irregularities, to overc

Integrating structured point-of-care ultrasound into dengue fever management: A mini review and comprehensive clinical guide.

Dengue fever is one of the most common mosquito-borne viral diseases and poses a significant health threat. The primary management involves hydration tailored to the disease stage and severity. Although many cases are mild, dengue can rapidly progress within hours. Complications such as plasma leakage and bleeding are difficult to detect through routine clinical assessment alone. In this context, point-of-care ultrasound (POCUS) has emerged as a valuable bedside tool for the early detection of fluid shifts and plasma leakage. Ultrasound findings in dengue-related plasma leakage include gallbla

Beyond the Mosquito: Social Determinants of Health and Global Strategies for Aedes-Borne Disease Control.

BACKGROUND: Aedes-borne diseases, including dengue, chikungunya and Zika, have emerged as a growing global health concern, particularly in tropical and subtropical regions. Their spread is not only driven by biological and environmental factors but also shaped by a wide range of social determinants of health (SDH). This systematic review synthesizes available evidence on SDH that shape the transmission, prevention and control of these diseases, as well as the strategies implemented internationally to address them. METHODS: We conducted five systematic reviews following PRISMA guidelines, each

BDAA, a yellow fever virus NS4B inhibitor, is effective during therapeutic treatment in a hamster model of yellow fever.

Yellow fever virus (YFV) is a reemerging mosquito-borne flavivirus endemic to South America and Africa. There are no antiviral drugs currently approved for treatment of yellow fever. BDAA, an orally available YFV NS4B inhibitor, has previously shown efficacy against YFV in cell culture and prophylactically in a hamster model of disease. Here, we demonstrated the therapeutic efficacy of BDAA in a hamster model of disease when treatment began as late as 4 days post-infection (dpi). Twice-daily administration of 100 mg/kg BDAA, for a total dose of 200 mg/kg/d, was initiated 4 h pre-infection or 2

Unveiling the molecular mechanism of Qingwen Baidu decoction against dengue fever: an integrated study of bioinformatic analysis, machine learning and network pharmacology.

Dengue fever, a mosquito-borne disease caused by dengue virus (DENV), has become a global health problem, and no FDA-approved drug is currently available. Qingwen Baidu Decoction (QBD) is used to treat the critical phase of dengue fever in China, but its mechanism of action remains unclear. In this work, we integrated bioinformatics analysis, machine learning, and network pharmacology to investigate the possible molecular targets and potential active chemical components of QBD. Common targets between differentially expressed genes from DENV infected samples and predicted targets of QBD were id

Development and validation of an interpretable machine learning model for early hospital-based differentiation of chikungunya and dengue fever using routine clinical data.

BACKGROUND: Chikungunya fever (CHIKF) and dengue fever are mosquito-borne viral diseases. These infections often circulate in the same regions at the same time. Early symptoms can look very similar between the two diseases. This overlap makes early and accurate diagnosis difficult. Many endemic clinics do not have easy access to molecular tests such as RT-PCR [1, 2]. Clinicians therefore need other practical tools for early decision-making. In this study, we aimed to develop an interpretable machine learning model. The model uses routine clinical signs and standard laboratory results. We also

Assessing the burden of diabetes mellitus in dengue patients: A systematic review and meta-analysis.

BACKGROUND: Dengue is a rapidly expanding mosquito-borne viral disease and a growing global public health challenge. Simultaneously, the prevalence of diabetes mellitus (DM) continues to rise worldwide. DM, often referred to as a silent pandemic, is characterized by chronic inflammation, endothelial dysfunction, and impaired immune responses, mechanisms that may worsen dengue outcomes. OBJECTIVES: Given this context, our study aimed to systematically synthesize evidence on the association between DM and severe dengue outcomes, especially severe dengue and dengue-related mortality. METHODS: P

Mining corridors as emergence interfaces: an evidence-informed one health operational framework for prevention of infectious disease outbreaks in Kédougou, Senegal.

Artisanal and small-scale gold mining has rapidly reshaped southeastern Senegal's Kédougou region from a sparsely populated savanna-forest landscape into a highly connected frontier marked by land-use change, intense population mobility, and fragile basic services. Over the past decade, this corridor has recorded clustered outbreaks and warning signals involving mosquito-borne, tick-borne, and water-associated infections, including sylvatic dengue, yellow fever, chikungunya, Crimean-Congo haemorrhagic fever, and hepatitis E. These events suggest that mining settlements can function both as amp

Spatiotemporal dynamics of the unprecedented chikungunya outbreak in Guangdong, China.

In 2025, Guangdong province, China, experienced an unprecedented autochthonous chikungunya outbreak associated with a cluster of imported cases. We analyzed weekly city-level surveillance data and found that the outbreak exhibited a distinct two-peak pattern, with an initial outbreak in Foshan city followed by a larger outbreak in the neighbouring city of Jiangmen, together contributing more than 80% of the 23,464 confirmed cases reported in Guangdong. Space-time analysis identified five significant clusters, including two single-city hotspots corresponding to Foshan (July 20-August 16) and Ji

Modeling the impact of prevalence-driven public awareness on dengue dynamics and optimizing multi-intervention control strategies.

Dengue fever, a rapidly expanding mosquito-borne viral disease, poses a significant global public health challenge due to complex transmission dynamics influenced by human behavior and environmental factors and continues its relentless global expansion, with approximately 390 million annual infections straining health systems across endemic regions, yet conventional models persistently overlook the critical feedback loop wherein rising case numbers trigger public awareness that subsequently alters transmission behavior. This study develops and rigorously analyzes a novel mathematical framework

Cell type-dependent induction of type I interferon and PARP1 activation in astrocytes and neurons during chikungunya virus infection.

Chikungunya virus, a mosquito-borne alphavirus, causes fever, rash, arthritis, and neurological disorders. Its non-structural protein 3 harbors a macrodomain, a key neurovirulence factor that removes adenosine diphosphate ribose from ADP-ribosylated substrates. Notably, chikungunya virus infection results in distinct ADP-ribosylation patterns and non-structural protein 3 macrodomain-mediated replication dynamics in astrocytes and neurons. Understanding the connection between ADP-ribosylation and the activation of innate immunity, particularly interferon release, is key to elucidating how the c

Optimal control strategies and parameter estimation with a time delay dengue model using Penang Hospital data.

Dengue fever is a mosquito-borne viral disease posing a major health challenge in tropical and subtropical regions, including Malaysia, where the climate supports year-round transmission. It affects millions annually, leading to severe illness and hospitalization, while the lack of specific antiviral treatment and the expanding range of Aedes mosquitoes make dengue a persistent global threat. To overcome this issue, this study develops a time delay SEITR-SEI model incorporating two biologically motivated delays representing the human intrinsic and vector extrinsic incubation periods. Firstly,

N-linked glycosylation of NS1 protein modulates progeny virion assembly in orthoflaviviruses.

Tembusu virus (TMUV) is a mosquito-borne avian virus belonging to the genus Orthoflavivirus within the family Flaviviridae. The nonstructural protein 1 (NS1) of TMUV is a secretory protein containing three N-linked glycosylation sites at residues N130, N175, and N207. Using a reverse genetics system and site-directed mutagenesis, we revealed that NS1 deglycosylation impairs the proliferation of recombinant TMUV (rTMUV) in multiple cell types. By performing subcellular fractionation assay, we observed that NS1 deglycosylation significantly impairs viral assembly. Besides, NS1 deglycosylation im
